Changes in gut microbiota with mushroom consumption (버섯 섭취와 장내 미생물 균총의 변화)

  • Mushroom consumption causes changes in the immune system and gut microbiota via the actions of mushroom probiotic components. β-Glucan structure-related substances suppress secretion of inflammatory mediators, and induce macrophage activation, enhancing immunity and immune function. Substances other than directly useful components can be metabolized into short-chain fatty acids by gut microbiota. These short-chain fatty acids can then induce immunity, alleviating various diseases. Substances used to stimulate growth of health-promoting gut bacteria, thereby changing the gut microbiota community are defined to be probiotics. Probiotic altered intestinal microflora can prevent various types of bacterial infection from external sources, and can help to maintain immune system balance, thus preventing diseases. Research into beneficial components of Pleurotus eryngii, Lentinula edodes, Pleurotus ostreatus, Flammulina velutipes, Auricularia auricula-judae, and Agaricus bisporus, which are frequently consumed in Korea, changes in microbiota, changes in short-chain fatty acids, and correlations between consumption and health contribute to our understanding of the effects of dietary mushrooms on disease prevention and mitigation.

옻나무 추출액의 급여가 육계의 생산성 및 장내미생물에 미치는 영향

  • 참옻나무 추출액이 육계의 생산성과 장내미생물 변화 및 영양소 소화율에 미치는 영향을 구명하고자 1일령 육계(Ross) 960수를 공시하여 평사에서 5주간 사양시험을 실시하였다. 증체량은 추출액 급여구가 첨가수준이 증가할수록 증가하는 양상을 보였으나 통계적 유의성은 인정되지 않았다. 사료섭취량 및 사료요구율 역시 차이가 없었다. 장내미생물 변화에서 맹장내 유산균, E. coli 및 Salmonella는 처리간 차이가 없었다. 영양소 소화율은 추출액 첨가구가 향상되는 결과를 보였으나 통계적 유의성은 인정되지 않았다.

  • Gut microbiota is a group of microorganisms that resides in the intestine and serves many important functions in human health. Using 16S ribosomal RNA sequencing analysis, a wide variety of bacteria in human gastrointestinal tract has been identified along with intriguing findings that there is a different bacterial composition among individuals. Fecal microbiota transplantation (FMT) is a procedure of stool transplantation from healthy donors to patients suffering from various diseases. Specifically, FMT is able to alter the composition of gut microbiota of recipients and therefore could be an effective treatment for the patients with gastrointestinal diseases including recurrent Clostridium difficile infection, inflammatory bowel disease, and irritable bowel syndrome. Here we review a list of human diseases related to gut microbiota disturbance and the case studies of FMT. We also summarize medicines and diagnostic tools that are under development. Therefore, gut microbiota can be a next generation's biotherapy for promotion of health and treatment of chronic diseases.

Fecal Microflora of Mice in Relation to Diet (식이에 따른 장내세균의 효소활성 및 장내세균층의 비교)

  • The effects of diet on the composition of fecal microflora in mouse and the aclivilies of several enzymes in the leces were investigated. Vegetarian dietary groups were found to contain about ten lines higher numbers of Locmbocillus and B$&bacterizml than animal dietary groups. An~rnal dietmy groups were found to contain about 5 tolo times higher numbers of anaerobic Closhidia and Bocieriocles than the vegeterian detary groups. Fccal microbial $\beta$-glucosidase, $\beta$-glucm'onidase, ii-yptophanase and orease activilies in ihe animal dietary groups were shown lo be 30 to 50% hgher than those in h e vegetarian detary groups.

Comparison of mice gut microbiota before and after fasting for a day (절식이 마우스 장내미생물에 미치는 영향)

  • In this study, we investigated the effects of fasting on gut microbiota of mice fed normal (CTL) or high-fat diets (HF). Mice were raised for 16 weeks and fasted for a day at the end of the experiment. Fecal samples were collected one day before and after fasting, which were analyzed using MiSeq. Our results showed that the species richness and evenness were decreased in fasted HF group, whereas no difference was observed for CTL groups. Moreover, HF fed mice gut microbiota showed different microbial communities after fasting, while CTL groups did not show microbiota shifts. Differential abundance analysis showed that fasting CTL group mice increased and decreased one operational taxonomic unit (OTU) in S24_7 and one OTU in Ruminococcaceae, respectively. On the other hand, fasting HF group mice decreased 10 OTUs and increased 3 OTUs most of which were classified to Ruminococcaceae. Our results suggest that fasting mice may affect the abundance of Ruminococcaceae species and effects of fasting seem to be more obvious for HF-fed mice compared to those of mice fed CTL-diet.

Effects of fermented coffee on human gut microbiota (발효커피가 사람장내미생물에 미치는 영향)

  • Fermented foods have been recognized as functional foods that provide health benefits, including the modulation of intestinal microbiota. Therefore, the aim of the present study was to examine the effects of coffee beans fermented with Lactobacillus plantarum and Bacillus amyloliquefaciens on healthy human gut microbiota. Fermentation increased the content of beneficial substances (i.e., flavonoids and polyphenols). The consumption of fermented coffee increased the occurrence of beneficial microorganisms such as fiber degraders and short-chain fatty acid producers, although no significant microbiota shifts were observed after the coffee consumption. The analysis of metabolic activities also showed no difference after the coffee consumption. Our study demonstrates that the consumption of the fermented coffee may increase some beneficial bacterial while remaining the gut microbiota and its activities.

