A review of the taxonomic and ecological characteristics of Korean mistletoe types (Viscum, Korthalsella, Loranthus and Taxillus) (한국산 겨우살이류(Viscum, Korthalsella, Loranthus and Taxillus)의 분류 및 생태학적 특성 고찰)

  • In general, studies of aerial parasitic plants known collectively as mistletoe have been carried out to investigate their ecological and agricultural characteristics. However, with the recently increased level of interest in medicinal resources, research on different types of Korean mistletoe has also increased. This study was carried out to review the work on the taxonomy and ecology of Korean mistletoe in preparation for the industrial use of these plants in the future. Mistletoe types are flowering plants belonging to Santalales, which exist in the form of parasites on the branches of trees or shrubs. In Korea, five taxa of four genera in two families of mistletoe exist: Viscum coloratum (Komarov) Nakai f. coloratum, Viscum coloratum (Komarov) Nakai f. rubroaurantiacum (Makino) Kitagawa and Korthalsella japonica (Thunb.) Engl. in Santalaceae, along with Loranthus tanakae Franch. et Sav. and Taxillus yadoriki (Sieb. ex Maxim.) Danser in Loranthaceae. As taxonomic studies of these species remain insufficient and given that the distribution ranges of these species are very wide, further observations pertaining to the morphological variations in each species are necessary. The distribution of mistletoes is known to be determined by the host specificity, the interval between the hosts, the environmental condition, the habits of the host plant, the eating characteristics of mediators in the area, and their habitat selection features.

Isolation of Endophytic Fungi Capable of Plant Growth Promotion from Monocots Inhabited in the Coastal Sand Dunes of Korea (사구에 서식하는 단자엽식물로부터 식물 생장 촉진 활성 내생 진균류의 분리)

  • Endophytic fungi predominantly inhabit grasses, and produce a variety of beneficial metabolites for plant growth, as well as help their hosts against pathogens and herbivores. Current study was focused on plant growth promoting activity of endophytic fungi inhabited in the roots of sand dune grasses. We collected 49 fungal isolates from the roots of four most common sand dune grasses and screened them for their growth promoting capacity. Results showed that 37 fungal isolates (75.5%) promoted plant height and shoot length of waito-c rice, 11 fungal isolates (22.5%) suppressed it, while 1 fungus (2%) showed no effect on the growth attributes. The fungal strain Gibberella fujikuroi, along with distilled water and Czapek broth medium, were taken as control for this experiment. It was concluded that a major proportion of endophytic fungi inhabited in the sand dune plants produce metabolites, and thus help in growth and development of the host plant.

Isolation of Pea and Soybean Nodule Bacteria and Assessment of their Nitrogen-fixing Capacities (완두 및 대두근류균(根瘤菌)의 분리(分離) 및 질소고정능력(窒素固定能力)의 비교(比較))

  • A series of experiments over three years was planned for practical application of rhizobia in farms and grass lands in Korea. This is the report for the studies of the first year mainly on the isolation and characterization of rhizobial strains, and on the assessment of their nodulation abilities and nitrogen fixation capacities. 1. Total number of 88 strains for soybean group and 22 st ra ins for pea and vetch group was isolated from nodules which were taken from legumes grown in Daekwanyong, Cheju and various places in Korea. 2. Morphological and cultural characteristics of the strains were studied, and attempts were also made to investigate their antigenic properties and to demonstrate lysogenic strains in these groups. The results were : i) the isolates varied in cultural characteristics on yeast mannitol broth and agar, and in degree of congo reel absorption ; ii) similarities in their antigenic properties were found between/among the strains: G-3/G-9/D216, G-20/G-52 in soybean group; iii) no lysogeny was found in the strains of these groups. 3. Plant infection tests by test tube and bottle method in light room were carried out to elucidate the ability of the strains to nodulate specific legumes and of the capacity of such nodules to fix atmospheric nitrogen. The isolates were grouped into non- invasive, ineffective, or effective to the legumes. Those strains which produced effective nodules, supporting similar level of growth as nitrate control, were: P-3, 4 and 8 in pea and vetch group; G-23, 27, and, D-216 in soybean group.

