Study on the Cultivation of Vitex rotundifolia According to Different Soils and Fertilization Methods (토양과 비배관리에 의한 순비기나무의 재배방법에 관한 연구)

  • This study is focused on the cultivation of seedlings and cuttings of roundleaf chastetree(Vitex rotundifolia L. f.) according to the sorts of soil and fertilization methods. The results are summarized as follows: When roundleaf chastetree seedlings(aged 1-0) and cuttings(aged 1-0) were cultivated in three different kinds of soil- sand, granite soil, and yellow silt- without any fertilization, 70% of the seedlings survived in sand and granite soil, and 35% did in yellow silt; 90% of the cuttings survived in granite soil and 80% did in sand and yellow silt. The general growth of both seedlings and cuttings was good in granite soil, and sand but relatively inferior in yellow silt. Seedlings showed good stem growth while cuttings had fine root growth. In case of cultivation of seedlings with different kinds and quantities of fertilizer adjusted and with granite soil as basic soil, seedlings and cuttings survived in the experimental section where only the organic fertilizer was used as basic fertilization; in contrast, seedlings grown in the experimental section were all dead where both inorganic and liquefied fertilizer were used. Cuttings showed considerably higher survival rates than seedlings at the experimental fertilization section; particularly, the survival rate of cuttings was twice as much as that of seedlings at the experimental section with low fertilization. This study results revealed that cuttings are desirable than seedlings in general rootage, growth, adaptability to soil, and cuttings have lower demand for soil fertility in time of growing roundleaf chastetree saplings.

Early Identification of Citrus Zygotic Seedlings Using Pollen-specific Molecular Markers (화분 특이적 마커를 이용한 감귤 교잡종 실생묘의 조기 동정)

  • This study was carried out to develop molecular techniques to allow the selection of zygotic seedlings in the early stage of the plant development. We identified 37 pollen-specific molecular markers from RAPD analysis and successfully used them for identification of the zygotic seedlings from various hybrid crosses. Three Satsuma mandarin cultivars ('Morita unshiu', 'Nangan 20' and 'Miyagawawase') were used as mother parents and seven cultivars ('Ponkan', 'Lee', 'Kinokuni', 'Shiranuhi', 'Tamnaneunbong', 'Shinyegam', and 'Sunburst' mandarins) served as pollen parents. PCR analysis showed that 2 primers could identify zygotic hybrid seedlings. Among them, an UBC-27 primer was used to identify the zygotic seedlings from hybrid crosses of "'Nangan 20' ${\times}$ 'Kinokuni'" mandarin, "'Nangan 20 ${\times}$ Ponkan'" mandarin and "'Miyagawawase ${\times}$ Sunburst'" tangerine. In total 29 out of 40 seedlings (73%), 9 out of 47 seedlings (19%), and 13 out of 45 (29%) were identified as zygotic seedlings, respectively. These results can show that the pollen-specific markers selected in this study can be used effectively for early identification of zygotic seedlings from Citrus hybrid crosses.

  • The purpose of this study was to evaluate the methods of identifying zygotic seedlings of crosses between 'Morita unshiu' (Citrus. unshiu Marc.) and 'Ponkan' (C. reticulata Blanco). In order to investigate the frequency and position of zygotic seedlings and to determine the efficiency of zygotic seedling identification, random amplified polymorphic DNA (RAPD) and sequence-related amplified polymorphism (SRAP) were performed using UBC (9, 27, 229, 230, and 254) primers and F4/R27, F7/R14, F12/R10, and F44/R62 primer sets, respectively. A total of 37 putative zygotic seedlings out of 55 individuals were selected by RAPD and SRAP. The F7/R14 primer pair showed a screening ability of 45.5% (25/55), whereas the primer UBC27 revealed the highest efficiency of zygotic seedling identification (50.9%, 28/55). When both UBC27 and F7/R14 were properly used for selection of hybridized seedlings of 'Morita unshiu' (C. unshiu Marc.) and 'Ponkan' (C. reticulata Blanco), screening efficiency was increased to 60% (33/55) for putative zygotic seedlings. Thus, it is possible to select putative hybrid zygotic seedlings in an accurate and effective manner by RAPD and SRAP.

