Effect of Temperature, Day length and Sucrose on Bulblet Enlargement of Korean Native Lilies In Vitro (자생나리의 기내 소인경 비대에 미치는 온도, 일장 및 자당의 영향)

  • Eight species of Korean native lilies were used as plant materials and investigated for the effect of temperature, day length, and sucrose on enlargement of bulblet in vitro. enlargement of bulblet was enhanced at $25^{\circ}C$ in most of Korean native lilies tested, meanwhile it was promoted at $20^{\circ}C$ in L. davuricum and L. hansonii. Moreover, enlargement of bublet was promoted under dark in L. concolor var. parthneion, L. distichum, L. amabile, and L. davuricum, to the contrary fresh weight was increased under longer day length in L. hansonii and L. maximowitzii. That of L. concolor var. parthneion and L. cernuum was not affected by light or dark condition. Fresh weight of the bulblets produced in vitro of most Korean native lilies tested was increased with increasing sucrose concentration and best at $120g{\cdot}L^{-1}$ sucrose in medium. However, enlargement of bublet was promoted by $60g{\cdot}L^{-1}$ sucrose in L. tsingtauense and L. cernuum and by $90g{\cdot}L^{-1}$ in L. davuricum. Bulblet enlargement tended to promote by lower rate of scaly leaf occurrence or separating bulblet and it was affected by occurrence of scaly leaf rather than separation of bulblet. However, occurrence of scaly leaf, separation of bulblet, and enlargement of bulblet were more significantly affected by genetic factors, species rather than culture conditions.

  • By analysis of pyruvic acid and sugar levels in bulbs, leaf sheaths, and leaves, and with regard to phyllotaxis, it was shown that the lower portion of each plant component had the greatest pyruvic acid content, the upper portion less, and the middle portion the lowest. When bulb scales were examined, pyruvic acid content was lowest in the second bulb scale from the papery scale. Pyruvic acid level increased toward the inner portion of the bulb. Pyruvic acid levels in leaf sheaths and leaves were greater than those in the bulb during the early stages of bulb growth. However, at harvest, the pyruvic acid content of bulbs increased slightly, whereas those of leaf sheaths and leaves decreased markedly. Tissue sugar contents were similar at the beginning of bulb growth. However, bulb sugar content increased greatly as the bulb grew, and sugar contents of leaf sheaths and leaves decreased. With respect to phyllotaxis, outer older leaves had the lowest total sugar and pyruvic acid contents. Young leaves showed a tendency to have less pyruvic acid and sugar compared with mature leaves.

The Effect of Culture Methods and Plant Growth Regulators on Bulblet Formation and Growth in Scale Segment Culture of Fritillaria thunbergii Miq. (패모 인편 배양시 자구 형성과 비대에 미치는 배양 방법과 생장 조절제의 처리 효과)

  • This experiment was carried out to establish a year-round production system of pathogen-free stock through micropropagation in Fritillaria thunbergii as medicinal bulbous plant. The effect of different types of culture method and plant growth regulators, activated charcoal and mannitol on bulblet formation and subsequent growth were investigated. The MS solid medium containing 1. 0 mg/L kinetin and 0. 3 mg/L NAA was effective on bulblet formation and propagation rate compared to liquid and suspension culture. Addition of activated charcoal at 0. 01% to 0. 1% in the medium promoted bulbing of cultured bulblets and shoot formation. Addition of 1% to 2% mainnitol in MS medium was effective on the formation of bulblet and subsequent growth of bulblets compared to control. In addition of inhibitors, $10{\sim}100\;mg/L$ B-9 and Chloromequat had effective to stimulate bulblet growth but their effects were not so much as mannitol treatment. ABA treatment had detrimental effect on survival rate of explant and bulblet formation.

Effects of Growth Regulators and Sucrose Concentrations on the Bulblet Formation through In Vitro Culture of Scale Segment in Nerine bowdenii (네리네(Nerine bowdenii)의 기내 인편배양시 자구형성에 미치는 생장조절제와 Sucrose 농도의 영향)

  • The twin-scale segments of nerine (Nerine bowdenii) were cultured to investigate the influence of NAA, BA and sucrose concentrations on in vitro bulblet formation. The formation of bulblets from twin-scale segments showed a good response both the percentage of bulblet formation and the number of bulblets per explant on MS medium supplemented with 1mg/L NAA and 2 mg/L BA. Formation of bulblet showed the highest efficiency on medium containing 30g/L, and the formation of bulblets was strongly inhibited on medium containing over 90g/L. When the twin-scale segments formed bulblets were subcultured three times to the same medium by 60 day subculture interval, the number of bulblets per explant was 6.5, 7.3 and 8.2 in order of first, second and third. The bulblets over 3mm in diameter were hypertrophied and rooted after transferring to the hormone-free MS medium. The plantlets over 50mm in height were successfully acclimatized in the soil mixed with the same volume of vermiculite and perlite, and the survival rate was over 95%.

