Apoptotic Effect of Phellodendri Cortex Water Extract on MIA PaCa-2 Cells (췌장암 세포주 MIA PaCa-2에서 황백 물 추출물에 의한 Apoptosis 유도 및 작용기전)

  • The purpose of this study is to investigate the apoptotic effect of Phellodendri Cortex water extract (PCWE) on pancreatic cancer cells and to find out the regulating mechanisms. Human-derived pancreatic cancer cell line, MIA PaCa-2 cells were treated by PCWE with various concentrations and the cytotoxicity was determined by MTT assay. The activation of Annexin V, DNA fragmentation, cell cycle arrest and caspase activation were observed to investigate the role of PCWE in pancreatic cancer cells. Also, to find out the regulating mechanisms, we examined the ROS production. The treatment of PCWE induced the cell death in both concentration and time dependent manner. The treatment of PCWE also increased the expression of Annexin V, DNA fragmentation, cell cycle arrest, and cleavage of caspase, which means cell-death PCWE induced was apoptosis but not necrosis. The ROS production was increased by PCWE treatment and the blockade of ROS inhibited the PCWE-induced cell death. These results could suggest that PCWE induced apoptosis via ROS release in pancreatic cancer cell.

Inhibition Effect of Chunglijagam-Tang on Invasion Activity of Human Lung Adenocarcinoma, A549 (청리자감탕(淸離滋坎湯)의 폐암 세포주 A549의 invasion activity 억제 효과)

  • By applying in vitro invasion assay model, we examined the anti-metatstastic effect of ChunghjagamTang(CLJGT). In 3H-thymidine incorporation assay, CLJGT treated groups showed the decreased DNA synthesis rate compared with control group. Gelatin zymogram assay showed that CLJGT decreases the gelatinolytic activity of MMP-9 from A-549, at the concentration of $800{\mu}g/ml$. We examined whether CLJGT inhibits the invasion of A-549 cells through the matrigel precoated transwell chamber. The results showed that CLJGT effectively inhibited the invasion of A-549 as compared with the control (+PMA) groups. From our research, part of mechanism underlying anti-metastastic effect of CLJGT was proven in vitro.

  • In an effort to develop a new therapeutic strategy for human gene therapy of solid ovarian tumor, we studied the expression of the p53 tumor suppressor Sene as well as regulatory subunits of cyclic AMP (cAMP)-dependent protein kinase in human ovarian carcinoma cells. Four cell lines (2774, Caov-3, SK-OV-3 and OVCAR-3) were selected for the analyses. The p53 transcript and protein were detected only in the 2774 cell line by Northern and Western Bnalysis. In the relatively fast growing cell line, SK-OV-3, the %rope 1 a regulstorv subunit (RIA of CAMP-dependent protein kinase was the highest among the four cell lines. The expression level of $RII\beta$ protein was low in the four cell lines examined. These results maw point to a direction to select the target gene(sl to be employed for gene therapy to control the ovarian cancer.

Inhibition of Cellular Proliferation by CURCUMAE LONGAE Rhizoma Extracts on MCF-7 (강황(薑黃) 추출물이 유방암 세포주 MCF-7 증식 억제에 미치는 영향)

  • Objectives : This investigation was undertaken to evaluate the anti-proliferation of CURCUMAE LONGAE Rhizoma extracts using MCF-7, human breast cancer cells. Methods : MCF-7 cells were cultured in Dulbecco's modified Eagle's medium/F12 supplemented with 10% fetal bovine serum and antibiotics. At varying times after extract treatment, cells were harvested with scraper and processed for analysis of proliferation, cytotoxicity. Results : The extract of CURCUMAE LONGAE Rhizoma strongly inhibits the proliferation of MCF-7 cells in a dose and time-dependent manner. Sulforhodamine B assay showed that the addition of ethanol extract of CURCUlVIAE LONGAE Rhizoma reduced the viability of MCF-7 cells in a dose-dependent manner. Conclusion : So, it can be concluded that CURCUMAE LONGAE Rhizoma have an inhibitive effect on MCF-7 human breast cancer cells.

  • Objectives : This study was carried out to investigate the inhibitive effects of cotton plant sectional extracts in cancer cell lines, Calu-6(human, Caucasian, lung, adenocarcinoma) and MCF-7(human, Caucasian, breast, adenocarcinoma). The incidence of cancer has been increasing even in korea due to the change of dietary life and westernization and becoming conspicuous as the disease threatening health. But cancer treatment have not been fully effective against the high incidence or low survival rate of most cancer. Methods : Calu-6 and MCF-7 cells were cultured and seeded in cell culture plates, respectively. And sectional extracts of cotton plant were treated to MCF-7 cells. Results and Conclusion : Sectional extracts of cotton plant showed no anti-proliferative effect on MCF-7 cells, but root and stem extracts showed strong anti-proliferative effects on Calu-6 cells. Fruit, leaf and flower extracts also showed anti-proliferative effects on Calu-6 cells but not so much like root and stem extracts. But seed extract showed no anti-proliferative effect on Calu-6 cells.

