Pathological Changes in Cultured Korean Catfish (Silurus asotus) Artficially Infected with Aeromonas veronii (Aeromonas veronii의 실험적 감염에 의한 양식 메기, Silurus asotus의 병리학적 변화)

  • Recently, a new disease showing symptoms such as epidermal exfoliation and muscular necrosis occurred in cultured Korean catfish. Although the mortality of fishes was low but the economic damages owing to loss of commercial value were severe. The authors isolated the causative agent from diseased fish and observed pathological changes both in naturally and artificially infected fish. The causative bacteria was identified as Aeromonas veronii. Subsequently we observed the daily death and pathological symptoms of artificially infected fish with Aeromonas veronii. Symptoms of artificially infected fish were similar to those of naturally infected fish and all fish died within 7 days after infection. Histopathological changes on the naturally infected fish revealed severe congestion and necrotic degeneration in the liver, spleen and kidney. Some bacterial aggregates with inflammatory degeneration were observed in the heart, and congestion and fibrosis in the lamina propria of digestive tube were predominant. In artificially infected fish, skin erosion and necrotic degeneration of muscle tissue around injected region were particularly manifested. Degeneration of hepatocytes in liver and hyalic degeneration around ellipsoids in spleen were partially observed. However, there were no predominant signs in digestive tube in artificially infected fish.

  • In the present study, we investigated the characteristic of growth and the tolerance of cold temperature in small yellow croaker and yellow drum. The results shows that the growth of yellow drum were faster than those of small yellow croaker with high food convergency efficiency. In condition factor (CF), the results shows that the values of yellow croaker and yellow drum were significantly higher in September, respectively. In mortality according to cold shock, two fishes was died under $4.7^{\circ}C$ in slow temperature descending test, but was both died under $9^{\circ}C$ in acute temperature descending test.

  • On May in 2008, mortality of anadromous Ayu Plecoglossus altivelis was observed on the Gangjeong river in Jeju. Major symptoms of the infected fish were mouth rot and skin ulcer. The causative agent was suspected as gliding bacteria. After culture on Shu-Shott and R2A media, we isolated bacterium belonging to the Flavobacterium from ayu with symptoms. As a result, the bacterium was identified as Flavobacterium succinicans JMFL55 by 16S rDNA sequence alignment with F. succinicans DSM 4002(98.27% similarity, GenBank accession NO. AM230492).

Red Tides in Mariculture Farms in Puksin Bay, Korea (북신만의 적조에 관하여)

  • Various scales of red tides have frequently occurred in the southern coastal waters of the Korean Penninsula since the late 1970's. Because most shellfish and finfish farms in Korea are located in the southern coastal waters, the impacts of red tides on the aquaculture have been increasing significantly. The Puksin Bay is one of the places where red tides have occurred almost every year since the early 1980's. During $1990\~1991$, mass mortalities of aquacultural species by the red tides were recorded. The causative organisms in this period were Leplocylindrus danicu(November '90 and June '91), Skeletonema costatum(December '90 and August '91), Nitzschia seriata(August '91), and Gymnodinium splendens(July '91). The maximum chlorophyll-a content was $265.7{\mu}g/l$ in the tides. Frequent red tides are associated with the eutropication of the bay. Some relationships between red tides' occurrence and europhication are herein discussed.

  • 본 논문에서 자동제어를 이용하여 양어업에 쓰이는 큰 수조를 관리하고, 수온과 탁도를 측장하고 관리자에게 수조 안에서의 환경 및 정보를 제공할 수 있는 스마트 수조에 대해서 기술한다. 기존의 양어업의 경우, 작업 연령의 고령화, 노동 인구 감소 등 심각한 문제가 발생되고 있어 양어업의 노동의 질이 현저히 감소 되고 있다. 본 프로젝트에서는 작업자의 부재 상황에서도 블루투스 무선통신을 활용하여 실시간으로 수조 환경에 대한 정보를 수신받을 수 있고, 수조를 자동화함으로써 급격한 기후변화로 인한 어류 및 어패류의 집단폐사에 대한 대비도 가능하다. 이를 구성하기 위해서 아두이노 메가 2560 메인으로 선택하여 많은 핀들을 연결 하였고, 블루투스 모듈을 통해서 실시간으로 정보를 수신받고, 간단한 조작이 가능하여 노동인구에 대한 문제를 해결할 수 있을 것이다.

