  • This experiment was conducted to investigate the effects of growth retardant uniconazole on the growth and flowering of Cymbidium Pine Clash 'Moon Venus' and Cym. Green Sour 'A One'. Reduction in leaf length of both cultivars was proportional to the amount (0-5g) of uniconazole and growth reduction of 'A One' was more remarkable than 'Moon Venus'. The inflorescence length was reduced 18.4% in 'Moon Venus' and 15.3% in 'A One' by 5g treatment of uniconazole. There was no significant difference in flowering time, number of inflorescences and florets and size of lower sepal regardless of uniconazole treatment. It was concluded that uniconazole treatment is effective in reducing the leaf growth of Cymbidium. However, as high concentration (5g) of uniconazole reduced the growth of inflorescence causing low quality, 1g was recommended as useful concentration.

  • This experiment was conducted to investigate the effects of a growth retardant uniconazole treatment time on the growth and flowering of Cymbidium Pine Clash 'Moon Venus' and Cym. Green Sour 'A One' in order to develop a substitutional technique on the highland cultivation. Leaf growth of both cultivars was more remarkably reduced by earlier uniconazole treatment time and growth reduction of 'A One' was more remarkable than 'Moon Venus'. Flowering time of 'Moon Venus' treated with uniconazole at May 7 and 27 was accelerated by 5 and 6 days respectively. Moreover, the number of inflorescences and florets showed a tendency to increase. However there was no difference in flowering time of 'A One' among treatments but there was a tendency to decrease in the number of inflorescences and florets by uniconazole treatment. In both cultivars, there was no difference among treatments in the length and width of lower sepal which represent the floret size.

Effect of a Combined Treatment with Uniconazole, Silver Thiosulfate on Reduction of Ozone Injury in Tomato Plant (Uniconazole 과 Silver Thiosulfate 의 복합처리가 토마토의 오존피해경감에 미치는 효과)

  • Studies were conducted to determine the combined effect of uniconazole [(E) -1-(4-chlorophenyl)-4, 4-demethyl 2-(1,2,4 triazol-1-yl)-1-penten-3-ol] and silver thiosulfate $[Ag {(S_2O_3)}^3\;_2-]$ (STS) on reduction of ozone injury in tomato plants(Lycopersicon esculentum Mill. 'Pink Glory'). Plants were given a 50ml soil drench of uniconazole at concentrations of 0, 0.001, 0.01 and 0.1 mg/pot at the stage of emerging 4th leaf. Two days prior to ozone fumigation, STS solution contained 0.05% Tween-20 was also sprayed at concentrations of 0, 0.3 and 0.6 mM. Uniconazole at 0.01 mg/pot and STS at 0.6 mM were effective in providing protection against ozone exposure(20h at 0.2ppm) without severe retardation of plant height and chemical phytotoxicity, respectively. Combined treatment with uniconazole, STS significantly reduced ozone injury at the lower concentration than a single treatment with uniconazole or STS. Uniconazole treatment reduced plant height, stem elongation and transpiration rate on a whole plant level and increased chlorophyll concentration. STS did not give any effect on plant growth and chlorophyll content but increased transpiration rate in non-ozone-fumigated plants. Ethylene production in the leaves of ozone-fumigated plants was decreased by uniconazole and STS pretreatment, but there was no protective effect on epinasty of leaves in uniconazole-treated plants. STS increased ethylene production in non-ozone-fumigated plants, but it significantly reduced the degree of epinasty and defoliation of cotyledons when plants were exposed to ozone. Uniconazole slightly increased superoxide dismutase and peroxidase activities. But STS showed little or no effects on such free radical scavengers. Day of flowering after seeding was shortened and percentages of fruit set were increased by uniconazole treatment. STS was highly effective on protecting reduction of fruit set resulting from ozone fumigation. These results suggest that combined use of uniconazole and STS should provide miximum protection against ozone injury without growth retardation resulting in yield loss.