  • Objecgtive : The aim of present study was to investigate inhibition effect of Whakijogyung-Tang(WJT) on the tumor cell lines. This study estimated the cytotoxicity of WJT about viability of S-180 and NlH3T3. Methods : The cytotoxicity of WJT about viability of cells were tested using a colorimetric tetrazoliun assay(MTT assay) Results and Conclusion : 1. Water extract of WJT had $IC_{50}$ of 863 ${\mu}g/ml$ in S-180 cell lines, but cytotoxicity of NIH3T3 was not significant difference compare with S-180. 2. n-Hexane fraction of WJT had similar cytotoxicity between S-180 and NIH3T3, but that could not have $IC_{50}$ in S-180 cell lines. 3. Ethyl acetate fraction of WJT had low degree cytotoxicity both S-180 and NIH3T3 cell lines. 4. Significantly, Butanol fraction of WJT had differenct citotoxicity between S-180 and NIH3T3. 5. $H_2O_2$ fraction of WJT had no cytotoxicity both S-180 and NIH3T3.

  • Objectives : The purpose of this study was to investigate inhibitory effect of Gagamtongsung-San(GTS) on the cancer. Methods : This study estimated the cytotoxicity of GTS about L1210 and NIH3T3. We used GTS extract distilled with water, n-Hexane, Ethyl acetate and Butanol. The cytotoxicitys of GTS about cancer cells and normal cells were tested using a colorimetric tetrazoliun assay(MTT assay). Results : The results of this study were obtained as follow ; l. Cytotoxicity of water extract of GTS in L1210 cell lines was significantly increased, compared with NIH3T3. 2. n-Hexane fraction of GTS had similar cytotoxicity between L1210 and NIH3T3, and that have similar $IC_{50}$ of water extract of GTS at 276 ${\mu}g/ml$ 3. Ethyl acetate fraction of GTS had low degree cytotoxicity both L1210 and NIH3T3 cell lines. 4. Butanol fraction of GTS had cytotoxicity between L1210 and NIH3T3. Significantly, Cytotoxicity of GTS in L1210 cell lines was significant increased. 5. $H_2O$ fraction of GTS had no cytotoxicity both L1210 and NIH3T3.

Anticancer and Antioxidant Effects of Saturated Fatty Acid against Skin Cancer Cell Lines (포화지방산의 피부암 세포주에 대한 항암 및 항산화 효과)

  • The anticancer and antioxidant effect of different lengths of saturated fatty acids was tested on NIH3T3 fibroblasts and human skin melanoma cellsn in this study. The cell existence rate and antioxidizing capacity and optic reservation of cells were observed. This saturated fatty acid was concentration-dependent. IC50 Concentrations in NIH3T3 fibroblasts, human skin melanoma cells and DPPH radical scavenging activity of fatty acid was increasing the order of carbochain length ; caprylic acid < lauric acid < palmitic acid < stearic acid. The reduction in cell number and morphological change in human skin melanoma cells was increasing the order of carbochain length ; caprylic acid < lauric acid < palmitic acid < stearic acid. These results suggest that carbochain length of fatty acid can be used as structure-activity relationships for anticancer and antioxidant.

  • The aim of this study was to examine the ameliorating effect of black ginseng on the growth of the HepG2 cell transplanted tumor in BALB/c nude mice. 27 male BALB/c nude mice (all six weeks old) were randomly divided into three groups: the control group, the first treatment group (HepG2300RG, using 300 mg/kg red ginseng), and the second treatment group (HepG2300BG, using 300 mg/kg black ginseng). The HepG2300BG in the HePG2 cells showed increased mean survival time than that of red ginseng group. The size and volume of the tumor in the 300BG group showed significant reduction compared to those of the HepG2300RG group (p<0.05). The body weight and liver weight of the HepG2300RG group was not significantly different with control and HepG2300BG group. The serum levels of ALT and AST in the HepG2300RG and HepG2300BG group were significantly lower than those of the control group. In conclusion, these results suggest that the black ginseng may have possible anti-tumor activities.

Estrogenic Activity of Leguminosae Species in Korea using MCF-7 Cells (유방암 세포주를 이용한 국내 자생 콩과식물의 에스트로겐 활성검색)

  • Leguminosae plants are known for its phytoestrogen constituents which play a major role in the prevention of osteoporosis, cancer and heart disease. In this study, the estrogenic activity of 158 samples from 58 species, 3 subspecies and 10 varieties of Leguminosae plants growing in Korea was evaluated. An estrogen, 17β-estradiol was used as a reference compound, and the potency of each sample was expressed in relative efficacy (%) compared to that of the reference by a reporter gene assay using MCF-7 cells. As results, the estrogenic activity of methanolic extracts of Phaseolus vulgaris var. humilis, Sophora flavescens, Lespedeza × robusta, Indigofera pseudotinctoria, Maackia amurensis, Glycine soja, Wisteria floribunda, Robinia pseudoacacia, Astragalus sinicus, Pueraria lobata, Lespedeza maximowiczii var. tomentella, Trifolium repens and Crotalaria sessiliflora showed similar to or higher at 100 ㎍/ml than the positive control at 10 nM. These findings can be a potential evidence for developing estrogen alternatives resolving various types of menopause symptoms with information on proper harvest season and usage plant part. To the best of our knowledge, the estrogenic activity of Lespedeza × robusta, Indigofera pseudotinctoria, Wisteria floribunda, Robinia pseudoacacia and Lespedeza maximowiczii var. tomentella is reported for the first time in this study.
