Efficacy and Safety Profile of TS-1 or TS-1/CDDP in Patients with Advanced Gastric Cancer (진행성 위암환자에 있어 TS-1 또는 TS-1/CDDP의 항암효과 및 안정성)

  • Purpose: Although several chemotherapy regimens used against advanced gastric cancer (AGC) have been studied extensively in an attempt to further improve the prognosis of patients, to date, no standard chemo-therapeutic regimens have been established. The aim of this study was to determine the anti-tumor efficacy and safety of TS-1 or TS-1 plus cisplatin (CDDP). Material and Methods: We treated 78 patients with AGC either with $80\;mg/m^{2}$ of TS-1 for 28 days, which was followed by a 2-week rest, or with $80\;mg/m^{2}$ of TS-1 for 21 days and $80\;mg/m^{2}$ of CDDP on day 8 every 5 weeks. Results: Tumor response rates in the neoadjuvant chemotherapy group and in the recurrent or post-palliative surgery group were 87.5% and 32.4%, respectively, and they were 28.6% and 48.4%, respectively, in the TS-1 group and the TS-1 plus CDDP group. The survival rates in the recurrent and the post-palliative surgery group were significantly different according to the degree of tumor response (P=0.0016), but the one-year survival rates according to the kinds of regimens (TS-1 or TS-1/CDDP group) were not significantly different. The incidences of grade 3 or 4 adverse effects in the TS-1 and the TS-1/CDDP groups were 14.3% and 36.8%, respectively. Conclusion: The anti-tumor efficacy and safety of TS-1 and TS-1 plus CDDP in Korean patients with AGC seemed to be high with modest adverse effects, thus suggesting the possible use of this regimen as a standard chemotherapy for gastric cancer.

Herbicidal Response and Germination Characteristics of Green kyllinga(Kyllinga brevifolia var. leiolepsis H.) Propagules (파대가리(Kyllinga brevifolia var. leiolepsis H.) 번식기관의 발아특성과 제초제에 대한 반응)

  • In this study, germination characteristics and herbicidal response of green kyllinga(Kyllinga brevifolia var. leiolepsis H.) were investigated. The storage method desirable for a rapid dormancy release was to keep the seed under low temp. and wetting condition for one to two months, or high temp($40^{\circ}C$) and drying condition for three months. The dormancy of rhizome was hardly observed. The optimum temperature for germination of seed and rhizome was around $30^{\circ}C$ and 16-$20^{\circ}C$, repectively. The germination of dormancy-breaked seed was completely dependent on light. Shoot emergence ratio(%) was decreased with increase of planting depth ; for example, only 18% of rhizome segments planted in the depth of 4cm under soil surface emerged above soil surface. Flooding at earlier growth stage resulted in significant decrease in shoot emergence as well as in dry weight. The germinablity of rhizome was almost lost as a decreased in fresh weight reached to 50%. Usually, green kyllinga was sensitive to herbicides such as bentazone, bensulfuron and benfuresate etc. which were known to be effective in Cyperaceae weeds, indicating that green kyllinga can be used as a representative plant in the screening of herbicides for Cyperus weeds.

Response of Colletotrichum sup. Causing Red Pepper Anthracnose to Protective and Ergosterol Biosynthesis-inhibiting Fungicides (보호용 살균제와 ergosterol 생합성 저해 살균제에 대한 고추 탄저병균의 약제 반응)

  • In 1999 and 2002, 130 and 258 isolates of Colletotrichum spp. causing red pepper anthracnose were obtained from infected red pepper fruits, respectively. Their responses to 4 protective and 3 ergosterol biosynthesis-inhibiting(EBI) fungicides were investigated by observing their mycelial growth on PDA incorporated with different concentrations of each fungicide. The Colletotrichum isolates obtained in 1999 showed higher $EC_{50}$ values than those isolated in 2002 against three protective fungicides such as dithianon, chlorothalonil, and propineb, whereas the response was reversed toward other protective fungicide, iminoctadine. On the other hand, the isolates of year 1999 were more resistant against three EBI fungicides such as tebuconazole, hexaconazole, and prochloraz than those of year 2002; the $EC_{50}$ values of the former were 1.2-4.4 times higher than those of the latter, The responses of the Colletotrichum isolates toward protective and EBI fungicides were fluctuated according to regions, where the infected fruits were collected. On the other hand, the resis tance of Colletotrichum isolates to protective fungicides increased during monitoring from July to September, However, their responses towards EBI fungicides were not changed.

