Mineralogy of Sea Sand Near Ongjingun through the Separation Processes (옹진군 해사의 선별공정에 따른 광물학적 특성)

  • Mineralogical study was carried out for heavy minerals in the sea sand near Ongjingun bay, Kyonggi-do separated using the gravity and magnetic separators. Ilmenite, zircon and minor monazite and garnet were valuable minerals with gangue minerals of quartz, K-feldspar, plagioclase, muscovite, hornblende, epidote and chlorite. Quantitative analysis with SIROQUANT program showed that the contents of ilmenite separated with the gravity separation (the shaking table separation), the 1st step magnetic separation (rare earth magnetic separation) and the 2nd step magnetic separation (the Eddy current magnetic separation) were increased into 0.8, 18.3, and 48.7%, respectively. The content of ilmenite, monazite and zircon were recalculated based on the chemical composition of the representative and heavy fraction products of raw sand, the 1 step and 2 step gravity separations, and the 1 step and 2 step magnetic separations. The content increased to 0.23, 0.55, 5.22, 16.17, and 44.99% in ilmenite, 0.11, 0.02, 0.16, 0.51, and 1.19% in monazite. Although the zircon content did not differ over the processes (0.13, 0.12, 0.11, 0.15, and 0.10%), the improved recovery of zircon is expected by applying sieving process because of its high content (27%) in the fine grain size fraction (< 140#) of the 2 step gravity separation.

  • Reconstruction of rabbit corneal epithelium was performed through dynamic culture method using self-manufactured amniotic membrane supporter and lyophilized amniotic membrane. Rabbit corneal epithelial cells were cultured and cryopreserved after isolation from limbus, and the cells could be proliferated by passage number 10. The basal layer was well formed, and the epithelium layer was constructed tightly by the increase of cell proliferation and differentiation by dynamic culture method than static culture. Thus, the reconstruction of the corneal epithelium using lyophilized amniotic membrane is considered to be a good in vitro model for transplantation of corneal epithelium to patients with a severely damaged cornea.

  • The powder preparation by using emulsion drying method, one of the chemical powder fabrication methods has the advantages; easy to control the chemical stoichiometry and to fabricate homogeneously fine particles. In the present study, the initial morphology and size distribution of the powder fabricated by using emulsion dry-ing method were controlled and were improved the homogeneity. By carefully controlling the mixing ratio of oil phase and aqueous solution and surfactant of preventing emulsion separation, the Bi(Pb)-Sr-Ca-Cu-O su-perconducting powders were prepared. The properties of the superconducting powder fabricated by this method and the microstructures and superconducting properties of the pelletized samples were investigated. The microstructures and electric properties of the tapes prepared by oxide powder-in-tube method were in-vestigated. The fabricated powder was spherical with less than 1$\mu$m but most of them was agglomerated with 2~5$\mu$m in size. The critical temperature of the pelletized sample annealed at 84$0^{\circ}C$ for 72 hours in oxygen par-tial pressure of 1/13atm in Ar atmosphere was 108K. And the critical current of the first and second annealed tapes in air prepared by oxide powder-in-tube process were 0.4A and 1.5A, respectively.

Applicability of a Photosynthetic Ciliate, Mesodinium rubrum MR-MAL01 -Usefulness as a Live Prey Species for the Marine Aquaculture of Bivalves- (광합성 섬모류 Mesodinium rubrum MR-MAL01의 응용성(1) 이매패류 종묘생산을 위한 먹이생물 유용성)

  • In a series of feeding experiments using the newly established the first laboratory strain of a temperate photosynthetic ciliate, Mesodinium rubrum MR-MAL01, direct evidence was obtained for ingestion of the cultured M. rubrum cells by the bivalves like the Korean scallop, Chlamys farreri and Manila clam. Ruditapes phil-ippinarum. Each experimental Korean scallop and Manila clam removed up to 9,590 and 23,200 cells min-1, respectively. Small particles almost identical to the ruptured cells of MR-MAL01 culture were observed in the gut of the experimental bivalves, although no intact M. rubrum cell was found. In a 28 days rearing experiment, Isoshrysis galbana (KMCC H-2), a microalga, supported better growth of small Manila clam spat (0.46 mm in shell length) than MR-MAL01 strain. For the large Manila clam spat (0.84 mm in shell length), however. MR-MAL01 strain was a better prey item. Mass culture methods for this photosynthetic marine ciliate may be developed for use as live feed in aquaculture of the spat and broodstock of bivalve and small-mouthed fish larvae.