Potential use of Bacillus amyloliquefaciens as a probiotic bacterium in abalone culture (북방전복, Haliotis discus hannai 에 대한 Bacillus amyloliquefaciens의 probiotic 효과)

  • In comparison to the numbers of such studies of fish, few studies have been carried out on the immunity, physiology and ecology of abalone, while studies on abalone disease are also extremely rare. Moreover, mass mortality of cultured abalone due to pathogenic bacteria has not been reported in the southern coast of Korea. However, Vibrio-like bacteria have been isolated from dead abalone, which indicates that a review is required in order to determine the cause of abalone mortality. Use of an antimicrobial agent to minimize the damage caused by disease in abalone farms is common, but the therapeutic effects are insignificant. Demand for probiotics has increased, but research on the development of probiotics for use in abalone culture is very rare. Therefore, the present study isolated KC16-2 from fermented kimchi soup and investigated the characteristics of the isolate as a candidate probiotic bacterium in abalone. KC16-2 was identified as Bacillus amyloliquefaciens (B. amyloliquefaciens KC16-2) based on its biochemical properties and 16S rRNA gene sequence. B. amyloliquefaciens KC16-2 showed inhibitory effects against the growth of various vibrios in vitro, and kept the numbers constant until four days after inoculation in marine water at a temperature of $15{\sim}25^{\circ}C$, indicating the possible use of KC16-2 as a probiotic, except in the winter. The growth of KC16-2 was inhibited by bile salt, but the numbers increased over time suggesting the bacteria were still alive in the abalone's digestive tract. Abalone fed with a diet including KC16-2 for 12 weeks showed good growth, but showed no significant differences from the control group. However, the mortality of the abalone supplied the probiotic diet was reduced to half that of the control group in a challenge test with Vibrio tubiashii. Therefore, we suggest that B. amyloliquefaciens KC16-2 could be used as a probiotic bacterium for control of the mortality of abalone caused by opportunistic pathogenic vibrios.

  • The effect of ammonia on survival and growth of the larval rockfish, Sebastes schlegeli was examined by a static renewal bioassay method. The tolerance of larvae to ammonia toxicity was more sensitive at the early larvae, but increased with the development of larvae. In 14 day-old-larvae after parturition, the mortality with treatment of each concentration of ammonia was 5% at control group and 0.0112 $NH_3$mg/l, whereas it was increased up to 27.5% at exposure group of 0.1230 $NH_3$mg/l with higher concentration. Regression equation between ammonium concentration(X) and mortality(Y) was followed; Y=0.516+3.482 X($r^2=0.4737$, P<0.01). The NOEC(no-observable-effect concentration) and LOEC (lowest-observable-effect concentration) to mortality compared to control group were 0.100 $NH_3mg/l$ and 0.1230 $NH_3$mg/l, respectively and chronic value(ChV) which is the geometric mean of the NOEC and LOEC was 0.1110 $NH_3$mg/l. Body length after 7-days exposure in control group, 0.0112 $NH_3$mg/l and 0.1230 $NH_3$mg/l were 7.8325mm, 7.700mm and 7.05mm, respectively. The NOEC, LOEC and chronic value(ChV) were 0.0335 $NH_3$mg/l, 0.0558 $NH_3$mg/l and 0.0432 $NH_3$mg/l, respectively.

  • In inland aquaculture, a lot of fishes often died cause of touch of L. carteri which is attached on the net cage. In this works, the experiment were carried out on classification of species, toxin test, and extermination with some chemicals. A bryozoa from net cage in lake Okjeong is classified into the Phylactolaemata, Lophopodella carteri Hyatt. A bryozoa broke out firstly on the 30th of June with water temperature up to $31^{\circ}C$ and it reattached on the net with water temperature below $30^{\circ}C$. Size of bryozoa colony is about $0.8{\times}0.6{\sim}1.7{\times}1.5cm$. Water column of attachment was 0~5m and the peak is 2~3m. In toxin test on the israeli carp, goldfish and catfish touched with bryozoa for 1 minute at $25^{\circ}C$ of water temperature, a dead fish appeared at 20 minutes after touched. Mortality was 90% for israeli carp and 100% for goldfish at 100 minuties after touched, but catfish was not died at all. It was supposed that the toxin is from nematocyst being around tentacles and this toxin act a deadly poison on a israeli carp and goldfish. In extermination test, the bryozoa treated with 300ppm of formalin, 5% of sodium chloride, 5ppm of malachite green, 200ppm of potassium permanganate, 1000ppm of potassium iodide, 10ppm of DDVP for minute respectively, all of them were not effected.

