Variation of Leucine Aminoeptidase Isozyme in Korean Land Races and Wild Soybeans (한국 재래 및 야생종 콩의 Leucine Aminopeptidase 변이)

  • A total 943 accession of soybeans (G. max) and 50 wild soybeans (G. soja) were examined for leucine aminopeptidase (LAP) isozyme variation by 5% polyacrylamide gel electrophoresis(PAGE) and isoelectric focusing(IEF) of pH 4~6.5. The Lap1*b by PAGE was the most common phenotype in both G. max and G. soja. The frequency of Lap1*b allele was observed to be higher in G. max(1.00) than in G. soja(0.96) of Korea. This result shows that G. max is fixed for Lapl*b allele at the Lap1 locus. LAP isozyme band type I and II were found using IEF of pH 4~6.5 in G. max and G. soja of Korea. Type I was observed from 92.8% in G. max and 92.0% in G. soja, and type II was discovered in 7.2% G. max and 8.0% G. soja. This result suggested the possibility to be found more various band types.

Seed Characteristics of Black Soybean Collections in Korea (수집재래 검정콩의 종실특성)

  • The improvement of superior black soybeans could be accomplished through maintenance and use of black soybean collections. A collection of 965 black soybean lines currently grown by farmers was made in 197 locations on spring, 1991. This study was carried out to evaluate them as useful germplasm. Seed characteristics of black soybean collections were evaluated for black soybean breeding for seed quality. The 100-seeds weight of 929 black soybean genotypes were distributed from 6.9 to 48.1 gram. The maximum and minimum 100-seeds weight were derived from Chungnam and Jeonbuk province, respectively. l00-seeds weight was heavier in middle and late maturity group, as maturity was later, whereas no tendency was shown in early maturity group. Length, width and thickness of the collected black soybean seeds were 9.5$\pm$1.5mm, 7.3$\pm1.3mm and 7.3$\pm1.2mm, respectively. Black soybean collections with yellow seed embryo was 72% and the others were green seed embryo.

  • Artemisw spp. have been known as natural health foods from ancient time and recently, Artemisia spp. have been developed as a preference food. In the paper Artemisia spp. collected from 28 local areas in 1993 were growth in condition shaded selected some suitable species for summer cultivation. Growth characteristics and chemical components of shade-cultivated were summerized by follows. 1. The volume per $m^2$ was 3.925g collected in Suncheon. Jeonnam which was most abundant, and 713g in Okcheon. Chungbuk was little. and there was difference at 5.6times. The volumes per $m^2$ collected in species were plentiful of 3,420g in Hwanghaessak, 3,173g in Meongcheonssuk and 3.l48g Siljebissuk. But there were little of 2,093g in Panssuk and 2,078g in Keunjebissuk. 2. Hwanghaessk, Siljebissuk and Meongcheonssuk were good seasoning, and leaf color was light green. and also comodities was show a good result. 3. Hwanghaessuk, which was excellent quality and quantity, was include many iron and magnesium, but content of copper was low. Meongcheonssuk and pangssuk were excellent in comodities. and content of protein was abundant, but contents of catcalcium, iron, and zink were low. 4. Hwanghaessuk and Siljebissuk were a great many quantity and taste, leaf color, quality and comodity were excellent. Therefore Hwanghaessuk and Siljebissuk were most suitable cultivated in shading the light.

Classification of Garlic Germplasms Based on Agronomic Characteristics and Multivariative Analysis (마늘 유전자원의 작물학적 특성과 다변량 분석에 의한 품종군 분류)

  • This research was conducted to investigate the genetic diversity and select useful accession with agronomic characteristics of garlic (Allium sativum L.). germplasms at Garlic Research Institute in Chungbuk Agricultural Research and Extension Service. Morphological diversity and relationships among 160 germplasms collected from 26 countries were assessed by methods of clustering and principal component analysis. Among 11 types of leaves and bulbs characteristics, emergence days of leaf showed the highest variation with coefficient of variation of 84.8%, and the bulb weight and the number of scales showed higher variability with 24.3%. Correlation analysis based on 11 quantitative traits showed that bulb weight and bulb length have very high positive correlation with bulb quantity. Plant height, leaf length, and number of leaves showed positive correlation with bulb weight as collections with better performance in growth produced large bulb with higher quality. The cluster analysis based on 5 principal components generated 6 clusters with an average distance of 1.6 among clusters. Domestic genetic resources were the largest with 36 species (22.5%) in group II.