Effects of Seed Soaking Treatment of Diniconazol on the Inhibition of Stretching of Tomato and Cucumber Seedlings (Diniconazol의 종자침지 처리가 토마토와 오이 플러그묘의 도장억제에 미치는 영향)

  • This study was conducted to find out the effect of diniconazole treatment by seed soaking on the stretching of tomato and cucumber seedlings. The emergence rate of tomato and cucumber seed was decreased as diniconazole concentration was higher and soaking period was longer. The emergence rate of tomato seedlings was lower as the soaking period was longer in $1mg{\cdot}L^{-1}$ concentration treatments, although there was no significant difference. The other concentration treatment (10, 50, and $100mg{\cdot}L^{-1}$) reduced emergence rate of tomato seedlings outstandingly in higher concentration and longer treated period, but 90% of treated seeds emerged at 10 days after sowing. As the concentration of diniconazol was higher and soaking period was longer, plant height and growth rate of seedlings were retarded. There covery of internodes growth was faster in lower concentration and shorter soaking period. The top and root fresh weight and dry weight of tomato and cucumber seedlings was not influenced by $1mg{\cdot}L^{-1}$ concentration, but these growth characteristics were reduced in higher concentration and longer soaking period. The dwarfed cucumber seedlings treated by diniconazol showed normal growth rate after planting and their male and female flower was set normally. These results suggest that the proper concentration and soaking period of diniconazol may be $10mg{\cdot}L^{-1}$ concentration for 12 hours in tomato seed, and $1mg{\cdot}L^{-1}$ concentration for 24 hours in cucumber seed.

The Apple Rootstock Transgenic M.26 (Malus pumila) with Enhanced Rooting Ability (발근력이 향상된 사과 대목 M.26 형질전환체)

  • The apple rootstock M.26 (Malus pumila) is very popular apple rootstock with semi-dwarf habit and the trees on M.26 produce high quality fruit at a young age. Since it is prone to poor prop ability to soil, however, young trees require staking in windy locations. The rolC gene was introduced into M.26 by Agrobacterium tumefaciens LBA4404 harboring pBI121 to obtain its transformants with dwarfism and enhanced rooting ability. One regenerated transgenic line was confirmed by polymerase chain reaction (PCR) analysis and Southern blot analysis of genomic DNA for the existence of rolC gene. The characteristics of transgenic line in vitro were not significantly different from non-transgenic line except for the active root formation and lateral root number. The rolC transgenic line showed reduced stem length and increased root number in vitro. Rooting ability was examined in the isolated greenhouse after mound layering. Compared to non-transgenic M.26, rolC transgenic line showed significantly higher rooting ability. The transgenic line did not show any other observable variation in shoot phenotype compared with non-transgenic line excepting increased branching

Effect of Drench Application of Growth Retardants on Growth and Development in Cut Lilies (절화 백합의 생장과 발육에 미치는 생장억제제의 관주처리 효과)

  • This study was carried out to investigate the effect of three different growth retardants for applying pot or bedding plant with 'Rialto', 'Elodie', and 'Raizan'. Ancymidol, uniconazole, and paclobutrazol were applied as a drench at various concentrations. In Oriental hybrid lily 'Rialto', no changes were observed not only plant height but also leaves and flowers in paclobutrazol treatment. But the growth of plant treated with ancymidol was inhibited above 30% compare to control regardless of concentrations. However, numbers of leaf and flower per plant were similar to those of controls. Plant height was similar to nontreated control in $1mg{\cdot}L^{-1}$ uniconazole treatment but that reduced about 30% in drench application of 5 and $10mg{\cdot}L^{-1}$ uniconazole. Asiatic hybrid lily 'Elodie' was not different in growth between application of paclobutrazol and nontreatment as 'Rialto'. While, in all ancymidol treatments, plant height was 50~60% shorter than that of control. Also, numbers of leaves were reduced and days to flowering were slightly delayed in $30mg{\cdot}L^{-1}$ ancymidol treatment as compared with the control. All traits except plant height of $1mg{\cdot}L^{-1}$ uniconazole were similar to control but there were differences of statistical significance between 5, $10mg{\cdot}L^{-1}$ uniconazole drench and nontreated control. Lilium ${\times}$ formolongi 'Raizan' like above two cultivars had no dwarf effect on height in paclobutrazol treatment. In drench application of ancymidol, plant height reduced over 60% than that of control regardless of concentrations. Especially, days to flowering were a little delayed with $30mg{\cdot}L^{-1}$ paclobutrazol treatment. Plant height of $10mg{\cdot}L^{-1}$ uniconazole was about 26% shorter than that of control, but other traits were no differences between uniconazole treatment and control. In result, the drench application of ancymidol and uniconazole was pretty effective for usage as pot lily without great changes of quantitative traits.

