Effect of Butylated Hydroxytoluene and 2-Acetylaminofluorene Administration and Microsomal Mixed Function Oxidase System in Young Rats fed different Fats (Butylated Hydroxytoluene첨가 식이 및 2-Acetylaminofluorene 투여가 식이지방을 달리한 쥐간의 Microsomal Mixed Function Oxidase계에 미치는 영향)

  • Sprague-Dawley male rats were fed the diet of p/s 4.0(soybean oil : I), p/s 0.08(Beef tallow : II) at the level of 15% fat until 8 weeks after weaning. I & II groups were divided into 4 sub-groups by diets with or without 0.3% butylated hydroxytoluene(BHT). 2-AAF was injected at the age of $5_{1/2}$, 6, $5_{1/2}$, 7 weeks. MFO system enzyme(cytochrome p-450, cytochrome p-450 reductase, cytochrome b5) activities and lipid peroxide were determined from isolated liver microsome. 2-AAF injected young rats had growth retardatiion. Lipid peroxide values were not influenced greatly by dietary fat, 2-AAF and BHT. Cytochrome p-450 contents were increased in I-BHT-AAF & II-AAF groups by 2-AAF and its contents were not affected by BHT. But cytochrome p-450 and cytochrome p-450 reductase were not increased in soybean oil diet ybean oil groups. Cytochrome b5 was not influenced by dietary fat, 2-AAF and BHT. Cytochrome p-450 and lipid peroxide, cytochrome p-450 reductase and cytochrome b5, which transfer to MFO system, appeared to have positive correlations(r=0.2474, r=0.2475, p<0.05) each other. This result suggests that MFO system metabolizing 2-AAF was influenced by dietary fats and BHT. 2-AAF induced growth retardation in young rats.

  • The aim of this study was to describe the NO$_3$ leaching in grassland ecosy-stems. Field study was performed in the southwestern district of Germany from 1991 to 1993. The study included 3 different slurry application, conventional slurry application, 50% reduced slurry application and without slurry application. On each plot were installed 3 ceramic cups at 120cm depth. Samples were collected twice a month and analysed for NO$_3$ colorimetrically using a Tecator Auto-Analyzer. Percolation was calculated as the difference between precipitation and potential evapotranspiration. The NO$_3$ concentration in soil water was remarkably variable during the year. The average NO$_3$ concentration during the experiment was the lowest (8.5mg/l) without slurry application and the highest with 240kg N/ha cattle slurry (25.3mg/l). the NO$_3$-N leached per year were respectively 29, 23, and 12kg/ha in case of 240, 120 and 0kg N/ha when cattle slurry was applied. The conventional feeding density of cattle husbandry in the district caused the NO$_3$ contamination in ground water. When the amount of cattle slurry applied was reduced to 50%, the NO$_3$ loading in ground water was reduced greatly. Therefore, the reduction policy of cattle slurry can positively contribute to the prevention of the NO$_3$-contamination in groundwater.

  • Toxicity of Machete on the mortality of the three different life stages, namely, hatching period, prelarval stage, and juvenile stage of carp(Cyprinus carpio Linne) was observed. The effect of the various exposure times on the mortality of the prelarval stage, and the effect of different temperatures on the toxicity during juvenile stage were also analysed. Hatchability and duration time of the carp were shown to be related to the concentration of Machete. In general, hatchability and duration time were shown to be decreased when the concentration of Machete was increased. For prelarval stage, 24hr and 48hr LC 50, and 24hr and 48hr EC 50 were observed to be 0.86mg/l and 0.43mg/l, and 0.35mg/l and 0.20mg/l, respectively. For juvenile stage, 24hr, 48hr, 96hr, and Incipient LC 50 were found to be 0.625mg/l, 0.365mg/l, 0.180mg/l, and 0.135mg/l, respectively. Apparently it was appeared that eggs were most resistant, and prelarvae were most sensitive to Machete among the three life stages. Although fish were removed from the Machete solution(3.2mg/l, and 1.8mg/l) prior to their death, continuous mortality was observed. Below 1.mg/l range, however, when concentration of Machete was lowered, the toxic effect of Machete on carp was reduced considerably even to the longtime exposure(24hours) Toxicity of Machete appeared to be related to water temperature, and it was observed that higher temperature reflected more toxicity.

