Classification of Strains of Ganoderma lucidum (Ganoderma lucidum의 계통분류(系統分類))

  • This study was carried out to investigate some morphological characters of fruit bodies of Ganoderma lucidum and to classify the fungus on the bases of the genetic character. Some of the isolates of the fungus which originally have the kidney-shaped fruit bodies produced the antler-shaped fruit bodies on artificial media and the latter characters were inherited. Pattern of the pileus surface and thickness of the pileus of the fruit bodies were also considered to be hereditary. Although morphology of the pileus margin and fruiting mode of the fungus were variable among the isolates, they were greatly influenced by environment conditions. Ganoderma lucidum could be classified into two groups and four strains according to the morphology of the fruit bodies on artificial cultivation media. Electrophoretic patterns of esterase, peroxidase, leucine aminopeptidase(LAP) and proteins of fruit bodies and mycelia of Ganoderma lucidum showed high genetic variation. Isozyme patterns of esterase of the mushroom mycelia were applicable for the classification of strains of the fungus. Patterns of proteins, leucine aminopeptidase and peroxidase did not indicate any genetic relation among the fungus strains.

Scheme for Verification Between Mobile Devices in a Service with Expiration Time by Using Zero-knowledge Proof (영지식 증명을 이용한 가입기간이 정해진 서비스에서 이동 통신 기기간 인증 기법)

  • As the mobile communication technology is developed, the services for communication between the mobile devices are provided, and the amount of usage is increasing tremendously. For the device-to-device communication, the device should be verified if it is a service member. The existing verification schemes include interactions with the third party, while this may cause the problems that the bandwidth is dissipated and the devices which are out of the communication range of the base station cannot communicate with other devices. To solve such problems, we propose a new scheme for verification between mobile devices without interaction of third party. For the proposed scheme, we develop and employ a new zero-knowledge proof protocol, which verifies the device's membership and its expiration time. Furthermore, the scheme guarantees privacy of the mobile device since it checks the encrypted verification message without decrypting, and protects replaying attack since it uses challenge-response method.

The effect of the mushrooms extract on the PSA expression in prostate cancer cells (버섯류 추출물이 전립선 암 세포 내 PSA 발현에 미치는 영향)

  • Mushroom have long been valued as highly nutritious and tasty foods in many societies throughout the world. It is known for biological activities including anti-inflammatory and anti-oxidative potential. However little is known about anti-cancer property. In this study, we investigated the anti-prostate cancer activity of mushrooms. For that, eight kinds of mushrooms such as, T. matsutake, S. crispa, G. lucidum-US, G. lucidum-AS, C. cardinalis-BR, G. frondosa, P. linteus, U. esculenta were extracted with hot water. Among them, three kinds of mushrooms including T. matsutake, G. lucidum-US and C. militaris-BR extracts inhibited prostate specific antigen (PSA) expression in prostate cancer cell, LNCaP. These results demonstrate that some of mushrooms inhibited PSA expression suggesting that the mushrooms might be a candidate for the treatment of prostate cancer.

Inhibitory effect of mushrooms extract on TNF-α/INF-γ induced-cytokine in human keratinocytes, HaCaT (버섯류 추출물의 피부각질세포(HaCaTcell) 내 염증성 사이토카인 억제효과)

  • Mushroom is known for anti-inflammatory and anti-oxidative potential. This study provides evidence that the inhibitory effect of mushroom on the expression of pro-inflammatory cytokines in human keratinocytes, HaCaT cells. To define the underlying mechanisms of action, tumor necrosis factor${\alpha}$/$IFN{\gamma}$-activated human keratinocytes model was used. Mushroom significantly inhibited the expression of cytokines in HaCaT cells. Taken together, the results demonstrate that mushroom inhibited inflammtion, suggesting that mushroom (DW extract: Grifola frondosa Cordyceps militaris), (Ethanol extract: Ganoderma lucidum, Lentinus edodes, Cordyceps militaris, Flammulina velutipes) might be a candidate for the treatment of skin inflammation.

An Anticoagulant Polysaccharide Isolated from Ganoderma lucidum (한국산 영지버섯에서 분리된 항응고성 다당)

  • The anticoagulant polysaccharide was screened from the ediable mushrooms. Among them, alkali extract of Ganoderma lucidum showed the highest activity in aPTT. A crude polysaccharide fraction (GL-I) was prepared from the 1N NaOH solution extract of Ganoderma lucidum followed by methanol-reflux, precipitation with ethanol, dialysis and lyophilization. GL-I inhibited the intrinsic pathway in blood coagulation pathway and exhibited concentration dependent anticoagulation effects. The anticoagulant activity of GL-I was decreased greatly by periodate oxidation, but was not changed by pronase digestion. These suggest that carbohydrate moiety may be related to the anticoagulant activity. GL-I consisted of glucose, galactose, fucose, xylose, mannose, arabinose in a molar ratio of 19.3:3.0:2.3:1.3:1.0:0.3. GL-I was partially purified on the DEAE-Toyopearl 650C(GL-IalongrightarrowGL-If) and on the Sephadex G-100(GL-Ic-ilongrightarrowGL-Ic-II).

