Effects of Rhodobacter sp. SA16 on Lettuce(Lactuca sativa L.) in Plastic Film House (시설 상추에 대한 Rhodobacter sp. SA16 처리 효과)

  • This experiment was conducted to determine the development of mixed organic fertilizer using photosynthetic bacteria and mass production of mixed microbial compound for the environment-friendly agriculture. Photosynthetic bacteria, Rhodobacter sp. SA16 was isolated from soil collected by plastic film house. The SA16 strain was identified based on 16S rDNA sequence analysis and it is closely related to Rhodobacter sp.(100% similarity). The mixed organic fertilizer using SA16 was made of $N-P_2O_5-K_2O=60-10-20\;g\;kg^{-1}$ with combined soybean cake, sesame cake, powdered blood, fish meal, powdered bones and red-yellow soil. The mixed organic fertilizer 0.45, 0.90 and 1.35 Mg $ha^{-1}$ application in Ihyeon series was treated based on soil testing for lettuce cultivation in plastic film house. These results showed that the yield was increased the 18 and 19%over control by the mixed organic fertilizer application 0.45 and 0.90 Mg $ha^{-1}$, respectively. In the physical properties of the soil, the porosity of mixed organic fertilizer 1.35 Mg $ha^{-1}$ treatment was highest at 58.8%. Our results clearly revealed that the organic fertilizer using Rhodobacter sp. SA16 and mass production of mixed strains could be a useful technology in pursuing environment-friendly agriculture.

Ozone Sensitivity of Physiological Indicators for Stress Evaluation in Four Families of Quercus aliena Blume (갈참나무 4가계에서 스트레스 평가용 생리 지표들의 오존 민감성)

  • Ozone sensitivity of physiological indicators and the difference of ozone tolerance on 4 families of Quercus aliena seedlings were investigated on the basis of the standardized physiological indicators. Photosynthetic parameters, photosynthetic pigment and malondialdehyde (MDA) content, and antioxidative enzyme activities were measured or analyzed from the leaves of Q. aliena seedlings at the end of ozone fumigation, and ozone tolerance indices among 4 families were calculated with the standardized physiological parameters. After ozone treatment, the reduction of carboxylation efficiency was observed in the leaves of four families, and their reduction were ranged from -24.1% to -56.9% of control seedlings. Photosynthetic pigment content differed significantly among 4 families and treatments. The reduction of total chlorophyll content showed the highest in family SU4 (-40.6%) and the lowest family US2 (-18.8%). Ascorbate-peroxidase (APX) activity showed significant difference among families and treatments, and increased as compared with control in three families, except for family US2. On the basis of the physiological indices, ozone tolerance of four families was ranked in the order of US1 > SU4 > US2 > SU1. In conclusion, photosynthetic parameters, pigment content and APX activity were recommended as appropriate indicators to assess the tolerance against ozone stress of Q. aliena.

Physiological and Biochemical Responses to Ozone Toxicity in Five Species of genus Quercus Seedlings (참나무속 5종의 오존 독성에 대한 생리생화학적 반응)

  • Physiological and biochemical changes of five species of genus Quercus exposed to ozone fumigation were investigated to assess their tolerance against ozone toxicity. At the end of 150 ppb ozone fumigation, chlorophyll contents, photosynthetic characteristics, malondialdehyde(MDA) and antioxidative enzyme activities were measured in the leaves of five Quercus species(Quercus acutissima, Q. aliena, Q. palustris, Q. serrata, and Q. variabilis). Chlorophyll and carotenoid contents, net photosynthesis and carboxylation efficiency decreased after ozone treatment, indicating that $O_3$-exposed plants underwent physiological inhibition. The reduction rate of total chlorophyll contents and carboxylation efficiency were respectively 15% and 34% for Q. aliena and 38% and 62% for Q. variabilis. The amount of MDA increased with the highest increase rate of 140% in Q. acutissima which also showed the highest increase rate(60%) of superoxide dismutase(SOD). Ascorbate peroxidase(APX) activity increased in Q. variabilis, Q. serrata and Q. acutissima by ozone treatment. Based on our results, ozone tolerance of the five Quercus species was ranked as Q. aliena>Q. palustris>Q. serrata>Q. variabilis>Q. acutissima. We concluded that chlorophyll contents, photosynthesis, MDA content and antioxidative enzymes were the important physiological markers for tolerance against ozone stress, which were closely related with one another.