The Effect of Root Exudate and Chemotaxis on Host Recognition in Soybean-Bradyrhizobium Symbiosis (대두(大豆)-근류균(根瘤菌) 공생(共生)에서 뿌리분비물(分泌物)과 화학주성(化學走性)이 숙주인식(宿主認識)에 미치는 영향(影響))

  • This study was carried out to research the effect of the chemotaxis of Bradyrhizobium japonicum KCTC 2422 and its mutant toward soybean root exudate and to elucidate the effect of the lectin of host specificity (Host Recognition) in soybean-Bradyrhizobium symbiosis. The results obtained were as follows: The homogeneities of the purified lectins from soybean and pea seed was ascertained chromatographically and electrophoretically. Gel electrophoresis of soybean lectin in the presence of sodium dodecyl sulfate appeared a single protein band, whereas pea lectin appeared two protein bands. Soybean lectin from 2 cultivars formed immunoprecipitin arcs at same position with anti-soybean lectin rabbit IgG, but pea lectin did not form immunoprecipitin lines with anti-soybean lectin rabbit IgG. Chemotactic responses of KCTC 2422, LPN-100 and LCR-101 toward proline in capillary assays were 3.1, 1.3 and 1.0-fold above background, respectively. The chemotactic responses of KCTC 2422, LPN-100, and LCR-101 toward Paldal crude root exudate in capillary assays were 3.5, 1.4 and 1.4-fold above background, respectively. The present work shows that B. japonicum and its mutants are capable of very different responses toward root exudate fraction. The chemotactic responses of KCTC 2422 was most with neutral fraction, least with anionic fraction and intermediate with cationic fraction. The nitrogenase activity of soybean nodule was shown in 15days after inoculation with LCR-101. However, we couldn't find out the nodules when soybean was inoculated with LPN-100. From these result we can suppose that the chemotaxis of Bradyrhizobium plays inportant the role of forming the nodule (host recognition) in the soybean-B. japonicum symbiosis.

Form and Embryonic Characteristics of Pedicularis hallaisanensis Seeds As Endangered Wild Species II-Class Using Host Plants (숙주식물을 활용한 멸종위기야생식물II급 한라송이풀 종자의 형태 및 발아특성)

  • This study was carried out to investigate the possibility of establishing a reproductive system for the seed of Pedicularis hallaisanensis, which is in the endangered wild species II class in Korea. The seed of P. hallaisanensis is egg-shaped, and the seed coat is dark brown. The embryo was identified as a dwarf type by the seed section. The seed length was $0.47{\pm}0.07mm$, width $0.16{\pm}0.006mm$, and thickness $0.12{\pm}0.01mm$. The weight of one seed was $0.0003{\pm}0.0001mg$, and 1000 seeds weighed $4.59{\pm}0.02mg$. The degree of seed viability was 75.33% by the tetrazolium (TZ) assay. The highest germination rate of P. hallaisanensis seed was 71% after 4 weeks of storage at $4^{\circ}C$. However, the germination rate tended to decrease gradually over a longer storage period. The germination rates after 6 or 8 weeks of storage at $4^{\circ}C$ were 64% and 60%, respectively. We used two host plants, Artemisia princeps and Dendranthema zawadskii, to determine the effect of host plants on P. hallaisanensis seed germination. The germination of P. hallaisanensis mixed with A. princeps or D. zawadskii started at 53.5 and 62.5 days after sowing, respectively. We did not find any germination 164 days postsowing with both host plants. When A. princeps and D. zawadskii were used as host plants for P. hallaisanensis seed germination, P. hallaisanensis seed germination rates were 45.5% and 19.5%, respectively. The average time to germination was 70.2 days for A. princeps, and 46.8 days for D. zawadskii.

Fluorometric Measurement of Acid Phosphatase Activity in the Angiosperm Parasite (Cuscuta cephalanthi) and its Host (Hedera helix) (기생피자식물(Cuscuta cephalanthi)과 그 숙주(Hedera helix)에서 acid phosphatase활동의 형광분석법에 의한 측정)

  • Acid phosphatase activities were analyzed in $\mu\textrm{g}$ tissue samples from an angiosperm parasite (Cuscuta cephalanthi) and its host plant (Hedera helix) by a fluorometric microtechnique. The apex and the coiling portion of the parasite axis exhibited greater enzyme activies than other portions of the hypha. Acid phosphatase activity in the haustorium was 2-3 times that in the hyphal axis. The vascular bundles of the normal host exhibited the greatest enzyme activity. The acid phosphatase activity in the host infected by the parasite decreased to the activity level of the haustorium.