  • The concept of innate immunity in insects which refers to the first line of host defense constitutes the humoral and cellular components which are involved in recognition and actively participate in the elimination of the intruding foreign micro- or macro-organisms. Several recent studies suggest that juvenile hormone (JH) modulates the cellular immune reactions in response to pathogen. In this study, pyriproxyfen (a JH agonist as an insect growth regulator) was tested in its any inhibitory effect on the immune reactions of the beet armyworm, Spodoptera exigua. To this end, five different hemocyte morphotypes of final instar S. exigua were identified by phase contrast microscopy. Plasmatocytes and granular cells, which constitute about 90% of the total hemocyte count, were prominently distinguished based on their basophilic/acidophilic nature using Giemsa stain. The role of pyriproxyfen on the functional ability of hemocytes was analyzed using FITC-labeled Providencia vermicola for the phagocytic potential of the hemocytes. Both granular cells and plasmatocytes exhibited phagocytosis behavior. Pyriproxyfen significantly inhibited the phagocytosis of both cell types, proposing its novel action as an immunosuppressant.

Sensitivity to Sterol Biosynthesis Inhibitors of Colletotrichum gloeosporioides Isolated from Persimmon in 2013 in Sangju, Gyeongsangbukdo (2013년 경북 상주지역 감나무로부터 분리한 Colletotrichum gloeosporioides 탄저병균의 스테롤 생합성 저해 살균제에 대한 감수성)

  • Colletotrichum gloeosporioides is one of the most serious pathogens of persimmon in Korea. In 2013, 67 isolates of C. gloeosporioides were isolated from infected fruits, leaf and twigs of persimmon (Diosprosi kaki) at Sangju, Gyeongsangbukdo and fungal responses against five fungicides (prochloraz manganese complex, tebuconazole, mancozeb+myclobutanil, fluquinconazole+prochloraz, and tebuconazole+tolyfluanid) were evaluated by their growth on the fungicide-medium. All isolates were inhibited mycelium growth on the medium with each recommended application concentration of flied. $EC_{50}$ (${\mu}g/ml$) of tebuconazole was from 0.02 to 1.04 and average was 0.31. $EC_{50}$ (${\mu}g/ml$) of prochloraz manganese complex was 0.02~0.23 average was 0.078. Average $EC_{50}$ values (${\mu}g/ml$) of Fluquinconazole+Prochloraz (FP) was 0.029. On the basis $EC_{50}$ (${\mu}g/ml$), the correlation coefficient (r) between tebuconazole and prochloraz manganese complex, prochloraz manganese complex and FP, tebuconazole and FP were 0.42, 0.44 and 0.27, respectively.

Effects of Walgookwhanhab-Bojoongikgitang administration on immune-function in Balb/c mice stressed by electric footshock (월국환합보중익기탕(越鞠丸合補中益氣湯)이 전기자극(電氣刺戟)스트레스를 받은 mouse의 면역기능(免疫機能)에 미치는 영향(影響))

  • The present experiments were designed to study the influence of WalgookwhanhabBojoongikgitang on immune functions of Balb/c mice under stress condition. WalgookwhanhabBojoongikgitang was orally administered to the mice for 15 days. On the 10th day the mice were immunized with sheep red blood cells (SRBC) and then subjected to electric footshock for 5 days (2 sessions a day, 11 footshocks a 31 min-session). The immune responses to SRBC were determined by means of hemagglutination and BIT cell populations in the spleen were studied by FACS analysis on the 16th day. The results were as follows. 1. After electric footshock, IDlce became sluggish and crowded to one side of the cage. Increased antibody titer for SRBC, increased B cell population, and decreased T cell population in the spleen were also observed. These results confirm that electric footshock caused stress inducing immunological and behavioral changes in Balblc mice. 2. Walgookwhanhab-Bojoongikgitang administration significantly antagonized the effect of electric footshock on the antibody titer for SRBC. As a result, antibody titers for SRBC in the mice treated with Walgookwhanga-Bojoongikgitang were maintained at the similar levels as those of control group mice even after the electric foots hock. 3. Walgookwhanhab-Bojoongikgitang administration also antagonized the effect of electric footshock on the BIT populations in spleen. As a result, Band T populations in the mice treated with Walgookwhanhab-Bojoongikgitang were maintained at the similar levels as in the control group mice even after the electric footshock. Taken together, Walgookwhanhab-Bojoongikgitang seem to help Balb/c mIce to maintain their humoral immune response and immune cell populations at a normal range under the stress conditions, suggesting its possible therapeutic use as a immune function modulator.