Physiological Responses of Rhododendron mucronulatum and R. indicum with Shading Treatment in Autumn Season (가을철 차광 처리에 따른 진달래와 영산홍의 생리적 반응)

  • This study was carried out to investigate the physiological responses of $Rhododendron$ $mucronulatum$ Turcz. and $R.$ $indicum$ (L.) Sweet seedlings with 0%, 35%, 55% and 75% shading of full sunlight in polyethylene film house. The shading treatments were performed during the late growth season for each species (from Sept. 9 to Nov. 5, 2008). The shading treatment was effective in reducing the daily temperature by 0.9 to $1.7^{\circ}C$ during September and by 0.8 to $1.7^{\circ}C$ during October. Before the shading treatments, the water content of $R.$ $mucronulatum$ and $R.$ $indicum$ amounted to 68.5% and 66.3%, respectively. The water contents of two species after 75% shading treatment period decreased to 66.2% (3.4% reduction) and 65.9% (0.6% reduction), respectively. Notably, both species had a similar tendency indicating less reduction rate of water content with 75% shading. $R.$ $indicum$ showed higher photosynthetic capacity with higher level of shading, and its photosynthetic capacity reached the highest level ($9.63{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$). On the other hand, shading-treated $R.$ $indicum$ showed higher intercellular $CO_2$ concentration, stomatal conductance and transpiration rate (55% shading > 35% shading > 75% shading) than non-treated ones. In addition, non-treated seedlings showed higher water use efficiency than treated ones. In particular, it was found that the leaf color of $R.$ $mucronulatum$ turned equivalent to purple under full sunlight, while its leaf color kept equivalent more to green with higher level of shading, as evidenced even in naked eyes. According to comprehensive analysis using Munsell Color Chart on potential leaf color variations of $R.$ $mucronulatum$ depending on the level of shading, it was found that relatively many leaf colors under full sunlight were equivalent to R (red) and Y (yellow) chart, while relatively many leaf colors with higher level of shading were equivalent to G (green) and Y chart, where the latter still showed green color.

Physiological Responses of Green Roof Plants to Drought Stress (건조스트레스에 따른 옥상녹화 식물의 생리적 반응)

  • This study evaluated the drought tolerance of Liriope platyphylla F.T.Wang & T.Tang, Dendranthema zawadskii var. lucidum (Nakai) J.H.Park, Hosta longipes (Franch. & Sav.) Matsum., Sedum sarmentosum Bunge and Zoysia japonica Steud. for an extensive green roof. In order to assess drought tolerance of green roof plants, several criteria were measured such as volumetric water content, leaf and soil moisture potential, chlorophyll a and b, chlorophyll fluorescence, photosynthesis, stomatal conductance, transpiration rate and antioxidants. The results of the drought tolerance measurement of green roof plants focused on the gradually withering of plants from lack of volumetric water content. D. zawadskii was the first to show an initial wilting point, followed by Z. japonica, H. longipes and L. platyphylla in order while S. sarmentosum showed no withering. It was concluded that H. longipes, L. platyphylla and S. sarmentosum were highly drought tolerant plants able to survive over three weeks. Furthermore, chlorophyll a and b were divided into two types: Type I, which kept regular content from the beginning to the middle of the period and suddenly declined, like H. longipes and Z. japonica; and Type II, which showed low content at the beginning, sharply increased at the middle stage and decreased, like D. zawadskii, L. platyphylla and S. sarmentosum. Volumetric water content and the amount of evapotranspiration consistently declined in all plant species. The analysis of chlorophyll fluorescence results that S. sarmentosum, which had relatively high drought tolerance, was the last to decline, while Z. japonica and S. sarmentosum withered after rapid reduction. At first, photosynthesis, stomatal conductance and transpiration rate showed high activity, but they lowered as the plant body closed stomata owing to the decrease in volumetric water content. Measuring antioxidants showed that when drought stress increased, the amount of antioxidants grew as well. However, when high moisture stress was maintained, this compound was continuously consumed. Therefore, the variation of antioxidants was considered possible for use as one of the indicators of drought tolerance evaluation.