Manufacturing and Sensory Characteristics of Jupjang Using Grains Fermented by Basidiomycetes (담자균 발효 곡물을 이용한 즙장의 제조와 관능적 특성)

  • The properties of various grains used for the solid-state cultivation of basidiomycetes were examined. The hydration time with cold water was 10 hours for malt soybean. The required hydration time for Job's tears, barley and wheat was 4, 6 and 12 hours, respectively, but the final moisture content ranged from 30 to 48 %, which was much less than the optimum moisture content for mycelial growth. For the mass cultivation of mycelia, the hydrated grains with cold water were placed in plastic bottles. The mycelial growth ra in e bottled grains was high in the early stage with inoculation of homogenized mycelium. The mycelium activity was maintained by adding sterilized water in the middle of the cultivation period. Jupjang underwent sensory evaluation to examine the possibility of utilizing basidiomycetes in functional foods. The grains fermented with Ganoderma lucidum was the best for Jupjang. The combination of malt soybean and Job's tears was the best for Jupjang. The acceptability of Jupjang was improved during the period of aging time.

Anti-aging & Skin Hydration Effects of Spore oil Extracted from Ganoderma lucidum (영지버섯에서 추출한 포자오일의 항노화 및 보습 효능)

  • This study evaluated the anti-aging activity with antioxidant, anti-inflammatory and moisture activity of Ganoderma lucidum spore oil(GLS). GLS increased DPPH radical scavenging activity in a dose-dependent manners. Anti-inflammatory assay measured the inhibitory effect of GLS on NO, TNF-α and IL-6 production in LPS-stimulated RAW264.7 cells. As a result GLS inhibited NO and pro-inflammatory cytokine, TNF-α, IL-6 production. Also using human fibroblast cell to the procollagen production analysis and COL1A1 mRNA expression level analysis for defining, and for AQP-3 mRNA expression level analysis, used human keratinocyte cell. GLS increased procollagen production and COL1A1, AQP-3 mRNA expression. Our results suggest that the GLS have potential anti-inflammatory and wrinkle improves, skin moisture effect.

Compatibility test of Ganoderma strains based on Di-mon mating (Di-mon 교배에 의한 여지버섯균주의 화합성 검정)

  • Compatibility test of the 115 world-wide collections of Ganoderma strains including 61 of Korean origin, were performed by di-mon mating with tester strains G. lucidum G001-1 ($A_1B_1$) and G. tsugae 1109-16 ($A_1B_1$). Unexpected results were found in that 75 strains, formerly considered to be G. lucidum exhibited clamp connections in mating with the tester 1109-16. This observation suggests that these strains require further examination prior to taxonomic classification. Interestingly, this compatibility could be confirmed more easily by the finding of distinct confrontation line between two strains on plate.

Protective Effect of Selected Amino Acids and Food Extracts on Ethanol Toxicity Decrement in Rat Liver (일부 아미노산과 식품 추출물의 에탄올 간독성에 대한 보호효과)

  • An rat liver enzyme test was carried out in order to investigate preventing effect of selested amino acids and some food extracts on ethanol induced liver toxicity in vitro. Solutions of aspartic acid, arginine, glutamic acid were prepared and treated on ethanol treated rat liver preparation. Protective effect of amino acids on lipid peroxidation was determined. Same experiments were conducted using aqueous extracts of Dried soybean sprout, Dried Alaskan pollack and Ganoderma lucidum. The TBA value indicating the lipid peroxidation decreased significantly (p<0.05) by addition of aspartate, glutamate and arginine, repectively at concentrations of $6.25{\sim}50\;{\mu}g/mL$. Similar results were observed by adding the aqueous extracts of Soybean sprout, dried Alaskan pollack and Ganoderma lucidum. The aqueous extracts added after ethanol treatment presemted more effect than added before the treatment.

Biological Activity of Methanolic Extract from Ganoderma lucidum, Momordica charantia, Fagopyrum tataricum, and Their Mixtures (영지버섯, 여주, 쓴메밀 및 혼합 시료의 메탄올 추출물의 생리활성)

  • The concentrations of bioactive material (polyphenolics and flavonoids) and the biological activities of antioxidative (DPPH [${\alpha},{\alpha}'$-diphenyl-${\beta}$-picrylhydrazyl] free radical scavenging activity, peroxidation of rat liver microsome, and Fe/Cu reducing power), ${\alpha}$-glucosidase inhibition, tyrosinase inhibition, and fibrinolytic activity were tested by in vitro experimental models using 70% methanolic extract from Ganoderma lucidum, Momordica charantia, Fagopyrum tataricum and their mixtures. The highest yield and the concentrations of polyphenolics and flavonoids were shown in the mixture extract. Mixture extract was shown to have the highest activities of ${\alpha}$-glucosidase inhibition and tyrosinase inhibition, fibrinolytic, DPPH free radical scavenging, and Fe/Cu reducing power in a concentration-dependent manner. From these results, mixture methanol extract was shown to have the most potent bioactive properties and to contain large amounts of biological materials such as polyphenolics and flavonoids. This mixture could be a good dietary supplement material candidate, such as for antidiabetic functional foods.