Persistence of the Insecticide Clothianidin in Paddy and Upland Soils (논 및 밭토양 중 살충제 Clothianidin의 잔류특성)

  • BACKGROUND: The current study purposed to analyse the dissipation levels of a neonicotinoid insecticide and clothianidin in paddy and upland soils and clarify the effects of soil moisture on degradation and persistence of the insecticide. METHODS AND RESULTS: In order to achieve the research purposes, clothianidin 8% SG was applied to the paddy and upland fields at the rate of 0.024 kg a.i./10a, while the analytical standard was treated at 0.25 mg/kg soil under laboratory conditions. Based on the multiple first-order kinetics, total clothianidin in soils was dissipated with $DT_{50}$ of 6.7-16.1 and 6.9-8.2 days in the paddy and upland fields, respectively, whereas the figures under the laboratory condition became larger showing 56.3 and 19.6 days. CONCLUSION: As affected by soil moisture, some differences in degradative pathways were observed. Flooding of soil caused evidently demethylation and delayed cyclization of a major metabolite, thiazolylmethylguanidine (TMG) and methylaminoimidazole(MAI), compared to the aerobic upland condition. More than 80% and 50% of the parent compound was dissipated by the 24th day after the final application in both soils and, transformation products had constituted most of soil residues after that.

Leaching and Adsorption of Flupyrazofos(KH-502) in the Soil (Flupyrazofos(KH-502)의 토양 중 용탈 및 흡착)

  • Adsorption, leaching, and retention of the Flupyrazofos(KH-502), a new active ingredient for insecticide, in the soils under laborarory and field conditions were investigated to provide the basic data for the safety use and to assess a secondary impact of this insecticide on soil and water environments. A significant power function relation was found between the adsorbed KH-502 and time, representing that 45% of the added KH-502 was adsorbed within 30 min. but a quasiequilibrium was reached after 6 to 12 hr with a slower adsorption. Adsorption phenomena followed th first-order kinetics and time required for 50% adsorption was 5.8 hr. The equilibrium adsorption isotherm was explained by the Freundlich equation and was classified as S-type. The amounts of KH-502 leached through the soil column (C) as compared to initial conc. ($C_0$) were very low and these relative concentrations ($C/C_0$) were 0.073 and 0.017 in SL and CL soils, respectively. The residual conc. of KH-502 in the surface soil was comparatively low and decreased with time. Half-lives of KH-502 in the surface soil was comparatively low and decreased with time. Half-lives of KH-502 under the field conditions were estimated to be 20 and 18 days in the SL and CL soils, respectively. The KH-502 cone, transported to the subsurface soils was extremely low. These results demonstrate that KH-502 has a low pollution risk potential to the surrounding environment as far as it is used following the recommended guideline.

  • DNA damage accumulates in cells as a result of exposure to exogenous agents such as benzopyrene, cigarette smoke, ultraviolet light, X-ray, and endogenous chemicals including reactive oxygen species produced from normal metabolic byproducts. DNA damage can also occur during aberrant DNA processing reactions such as DNA replication, recombination, and repair. The major of DNA damage affects the primary structure of the double helix; that is, the bases are chemically modified. These modification can disrupt the molecules'regular helical structure by introducing non-native chemical bonds or bulky adducts that do not fit in the standard double helix. DNA repair genes and proteins scan the global genome to detect and remove DNA damage and damage to single nucleotides. Direct reversal of DNA damage, base excision repair, double strand break. DNA repair are known relevant DNA repair mechanisms. Four different mechanisms are distinguished within excision repair: direct reversal, base excision repair, nucleotide excision repair, and mismatch repair. Genetic variation in DNA repair genes can modulate DNA repair capacity and alter cancer risk. The instability of a cell to properly regulate its proliferation in the presence of DNA damage increase risk of gene mutation and carcinogenesis. This article aimed to review mechanism of excision repair and to understand the relationship between genetic variation of excision repair genes and head and neck cancer.

Tolerance of Apple Blue Mold (Penicillium expansum) to Various Fungicides (사과 푸른곰팡이병균의 각종 살균제에 대한 내성)

  • Responses to the 14 fungicides added in PSA of penicillium expansum isolated froms the four major apple growing areas in Korea were examined. Degrees of the fungal tolerance were various with little difference by area and with great difference by kind and concentration of fungicide. Fall isolates of the fungus grew at higher fungicide concentration than did the spring isolates. $ED_{50}$ and MIC to mycelial growth of genomyl, garbenda, polydong, polyoxin and thiophanate methyl were $0.1{\mu}g-41.7{\mu}g/ml\;and\;100{\mu}g-2,250{\mu}g/ml,$, respectively, indicating high fungicide effects on the fungus isolated from decayed apples in storages and infected apples in orchards with low fungal tolerance; of captafol, captan, iprodione and mancozeb were $2.6{\mu}g - 750{\mu}g/ml\;and\;638{\mu}g-40,500{\mu}g/ml$, respectively, indicating medium degree of both fungicidal effects and fungal tolerance; and of chlorothalonil, folpet, oxidong, propineb and triademefon were $27.8{\mu}g-8,354{\mu}g/ml\;and\;2,625{\mu}g-150,357{\mu}g/ml$, respectively, indicating low fungicide effects and high fungal tolerance. Fungicidal inhibitory effects on conidia formation exhibited a similar trend as on mycelial growth whereas those on conidia germination showed an opposite trend to the mycelial growth with the exception of polyoxin and triademefon.