Report on the Genus Cordyceps and Its Anamorphs Collected in Korea (한국자생 동충하초의 채집 및 자원보고)

  • Morphological characteristics of Cordyceps species and its allies collected in Korea were clarified. Through the survey conducted from June 1999 to October 2002 in 19 mountains in Korea, 667 samples of entomogenous fungi were collected. Cordyceps and its allies of 17 species of 5 genera were identified as Cordyceps gracilioides, C. japonica, C. longissima, C. martialis, C. militaris, C. myrmecophila, C. nutans, C. pruinosa, C. sphecocephala, C. tricentri, Hirsutella nutans, Paecilomyces cicadae, P. farinosus, P. tenuipes, Paecilomyces sp., Shimizuomyces paradoxa, Tilachlidiopsis nigra. The fungi with insect hosts have been collected mainly in the place of shade or mosses near brooks and streams that had high humidity. Overall the frequenct of fungal infection in natural ecosystem was relatively low as few as 10 collections per each species. However, many species were found in terms of the few number of colleciton sites with seasonal limitations. Occurrence of the fungi in Jeju island remote from inland of the Korean peninsula were diverse in their species due to the varied weather of vertical distribution following the altitude. Three most common species were C. nutans, P. tenuipes and C. militaris, mainly found early in August when the relative humidity and temperature were high, of which C. nutans occupied the highest frequency consisting of 65% in total collections. Neither variation in ascomata arrangement in stromata nor developement of secondary spores was recognizable, while the number, shape and colour of stromata varied with insect hosts and weather conditions.

Comparison of Genetic Diversity of Saxifraga Species Distributed in the Arctic Svalbard and Korea (북극권 Svalbard 지역과 한국에 분포하는 Saxifraga 속 식물의 유전적 다형성 비교)

  • The species in genus Saxifiraga distributed in circumpolar arctic are taxonomically difficult to study. RAPD analyses were performed to compare the genetic diversity of the 16 Saxifrages originated from the Norwegian Arctic Svalbard and Korea. The 12 accessions of URP primers were tested and 4 of which showed polymorphism were selected. Total 79 (44.8%) DNA bands were scored and analyzed by UPGMA cluster analysis. The results indicated that all of the 9 Saxifraga species from Svalbard showed high genetic diversity than those from Korea. The Similarity matrix and cluster analyses indicated that the Saxifraga species from Svalbard and Korea can be divided into two different subgroups. RAPDs of the Saxifraga species of Korea showed higher homologous patterns than those of Arctic Saxifrage. Among the Saxifraga species, we found that the morphological similarity reflects the genetic similarity. The geographic distance, clonal reproduction, and environmental condition may contribute the high level of genetic diversity between Saxifraga species from the two isolated regions.

Characterization of Agronomic Traits and Comparison of Growth Characteristics by Different Cultivation Seasons in Buckwheat Germplasm (메밀 유전자원 특성평가 및 재배시기별 생육특성 비교)