Flying Aphid Population at the Horticultural Experiment Station, Suweon (원예시험장 주변의 진딧물)

  • This survey was aimed to accumulate basic data of aphid population at the Horticultural Experiment Station at Suweon. The yellow pan traps were setted at five locations (Fig.1.), and ran from May 1 to October 31. 1970. About one hundred and twenty species of aphids were trapped, including 24 species of plant vims vectors. Of these, dominant species were as follows: (Asterisk shows virus vector) Aphid species No. of catches * Aphis spiraecola PATCH 2,635, * Aphis craccivora KOCH 2,377, * Myzus persicae SULXER 2,111, Capitophorus hippophaes javanicus H.R. LAMBERS 2,051, Anoecia fulviabdominalis SASAKI 1,480, * Aphis gossypii GLOVER 867, * Macrosiphum avenae FABRICIUS 859, Cervaphis quercus TAKAHASHI 692, * Lipaphis erysimi KALTENBACH 645, Pleotrichophorus chrysanthemi THEOBALD 489, The above 10 species consisted $76.5\%$ of total catches and the 24 vector species consisted $55.5\%$. The curve of the seasonal occurrence of flying aphids at Horticultural Experiment Station shows bimodal, typical for the temperate region. The total number of trapped aphids at the Station from May to September, 1970, were less than that of average yearly catches at the College of Agriculture from 1967 to 1970. Thi, low numbers at Horticultural Experiment Station may attribute to the frequent spraying of insecticides from Spring to Summer on growing crops there. But the aphids population increase suddenly in the middle of October. This might be resulted from cease of insecticide applications and migration of aphids from summer host to winter host plants.

Structure Elucidation of New Cochlioquinol Derivatives from Pathogenic Fungus Bipolaris cynodontis (식물 병원균 Bipolaris cynodontis로부터 분리한 새로운 Cochlioquinol 유도체의 구조 분석)

  • Three active compounds were isolated from the culture of a plant pathogenic fungus, Bipolaris cynodontis. The structure elucidation of these compounds was accomplished by 2D NMR techniques, such as $^1H-^1H$ and $^{13}C-^1H$ COSY, COLOC, HMBC and rotating frame NOE(ROESY). Compounds were found as derivatives of cochlioquinone and cochlioquinol that were previously isolated as phytotoxins from B. bicolor and B. cynodontis, respectively. The compounds showed phytotxicity against Italian ryegrass, one of the host plants of B. cynodontis.

Ecological Study on Arbuscular Mycorrhizae(AM) at Coastal Reclaimed Lands (해안(海岸) 간척지(干拓地)에서 Arbuscular Mycorrhizae(AM)에 관한 생태학적(生態學的) 연구(硏究))

  • The symbiotic activities of arbuscular mycorrhizal fungi(AMF) such as spore density, symbiotic intensity and vesicle density, phytomasses of higher plants such as Calamagrostis epigeios, Imperata cylindria, Artemisia scoparia, Aster tripolium and Sonchus brachyotus and seasonal change of the AMF activities, electric conductivity and zinc contents in plant and soil were determined in the rhizospheres of higher plants at abandoned old coastal reclaimed lands, where constructed in 12 and 30 years ago. If plants of reclaimed land classified to salinity, symbiotic activities of AMF were high in order of obligate halophyte, facultative halophyte and glycophyte. Also, those plants classified to life form, symbiotic activities of AMF were high in order of annual, biennial and perennial plants. Seasonal variation of spore density, one of symbiotic activities showed that the plateau density maintained continuously from the end of growing season of the higher plants to next spring. For this reason, it regarded that reproduction of AMF spore would be formed in autumn, when the higher plants will be developed. Seasonal change of symbiosis intensity, other symbiotic activities, however, showed that the highest symbiosis intensity occurred in spring and summer but the lowest in autumn. In relationships among symbiotic activities, spore density was directry proportional increase of symbiosis intensity. Moreover, phytomass of higher plants also was directly proportional to increase the spore density as well as symbiosis intensity. Vesicle density, however, did not any correlation with the phytomass, spore density and symbiosis intensity. From these results, it can know that both spore density and symbiosis intensity are strongly possible to use as the measure of symbiotic activity owing to symbiosis of tho-AMF, the more absorption of zinc by the higher plants carried out the less concentration of zinc in the soil.

Chloroplast Genetic Transformation in Higher Plants: An Encounter Between Prokaryote and Eukaryote (고등식물의 엽록체 형질전환: 원핵생물과 진맥생물의 조우)

  • Chloroplasts are believed to be descended from certain cyanobacteria, which were taken up by phagocytosis into a host cell and lived there in a symbiotic relationship. In contrast to the current static concept on the chloroplast genome, its dynamism has been recently demonstrated: the chloroplast genome is active in intramolecular homolgous recombination, producing subgenomic circles when it obtains homolgous sequences via genetic transformation. Chloroplast tranformation in higher plants provides many advantages over nuclear transformation that include higher expression levels of transgenes, polycistronic expression of transgenes, and maternal transmission of transgenes. Tobacco has been used as a model for chloroplast genetic transformation. However, it is recently possible to transform the chloroplasts of other major food and economic crops including rice, soybean, and cotton. Chloroplast-transformed crops will be able to replace bioreactors using microorganisms for production of value-added proteins in future.
