Effect of control measures on the contamination and growth inhibition of Listeria monocytogenes in Flammulina velutipes (팽이버섯 재배 농가에서 Listeria monocytogenes 오염과 성장억제를 위한 관리기술 효과)

  • The consumption of Flammulina velutipes mushroom imported from Korea has been associated with the cases of listeriosis in the United States, Canada, and Australia. We investigated the effect of sanitizing the plastic wrapper (used in packaging F. velutipes) with slightly acidic electrolyzed water (SAEW) and ultraviolet C waterproof light-emitting diode (UVC-W-LED) on reducing the Listeria monocytogenes. Further, the effect of UVC-LED on L. monocytogenes growth in F. velutipes at different storage temperatures (2, 4, and 10℃) was determined. The combined (SAEW+UVC-W-LED) treatment for 5-10 min reduced 99.9% of bacterial population from the contaminated plastic wrapper. In addition, the UVC-LED treatment for 3 min reduced the L. monocytogenes concentration in F. velutipes by 0.47 log CFU/g. Moreover, the growth of L. monocytogenes in the treated mushrooms was slower than that of the untreated (control) ones. L. monocytogenes concentration in F. velutipes increased over 3 log CFU/g at 2℃ and 10℃ for 60 and 10 days, respectively. The growth of L. monocytogenes at the bottom of mushrooms was faster than that at the top at both the temperatures. These results indicate that the combined SAEW+UVC-W-LED treatment of plastic wrappers and the UVC-LED treatment of mushrooms can be used as potential hurdle technologies to control the risk of L. monocytogenes in mushrooms prior to packaging at farms.

  • Poly-lactic acid (PLA) is the most promising polymer in additive manufacturing as an alternative to acrylonitrile butadiene styrene (ABS). Since it is produced from renewable resources such as corn starch and sugar beets, it is also biocompatible and biodegradable. However, PLA has a couple of issues that limit its use. First, it has a comparatively low glass transition temperature of around 60 ℃, such that it exhibits low thermal resistance. Second, PLA has low impact strength because it is brittle. Due to these problems, scientists have found methods to improve the crystallinity and ductility of PLA. Polyethylene glycol (PEG) is one of the most studied plasticizers for PLA to give it chain mobility. However, the blend of PLA and PEG becomes unstable, and phase separation occurs even at room temperature as PEG is self-crystallized. Thus, it is necessary to investigate the optimal mixing ratio of PLA-PEG at the molecular scale. In this study, molecular dynamics will be conducted with various ratios of L-type PLA (PLLA) or DL-type PLA-PEG (PDLA-PEG) systems by using BIOVIA Materials Studio.

Predictive Equation of Dynamic Modulus for Hot Mix Asphalt with Granite Aggregates (화강암 골재를 이용한 아스팔트 혼합물의 동탄성 계수 예측방정식)

  • The presented work provided a predictive equation for dynamic modulus of hot mix asphalt, which showed higher reliability and more simplicity. Lots of test result by UTM at laboratory has been used to develop the precise predictive equation. Evaluation of dynamic modulus for 13mm and 19mm surface course and 25mm of base course of hot mix asphalt with granite aggregate and two asphalt binders (AP-3 and AP-5) were carried out. Superpave Level 1 Mix Design with gyrator compactor was adopted to determine the optimum asphalt binder content (OAC) and the measured ranges of OAC were between 5.1% and 5.4% for surface HMA, and around 4.2% for base HMA. The dynamic modulus and phase angle were determined by testing on UTM, with 5 different testing temperature (-10, 5, 20, 40, & $55^{\circ}C$) and 5 different loading frequencies (0.05, 0.1, 1, 10, 25 Hz). Using the measured dynamic modulus and phase angle, the input parameters of Sigmoidal function equation to represent the master curve were determined and these will be adopted in FEM analysis for asphalt pavements. The effect of each parameter for equation has been compared. Due to the limitation of laboratory tests, the reliability of predictive equation for dynamic modulus is around 80%.

