A Potent inhibitor of trypsin and other various serine proteases including chymotrypsin, elastase, kallikrein, plasmin and subtilisin, has been purified to homogeneity from chick skeletal muscle by convendonal chromatographic procedures. The Inhibitor has an apparent molecular weight of 66, 000 dalton as determined by gel filtration. When the purified inhibitor was electrophoresed in the presence of sodium dodecyl sulfate, there appeared rwo protein bands having molecular weights of 66, 000 and 64, 000 dalton. The 64, 000 dalton protein seems to be the product of 66, 000 dalton protein by a lin'ited proteolysis during the purification procedure or in viuo. Thus, it seems to consist of a single polypeptide. The inhibitor appeared to be glycoprotein and have an isoelectric point of 7.4. It contains relatively large amount (8.33 mole%) of cysteine residues.

Comparative thickening and dewatering characteristics of municipal wastewater sludge were investigated in terms of separated and combined treatment of primary and secondary BNR sludge. Also, various conditioning methods such as cation polymer addition, steam and ultrasonication treatment were examined to improve dewaterability of sludge. The dewaterability was measured by using specific resistant test, wedge zone simulator and centrifuge. The result of the sludge thickening test revealed that separated thickening was better in terms of solids recovery and supernatant quality. Particularly, the thickening of primary sludge with high solids (about 3.5% TS) showed very poor solid separation. The addition of cation polymer showed better conditioning characteristic for dewatering and the optimal polymer dosage was 0.26% for primary sludge, 0.43% for secondary sludge and 0.38% for combined sludge. Contrary to the result of the thickening, the dewatering test revealed that dewatering of the combined sludge is better than that of separated sludge, representing better solids separation and filtrate quality. The polymer addition was essential to improve dewaterability in filter (belt) press type dewatering but it was inefficient for the dewatering of secondary sludge only. The centrifuge type dewatering showed better performance and the dewaterability was slightly improved when the polymer was added. Based on the results of this research a sustainable sludge treatment process, particularly in terms of the recycle water quality and solids recovery, was proposed.

The current research reviewed the design conditions that would maximize the efficiency of the hybrid collector that combines in one unit "cyclone-inertial impaction-bag filter". The computational analysis for the shape of cyclone entry predicts that a design that installs the guide vane at the entry of the tangential type cyclone brings a high-rpm and powerful vortex, very effective in promoting the deflection of coarse particles from the streamline at the cyclone. As the lower part of the cyclone is venturi-shaped, however, a strong flow downward of 4 to 5 m/sec persists through the lower part of the hopper, revealing the likely reentrainment of collected dust. And the removal of the venturi at the lower part of the cyclone would solve the problem of the reentrainment of collected dust. The acceleration of the flow velocity through the adjustment of the gap of the collision baffle would increase the effect of collision, but as the interference with the dust separation is expected, the original design should be kept for the baffle.
A disease that manifested severe anemia of the gills occurred in the olive flounder (Paralichthys olivaceus), which was cultured for 5-6 months with a recirculation water system in the laboratory. Microscopic observations showed immature parasite in the gills and mature adults in the oral cavity. The matured parasite was 5.60-9.32 (7.42) mm in total length, with 4 pairs of clamps on the pedunculated haptor, which was separated from the body proper by a long isthmus. From the morphological observations of the larvae, it was identified as Neoheterobothirum hirame belonging to the monogenea. The average hematocrite of infected fish was 10.3 ± 2.8%, significantly lower than that of normal flounder 31.4 ± 4.2%. Histopathologically, fish infected with N. hirame exhibited reduced numbers of erythrocytes in blood vessels of the gill filament and lamellae, reduced red pulp area of the spleen, and hepatocyte atrophy. From the above results, the cause of severe anemia in olive flounder was identified as N. hirame infection. This study shows an example where the growth of pathogens can be a problem in the recirculation system due to low water exchange rate.
