Several solvents were used to fractionate an extract obtained from the chapped root bark of Ulmus davidiana Planchon. The each fractionary part was condensed under reduced pressure and then examined to investigate the inhibitory effect on MMPs by modified gelatin zymography, where EA fraction showed the inhibition effect on the activity of MMPs. A compound showing inhibition effect on the MMPs was isolated and purified from EA fraction. Under IR, $^1H$- and $^{13}C$- NMR analyses it is very close to a catethin. This substance showed 48% inhibition effect on measurement of MMP-9 activity at 5 mM and 43% at 10 mM. To verify the effect of this substance on cells, human hepatoma, SK-Hep-1 cells as a cancer model, and Chang liver cells as a normal model were selected. MTT assay was performed to examine the cell viability by treatment of $1{\mu}L/mL$ of the purified substance on cells. The purified substance showed negligible toxicity on human liver cell line.
Natural anti-oxidative compounds have important disease prevention and food preservation properties, in addition to anti-bacterial, anti-inflammation, anti-cancer, and skin whitening effects. High-performance liquid chromatography (HPLC), with an ultra vilolet (UV) detector coupled to a reverse phase C18 column and an online measurement system for 1, 1-diphenyl-2-picryl hydrazyl (DPPH) radicals, was used to search for potent antioxidative compounds in crude extracts. The online HPLC-DPPH assay was then applied to confirm antioxidative compounds in water extracts from Radix of Pueraria lobata, Rhizoma of Zingiber officinale, Fructus of Chaenomeles sinensis, Cortex of Ulmus pumila, and Radix of Astragalus membranaceus. To determine the yields of the extracts, the Brix% of each extract solution was measured using a refractometer. When the relative DPPH radical scavenging ability values of the water extracts were compared with those of a positive control (ascorbic acid), the water extracts of P. lobata, C. sinensis, and U. pumila were 7.77%, 4.71%, and 4.19%, respectively. The results suggest that this method provides a useful assay for rapid measurement of DPPH radical scavenging abilities and conformation of antioxidative compounds in natural products. Moreover, it can reduce the time spent on the separation of active compounds from natural materials, such as medicinal plants, in addition to the use of reagents for separation.

The fruit or root of Ulmus davidiana var. Japonica and its extract has traditionally been used as anti inflammatation medicines as well as a remedy of the digestive ulcer for a long time. Ulmus davidiana var. Japonica has also been used to cure palliative and diuresis, because it contains lot of mucus and tannin. In this study, we examined the moisturizing effect and anti-inflammation effect of polysaccharides extracted from Ulmus davidiana var. Japonica to investigated whether it can be used as a cosmetic ingredient or not. The polysaccharides were extracted from Ulmus davidiana var. Japonica. Then the hydrolyzed was obtained through the hydrolysis. It has been ascertained that the polysaccharides we mainly composed of rhamnose, galactose, and glucose through HPLC experiments, and it has 20,000 of the molecular weight and 89.3 dL/g of intrinsic viscosity In a moisturizing test of measuring water loss in a desiccator and moisture content by Comeometer CM820, Ulmus davidiana root extract showed a similar moisturizing effect as hyaluronic acid. And its anti-inflammation effect on carrageenin-induced edema evaluated by measuring the thickness of swelling over paw induced by carrageenin (25 uL of $2\%$ carrageenin saline injection), the $\%$ swelling over paw treated with Ulmus davidiana root extract was much less than that of paw treated with carrageenin only. The anti-inflammatory effect of Ulmus davidiana root extract was almost similar to that of ketoprofen. These results can say that Ulmus davidiana root extract can be effectively used as a cosmetic ingredient.
This study was carried out for the functional investigation of the $Ulmus$$pumila$ L. extracts for use in functional-food processing. Extracts of $Ulmus$$pumila$ L. were obtained using distilled water and 70% ethanol, and the extracts were tested for their electron-donating ability, SOD-like activity, nitrate-scavenging ability, and anticancer (MDA and A 549 cells) activity. The extraction yields of the water and ethanol extracts were 12.7 and 12.0%, respectively; the polyphenol contents were $623.5{\pm}2.4$ and $710.5{\pm}2.1$ mg/100 g; the electron-donating ability was high in proportion with the density; and the water extract was higher than the ethanol extract (76 and 64%, respectively) at 1,000 ppm. In all the 1,000 ppm densities, the SOD-like activity of the water extract was far higher than that of the ethanol extract (53 and 38%, respectively), and the nitrite-scavenging ability of the ethanol extract was higher than that of the water extract (47 and 43%, respectively). As for the anticancer ability at 1,000 ppm, it was 62% in the water extract and 42% in the ethanol extract in the MDA cell, and 60% in the water extract and 45% in the ethanol extract in the A 549 cell. Thus, the proliferation inhibition ability of the water extract against cancer cells was found to be far higher than that of the ethanol extract (60 and 45%, respectively).
We prepared extracts from Ulmus davidiana (root, Korean source; URK) and Ulmus davidiana (bark, Korean source; UBK). URK extracts obtained with all tested solvents showed the highest antioxidant effects on fish oils. Both treatments containing 0.1% (v/v) extract from URK and UBK each showed that peroxide values of 30 meq/kg were maintained for 6 h and levels of 40 meq/kg were apparent for up to 18 h, indicating that antioxidative activity seemed to sustain during all tested time periods. Compared with commercial antioxidants, butanol and methanol extracts diluted to 0.05% (v/v) had similar antioxidative effects. Water and butanol UBK extracts diluted to 0.1% (v/v) both showed the highest antioxidative activities. After addition of metal ions, methanol and butanol URK extracts diluted to 0.1% (v/v) showed enhanced antioxidative activity. UBK ethanol extracts displayed superior antioxidative activity and a constant peroxide value throughout storage. However, in the case of Perilla oil, $\alpha$-tocopherol which is known as a natural antioxidant did not show any antioxidative activity except in the BHT. Methanol and butanol URK extracts diluted to 0.2% (v/v) showed superior antioxidative activities throughout the experiment. A methanolic UBK extract (0.2%, v/v) also had a similarly increased antioxidative effect. In tests involving addition of metal ions to all extracts, the methanolic UBK extract (0.2%, v/v) showed excellent antioxidative activity. When lard was tested, antioxidant levels did not differ significantly among extracts prepared using four different solvents at either 0.05% or 0.1% concentrations (both v/v). Addition of metal ions at levels of 0.05% or 0.1% (w/v) to these extracts had no significant additive effect on oxidation.
To increase the fibrinolytic activity and production of mycelium, extracts of 7 plant species were supplemented to the growth media of Armillaria mellea, and mycelial growth and enzymatic activity in the mycelium extracts of A. mellea were estimated. The mycelial production of A. mellea was slightly increased by adding ASH-R, UDVN or RGR extract, whereas KG extract significantly affected the growth. Supplement of ASH-S, UDVN and RGR extracts increased proteolytic activity from 36.8 to 46.1% Fibrinolytic activity was increased to $50{\sim}65%$ by supplement with RVS, ASH-S and RGR extracts, respectively. Enzyme extracts of the fungus grown with RGR extract supplement degraded all chains of fibrinogen within 2 hours, whereas control was required 3 hours. Degradation of fibrin fragments by the enzyme extracts was also observed through microscopy.
Antioxidative and antibacterial activities of endemic plants extracts in Korea were investigated. Hydrogen radical scavenging activity was 99.72% in an elm tree, 99.725 in a Job's tears, 99.575 in an eggplant, and 94.025 in a barrenwort. Antibacterial activity of ethanol extract from 23 different species of wild plants were determined. The gull nut was showed the strongest antibacterial activities (16.0-19.0 mm) and also showed high antibacterial activities in a pine neddles, a gulmyungja, a wild ginger plants, a ginko (9.5-11.5 mm). Only extracts of Saurrranceae extract showed the antibacterial activity in Bacillus subtilus, Stapylococcus aureus, Escherichia coli, Samonella entetotidis. An antibacterial activities were showed weakly in an esungcho and an eggplant. A ground ivy showed Antibacterial activity on Stapylococcus aureus. A barren wort showed in antibacterial activity on Stapylococcus aureus, and E. coli. This results were suggested that many edemic plants resources contains antioxidative and antibacterial substances.

