Quality Characteristics and Antioxidant Activity of Prickly Pear Cactus Cladodes (손바닥 선인장 엽상경의 품질 특성과 항산화 효과)

  • Prickly pear cactus cladodes were extracted with hot water and 70% ethanol, followed by fractionation with n-hexane (HF), ethyl acetate (EF), n-butanol (BF), and distilled water. Total phenolics and total flavonoid contents as well as antioxidative and anti-inflammatory activities were then measured. Total phenolic contents were 784, 452, and 220 mg gallic acid equivalents (GAE)/g, whereas total flavonoid contents were 214, 76, and 113 mg quercetin equivalents (QE)/g in EF, BF, and HF, respectively. DPPH and ABTS radical scavenging activities ($IC_{50}$) were 103 and $105{\mu}g/mL$ in EF, 359 and $379{\mu}g/mL$ in BF, and 469 and $605{\mu}g/mL$ in HF, respectively. Oxygen radical absorbance capacity was highest at $391{\mu}M$ TE in EF (in decreasing order of $117{\mu}M$ TE in BF and $64{\mu}M$ TE in HF), whereas superoxide anion radical scavenging activity ($IC_{50}$) was highest at $40{\mu}g/mL$ in EF (in decreasing order of $69{\mu}g/mL$ in BF and $98{\mu}g/mL$ in 70% ethanol extract). Inhibitory activity ($IC_{50}$) of nitric oxide (NO) production induced by LPS-activated RAW264.7 cells was highest at $62{\mu}g/mL$ in HF (in decreasing order of $104{\mu}g/mL$ in EF and $465{\mu}g/mL$ in BF). The selectivity index (ratio of inhibitory activity of NO production to cell cytotoxicity) was highest at 4.63 in EF (in decreasing order of 3.37 in HF and 2.14 in BF). In conclusion, EF showed potent antioxidant and anti-inflammatory effects with high phenolic and flavonoid contents.

Antioxidant Activities of Ipomoea batatas L. Lam. (Purple Sweet Potato) Extracts Cultured in Korea (국내산 자색고구마(Ipomoea batatas L. Lam.) 추출물의 항산화활성 평가)

  • In this study, the antioxidative effects for the extracts of purple sweet potato (Ipomoea batatas L. Lam.) were investigated. The purple sweet potato was extracted with 70% ethanol and the ethyl acetate fraction was obtained from the extracts. The yields of extract and ethyl acetate fraction were 39.2% and 3.49% per dried powder, respectively. To confirm the antioxidative effects of the extracts, free radical scavenging activities (1,1-diphenyl-2-picrylhydrazyl), total antioxidant capacity by luminol-dependent chemiluminescence assay and the protective effects against reactive oxygen species (ROS) in erythrocytes were measured. Free radical scavenging activities ($FSC_{50}$) of the 70% ethanol extract and ethyl acetate fraction were $90.16{\mu}g/mL$ and $7.69{\mu}g/mL$, respectively. The free radical scavenging activity of ethyl acetate fraction was higher than that of (+)-${\alpha}$-tocopherol ($8.98{\mu}g/mL$). Total antioxidant capacities ($OSC_{50}$) of the 70% ethanol extract and ethyl acetate fraction were $5.75{\mu}g/mL$ and $1.92{\mu}g/mL$, respectively. The capacity of ethyl acetate fraction was similar to L-ascorbic acid, known as a prominent water soluble antioxidant ($1.50{\mu}g/mL$). The cellular protective effects of the ethyl acetate fractions on the $^1O_2$-induced cellular damage of human erythrocytes were increased in a concentration dependent manner ($5{\sim}50{\mu}g/mL$). The ${\tau}_{50}$ value in $5{\mu}g/mL$ was 45.6 min which was higher than that of (+)-${\alpha}$-tocopherol in all concentrations. These results indicate that the ethyl acetate fraction of purple sweet potato (I. batatas) has the excellent antioxidative capacity and could be applicable to anti-aging cosmeceutical ingredients for skin aging inhibition.