Dewatering process to remove water from pigment sludge was important in the diverse aspects of the improvement of product quality, curtailment of the drying cost and the transportation. It was difficult to dewater pigment particles with the mechanical forces because the size was fine under $5{\mu}m$. Thermal filter press dewatering equipment composed of squeezing plate and a fixed heating plate was developed to improve the dewaterability of pigment the sludge as supplying the heat from the fixed heating plate to the cake. Several tests that estimate the dewaterability for pigment sludge as with or without squeezing process and the difference of dewatering time was conducted with this equipment. Dewaterability of thermal dewatering under squeezing process was increased about 20% compared with non squeezing process. Under squeezing process, thermal dewatering tests changing dewatering time with 70 and 80 minute were conducted respectively. The water content of cake was more reduced at dewatering time of 80 minute compared with 70 minute, and dewatering velocity was also decreased, which caused the productivity of thermal filter press to drop. It was observed that clogging of filter cloth didn't almost occur because the liquid was discharged from cake layer easily. In this research, it was resulted that the squeezing process and long dewatering time were effective to improve the dewaterability of pigment sludge. So, this thermal filter press equipment was useful for dewatering the fine particle sludge like pigment.
In order to reuse and concentrate the sewage, a forward osmosis using fertilizer as draw solution was applied. Sewage-1, which is the supernatant after settling for 30 minutes for the primary settling basin influent, and Sewage-2, which is the supernatant after settling for 30 minutes for the effluent, and Sewage-3, which is the filtrate filtered through a $1{\mu}m$ cartridge filter for the effluent were tested. Eight draw solutions of $NH_4H_2PO_4$, KCl, $KNO_3$, $NH_4Cl$, $(NH_4)_2HPO_4$, $NH_4NO_3$, $NH_4HCO_3$, and $KHCO_3$ were used in consideration of osmotic pressure, solubility and pH. In the case of Sewage-3, the permeate flux was almost similar to that of the discharge water of the sewage treatment plant, and was larger than that of Sewage-1 and Sewage-2. $NH_4H_2PO_4$ was the smallest, and $NH_4NO_3$ was the largest in the specific reverse solute flux. $NH_4H_2PO_4$ was found to be most useful for the reuse and concentration of sewage because it contains nitrogen and phosphorus, which are the major components of fertilizer, as well as low specific reverse solute flux. When $NH_4H_2PO_4$ was used as the draw solution, the concentration factor after 24 hours for Sewage-3 was 1.72.
This study was carried out to examine the nitrogen removal rate of a subsurface-flow treatment wetland system which was constructed on floodplain of the Kwangju River from May to June 2001. Its dimensions were 29m in length, 9m in width and 0.65m in depth. A bottom layer of 45cm in depth was filled with crushed granite with about $15{\sim}30\;mm$ in diameter and a middle layer of 10cm in depth had pea pebbles with about 10 mm in diameter. An upper layer of 5 cm in depth contained course sand. Reeds (Phragmites australis) were transplanted on the surface of the system. They were dug out of natural wetlands and stems were cut at about 40 cm height from their bottom ends. Water of the Kwangju River flowed into it via a pipe by gravity flow and its effluent was funneled back into the river. The height of reed stems was 44.2 cm in July 2001 and 75.3cm in September 2001. The number of stems was increased from $80\;stems/m^2$ in July 2001 to $136\;stems/m^2$ in September 2001. Volume and water quality of inflow and outflow were analyzed from July 2001 through December 2001. Inflow and outflow averaged 40.0 and $39.2\;m^3/day$, respectively. Hydraulic detention time was about 1.5 days. Average nitrogen uptake by reeds was $69.31\;N\;mg/m^2/day$. Removal rate of $NO_3-N$, $NH_3-N$, T-N averaged 195.58, 53.65, and $628.44\;mg/m^2/day$, respectively. Changes of $NO_3-N$ and $NH_3-N$ abatement rates were closely related to those of wetland temperatures. The lower removal rate of nitrogen species compared with that of subsurface-flow wetlands operating in North America could be attributed to the initial stage of the system and inclusion of two cold months into the six-month monitoring period. Increase of standing density of reeds within a few years will develop both root zones suitable for the nitrification of ammonia and surface layer substrates beneficial to the denitrification of nitrates into nitrogen gases, which may lead to increment in the nitrogen retention rate.
