Deutzia paniculata is an endemic species, which is geographically restricted within southern part of Korea. Four populations of D. paniculata were sampled across its natural range, from the smallest population, Mt. Dalum, which held less than 100 individuals, to the largest, Mt. Unmum, over 3,500 individuals. Artificial pollination study showed that D. paniculata had an obligate outcross breeding system. Major pollinators were two bee species, Lasioglossum exiliceps and Allograpta balteata (de Geer). The breeding system and patterns of allozyme variation of D. paniculata were investigated to understand the population biology and to explain on reserve designs and management proposals relevant to this species. D. paniculata held relatively low genetic variation at the eight allozyme loci surveyed. Measures of genetic variation in this species alleles per locus ($A_s=1.33$), proportion of polymorphic loci (P=23.85%), and expected heterozygosity ($H_{es}=0.110$) were similar to values reported for endemic species. Mt. Dalum population (DAL) was composed with one clone based on allozyme data. Individuals of D. paniculata were frequently included in root connected clusters. Population genetic structure between and within four populations was probably the result of shrinking effective population size and the extinctions of intervening populations. For the conservation of genetic diversity, maximum number of different genotype need to be protected based on genetic structure and mating system.

We investigated the genetic diversity of an endemic rare species, Forsythia ovata Nakai by examining 93 ISSR amplicons in 84 individuals distributed among five populations. The overall percentage of polymorphic ISSR amplicons was 54.8% and mean number of amplicons per ISSR primer was 6.6. The amount of genetic diversity was relatively lower than other shrub species. The Mt. Seokbyeong and Mt. Seorak B populations had the highest level of genetic diversity. Although the Seokgae-jae population had the lowest level of genetic diversity, the population was genetically the most distinctive from the other populations. About 30.6% of the total variation was allocated between five populations, which was slightly higher than other shrub species. Such a pattern of genetic variation may have resulted from the limited distribution and small population sizes of F. ovata. The UPGMA dendrogram based on Nei's genetic distance showed some decisive geographic patterns. These results suggest that, in addition to the preservation of the natural stands, the conservation of larger number of populations with small number of individuals per population is more effective for the dynamic ex situ conservation and for maintaining the genetic diversity of F. ovata than smaller number of populations with large number of individuals.
Genus Phyllostachys is a long-lived woody species primarily distributed throughout South East Asia. Many species of this genus has been regarded as medically and ecologically important in the world. We evaluated representative samples of the four taxa with RAPD to estimate genetic relationships within the genus Phyllostachys. The percentages of polymorphic loci were 8.9-33.3% at the species level. P. bambusoides was found to show lower genetic diversity (H=0.018) than other species. Total genetic diversity ($H_T$) was 0.315, genetic diversity within populations ($H_S$) was 0.043, the proportion of total genetic diversity partitioned among populations ($G_{ST}$) was 0.659 and the gene flow (Nm) was 0.0263. As some Korean populations were isolated and patchily distributed, they exhibited low levels of genetic diversity. The four taxa of the genus Phyllostachys analyzed were distinctly related to a monophyletic. P. nigra var. henonis. Stapf was found to be more closely related to P. pubescens than to P. nigra. P. bambusoides was quite distinct from the remaining species.
In order to shed some light on the historical change of biodiversity concepts, this paper reviews the science and technology studies (STS) literature on the history of biological research on human genetic diversity. By doing that, I show how the notion of genetic diversity in the human population - from "race" to "population" to "biogeographical ancestry" - has changed with methodologies and techniques over the last hundred years. In the meantime, I point out contexts and situations, despite conceptual and methodological developments, that show that current human genetic diversity research is slipping into the past mistakes of scientific racism. This article offers biodiversity researchers an opportunity to consider their own scientific practices on classifying species more reflectively.