Development of the Eggs and Pre-Leptocephalus Larvae by Natural Spawning of Artificially-Matured Japanese Eel, Anguilla japonica (인위적 성숙 유도된 뱀장어 Anguilla japonica의 자연산란에 의한 난발생과 Pre-leptocephalus 자어의 발달)

  • Embryonic and pre-leptocephalic larvae development of the eel, Anguilla japonica, are described following natural fertilization in the indoor tank of $23^{\circ}C$ water temperature. Following a routine hormone treatment technique for the brood stock, female eels were artificially matured by weekly intramuscular injections of salmon pituitary extracts (SPE) at a dosage of 20 mg/kg body weight (BW) for a total of 10-14 doses to induce ovarian maturation, while male eels received weekly intramuscular injections of human chorionic gonadotropin (HCG) at a dosage of 1 IU/g BW for a total of 6-10 doses to induce testicular maturation in a separate aquarium and induced natural spawning. Fertilized eggs of about 1.0 mm in diameter were pelagic and showed a typical discoidal cleavage. Hatching occurs 38 hrs after fertilization at a water temperature of $23^{\circ}C$. The newly hatched larvae measured about 3.0 mm in total length and the number of myomeres averages 42. Their mouths and anuses were opened at 4.5 days and the yolk sacs of the pre-leptocephalic larvae were almost absorbed at 6.5 days after hatching. Pre-leptocephalic larvae survive for 14.5 days. At this time they are $5.87{\pm}0.25mm$ in total length and have about 98 myomeres. However, morphological characterization of embryonic and pre-leptocephalic larvae were not different between natural fertilization and artificial fertilization by the dry method.

Morphological and Skeletal Development of the Larvae and Juveniles of the Slender, Ilisha elongata(Bennett)(Teleostei: Clupeidae) (준치, Ilisha elongata(Bennett) 자치어(仔稚魚)의 형태 및 골격발달)

  • The purpose of the present study was to describe the morphological and skeletal development of the larvae and juvenile of Ilisha elongata(Bennett). The larvae(mean 5.02 mm in total length, TL) opened the mouth and anus with 36~38+15~16=51~54 myotomes, and anus located about 73.0% of TL. The clavicle of shoulder girdle was ossified. The larvae(mean 7.82 mm in TL) have all fin-fold differentiated and its caudal notochord began to flex. Parasphenoid of cranium and dentary of the lower jaw were ossified. The caudal notochord of the larva(mean 9.84 mm) was flexed $45^{\circ}$, and anus located about 67.0% of TL. The body height of the larvae(mean 23.32 mm in TL) were 18.0~20.0% of TL, and the aggregate numbers of all fin rays were completed. Ossification was completed in the larva of mean 26.52 mm in TL.

Development of Eggs, Larvae and Juvenile of the Korean Bullhead, Pseudobagrus fulvidraco (Richardson) reared in the laboratory (동자개의 형태발달과 성장특성)

  • The early developmental stages, growth and morphological changes of the Korean bullhead, Pseudobagrus fulvidraco, were studied from a series of reared specimens. Details of the early developmental stages are illustrated with special reference to morphological transformations. Egg and sperm of Korean bullhead were obtained from mature adults under hormonal treatment, fertilized artificially, and incubated in the aquarium. The incubation period of fertilized eggs was 55 to 66 hours at a temperature of 24.9${\pm}$0.34$^{\circ}$. Larvae were fed successively with Artemia salina and Daphnia magna for 2 to 15 days and artificial food after 20 days. Fertilized eggs were adhesive and spherical with a diameter of 2.04mm(n = 100). The mean total length of newly hatched larvae was about 4.92${\pm}$0.33 mm. Mouth opening occurred on one-day-old yolk-sac larvae, and initial feeding was observed on the third day after hatching. The morphological transitions from larvae to juvenile and juvenile to young stages occurred when the fish reached about 17 mm in total length (about 13days after hatching) and about 32 mm in total length (about 30 days after hatching), respectively. Many changes in proportion of body parts to total length were observed at about 7~8 mm and 30~32 mm, corresponding to the transformations from larvae to juvenile and from juvenile to young, respectively. In comparing relative growth of each body part against total length, those characteristics related to head parts showed positive growth in the pre-larval stages, while those concerning mobile abilities showed positive growth in the post-larval stage.