Antioxidant, Antimicrobial and Cytoprotective Effects of the Extract and Its Fraction Obtained from Rhizomes of Belamcanda chinensis (L.) DC (범부채 뿌리 추출물 및 분획물의 항산화, 항균 및 세포 보호 효과)

  • In this study, we investigated antioxidant, antimicrobial and cytoprotective effects of 50% ethanol extract and ethyl acetate fraction from rhizomes of Belamcanda chinensis (L.) DC. 1,1-Diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activities ($FSC_{50}$) of the 50% ethanol extract and ethyl acetate fraction were 621.5 and $253.0{\mu}g/mL$, respectively. Total antioxidant capacities ($OSC_{50}$) of the extract and fraction were 13.6 and $3.0{\mu}g/mL$, respectively. Minimum inhibitory concentrations (MIC) of the ethyl acetate fraction for Staphylococcus aureus and Candida albicans were 156, $1,250{\mu}g/mL$, respectively, indicating similar or higher levels of those of using methyl paraben. Cytoprotective effects of the 50% ethanol extract against $^1O_2$-induced cellular damage (${\tau}_{50}$) showed in a dose dependent manner at 4 to $64{\mu}g/mL$. ${\tau}_{50}$ of the 50% ethanol extract, ethyl acetate fraction and (+)-${\alpha}$-tocopherol at $16{\mu}g/mL$ were 36.4, 45.0 and 45.8 min respectively, and the ethyl acetate fraction showed cytoprotective effects similar to (+)-${\alpha}$-tocopherol. In ultraviolet B radiation-induced HaCaT cell damage, the ethyl acetate fraction decreased intracellular reactive oxygen species (ROS) up to 45.9% at $8{\mu}g/mL$. Also in $H_2O_2$-induced HaCaT cell damage, the ethyl acetate fraction significantly increased the cell viability at $0.5{\sim}8.0{\mu}g/mL$. As a result of chemical analyses of the ethyl acetate fraction, the presence of flavonoids and polyphenol such as irisflorentin, irigenin, tectorigenin, resveratrol, iridin and tectoridin were identified. In conclusion, the extract/fraction from rhizomes of B. chinensis can be applied as a natural antioxidant and antimicrobial material to cosmetics.

  • Tissue thromboplastin (tissue factor), a membrane bound glycoprotein is an important initiating factor in blood coagulation cascade, which leads to the formation of thrombin by activating both factor X and IX. Activation of blood coagulation by TF is essential for blood injury, and stimulates the blood coagulation in myocardial infarction, cancer and blood coagulatory diseases. High density lipoprotein, apolipoprotein A-II were known to be biological TF inhibitors. Recently, studies on search for TF inhibitors from natural products have been active in Korea. Among the edible mushrooms screened for inhibitory activities on the TF, Lentinus edodes showed the most strong activity, followd by Agaricus bisporus and Ganoderma lucidium. And the fractionation of the above mushrooms with the chloroform ($CHCl_3$) and ethylacetate (EtOAc) was done and evaluated for the inhibitory activities on TF. In Ganoderma lucidium, $CHCl_3$ fraction and $H_2O$ layer were not active, but EtOAc fraction exhibited a strong inhibitory activity on TF and the $IC_{50}$ value was $1.07{\times}10^{-4}\;g$. In the case of Agaricus bisporus, there were no inhibitory activities on the TF in all of the fractions. $CHCl_3$ fraction and $H_2O$ layer of Lentinus edodes did not show inhibition on the TF but EtOAc fraction showed strong inhibition on the TF, and the $IC_{50}$ value was $7.70{\times}10^{-4}\;g$.

Studies on the Composition and Antioxidative Effect of Leaves form Korean Rosa davurica Pall. (한국산 생열귀나무(Rosa davurica Pall.) 잎의 성분 및 항산화 효과에 관한 연구)

  • The objective of this study was carried out to investigate nutritional characteristics and biological activities effects of Korean leaf of Rosa davurica Pall. in vitro. They were extracted with methanol and then further fractionated to n-hexane, chloroform, ethyl-acetate, n-butanol and water from methanol extracts. Methods of the antigenotoxic used in this experiment were UVA/UVB absorption property and DPPH radical scavenge. The proximate compositions of leaves of Rosa dauvrica Pall were 67.5% of crude Moisture, 0.7% of crude fat, 6.8% of crude protein, 6.1% of crude ash, and 20.8% of crude fiber. The major minerals were K (1637.2 mg%), Ca (219.5 mg%), P (182.1 mg%), and Mg (135.1 mg%). Most of the fractions of methanol extract which leaves of Rosa dauvrica Pall. have strong absorbency at UVB region (308 nm) and UV A region (350nm). These fractions have a good absorbency property as synthetic filter and could be served as substitutes for synthetic UV sunscreen agents. All fractions (n-hexane, ethyl-acetate, n-butanol and water) from methanol extracts except chloroform fraction exhibited DPPH radical scavenging activity with IC$\_$50/ of 35.3, 6.0, 14.0, and 18.0 $\mu\textrm{g}$/mL.