  • Recent attention has been drawn to materials that undergo reversible expansion and contraction in response to external stimuli, leading to morphological changes. These materials hold potential applications in various fields including soft robotics, sensors, and artificial muscles. In this study, a novel material capable of responding to high temperatures for protection or encapsulation is proposed. To achieve this, liquid crystal elastomer (LCE) with nematic-isotropic transition properties and polyimide (PI) with high mechanical strength and thermal stability were utilized. To utilize a solution process, a dope solution was synthesized and introduced into micro-printing techniques to develop a two-dimensional pattern of LCE/PI bilayer structures with sub-millimeter widths. The honeycomb-patterned LCE/PI bilayer mesh combined the mechanical strength of PI with the high-temperature contraction behavior of LCE, and selective printing of LCE facilitated deformation in desired directions at high temperatures. Consequently, the functionality of selectively and reversibly encapsulating specific high-temperature materials was achieved. This study suggests potential applications in various actuator fields where functionalities can be implemented across different temperature ranges without the need for electrical energy input, contingent upon molecular changes in LCE.

Growth of Fe3O4 Particles and Their Magnetic Properties (Fe3O4 분말제조와 자기적 특성)

  • Fe$_3$O$_4$ particles, prepared by a sol-gel method, were examined for their structural characteristic, particle shapes and sizes, and their magnetic properties. Two different chemical compositions (using a mol rate Fe$^{2+}$/Fe$^{3+}$ = 1/2 and only Fe$^{2+}$) and 2-methoxyethanol were used for making proper solutions. And the solutions were refluxed and dry in a dry oven and the samples were fired at 200$\sim$600$^{\circ}C$ in the N$_2$ atmosphere. The formation of single-phased spinel ferrite powders was identified with the X-ray diffraction measurement as they were fired at above 250$^{\circ}C$. The result of scanning electron microscopy measurement showed the increase of annealing temperature yielded the particle size increased. The magnetic transition was observed using the Mossbaur spectroscopy measurement. As the ferrite, prepared with the chemical composition (Fe$^{2+}$/Fe$^{3+}$ = 1/2), was fired at 250$^{\circ}C$, 78% of the ferrite had a ferrimagnetic property and 22% of the ferrite was non-magnetic. In case of preparing the sample with only Fe$^{2+}$ and annealed at 200$^{\circ}C$, it had a single phased spinel structure but its particle size was too small to be ferrimagnetic. The annealing temperature above 250$^{\circ}C$ made powders a spinel structure regardless of the preparation method. They had a typical soft magnetic property and their saturation magnetization and coercivity became larger as the annealing temperature increased.

Luminescence Characterization of SrAl2O4:Ho3+ Green Phosphor Prepared by Spray Pyrolysis (분무열분해법으로 제조된 SrAl2O4:Ho3+ 녹색 형광체의 발광특성)

  • $Ho^{3+}$ doped $SrAl_2O_4$ upconversion phosphor powders were synthesized by spray pyrolysis, and the crystallographic properties and luminescence characteristics were examined by varying activator concentrations and heattreatment temperatures. The effect of organic additives on the crystal structure and luminescent properties was also investigated. $SrAl_2O_4:Ho^{3+}$ powders showed intensive green emission due to the $^5F_4/^5S_2{\rightarrow}^5I_8$ transition of $Ho^{3+}$. The optimal $Ho^{3+}$ concentration in order to achieve the highest luminescence was 0.1%. Over this concentration, emission intensities were largely diminished via a concentration quenching due to dipole-dipole interaction between activator ions. According to the dependence of emission intensity on the pumping power of a laser diode, it was clear that the upconversion of $SrAl_2O_4:Ho^{3+}$ occurred via the ground state absorption-excited state absorption processes involving two near-IR photons. Synthesized powders were monoclinic as a major phase, having some hexagonal phase. The increase of heat-treatment temperatures from $1000^{\circ}C$ to $1350^{\circ}C$ led to crystallinity enhancement of monoclinic phase, reducing hexagonal phase. The hexagonal phase, however, did not disappear even at $1350^{\circ}C$. When both citric acid (CA) and ethylene glycol (EG) were added to the spray solution, the resulting powders had pure monoclinic phase without forming hexagonal phase, and led to largely enhancement of crystallinity. Also, N,N-Dimethylformamide (DMF) addition to the spray solution containing both CA and EG made it possible to effectively reduce the surface area of $SrAl_2O_4:Ho^{3+}$ powders. Consequently, the $SrAl_2O_4:Ho^{3+}$ powders prepared by using the spray solution containing CA/EG/DMF mixture as the organic additives showed about 168% improved luminescence compared to the phosphor prepared without organic additives. It was concluded that both the increased crystallinity of high-purity monoclinic phase and the decrease of surface area were attributed to the large enhancement of upconversion luminescence.