  • The re-crystallization process for recovering granular crystalline gypsum from phospho-gypsum have proposed. The process consists of two stages; at the first stage, needle-like fine gypsum is recovered with under-product of 325 mesh wet screening after the speedy hydration of dehydrated phospho-gypsum, and at the second stage, granular crystalline gypsum is recovered with upper-product of 325 mesh wet screening after dehydration and re-crystallization of the needle-like fine gypsum in $Na_2SO_4$ solution. This paper is mainly focused on the influence of dehydration time, cooling speed of temperature and re-crystallization temperature on the recovery of granular crystalline gypsum. And the re-crystallization velocity of needle-like fine gypsum at room temperature and the variation of $Ca^{2+}$ concentration of gypsum slurry during over all re-crystallization process were also discussed.

Mineralogical and Physico-chemical Properties of Fine fractions Remained after Crushed Sand Manufacture (국내 화강암류를 이용한 일부 인공쇄석사 제조과정에서 생기는 스러지의 광물.물리화학적 특성)

  • Artificially crushed sands occupy approximately 30 percent of the total consumption in South Korea. The demand for the crushed sands is expected to rise in the future. Most manufacturers use granitic rocks to produce the crushed sands. During the manufacturing process, fine fractions (i.e., sludges or particles smaller than 63 microns) are removed through the process of flocculation. The fine fraction occupies about 15% of the total weight. The sludges are comprised of quartz, feldspars, calcite, and various kinds of clay minerals. Non-clay minerals occupy more than 75 percent of the sluges weight, according to the XRD semi-quantification measurement. Micas, kaolinites, chlorite, vermiculite, and smectites occur as minor constituents. The sludges from Jurassic granites contain more kaolinites and $14{\AA}$-types than those from the Cretaceous ones. The chemical analysis clearly shows the difference between the parent rocks and the sludges in chemical compositions. Much of colored components in the sludges was accumulated as the weathering products. Particle size analysis results show that the sludges can be categorized as silt loam in a sand-silt-clay triangular diagram. This result was for her confirmed by the hydraulic conductivity data. In South Korea, the sludges remained after crushed sand production are classified as an industrial waste because of their impermeability, and which is caused by their high silt and clay fractions.

Strain Development for the Over-production of Alkaline Protease from Vibrio metschnikovii by Molecular Evolution (분자진화 기술을 통한 Vibrio metschnikovii 유래 고활성 알칼리성 단백질 분해효소 생산균주 개발)

  • Alkaline protease-overproducing strains of Vibrio metschnikovii were developed by using the molecular evolution from the classical mutants V. metschnikovii L12-23, N4-8, and KS1. Each vapK (Vibrio alkaline protease K) was obtained from the genomic DNAs of mutants by PCR to carry out the DNA shuffling. The modified vapK-1 obtained by DNA shuffling was used again as a template for the error-prone PCR to make the vapK-2. Both genes were cloned in the plasmid pKF3 to construct the recombinant plasmids which have one or two copies of the modified genes. The recombinant plasmids were back-transformed to V. metschnikovii KS1 to construct recombinant V. metschnikovii that expresses the alkaline protease. About 3.9-fold more protease activity was measured in the strain which has the plasmid containing two copies of vapK-2 when compared to strain KS1. When compared to wild type V. metschnikovii RH530, 43-fold more activity was achieved. Comparison of amino acids among vapK, vapK-1, and vapK-2 revealed that the active sites was highly conserved and not changed. However, many amino acids except the active sites were changed. These results suggested that the changes in amino acids might play an important role in the increase of protease activity by allowing the easy access of substrate to active sites of the protease. The fermentation of alkaline protease from the V. metschnikovii KS1 harboring the plasmid that contains two copies of vapK-1 showed the possibility of this strain to be used as industrial producer.