Micropropagation of Lillium Oriental Hybrid 'Casa Blanca' using Bulblet Sections with swollen Basal Plate in Bioreactor (생물반응기에서 저반부가 비대된 자구 절편체에 의한 오리엔탈 나리 'Casa Blanca' 의 대량증식)

  • A series of studies were carried out to establish micropropagation system, using airlift bioreactors (ebb $\varepsilon$ flood type, 5 L), of Lilium oriental hybrid 'Casa Blanca'. The bulblets with swollen basal plate were formed from bulb scales, then proliferated to bulblet clusters with swollen basal plate. Finally normal bulblets were formed from the sections. Bulblet formation and proliferation with swollen basal plate were not accomplished entirely in liquid culture of 5 L airlift bioreactors, but leafy bulb scales grew vigorously. Bulblet clusters with swollen basal plate were proliferated by periodic immersion culture. Bulblet proliferation was not affected by light, but scale leaves grew under light. MS medium containing 2.0 mg/L benzyl adenine (BA) and 0.3 mg/L indole acetic acid (IAA) was favorable to the bulblet proliferation with swollen basal plate. In liquid culture of 5 L bioreactors, bulblets from bulblet sections with swollen basal plate grew vigorously on MS medium with 70 g/L sucrose. It was effective for bulblet growth to replace the new medium after 8 weeks in culture during 16 weeks of cultural period. 15 g injection of bulblet sections as a cultural material was suitable for bulblet growth in 5 L bioreactors.

On Improvement of Garlic Productivity by Inactivation of Virus in Garlics (마늘 Virus 불활성화에 의한 생산성 향상에 관하여)

  • The effect of heat or chemotherapeutant treatment on the mosaic virus infected garlic(Allium sativum L.) scales and that of chemotherapeutant added to the culture medium were summarized as following. The treatment of the virus infected garlic scales at $37\~57^{\circ}C$ for 35 days to one hour in water or in air shelved no effort of inactivating the virus. Although treatment of the garlic scales at $62\~72^{\circ}C$ for 90 to five minutes reduced the mosaic symptom on the leaves of the garlic plants grown after the heat treatment, it reduced the growth vigor of the plants so greatly that complete inactivation of the virus in garlics was not feasible. The mosaic symptom on the leaves of garlic plant was reduced when the infected garlic scales were grown after 24 hours soaking in $10\~50\;ppm$ Malachite Green, 2,4-Dichlorophnoxy Acetic Acid, or in $20\~100\;ppm$ Quinhydron. These chemotherapeutants, however, inhibited the growth of garlic plant at the high concentration. Garlic scales soaked in $10\~50\;ppm$ Naphthyl Acetic Acid showed the least mosaic symptom without its complete extiction on the garlic leaves. When incorporated into the modified Murashige-Skoog's medium, $0.5\~l.5\;ppm$ Naphthyl Acetic Acid could inactivate the mosaic virus in newy developed garlic plants showing no mosaic symptom on the leaves, no inclusion bodies and intact nuclei in the leaf tissue cells.

Production of Seed Garlic by Sawing Bulbils of Southern Type Garlic in Jeju Island (제주지역에서 난지형마늘 주아 파종에 의한 종구 생산)

  • This study was conducted to evaluate cultivation to save the production method cost of warm type garlic. The optimum system for producing excellent seed bulbs by using bulbils of garlic has been required. The bigger size bulbils induced fast growth and lower rate of non-cloved bulbs. The ratio of non-cloved bulbs was the highest in the 0.1 g bulbils sowing of 'Namdo' and was low in the big size bulbils sowing. The bulbil growth rate of 'Deaseo' was much faster than that of 'Namdo', but most of 'Deaseo' bulbils showed low rate of non-cloved bulbs and small size scales in bulbs. The higher productivities of "Namdo" bulbils appeared at the September 11th sowing time, but 'Deaseo' bulbils produced small size non-cloved bulbs and got the very low ratio of non-cloved bulb production in all of the treatment, sowing that Jeju Island could not produce the non-cloved bulbs with 'Deaseo' garlic bulbuls. The growth rate such as leave number, leave length, bulb and clove size in the sowing distance of big size bulbils increased as the sowing distance was wider, but the rate of non-cloved bulbils decreased regardless of treatment. More than 3 g clove in the $10{\times}15cm$ distance of bulbs was produced resulted in sowing clove production potentials by using big size bulbil.