Effects of Growth Retardants B995 and CCC on the Growth arid Yield of Soybean (생장 조절제(B995.CCC) 산포가 대두의 생육 및 수량에 미치는 영향)

  • The effects of retardants B995 and CCC on the growth and yield of soybean cultivar, kwanggyo, were investigated. The 3 application times(4th, 5th and 6th trifoliate leaf stage) combined with 4 concentrations (500, 1000, 2000 and 3000 ppm) were treated, and the retardant solutions were sprayed on the foliage. Only stem length in retardants, rate of podding in application times, and, in concentrations, stem length, number of branch, number of pod per plant and grain yield, were significant. In the inhibition of stem elongation, B995 was more effective than CCC, and rate of podding was the highest in the 4th leaf-spraying. The highest concentration, 3000 ppm, was the most effective in the inhibition of stem elongation and in the increase of number of branch and pod, rate of podding, and in yield. The number of node of mainstem, dry stem weight, chlorophyll and 100 grain weight were not significant with any treat. Yield had positively correlated with number of branch and pod, and rate of podding but negatively with stem length. The retardants was thought to be considerably conducive to the increase of yield.

Effect of Pinching Time and Plant Growth Retardants on Growth Control of Ardisia pusilla in an Ebb and Flow System (Ebb and Flow 시스템에서 적심시기와 생장조절제 처리에 의한 산호수의 생장조절)

  • This study was conducted on the effect of plant growth retardants (PGRs) on growth of potted Ardisia pusilla and Ardisia pusilla var. variegata grown for a short term in an ebb and flow system. Plants were planted in 10 cm plastic pots, containing a mixture of peat moss and perlite (1 : 1, v/v) on 30 June 2005 and were grown until 23 Sep. 2005. The PGRs used were 50, 200, 350, 500 $mg{\cdot}L^{-1}$ daminozide (B- 9), 10, 40, 70, 100 $mg{\cdot}L^{-1}$ paclobutrazol (Boundy), and 5, 15, 25, 35 $mg{\cdot}L^{-1}$ ethephon (Florel). On every irrigation, the nutrient solution containing PGRs was supplied at a 2 cm depth and was kept for 15~20 minutes. The surplus nutrient solution was drained back into the tank for next irrigations. In both cultivars, paclobutrazol gave the most pronounced effect in inhibition of stretchiness. Ardisia pusilla, which was pinched just before initiation of paclobutrazol treatment, resulted in the greatest inhibition of shoot elongation. Daminozide treatments above 200 $mg{\cdot}L^{-1}$ also gave reduction of shoot length. However, ethephon treatments gave no dwarfing effect in Ardisia pusilla. Inhibition of stretchiness was observed even in the lowest concentration treatments. Among the PGRs, paclobutrazol was the most effective in suppressing plant stretchiness.

Several Treatments of Veronica rotunda Nakai for Pot Cultivation (분화 재배를 위한 둥근산꼬리풀의 몇 가지 처리)