Studies on the Grassland Development in the Frest VII. Effect of shading degrees on the crop growth rate, net assimilation rate and photosynthtic active of main grasses (임간 초지 개발에 관한 연구 VII. 차광정도가 주요목초의 개체군생장향, 순동화율 및 광합성능력에 미치는 영향)

  • This experiment was carried out to investigate the effect of shading degrees (0:full sunlight, 25, 50 and 75%) on the crop growth rate (CGR), net assimilation rate (NAR) and photosynthetic activity of grasses for obtaining the basic data on the development, management and utilization of pasture in the forest. For the test, different artificial shading houses were established and pasture species used in this study were orchardgrass, timothy, perennial ryegrass and ladino clover. The experiment was performed at Livestock Experiment Station, Suwon, in 1984-'85. The results obtained are summarized as follows. 1. The highest CGR was obtained at 0% (full sunlight) and 25% of shading, followed by 50 and 75% of shading degrees, in that order. CGR of orchardgrass and perennial ryegrass were higher than those of timothy and ladino clover. 2. NAR was significantly decreased as the shading degrees increased. The highest NAR was observed at perennial ryegrass plot. 3. The photosynthetic activity during summer season was the highest at 25% of shading, followed by 50, 0 and 75% of shading degrees. While, the photosynthetic activity during fall season was the highest at 25% of shading, followed by 50, 0 and 75% of shading degrees, in that order. Those of orchardgrass and perennial ryegrass were remarkably higher than that of timothy. 4. A significant positive correlation was obtained in the relationship between CGR and NAR of main grasses, regardless of pasture species and investigated date.

Seasonal Variations of Sediment Oxygen Demand and Denitrification in Kanghwa Tidal Flat Sediments (강화도 갯벌 퇴적물의 산소요구량과 탈질소화의 계절 변화)

  • Seasonal variations of remineralization and inorganic nitrogen removal capacity were measured from Dec. 2001 to Apr. 2004 in a tidal flat located in south-western pan of Gwanghwa island, Korea by measuring the sediment oxygen demand (SOD) and denitrification. SOD was higher in muddy sediment (Dong-Mak; three year average=$683;m^{-2}d^{-1}$) than sandy sediment(Yeocha; three year average=$457;m^{-2}d^{-1}$). The SOD was high in summer and tended to be lower in winter. During the sediment incubation in Apr. 2002, production of oxygen from sediment was observed implying active benthic photosynthesis. Denitrification was also higher in muddy sediment (Dong-Mak: $5.4;m^{-2}d^{-1}$) than sandy sediment (Yeocha; $3.4;m^{-2}d^{-1}$). The denitrification rate corresponds to the carbon remineralization rate of 9.3 and $5.9\;mg-C\;m^{-2}d^{-1}$ in Dong-Mak and Yeocha, respectively. The denitrification rates were lower compared to rates observed in other coastal area $(0{\sim}200\;{\mu}mole\;m^{-2}h^{-1})$. Although Kwanghwa tidal flat sediments are replete in organic matter, remineralization activity seems to be limited by the availability of labile organic matter. The Kwangwha tidal flat may have potential to effectively remove large load of organic matter. Net remineralization rates were 196 and $132\;mg-C\;m^{-2}d^{-1}$ in Dong-Mak and Yeocha, respectively.

A Study on the Production Structure and Biomass Productivity of Quercus variabilis Natural Forest (굴참나무천연림(天然林)의 생산구조(生産構造) 및 물질생산력(物質生産力)에 관(關)한 연구(硏究))

  • Growth and biomass production of natural stands of Quercus variabilis in relation to tree density were studied to obtain basic guide lines for future tending operation. Two natural stands of Quercus variabilis located at 900m (A stand: 6,600trees/ha, $15.84m^2/ha$, $\frac{19}{17-20}$) and 800m (B stand: 4,300trees/ha, $16.65m^2/ha$, $\frac{20}{17-21}$) elevation in Sancheong, Kyongnam Province were selected for the comparative study and following results were obtained through a sample plot method. After diameter of individual trees in the sample plots was measured, twelve average trees from each diameter class were cut felled to measure dry weight of $W_S$, $W_B$, $W_L$, $W_{Ba}$, and standing biomass and biomass production rates by a allometrior regressions related to $D^2H$. Vertical distribution of leaves along the stems indicated that photosynthesis was carried out 2.2m above the ground in Stand A and 1.2m in Stand B. Maximum photosynthesis was located 4.2m and 6.2m above the ground in Stand A and B, respectively. Leaf area index was 4.25ha/ha for Stand A, and 3.89ha/ha for Stand B. Above-ground standing biomass was 49.51 ton/ha for Stand A and 59.20 ton/ha and net annual production was 6.75 ton/ha/yr. for Stand A and 8.99 ton/ha/yr. for Stand B. The ratio of net annual production to standing biomass was 17.5% for Stand A and 16.7% for Stand B. Net assimilation rate was 2.75kg/kg/yr. for Stand A and 3.58kg/kg/yr. for Stand B. Stem wood production rate was 1.46kg/kg/yr. for Stand A and 2.09kg/kg/yr. for Stand B. Bark production rate was 0.60 kg/kg/yr. for Stand A and 0.34kg/kg/yr. for Stand B. Above data indicated that Stand B utilized growing spaces and sites more efficiently than Stand A. It is concluded chat productivity of natural stands of Quercus variabilis can be enhanced through optimization of basal areas and number of tree per hectare and that sound management of natural oak stands should be based on systematic sampling of the area for periodic productivity estimation.