Effects of Water Temperature on Egg Development, Hatching and Laval Growth Rearing of the Pacific Cod Gadus macrocephalus (태평양 대구 Gadus macrocephalus 채란, 부화 및 자어성장에 미치는 수온의 영향)

  • We investigated egg collection and the effects of water temperature (4, 7, 10, 13 and $16^{\circ}C$) on egg development, hatching and larval growth of Pacific cod Gadus macrocephalus under laboratory conditions. Fertilized eggs were round in shape ($1.01{\pm}0.03\;cm$) and adhesive to nylon nets. Fertilization rate was 68% by wet method. The time of egg development was negatively proportional to water temperature with the range of $4^{\circ}C$ to $13^{\circ}C$. Eggs hatched only at $7^{\circ}C$ after 288 hours of fertilization and $10^{\circ}C$ after 192 hours. Hatching rate was highest as 65% at $7^{\circ}C$ followed by $34.4^{\circ}C$ at $10^{\circ}C$. Survival rate was 18.3% at $7^{\circ}C$ and 5.2% at $10^{\circ}C$.

  • Larvae of the freshwater shrimp, Macrobrachium nipponense were reared in the laboratory at constant condition $(25^{\circ}C,\;7\%o)$, and their feeding rate, oxygen consumption rate, and growth rate were measured in regular intervals of time during larval development. Regression equations describing rates of feeding, growth and respiration as functions of time during individual larval molt cycles were inserted in a simulation model in order to analyse time-dependent patterns of variation as well as in bioenergetic efficiencies. Absolute values for feeding, growth, respiration and assimilation showed clear changes during the molt cycle, The absolute and specific values of respiration (R: R/C) showed small variation during the individual molt cycles. Significance of respiration in relation to growth (G) increased within the carbon budget, respiration rate (R/C) outbalanced growth rate (G/C) in late premolt. When the portion of metabolizable carbon is respired (R/G), metabolic coefficient was < 1 (i.e. R$(K_2)$ decreased concurrently, In cumulative carbon budget, total feeding was $491.54\;{\mu}g$ C/ind., assimilation was $85.3\%$, respiration was $47.7\%$, and growth was $37.6\%$ from hatching to postlarval stage.

  • For disposal food wastes which is about 30% portion of tatal organic wastes, vermicomposting is more environmental-friendly treatment than lanfill, incineration, etc. Recently, the interest has been increasing but there are many problems on management of vermicopmposting in field especially. This study was conducted to investigate an optimal factors, which are limit NaCl conc., the mixing rate of food wastes and casting for growth of an earthworm related to efficient vermicomposting. The limit conc. of NaCl was 0.5% and in case of feeding food wastes to earthworms as a prey only, most earthworms were dead in a few hours due to excessive degradation of organic materials and high NaCl conc. However as feeding with the casting of proper mixing rate(3:7), most earthworms were survived until finishing composting. It was investigated the increaser degree of matureness of food wastes, the higher conc. of NaCl, therefore for efficient vermicomposting, it seemed proper mixing rate of food wastes and the casting is better than matureness of food wastes. and the most suitable mixing rate was 3:7 food wastes and the casting.

Effects of Dietary Methionine Level on Lipid Peroxidation and Hepatic Morphology in Rat (식이중의 Methionine첨가수준이 흰쥐의 체내 지질 과산화와 간조직 형태에 미치는 영향)

  • The effect of dietary methionine level on lipid peroxidation of rats was studied. Rats were fed vitamin E- selenium- deficient diet or diet supplemented with various levels (0.3, 0.6, 0.9%) of methionine. In rat fed MF diet, body weight gain and feed efficiency ratio were decreased compared with those of control rats, but reversed by supplementation with 0.3 and 0.6% methionine. Lipid peroxide levels in plasma and hepatic mitochondrial fraction of MF group rats were significantly higher than those of control rats. However, supplementation with 0.6% methionine modified this increment. GSH-Px activity was decrased to varying degrees in erythrocyte and hepatic mitochondrial fraction from rats fed MF diet. Methionine supplementation did not affect induction of this enzyme activity. Examination of hepatocytes by electronmicroscopy showed that Influence of vitamin E, selenium, and methionine deficiency was mainly characterized by lipid droplets, swollen mitochondria and microvilli destruction. Supplementation with various levels of dietary methionine modified these changes to some extent. The results of this experiment indicated that MF diet causes significant change in lipid peroxide level, GSH-Px activity and morphology of rats which these changes may lessen by supplementation with 0.6% methionine.