The evaluation of functional active ingredients and dyeing effect on cotton fabric using 16 types of natural products water extracts and 50% ethanol extracts, there were differences in color intensity, but 6 types of red, 5 types of yellow, 2 types of orange, and 3 types of green were observed. In general, the color of the 50% ethanol extract was high and clear. As a result of dyeing by immersing cotton gauze in water extracts and 50% ethanol extracts of these natural products for 2 days, the dyeing effect was generally good, and it was observed that there was little fading decrease in color even after drying at room temperature and opening and storing. The pH of the water extracts of 16 natural products ranged from neutral to slightly acidic, with no side effects on the skin. For DPPH free radical inhibition antioxidant effect, the effect of 50% ethanol extract for each sample was better than that of water extract. As for the polyphenol content, the content of the 50% ethanol extract tended to be higher than that of the water extract. Therefore, the purpose of this study was to investigate the correlation between the antioxidant activity of 16 natural extracts and the prevention of female vaginitis.
Objectives : This study was to investigate the inhibitory effects of Ulmus davidiana on the generation of peroxynitrite $(ONOO^{-})$, nitric oxide (NO) and superoxide anion radicals $(O_{2}^{-})$ in the endothelial cells of rat vessels. The effects of Ulmus davidiana on the expression of inflammation-related proteins, $NF-{\kappa}B$ (p50, p65), COX-2, and iNOS, were examined by western blotting. Methods : For this study, fluorescent probes, namely dihydrorhodamine 123 (DHR 123), 4,5-diaminofluorescein (DAF-2) and 2',7'-dichlorodihydrofluorescein diacetate (DCFDA) were used. Western blotting was performed via using anti-$NF-{\kappa}B$ (p50, p65), anti-COX-2, and anti-iNOS, respectively. Results : Ulmus davidiana inhibited the generation of $ONOO^{-}$, NO and $(O_{2}^{-})$ in the lipopolysaccharide (LPS)-treated endothelial cells of rat vessels in vitro. Ulmus davidiana inhibited the expression of COX-2 and iNOS genes by means of decreasing the $NF-{\kappa}B$ activation. Conclusions : These results suggest Ulmus davidiana is effective on inhibiting the generation of $ONOO^{-}$, NO and $O_{2}^{-}$, and that therefore it might have a potential role as a treatment for the inflammatory process and inflammation-related diseases.

The bark of the root and stem of Ulmus davidiana var. japonica has been used as a traditional Korean medicine to treat inflammatory disorders. This plant reportedly shows antioxidant, anticancer, and anti-inflammatory effects. In this study, we investigated the protective effects of Ulmus davidiana var. japonica ethanolic extract (UDE) on UVB irradiation-induced wrinkle in hairless mice. We evaluated for their free radical-scavenging activities against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and the anti-elastase activities, and for their anti-matrix metalloproteinase-1 (MMP-1) activity in human skin fibroblast cells. In the wrinkle measurement and image analysis of skin replicas, the results showed that UDE significantly inhibited wrinkle formation caused by chronic UVB irradiation. These results suggest that UDE has anti-wrinkle activity.