Antimutagenic and Cytotoxic Effects of an Ethanol Extract of Buckwheat Sprout (메밀싹 에탄올 추출물의 항돌연변이원성 및 암세포 성장억제 효과)

  • This study was performed to determine the antimutagenic and anticancer effects of ethanol extract of buckwheat sprout using Ames test and SRB assay, respectively. An ethyl acetate fraction (200 ${\mu}/plate$) from the ethanol extract of buckwheat sprout showed inhibition rate of 80.6% against the mutagenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in Salmonella typhimurium TA100 strain. Also the ethyl acetate fraction (200 ${\mu}/plate$) showed higher antimutagenic activity than other fractions against the mutagenesis induced by 4-nitroquinoline-1-oxide (4NQO) in Salmonella typhimurium TA98 and TA100. In addition, the ethyl acetate fraction (200 ${\mu}/plate$) showed high antimutagenic effect of 80.9% and 85.9% against the mutation of TA98 and TA100 strains induced by 3-amino-1,4-dimethyl-5H-pyrido-(4,3-b)indol (Trp-P-1), respectively. The cytotoxic effects of each solvent fraction from the ethanol extract of buckwheat sprout against human cancer cell lines including lung carcinoma (A549), gastric carcinoma (AGS), breast adenocarcinoma (MCF-7), hepatocellular carcinoma (Hep3B), and colon adenocarcinoma (Colo 205) were investigated. The ethyl acetate fraction of buckwheat sprout ethanol extract at the concentration of 1.0 mg/ml showed strong cytotoxic activities of 70.3, 94.8, 79.6, 82.3, and 73.2% against A549, AGS, MCF-7, Hep3B and Colo 205 cancer cell lines, respectively.

A Study of the Whitening Activities of Magnolia obovata Bark Ethyl Acetate Fractions as Cosmetic Ingredient (화장품 소재로서 후박 에틸아세테이트 분획물의 미백활성에 관한 효과)

  • EtOAc fractions of Magnolia obovata (M. obovata) Bark extracts were studied for the potential ingredient as a safe and effective whitening cosmetic material. The concentration of active substances honokiol was determined by HPLC. In vitro, the fractions reduced the extracellular and intracellular melanin contents in B16F10 cells in dose dependently and inhibited extracellular melanin secretion ($IC_{50}=11.05{\mu}g/mL$). The $12.5{\mu}g/mL$ treatment of maximum concentration effectively inhibited up to about 60% to the amount of extracullular melanin. Also, the $12.5{\mu}g/mL$ treatment of maximum concentration effectively inhibited up to about 59% to the amount of intracullular melanin ($IC_{50}=10.85{\mu}g/mL$). The $IC_{50}$ value of ${\alpha}-arbutin$ used as a positive control was $59.99{\mu}g/mL$. So, EtOAc fractions of M. obovata Bark extracts showed whitening effect when compared with the non-treatment group. In case of in vivo study, Cosmetic cream with EtOAc fractions of M. obovata Bark extracts was approved by Ethics committee of KDRI (IRB number: KDRI-IRB-1537). As a result in progress for skin sensitization as well as assessment of skin irritation through repeated patch test, skin allergens was identified as non sensitizing agents. Also, cosmetic cream with EtOAc fractions of M. obovata Bark extracts showed significant topical whitening effect and reliable skin safety when compared with the non-treatment group. In conclusion, EtOAc fractions of M. obovata Bark extracts may be a useful cosmetic ingredient for effective skin whitening.