This study employed inter-simple-sequence repeat (ISSR) to assess genetic variation among 189 individuals representing 10 populations (nine in Korea and one in Japan) of Millettia japonica, which has recently been lifted from the endangered species of Korea. The calculated Shannon's information index value (I = 0.2689) of the species was appreciable and was higher than other endangered leguminous woody taxa. Gochang (I = 0.2968), Namhae (I = 0.2951), and Mt. Toham (I = 0.2823) populations showed relatively high genetic diversity, whereas the Kyushu (in Japan) population (I = 0.2487) exhibited the lowest. The results of an analysis of molecular variance indicated that 86.49% of the diversity was attributed to within populations, and 13.51% to differences among populations, suggesting that M. japonica populations do not have significant geographic differentiation and that the gene flow between populations exists to some extent (Nm = 1.8446). Continuous habitat monitoring should be conducted to conserve genetic diversity of M. japonica, particularly for those populations with relatively high genetic diversity. Selection of many individuals from the populations in Gochang, Namhae, and Mt. Toham is thought to be an appropriate strategy for ex situ conservation of M. japonica in Korea.
Olive flounder (Paralichthys olivaceus) is one of the most popular farmed fish in Korea. Genetic variability of the fish was investigated by means of microsatellite DNA markers. All of the 8 microsatellite loci were analyzed in this study. For the confirmation of genetic variation during a shift in generation, microsatellite variability was compared within the same hatchery strains but produced in different spawning years. When genetic variability of farmed flounders produced in 2006 and 2007 was compared with that of 2003, a marked reduction of genetic variability was observed in the 2006 and 2007 populations. Mean number of alleles per locus and expected mean heterozygosity decreased from 9.75 and 0.796 (in 2003 population) to 7.78 and 0.785 (in 2006 population), respectively. Moreover, we have observed the distortion of allele frequency. These results show that reduced genetic variability of farmed olive flounder in processed generation has lower numbers of alleles and genetic variability than these of wild fish. Our results suggest that to have a sustainable aquaculture of this species, there is need for scientific broodstock management based on genetic variation and more intensive breeding practices to improve genetic diversity and to avoid detrimental inbreeding effects.
The epidemics of rice blast which occurred in south parts of Korea during the period from 1999 to 2001 and damaged several high quality rice cultivars developed using "Milyang 95" and/or "Milyang 96" as a parent. Genetic diversity of 23 rice cultivars including "Milyang 95" and it's relatives was assessed using 54 simple sequence repeats (SSR) markers reported to be linked to major blast resistance genes. Fifty-four SSR markers representing fifty-seven loci in the rice genome detected polymorphism among the 23 cultivars and revealed a total of 170 alleles with an average of 3.0 alleles per primer, The number of amplified bands ranged from 1 to 7. Several SSR markers including RM249, RM206 and OSR20 were informative for assessing the genetic diversity of relatively closed japonica rice cultivars. The 23 cultivars were classified into four groups by cluster analysis based on Nei's genetic distances, and the cultivars developed from same parents showed a tendency to cluster together that is consistant with genealogical information. High quality rice cultivars, Daesanbyeo, Donganbyeo, and Milyang 95 belonged to the same cluster, At the loci, RM254 and OSR32, all of the cultivars derived from the crosses using "Milyang 95" shared same alleles, suggesting that these japonica cultivars might carry alleles that are identical by descent. AFLP markers were employed to detect genetic diversity and genetic relationship among 26 accessions of foxtail millet collected from Korea. Analysis of 26 accessions of foxtail millet with nine AFLP primer set combinations detected a total of 170 bands, of which 145 (85.3%) showed polymorphic at the species level. The phenotypic diversity (Hs) calculated for the nine primer combinations ranged from 1.84 to 6.8, with an average of 3.85. The average phenotypic diversity values were 3.39 and 2.99 for accessions collected from Gangwon province (Group 1), and accessions collected from the other areas including Gyeonggi province (Group 2), respectively. In the cluster analysis of 26 accessions, two major groups were recognized at 73% genetic similarity. Group I includes 13 accessions, which were collected in Gangwon province, and 1 accession, which was collected in Gyeonggi province, with genetic similarity of 76.8%. Group II includes two accessions, which were collected in Gangwon province, and 10 accessions, which were collected in the other areas with genetic similarity of 78.9%. The presenting data on the assessment of genetic diversity and genetic relationships among Korean accessions of foxtail millet will be helpful for efficient collection or conservation strategy of foxtail millet germplasm in Korea.