Comparative Analysis of Bacterial Diversity in the Intestinal Tract of Earthworm (Eisenia fetida) using DGGE and Pyrosequencing (DGGE 방법과 Pyrosequencing 방법을 이용한 지렁이 장내미생물의 다양성 분석)

  • The beneficial effects of Eisenia fetida on soil properties have been attributed to their interaction with soil microorganisms. The bacterial diversity of the intestinal tract of E. fetida was investigated by culture-dependent and culture-independent methods including denaturing gradient gel electrophoresis (DGGE) and pyrosequencing analyses. In a pure culture, Lysinibacillus fusiformis (51%), Bacillus cereus (30%), Enterobacter aerogenes (21%), and L. sphaericus (15%) were identified as the dominant microorganisms. In the DGGE analyses, B. cereus (15.1%), Enterobacter sp. (13.6%), an uncultured bacterium (13.1%), and B. stearothermophilus (7.8%) were identified as the dominant microorganisms. In the pyrosequencing analyses, Microbacterium soli (26%), B. cereus (10%), M. esteraromaticum (6%), and Frigoribacterium sp. (6%) were identified as the dominant microorganisms. The other strains identified were Aeromonas sp., Pseudomonas sp., Borrelia sp., Cellulosimicrobium sp., Klebsiella sp., and Leifsonia sp. The results illustrate that culture independent methods are better able to detect unculturable microorganisms and a wider range of species, as opposed to isolation by culture dependent methods.

Effect of Dandelion (Taraxacum Officinale) Extracts on the Intestinal Microorganisms of Streptozotocin-Induced Diabetic Rats (민들레추출물이 당뇨유발 흰쥐의 장내 미생물 균총에 미치는 영향)

  • This study was designed to test the effect of dandelion (Taraxacum officinale) extracts on the intestinal microorganisms of streptozotocin-induced diabetic rats. Male Wistar rats were divided into normal, diabetic control and dandelion extract groups. The extracts were prepared by water, ethylacetate and ether from leaf and root, respectively. Diabetes was induced by injecting streptozotocin (55 mg/kg BW, i.p.) in citrate buffer. The extract was supplemented in 11.45g of raw dandelion / kg diet for 50 days. The growth of Lactobacillus was more enhanced in dandelion leaf-water and leaf-ethylacetate extract group than that of the diabetic control group, whereas the growth of E. coli decreased. Results indicate that the dandelion extracts would be effective to improve the intestinal microorganisms.

옻나무 추출액의 급여가 산란계의 생산성 및 장내미생물에 미치는 영향

  • 참옻나무 추출액이 산란계의 생산성과 계란품질, 장내미생물 변화 및 영양소 소화율에 미치는 영향을 구명하고자 산란졔 Isa brown 57주령 480수를 공시하여 8주간 케이지에서 사양시험을 실시하였다. 산란율, 산란량 및 사료요구율은 참옻나무 추출액 5000 ppm 급여구가 산란율이 무첨가 대비 유의적으로 향상되었으며, 전반적으로 추출물 급여구의 산란율이 증가하였다. 평균난중과 사료섭취량은 처리간 차이가 없었다. 난각강도, 난각후도는 추출액 급여구가 무첨가 대비 유의적으로 개선되었으며, 난황색 및 Haugh unit은 전 기간에서 추출액 첨가구가 개선되는 경향을 보였다. 맹장내 유산균과 E. coli는 처리간 차이를 보이지 않았으며, Salmonella는 무첨가 대비 추출액 급여구가 약간 낮은 경향을 보였으나 통계적 유의성은 인정되지 않았다. 영양소 소화율은 옻나무 추출액 급여구가 무첨가 대비 전반적으로 향상되는 결과를 보였다.

Differences in fecal and cecal microbiota in C57BL/6J mice fed normal and high fat diet (고지방 식이 조절에 따른 C57BL/6J 마우스의 분변과 맹장에서 나타나는 미생물생태 차이)

  • A number of studies have been conducted to prevent obesity due to the worldwide increasing rate of obesity and its adverse effects on our health. Recently, a relationship between obesity and gut microbiome has been reported. Fecal and cecal microbiota are generally targeted for examining the gut microbiome during dietary interventions. There is, however, no common understanding on which microbiota and how results elucidated from the data would differ. In this study, we conducted dietary induced obesity study and compared fecal and cecal microbiota affected by dietary interventions. Normal Diet and high fat diet were fed to 6 weeks old mice for 12 weeks, and 16 S rRNA genes amplified from fecal and cecal DNA were sequenced using MiSeq. Our results show that the 𝛼-diversity showed significant differences between the dietary interventions as well as cecal and fecal microbiota. The difference in the taxonomic compositions between cecal and fecal microbiota had become clearer at the family and genus level. At the genus level, Faecalibaculum and Lactobacillus were more abundant in the cecal and fecal microbiota, respectively. In general dietary intervention studies, dietary effects are more significant than type difference. However, the microbiota analysis results should be interpreted carefully, considering both diet and samples (feces/caecum).