Protective Effect of Citrus unshiu Peel Extract on Ethanol-Induced Fatty Liver in Rats (흰쥐에서 감귤과피 추출물의 알코올성 지방간 개선 작용)

  • This study investigated whether or not Citrus unshiu peel extract (CPE) affects fat accumulation in livers of rats fed an ethanol-containing liquid diet. Initially, male Sprague-Dawley rats were housed individually in stainless steel, wire-bottomed cages with free access to a Lieber-Decarli control liquid diet. Rats were divided by body weight into three groups of eight each: one group of rats was fed the Lieber-Decarli control liquid diet devoid of ethanol (control), another was fed the Lieber-Decarli ethanol diet (ethanol), and third was fed the same ethanol diet except containing CPE. All three groups were fed their respective diets for 6 weeks. Serum and liver lipids were analyzed and liver histology performed. Body weight did not differ among the groups over the 6-wk duration. Histology images showed that CPE administration significantly improved fat accumulation in livers, which was induced by ethanol diet. Serum levels of transaminases and lipids also were reduced by CPE consumption. Taken together, the results indicate that CPE may protect ethanol-induced fatty liver by lowering fat accumulation in both the liver and blood. The protective effects of CPE appear to be due to its phenolic contents.

A study of metabolic effect in high and low fat diet on Albino Rat (지방 함량에 따르는 흰쥐의 체내 대사 연구)

  • The present study was undertaken to determine the metabolic effect of various levels of fat in the diet. Forty males and the same number of females weighing $35{\pm}29$ were divided into three experimental groups and one control group, 10 rats each in both sexes. The dietary lipid contents were included in three levels, 2% as low, 30% as high and fat free diet in order to reflect the lipid consumption of present Korean diet. 20% sugar casein diet were employed as standard for control animals. This study was carried for 16 weeks. After these period animals were sacrificed to collect the internal organs and blood samples by heart puncture. In the result of this study, high fat diet group is lower than low fat diet group in the body weight gain, food efficiency ratio, protein efficiency ratio, urinary nitrogen and organ nitrogen contents were same trend but there is no significant difference in these respects. It was noteworty that high, and free fat diet group revealed more glucose total protein, albumin and globulin contents in the serum than other compared groups. It can be concluded that fat content whether low or high are free in the diet did not show significant effect on body nitrogen metabolism. But 30% high fat diet increased the total lipid and total cholesterol contents in the liver and the serum. This fact can be interpreted that 2% low fat diet and fat free diet do not need to increase the amount of fat content in the diet as high as 30% fat diet. This result might indicate the one possible reason to decide the recommanded dietary fat levels in Korean diet.

Effect of Monascus koji on Blood Pressure and Serum Cholesterol Composition of SHR by Chronic Dietary Administration (홍국 장기투여가 SHR의 혈압 및 혈중 콜레스테롤 조성에 미치는 영향)

  • The antihypertensive and cholesterol- lowering effects of Monascus koji (M. koji) prepared with Monascus ruber IFO32318 were examined in spontaneously hypertensive rats (SHR). Control was fed a normal diet and groups I, II or III were fed diets containing 0.03%, 0.1% or 0.3% M. koji for 8 weeks, respectively. After 8 weeks, all animals were fed normal diets in the following 2 weeks. The blood pressure of rats fed M. koji added diets were significantly attenuated as compared with control and the diet containing high concentration of M. koji had a tendency of stronger antihypertensive effect. These differences lasted throughout the experimental period when they were fed experimental diet. For 2 weeks after the 8 weeks of experimental diet all groups were fed the same normal diet and the differences of blood pressure caused by M. koji disappeared. In rats fed the M. koji added diet, the serum total cholesterol (TC), HDL cholesterol (HDL-C), LDL cholesterol (LDL-C) were not differed compared with control, but VLDL cholesterol (VLDL-C) was significantly lowered. M koji also significantly decreased serum risk factors, both TC/HDL-C ratio and non HDL-C/HDL-C ratio. Consequently, it is suggested that M. koji may play an important role to attenuate hypertension and to improve serum lipid Profiles.