Differences in Seed Vigor, Early Growth, and Secondary Compounds in Hulled and Dehulled Barley, Malting Barley, and Naked Oat Collected from Various Areas (맥종별 주산지와 재배한계지 수집종자의 활력, 초기생장 및 이차화합물 차이)

  • The purposes of this study were to determine how changes in temperature affect germination rates and growth of hulled and dehulled barley, malting barley, and naked oat plants, and to measure chlorophyll content, photosynthetic efficiency, and secondary compounds (total phenol, total flavonoid, and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity) in plants grown at 13℃ or 25℃). Various types of barley seeds were collected from areas with ideal conditions for barley cultivation, hereinafter referred to as IA, and also from areas where barley cultivation is more difficult due to lower temperatures, hereinafter referred to as LTA. Seeds were tested for seed vigor. While there were significant differences in the electrical conductivity values between seeds collected from certain specific areas, no significant differences were evident between IA and LTA seeds, regardless of the type of barley seed. When plants were grown at 25℃, there were no significant differences in germination rates, plant height, root length and shoot fresh weight between plants originating from IA and LTA. However, there were differences in the measured parameters of some specific seeds. Similarly, under the low temperature condition of 13℃, no differences in the emergence rate, plant height, and shoot fresh weight were evident between plants originating from IA or LTA, regardless of the type of barley. However, there were differences between some specific seeds. One parameter that did vary significantly was the emergence date. Hulled barley and malting barley emerged 5 days after sowing, whereas naked oats emerged 7 days after sowing. There were no differences in the chlorophyll content and photosynthetic efficacy, regardless of the type of barley. There were no significant differences in total phenol, total flavonoid content, and DPPH radical scavenging activity between plants originating from IA and LTA, regardless of the type of barley. However, there were differences between some specific seeds. In particular, for malting barley the total flavonoid content differed in the order of Gangjin > Changwon > Haenam = Jeonju > Naju. The results indicate that crop growth, yield and content of secondary compounds in various types of barley may be affected by climate change.

The Induction of Secondary Toxic Substances and Antioxidative Enzymes by $SO_2$ Fumigation in Foliage Plants (관엽식물에서 $SO_2$ 처리에 의한 이차독성물질 생성 및 항산화효소의 유도)

  • To study whether the secondary toxic substances such as ethylene and reactive oxygen species(ROS) are induced by air pollutants in foliage plants, $SO_2$ was fumigated to Pachira aquatica, Spathiphyllum patinii, and Hedera helix. $SO_2$ was controlled to $1\;{\mu}L/L$ and then fiumigated to plants for 2 days(8 hrs/day). It resulted in visible injury in P. aquatica and H. helix while no symptom appeared in S. patinii. Photosynthetic rate and water use efficiency were most remarkably reduced in P. aquatica compared to other two species whereas least in S. patinii. Two days after $SO_2$ fumigation, ethylene evolution was quantified to 23.56, 10.43 and 4.79 nL/g/h in P. aquatica, H. helix and S patinii, respectively. On the other hand, antioxidative enzymes were clearly activated by $SO_2$ treatment in all tested plant species implying ROS production. In conclusion, we could suggest that ethylene and ROS have been intimately related to the defense mechanism against $SO_2$ and their induction degree increased with plant susceptibility to $SO_2$. Furthermore, it was found that S. patinii was tolerant and P. aquatica sensitive to $SO_2$ on the basis of antioxidative enzyme activity and ethylene evolution.