The fresh water prawn, Macrobrachium rosenbergi (De Maan) is a very common species in Info-pacific region inhabiting both fresh and brackish waters in low land areas, and especially abundant in the lower reaches of most rivers which are influenced by seawater, It is one of the largest and commercial species of genus Macrobrachium. As a place of the researches to clear the possibilities of transplantation and propagation of this species in the Far East region the larval development, growth, optimum temperature and metamorphose rate up to first post larvae in aquaruim are cleared under the conditions of salinity $6.58-7.05\%_{\circ}$ Cl, pH 8.0-8.2, the rate of flow 0.3 liter per minute and illumination 3000 lux. Temperature ranged from 27.5 to $28.7^{\circ}C$ during the period of earlier larval development. For the study oil the relationship between temperature and metamorphose rate from zoea to first post larvae, the temperatures in experimental tank were $22.2^{\circ}C\pm1$, $26.1^{\circ}C\pm0.85$, $28.1^{\circ}C\pm0.34$, $30.4^{\circ}C\pm0.66$, $33.7^{\circ}C\pm0.66$, $33.7^{\circ}C\pm0.38$ and $36.8^{\circ}C\pm0.26$. During the work, food used for the larvae was Artemia salina nauplius in the filter-circulation aquariums. This species metamorphosed to the first post-larvae through eleven zoea stages, and the characters of each larval stage are described and optimum temperature for metamorphosis rate and survivals to the first post larvae is $28.1^{\circ}C\pm0.34$.
In order to establish the method to prepare and use the sulfur dioxide gas generating agent in low temperature storage of grape fruits, the $SO_2$ generation capacity from various sulfite compounds was investigated, and the method to regulate $SO_2$ gas generation and to detect the gas was developed. The conidial germination and mycelium growth of Botrytis cinerea which causes gray mold disease during grape fruit storage was completely inhibited at the $SO_2$ gas concentration of 400 ppm and 3200 ppm, respectively. Sodium hydrosulfite generated the most amount of $SO_2$ gas among the investigated 5 different sulfite or bisulfite compounds. By adjusting the number of pinholes on packaging film of the compound or by adding pH adjusting agent, e.g. citric acid or phosphates, it was possible to regulate the amount and duration of $SO_2$ gas generation from the compound. Because malachite green was quantitatively discolored by $SO_2$ gas, the solution or impregnated paper with the compound could be practically utilized as a indicator detecting $SO_2$ gas. Finally, when Muscat Bailey A grape was stored at low temperature with $SO_2$ gas generating agent, the disease incidence was reduced after storage.
In Korea, there are 1,474 thousand pumping wells nationwide which account for about 12% of total water use in 2012. As much as 39 hundred million tons of groundwater were used while 333 hundred million tons of total water were supplied in 2012. Because the water management authority projects that water demand will exceed supply by 2021, the authority is planning to extensively expand groundwater use in accordance with economic feasibility. Using the basic frameworks of cost-benefit analyses of the World Bank and the US Environmental Protection Agency (US EPA), the objective of this study is to examine the costs and benefits of the expansion of Korea's groundwater extraction through pumping wells. We conclude that the BC ratio of the groundwater pumping wells is 2.98. This signifies that the benefits are 2.98 times higher than the costs. The benefits include use and non-use values of pumping wells while the costs include the installation and maintenance of new wells, in addition to the restoration and pollution costs of abandoned wells, as well as fees for water quality tests, etc.