  • 메밀은 마디풀과(polygonaceae)의 메밀속에 속하는 일년생 초본으로 야생종을 포함하여 20여 종이 확인되고 있다. 현재 재배되고 있는 메밀 재배종은 보통메밀(Fagopyrum esculentum Moench)과 쓴메밀(Fagopyrum tataricum Gaertn., 타타리메밀) 등 두 종이 주류를 이루고 있으며 우리나라에서는 보통메밀이 주로 재배되어 왔다. 본 연구는 농업유전자원센터에서 보유중인 보통메밀 341점과 쓴메밀 38점을 봄, 여름에 파종하여 특성조사를 수행하고 재배시기별 생육특성을 비교하기 위해 수행되었다. 봄, 여름 파종은 각각 2017년 4월 14일과 8월 17일에 전주 소재 농업유전자원센터 포장에 하였으며 줄기색, 엽색, 엽형, 개화기, 성숙기, 꽃색, 종피색, 종자모양, 주경절수, 총분지수, 백립중 등 11개 농업형질을 조사하였다. 봄 파종시 개화기는 대부분 5월 25일 전후였으며 성숙기는 6월 말부터 7월 중순까지 분포하였고 여름 파종시는 9월 15일 전후 개화하여 10월 말에 성숙하였다. 줄기색은 홍색의 비율이 높았고 엽색은 연녹색과 녹색이 대부분이었으며 엽형은 화살촉형이 90% 이상이었다. 꽃색은 흰색이 대부분이었지만 연녹색 자원도 40점 조사되었으며 종피색은 연갈색이 70%를 차지하였다. 종자 생산과 관련된 주경절수와 총분지수는 봄 파종과 여름 파종에서 많은 차이를 나타냈으며 봄 재배시 영양생장이 왕성하여 가을 재배보다 많은 주경절수와 총분지수가 조사되었다. 그러나 백립중에서는 여름 파종이 봄 파종보다 무거운 경향을 나타내어 주경절수, 총분지수와 백립중 간 부의 상관관계를 추론할 수 있었다. 일반메밀 유전자원을 국내 수집지별로 나누어 조사한 결과 주경절수와 총분지수는 봄, 여름 파종 모두에서 전남 수집자원이 가장 많았으며, 강원, 경남, 경북, 전북, 충북 수집자원과 유의성있는 차이를 나타내었다. 강원, 충북 수집자원은 제일 낮은 주경절수와 총분지수를 보였으나 백립중은 가장 높게 조사되었다.

  • 본 연구에서는 인터넷에 존재하는 다양한 XML 문서에서 의미정보를 검색하기 위한 의미정보수집 이동에이전트 시스템을 개발하였다. 이동에이전트는 실행 프로그램이 이기종 분산 환경의 네트웍에서 자율적으로 이동 및 반응하며 실제로 데이터가 존재하고 있는 장소로 이동하여 목적을 수행한다. 의미 정보수집 이동에이전트 시스템의 연구를 위하여, 정보를 개념화하고 포괄적 DTD를 자동으로 생성할 수 있는 DTD생성기를 개발하였으며 의미정보를 추론할 수 있는 추론알고리즘을 연구하였다. 개발된 의미정보수집 이동에이전트 시스템은 정보가 존재하는 원격지 사이트에 파견되어 비정형 XML 문서를 대상으로 포괄적 DTD와 추론엔진을 이용하여, 의미정보를 추출하고 전송하는 임무를 수행한다. 따라서, 의미정보수집 이동에이전트 시스템을 이용한 정보수집은 정보의 질을 향상시키고 네트워크의 부하를 감소시킨다.

Genetic Variation of Cytochrome P450 Genes in Garlic Cultivars (마늘유래 Cytochrome P450 유전자의 변이 분석)

  • Wound inducible P450-Esg cDNA, one of cytochrome P450 gene family, was isolated from shoot of Euiseong garlic cultivar. P450-Esg cDNA possesses highly conserved heme-binding domain in the nucleotide sequence, and 1,419 bp of open reading frame (ORF) coding of 473 amino acids. Based on the nucleotide sequence analysis of P450-Esg homologous from twelve garlic cultivars, two domains, one domain between 472 to 510 bp, and the other between 1,210 to 1,249 bp from start codon (ATG), showed various nucleotide polymorphism among cultivars. Sequence of heme-binding domain in P450-Esg homologous, which is located at the domain between 1,210 to 1,240 bp from start codon, showed various nucleotide polymorphism as well as amino acid sequence polymorphism among twelve garlic cultivars. Anther domain, between 472 to 510 bp from start codon, showed exactly same amino acid sequence in the twelve garlic cultivars, but there were various single nucleotide polymorphism to the cultivars.