Inheritance of Fruit Texture Traits in Oriental Pear (Pyrus pyrifolia var. culta Nakai) (동양배 과실의 육질 관련 형질의 유전분석)

  • To improve the breeding efficiency of oriental pear, the inheritance of major fruit traits were analyzed using 4,035 seedlings of 15 cross combinations combined with 13 cultivars. Yearly variation of parental cultivars, broad-sense heritability of the traits, and frequency of seedlings with commercially acceptable characteristics by the mid-parent values were studied. Despite the heritabilities of traits related with pear the fruit texture were different depending on cross combination, the average narrow-sense heritability ($h^2$) for firmness, flesh hardness, flesh density and grit were over 0.6, a relatively high value. However fruit firmness showed different heritability among the cross combination and its value of cross combination between 'Niitaka' and 'Choju' was very low as 0.11. Positive correlation was not observed on fresh hardness, fresh density and grit but observed on firmness between mid-parent value and average value of offspring. Regression of offspring on firmness, fresh hardness, fresh density and grit between mid-parent value and average value of offspring were 0.778, 0.343, -0.273, 0.313, respectively. Frequency distributions of fruit texture in 15 cross combinations of pear seedling was influenced by parental characteristics. When fruit firmness was estimated as low, low ${\times}$ high and high ${\times}$ high values in parental fruits, firmness in their offspring exhibited low, medium and high, respectively. In addition, when parental grit by sensory evaluation were light ${\times}$ light and light-medium ${\times}$ medium-heavy, most of offsprings showed light and medium grit, respectively.

Comparison of Physiological Characteristics, Stomata and DNA Content between Seedling and 5-year-old Somatic Plant (Somatic Embryo Derived-plant) in Liriodendron tulipifera (백합나무 5년생 실생묘 및 체세포묘 (체세포배 유래 식물체) 간의 생리적 요인, 기공 및 DNA 함량 비교)

  • Field performance of somatic plants (somatic embryo derived-plants) of yellow-poplar (Liriodendron tulipifera) produced from somatic embryogenesis was compared with that of seedlings at age 5. In comparison of photosynthetic rate (seedling, $10.67{\mu}mol$ $CO_2m^{-2}s^{-1}$; somatic plant, $9.04{\mu}mol$ $CO_2m^{-2}s^{-1}$), stomatal conductance rate (seedling, 0.2 $H_2Om^{-2}s^{-1}$; somatic plant, 0.166 $H_2Om^{-2}s^{-1}$) and respiration rate (seedling, 1.71 mmol $H_2Om^{-2}s^{-1}$; somatic plant, 1.513 mmol $H_2Om^{-2}s^{-1}$), no significant differences were found between plants. The seedlings were a little higher in comparison of stomatal density (seedling, $23.33/mm^2$; somatic plant, $22.43/mm^2$), length (seedling, $25.83{\mu}m$; somatic plant, $23.46{\mu}m$) and width (seedling, $15.87{\mu}m$; somatic plant, $15.3{\mu}m$). In comparison of DNA content of the leaves using flow cytometry, no differences in ploidy level were found between the seedlings and somatic plants.

광과 온도가 미국자리공(Phytolacca americana L.) 실생 출현과 종자 발아에 미치는 영향

  • 분포가 확대되어 가고 있는 미국자공의 서식지 환경요인과 발아특성을 조사하였다. 그 결과 광의 구배에 따른 기온의 차는 없었으나, 지온의 일교차는 상대조도에 따라 아주 다르게 나타났다. 실생 출현은 100%에서 150개체가 출현하였고, 33%에서는 80개체가 출현하였지만 8%에서는 출현하지 않았다. 2002년 채집되어진 종자는 광조건에 관계없이 항온에서 거의 발아하지 않았으나 광조건하의 변온에서는 발아율이 70%, 암조건하의 변온에서는 25%의 발아율이 보였다. 미국자리공의 발아는 광조건보다 지온변화에 보다 직접적으로 영향을 받는 것으로 판단된다.