Effects of Sowing Date on Grain Filling and Related Traits in Winter Barley (파종기 차이가 보리의 등숙과 등숙관련 형질에 미치는 영향)

  • Experiments were conducted in the research field of the Wheat and Barley Research Institute with three barley cultivars, Olbori, Kangbori and Suwon 18, which showed different growth characteristics. The highest rates of daily dry matter accumulation in grains were 1.03 1.94 mg / grain / day in early and optimum sowings and 0.88 1.88 mg / grain / day in late sowings, which occurred around 20~30 days after heading in early and optimum sowings and 15~20 days after heading in late sowings. Grains reached their maximum weight by 40 days after heading in early and optimum sowings and 35 days after heading in late sowings. Total sugar content in grains followed a pattern of linear increase immediately after heading, but it started to decrease around 20 days after heading. On the other hand, starch content continued to increase until maturity. The contents of both components were high in the order of 'Olbori' > 'Kangbori' > 'Suwon 18', but they did not respond in a regular pattern to different sowings. 1,000 grain weight showed highly significant positive correlation(r=0.767$^{**}$) with the duration of grain growth, but it had negative correlations with the average (r=-0.548$^{**}$) or the sum (r=-0.595$^{**}$) of post-anthesis daily mean temperature.ature.

Structural Characteristics and Physical Properties of Wild Silk Fibres; Antheraea pernyi and Antheraea yamamai (야잠사의 구조특성 및 물리적 성질)

  • The structural characteristics of Antheraea yamamai and Antheraea pernyi silk were investigated by using x-ray diffraction method, IR spectroscopy and polarizing microscopy. The amino acid composition, fiber density, thermal decomposition temperature and glass transition temperature were also measured for relating these physical properties to the structure in comparison with those of Bombyx mori silk fiber. There was no significant structural difference between A. yamamai and A. pernyi silk fiber on an examination of x-ray diffraction curve and IR spectrum. Both of these wild silk fibers showed double diffraction peaks at the Bragg angle 2Θ16.7˚ and 20.5˚by x-ray diffraction analysis as well as IR absorption peaks for the bending vibration of specific groups related to ala-ala amino acid sequence. On the other hand, the x-ray diffraction curve and IR spectrum of Bombyx mori silk fiber are different from those of wild silk fibers, indicating different crystal structure as well as amino acid sequences. It showed under the polarizing microscope examination that the birefringence and optical orientation factor of wild silk fibers are much lower than those of B. mori silk. Also, the surface of degummed wild silk fibers was characterized by the longitudinal stripes of microfibrils in the direction of fiber axies. The amino acid composition, which is strongly related to the fine structure and properties, was not significantly different between these two wild silk fibers. However, the alanine content was somewhat less and polar amino acid content more for A. yamamai. As a result of fiber density measurement, the specific gravities of B. mori, A. pernyi and A. yamamai were 1.355~1.356, 1.308~1.311, 1.265~1.301g/㎤ in the order, respectively. The calculated crystallinity(%) was 64% for B. mori and 51~52% for wild silk fibers, which showed same trend by IR method in spite of somewhat higher value. The thermal decomposition behaviour was examined by DSC and TGA, showing that the degradation temperature was in the order of B mori, A. prernyi and A. yamamai at around 350$^{\circ}C$. It was also observed by TGA that the decomposition seems to proceed step by step according to their specific regions in the fiber structure, resulting the difference in their thermal stabilities. The glass transition temperature was turned out to be 220$^{\circ}C$ for B. mori, 240$^{\circ}C$ A. yamamai and 255$^{\circ}C$ A. pernyi by the dynamic mechanical analysis. It is expected that the chemical properties are affected by the dynamic mechanical behavior in accordance with their structural characters.