Establishment of in Vitro Culture and Effect of Chilling Treatment of Mother Bulb On Bulblet Formation in Fritillaria thunbergii Miq. (패모의 초기배양과 모구의 저온처리가 자구형성에 미치는 영향)

  • This experiment was conducted to obtain basic information for the establishment of in vitro initial culture system in Fritillaria thuubergii Miq. Methods of surface sterilization of scale segments as explant and effect of antibiotics added into the culture medium on contamination of explant and chilling treatment of mother bulb on bulblet formation were investigated. Portent of contamination of cultured scale segments was significantly higher in the outer scale segments which were unsuitable as initial culture explant than inner scale segments. Contamination of explants taken from inner scale of bulb was reduced by surface sterilizing explants in the solution of $4{\sim}5%$ sodium hypoclorite for $10{\sim}15$ mimutes. Addition of antibiotics such as kanamycin, vancomycia cefotaxim, agrirnycin and agreptomycin and dithane as fungicide and$lncyte^{tm}$ into MS medium was effective to reduce bateriological contamination, but did not work to control fungi. It had effective to delay the degree of contamination caused by fungi and bacteria haboring in cultured explants. Bulblet formation from cultured scale segments was promoted by dry storage for $2{\sim}4$ weeks or moisture storage of mother bulbs for $4{\sim}6$ weeks at $10^{\circ}C$ before excision of explants. Addition of kinetin into medium could not exerted for the bulblet formation from the scale segment of dry storaged bulb compared to control. But explant taken from 6 week moisture storaged bulb formed more than 10 bulblets per explant on the medium containing $3{\sim}5mg/L$ kinetin.

  • To investigate the effects of MS medium strength and nitrogen concentration on bulblet formation from bulblet scale segment culture and bulblet growth from bulblet of Lilium oriental hybrid 'Casa Blanca', asiatic hybrid 'Mona', and longiflorum hybrid 'Hinomoto', 0.5∼2.0 strength of MS salts and 30∼120 mM nitrogen concentrations of MS medium were examined in vitro. The number of bulblets from bulb scale segment was favored in the strength of 0.5∼1.0 strength of MS salts or 30 mM total nitrogen concentration of MS medium in three cultivars. But the growth of bulblets formed in vitro was promoted in the 2 strength of MS medium or hight concentration of total nitrogen of MS medium up to 120 mM in three experimented cultivars.

Growth environment and nutritional evaluation of native Allium victorialis var. platyphyllum in Ulrung island (야생 산마늘의 생육 환경과 영양 평가)

  • In Ulrung floral community the growing environments and nutritive values of wild garlic was investigated. The wild garlic, grown for $3{\sim}4\;years$ in community area, were able to develope bulbs. The bulbs with 2 leaves had the highest ratio of propotion as 75.1% of share among the distribution of bulbs with completely developed leaves and 1 leaf-bulbs and 3 leaves-bulbs had 19.6% and 5.3%, respectively. Also the 2 or 3 leaves-bulbs indicated the better growth states in the part of shoot and bulbs than 1 leaf-bulbs, and there was a significant defferences in each growth between 2 leaves-and 3 leaves-bulbs. On the wild garlics we cannot find out insect injuries. However, there was deseases with symptoms of leaf-spots and lesion from May to June. At first it made a small spots on the leaves, followed by senescence at arrounding area of spots, and then it expanded to whole leaf. This pathogenic bacterium was supposed to be a member of Aristastoma. The contents of crude protein, fat, and carbohydrate in each portion of wild garlics decresed in the order of shoot, expanded leaves and bulbs. On the contrary, crude fibers increased in the same order and so it is not suitable for human consumption nutritional value. However, because the shoots of wild garlics have more than the other Allium sp., the shoots will be a good quality food stuff.