Evaluation of reverse transcriptase loop-mediated isothermal amplification (RT-LAMP) assay for detection of infectious hematopoietic necrosis virus (IHNV) (Infectious hematopoietic necrosis virus (IHNV)-검출 Reverse transcriptase loop-mediated isothermal amplification (RT-LAMP) 법의 평가)

  • A reverse transcriptase loop-mediated isothermal amplification (RT-LAMP) assay was evaluated to monitor infectious hematopoietic necrosis virus (IHNV) from artificially infected rainbow trout Oncorhynchus mykiss. The cumulative mortalities of fish challenged with IHNV at $10^{6.5}\;TCID_{50}$/fish, $10^{5.5}\;TCID_{50}$/fish and $10^{4.5}\;TCID_{50}$/fish were 40%, 0% and 0%, respectively. Dead fish and survivors at 16 and 28 d post-challenge in each group were employed for IHNV detection by RT-LAMP assay and virus isolation using BF-2 cells. IHNV from $10^{4.3}$ to $10^{6.8}\;TCID_{50}/ml$ was isolated from all the dead fish and also detected in all of the examined dead fish by RT-LAMP assay. In survivors at 16 d, 60% (3/5 fish, $10^{2.8}-10^{5.05}\;TCID_{50}/ml$), 20% (1/5 fish, $10^{1.05}\;TCID_{50}/ml$) and 60% (3/5 fish, $10^{1.05}-10^{4.8}\;TCID_{50}/ml$) were found to be IHNV-positive by virus isolation in fish challenged with IHNV at $10^{6.5}\;TCID_{50}$/fish, $10^{5.5}\;TCID_{50}$/fish and $10^{4.5}\;TCID_{50}$/fish, respectively, while 20% (1/5 fish), 0% (0/5 fish) and 20% (1/5 fish) were IHNV-positive by RT-LAMP assay. No IHNV was detected in the survivors at 28 d and control fish. These results indicate that the RT-LAMP assay is useful for detection of IHNV in diseased fish although it is not enough to monitor virus in IHNV-survivors.

Toxicity of Chloramine-T on the Flounder, Paralichthys olivaceus, and Bactertcidal Activity Against Fish Pathogenic Bacteria and Blue Green Algae, Tetraselmis suecica (Chloramine-T의 넙치에 대한 독성과 어병세균 및 Blue Green Algae에 대한 살균 효과)

  • CT(Sodium N-chloro-para toluenesulfonamide, Chloramine-T) known to be a strong oxidative agent was investigated to use as a candidate of disinfectant in the marine aquaculture industry by the analysis of the bactericidal activity against different aquatic microorganisms. One hundred percent mortality appeared at and above 16 ppm CT in sea water in flounder, the predominant species in the marine aquaculture of Korea, when exposed for 48hr. However, bactericidal activity was appeared to be very effective, and all different species of the fish pathogenic bacteria exposed to CT of less than 2 ppm in sea water were dead within 15 min. It allowed us to confirm that CT could be a very effective disinfectant to protect the spread of fish pathogenic bacteria derived from diseased fish or sea water in marine aquaculture. High concentration(10 ppm) with longer exposing time(24 hrs) was required for the algicidal activity of CT, at which concentration might induce the acute toxicity against fish, however, restricted the expanded use of CT for the elimination of zooplanktons at phytoplanktons in marine farms. Moreover the bactericidal activity of CT inhibited almost completely in the present of more than 10 ppm of organic materials pointed out that quality of the culturing sea water should be considered carefully for the application CT to the aquatic farms as a therapeutic agent.