Effect of Solvent Fractions from Methanol Extract of Doenjang on Inhibition of Growth and DNA Synthesis of Human Cancer Cells. (인체 암세포 성장 및 DNA 합성 억제에 미치는 된장 분획물의 영향)

  • Growth and DNA synthesis inhibitory effects of doenjang methanol extract and its solvent fractions on AGS human gastric adenocarcinoma cells, Hep 3B human hepatocellular carcinoma cells, HT-29 human colon cancer cells and MG-63 human osteosarcoma cells were studied. The treatment of doenjang methanol extract ($ 200{\mu}g/ml $) with the AGS, Hep 3B, HT-29 and MG-63 cancer cells after 6 days of incubation inhibited the growth of cancer cells by $32\%$, $51\%$, $84\%$ and $33\%$, respectively. To separate active compounds of doenjang, doenjang methanol extract was fractionated with dichloromethane, ethylacetate, and buthanol. Among the solvent fractions, the dichloromethane and ethylacetate fractions showed the highest growth inhibitory effects on various cancer cells. For example, the dichloromethane and ethylacetate fractions ($200a{\mu}g/ml$) sig-nificantly inhibited the growth of various cancer cells by $89\∼96\%$ and$62\∼86\%$, respectively. DNA synthesis of AGS and Hep 3B cancer cells was significantly inhibited by adding dichloromethane fraction ($200{\mu}g/ml$) up to $94\%$ and $80\%$, respectively. Similarly, the ethylacetate fraction ($ 200\mug/ml $) showed a $ 95\% $ inhibition rate of DNA synthesis in AGS cells. These results suggest that the dichloromethane and ethylacetate fractions have specific active compounds, which will explain this anticancer effect of doenjang.

Antimicrobial Activity of Fractional Extracts from Houttuynia cordata Root (어성초(Houttuynia cordata) 뿌리에서 추출한 순차분획물의 항균활성)

  • The solvent extracts of Houttuynia cordata root, which were extracted by using several solvents with different polarities, were prepared for utility as a natural preservatives. The antimicrobial activity was investigated by disc diffusion method against 22 microorganisms consisting of food borne pathogens, food poisioning microorganisms and food-related bacteria. The extraction yields were 15.7%, 3.7%, 0.13%, 0.5% and 5.9% in ethanol, chloroform ethylacetate, butanol and aqueous fractions, respectively. Antimicrobial activities were shown in ethanol, ethylacetate and butanol fraction of Houttuynia cordata root. However chloroform and aqueous fractions showed weak antimicrobial activity against the tested bacteria. Among the four fractions, ethylacetate fraction showed the strongest antimicrobial activities against microorganisms tested, such as B. megaterium, E. coli, K. pneumoniae and S. typhimurium. The polyphenolic compounds widely occuring in the traditional medicine plants have been reported to possess high antimicrobial activity. The polyphenolic compound in ethylacetate and butanol fraction were 35.9% and 16.0%, ethanol, chloroform and aqueous fraction were 5.0%, 2.3% and 1.7%, respectively. There are some relationship between antimicrobial activity and polyphenol content in natural plants. The ethylacetate fraction could be suitable for the development of a food preservative.

Antioxidant Activity of Solvent Fractions from Distylium racemosum in Jeju (제주 자생 조록나무 분획물의 항산화 효과)

  • Anti-oxidant activity of 600 medicinal plants from Jeju was analyzed. Extracts from the leaves of Distylium racemosum have the highest anti-oxidant activity. D. racemosum is an oriental medicinal plant belonging to the Hamamelidacea and grows in the wild in Jeju. This study was conducted to evaluate the antioxidant and cell viability of different fractions (n-hexane, methylene chloride, ethyl acetate, buthanol, DW) from D. racemosum. Anti-oxidant activity was measured using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity test and total phenolic content. Cell viability was determined by 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) cell viability assay on HepG2 and A549 cells. Among various extracts, the ethyl acetate fraction showed the highest DPPH radical scavenging activity, reaching approximately 93% at 0.5 mg/mL, higher than that of quercetin used as a positive control. Ethyl acetate fractions showed the highest total phenolic content at 505 mg GAE/g. The phenolic content of each extract showed association with DPPH radical scavenging activity. The ethyl acetate extracts were resistant against hydrogen peroxide ($H_2O_2$) treatment in the MTT cell viability assay and showed a higher cell protective effect than other fraction extracts. These results suggest that the ethyl acetate fraction might be a source of anti-oxidants of D. racemosum.