Efficiency of Sex-linked Molecular Markers for the Selection of Seedlings Bearing Male Flowers in Persimmon (Diospyros kaki Thunb.) (감 수꽃 착생 실생개체 선발 마커의 효율성 검정)

  • Persimmon flowers are fruit-bearing female, pollen-bearing male, or hermaphrodite, containing both a pistil and a stamen. Using prominent PCNA persimmons as male parents is very important for breeding programs, as the selection procedure for new cultivars bearing male flowers requires a long time and a large field in a traditional crossbreeding method. To improve breeding efficiency through early selection of male flower-bearing plants at the seedling stage, analysis was performed on 88 major cultivars whose gender expressions are known, using two male flower selection markers recommended by Akagi et al. The OGI locus marker and DISx-AF4S marker results showed that 83 and 72 cultivars, respectively, matched in terms of gender expression and marker analysis. For the OGI locus marker, 890 plants were selected from 2,509 seedlings obtained from crossbreeding with the mother plant "Migamjosang," which was the breeding cultivar. Comparing the gender expression of the flowers and the marker with 1,186 crossbred seedlings, excluding the unfertilized and dead plants, inconsistencies were found in 401 plants (33.8%). For the DISx-AF4S marker, 636 plants were selected from 889 seedlings obtained from 12 cross-combinations. The results of the sex expression and marker analysis were compared to 379 plants, excluding the unfertilized and dead plants, and inconsistencies were found in 247 plants (65.2%). These results indicate that the examined DISx-AF4S and OGI locus markers would not be suitable for utilization in the breeding field.

A Study on the Tolerance to the Soil Properties and Water Contents of Vitex rotundifolia Seedlings for Extension of Rehabilitation Plant (순비기나무의 녹화소재 이용성 확대를 위한 토양 및 토양수분 적응성에 관한 연구)

  • This study was carried out to survey the usability of Vitex rotundifolia as an afforestation and landscaping plant on destroyed slopes, sandyhills of sea boards, and places in need of landscaping. The growth characteristics of seedlings and rooted cuttings of Vitex rotundifolia in soils of varying properties and water content ere studied. In three test soils, with a water content of 5% each, the survival rate was more than 70ole for seedlings and more than 40% for rooted cuttings. This demonstrates that Vitex rotundifolia is a very strongly xeric tree species. The length of stem, the diameter of root stock, leaves, and the length of root increased in all the three types of soil as the quantity of soil water increased. The difference of the growth in response to the water content of the soil was most pronounced in the length of the stem. The growth of seedlings was most notable in the weathered granitic soil. The sea sand and the red silt loam came second and third, respectively. However, there was no major difference in the three test soils. Thus, Vitex rotundifolia appears well-suited to several soil properties and soil water quantities, which makes it very useful as an afforestation and landscaping plant in various sites.

Variation of Growth Characteristics in Natural crossed Population of Paeonia lactiflora Pallas (자연교잡 실생작약 집단의 생육특성 변이)

  • This study was carried out to find growth characters and distribution of above and underground in natural crossed population of Paeonia lactiflora Pallas. Sprouting time was 1 April to 15 April, and distribution of stem length ranged from 21 to 80cm, and 3 to 18 in stem number. Distribution of root length ranged from 12 to 59cm, root number per plant was 73 to 4, root yield per plant was highly varied by 37 to 882g, and larger root above 750g was occupied by 7.1%. Paeoniflorin content was 1.9 to 10.29% and plants containing higher paeoniflorin was 14.3%. Root length and yield had highly positive correlation. Root diameter and stem length was also positively correlated.