Mineralogy and Geochemistry of Ultramafic Rocks from the Singok Area, Western Part of Chungnam (충남 서부 신곡 지역에 분포하는 초염기성암의 광물조성 및 지구화학)

  • In the Singok area, western part of Chungcheongnam-Do, two ultramafic ma~ses, Singok mass and Kaewol mass, occur as isolated lenticular bodies in the Precambrian Kyeonggi gneiss complex. The masses extend for several hundred meter to NNE direction, parallel to the main fault line of this area. The rocks are dunite and harzburgite, but partially and absolutely serpentinized. They dominantly show porphyroclastic and recrystallized textures with equigranular-mosaic and protogranular textures. In spite of differences among the alteration and metamorphism, the ultramafic masses are characterized by varying amounts of high fosteritic olivine ($Fo_{0.88-0.93}$), magnesian pyroxene ($En_{0.93-0.97}$), and tremolitic to tschermakitic hornblende with minor spinel, serpentine, chlorite, calcite, magnetite, phlogopite and talc. It is compared with adjacent gneiss complex containing amphibole, biotite, plagioclase, alkali-feldspar and quartz. Geochemically, these rocks show high magnesium number (Mg>90.38), and transitional element (Ni=595-2480, Cr==IOlO-4400, Co=36-120 ppm), low alkali element ($Na_{2}O$<0.3, $K_{2}O$<0.11, $Al_{2}O_3$<2.95 wt%) and depleted incompatible element contents, which is compared with adjacent rocks (Mg < 83.69, $Na_{2}O$=1.02-3.42 wt%, $K_{2}O$=O.67-5.65 wt%, $Al_{2}O_3$=9.15-16.86 wt%, Ni < 435 ppm, Cr < 1440 ppm, Co<59 ppm, enriched incompatible element contents). Overall characteristics of ultramafic rocks from the Singok and Kaewol masses are similar to the those of adjacent ultramafic bodies in Chungnam with worldwide orogenic related Alpine type ultramalic rocks. Calculated geothermometries suggest that the ultramafic rocks have experienced metamorphism in the condition ranging from the greenschist facies to granulite facies.

Replacement of Saline Water through Injecting Fresh Water into a Confined Saline Aquifer at the Nakdong River Delta Area (염수로 충진된 낙동강 델타지역 피압대수층에서 담수주입에 의한 염수치환 연구)

  • We performed injection tests in a deep-seated confined aquifer to assess the potential of artificial recharge as a means of preventing saltwater contamination, thereby securing groundwater resources for the Nakdong Delta area of Busan City, Korea. The study area comprises a confined aquifer, in which a 10-21-m-thick clay layer overlies 31.5-36.5 m of sand and a 2.8-11-m-thick layer of gravel. EC logging of five monitoring wells yielded a value of 7-44 mS/cm, with the transition between saline and fresh water occurring at a depth of 15-38 m. Above 5 m depth, water temperature is 10-15.5℃, whereas between 5 and 50 m depth the temperature is 15.5-17℃. Approximately 950 m3 of fresh water was injected into the OW-5 injection well at a rate of 370 m3/day for 62 hours, after which the fresh water zone was detected by a CTD Diver installed at a depth of 40 m. The persistence of the fresh water zone was determined via EC and temperature logging at 24 hours after injection, and again 21 days after injection. We observed a second fresh water zone in the OW-2 well, where the first injection test was performed more than 20 days before the second injection test. The contact between fresh and saline water in the injection well is represented by a sharp boundary rather than a transitional boundary. We conclude that the injected fresh water occupied a specific space and served to maintain the original water quality throughout the observation period. Moreover, we suggest that artificial recharge via long-term injection could help secure a new alternative water resource in this saline coastal aquifer.