  • 둥근산꼬리풀(Veronica rotunda Nakai)은 다년생 초본으로 높은산 정상에서 자라며, 초장은 10~20 cm, 잎의 길이는 2~4 cm 정도이다. 또한, 기부의 잎은 엽병이 불분명하고, 줄기에는 털이 없는 것으로 알려져 있다. 관상소재로 이용되고 있으나, 식물정보는 매우 부족하며, 활용하기 위해서는 재배 연구와 식물에 대한 정보 수집이 필요하다. 이에 본 연구는 둥근산꼬리풀의 관상용 재배 시 적정 차광, 기비, 추비, 왜화제 및 적심 등의 처리 정도를 알아보고자 수행하였다. 재배방법의 확립을 위하여 선행연구에서 육묘한 유묘(2018년 5월 15일에 파종하여 생산)를 7월 16일에 정식하여 10월 15일까지 약 14주 동안 재배하였다. 공통조건은 플라스틱 화분 10호(이색포트, $9{\times}9cm$)에 원예상토를 충진하고, 200구 트레이에 셀 당 4립씩 파종하여 생산된 1셀을 정식하였다. 차광 정도 실험은 0, 55, 75% 차광막을 이용하였으며, 기비는 싱싱코트를 0, 10, 20알 처리하였고, 추비는 hyponex (N:P:K, 4:6:6)를 0, 1000, $2000mg{\cdot}L-1$의 농도로 엽면시비 하였다. 또한, 관상효과 증가를 위해 왜화제는 diniconazole, daminozide를 농도별(각 0, 1000, $2000mg{\cdot}L-1$)로 처리하였고, 적심은 4주차에 1회 처리 유무의 효과를 비교하였다. 연구의 결과, 둥근산꼬리풀에 기비 10알 처리는 생육에 유의적인 차이는 보이지 않았으나, 20알 처리는 무처리와 10알 처리에 비해 절간장, 경직경이 증가하는 결과가 나타났다. 추비는 절간장을 증가시켰으나, 측지수와 마디수는 유의적으로 감소시켰다. 차광처리별로는 55, 75% 차광 모두 무차광에 비해 측지수, 마디수, 엽수 및 개화율을 크게 감소시키는 결과를 보여, 둥근산꼬리풀는 광이 충분한 곳에서 재배하는 것이 적합한 것으로 생각된다. 왜화제의 처리는 종류와 농도에 관계없이 둥근산꼬리풀은 측지수, 마디수, 엽수를 감소시키는 결과를 보였다. 또한 초장이 짧아지고 경직경은 두꺼워졌으나, 유의적인 차이는 없었다. 종류별로는 daminozide $2000mg{\cdot}L-1$ 처리는 무처리에 비해 개화율이 증가하여, 둥근산꼬리풀의 분화용 재배시 daminozide $2000mg{\cdot}L-1$를 처리하는 것이 효과적이다. 적심처리는 무처리에 비해 전반적인 생육 및 개화를 억제하는 결과가 나타났다. 결론적으로 둥근산꼬리풀의 분화용 재배시 광량이 충분한 곳에서 재배하고, 기비와 추비는 절간장을 증가시키며, 측지수, 마디수, 엽수에 큰 차이가 없거나 감소하는 결과를 보여 관상용 재배에 적합하지 않은 것으로 판단된다. 추가적으로 daminozide $2000mg{\cdot}L-1$를 처리하며, 적심처리를 하지 않는 것이 좋을 것으로 생각된다.

Effect of Inoculation Concentration on Pathogenicity, Development, Propagation and Body Length of Entomopathogenic Nematode, Steinernema arenarium (Nematoda: Steinernematidae) (접종농도가 곤충병원성선충 Steinernema arenarium (Nematoda: Steinernematidae)의 병원성과 발육 증식 및 체장에 미치는 영향)

  • Effect of inoculation level on pathogenicity, development, and propagation of entomopathogenic nematode, Steinernema arenarium was investigated using the last instar of great wax moth, Galleria mellonella. Pathogenicity of S. arenarium was higher with increasing inoculation level representing 82% at the rate of 5 infective juveniles (IJs) while >98% at the rate of >10 IJs. The number of IJs penetrated into the host was 2.7, 5.0, 7.4, and 12.2 at the rate of 5, 10, 20, and 40 IJs, respectively while 24.3 at the rate of 80 IJs and 40.2 at the rate of 160 IJs. Inoculation level did not affect female adult size (4,616 to 6,444 ${\mu}m$) while affected male adult size (1,600 to 1,934 ${\mu}m$). The rate of stunted female adults was 70.2% at the inoculation level of 80 IJs and 63.7% at the inoculation level of 160 IJs. The number of progenies was 20,431, 26,696, 47,943, 50,516, 58,701, and 74,235 at the rate of 5, 10, 20, 40, 80, and 160 IJs, respectively. The body lengths of IJs were different depending on inoculation level ranging from 636 to 1,496 ${\mu}m$.
