Analysis Genetic Similarity of Gentiana scabra var. buergeri by Randomly Amplified Polymorphic DNA (RAPD를 이용한 용담의 유전적 유사도 분석)

  • Randomly amplified polymorphic DNA(RAPD) analysis was applied to detect the molecular polymorphisms in Gentiana scabra var. buergeri. A high level of molecular variability was found among wild plants and cultivars. In genetic analysis, eight of twenty primers were selected and 54 amplification products ranged 2. 2 to 0.2 kb were compared. Twenty - nine amplified products showed polymorphic, while five were monomorphic. Twenty of line specific bands were found. In genetic similarity and cluster analysis using PCR products, three wild plants collected from Naejangsan, Daedunsan and Keojedo and one cultivar Seochunjaerae were grouped into one cluster, while cultivar Jinbujaerae and Japanese line separated into another clusters, respectively. The identification of DNA polymorphisms by the RAPD technique will facilitate the selection of the lines from different origin.

Study on Physiological and Ecological Characteristics of Collective Varieties on Elephant food ( Amorphophallus Konjac K.) (구약감자 수집종의 생리생태적 특성)

  • This research was performed to understand physiological and ecological characterisities and to de stable production by the way of the establishment of cultivation in elephant food. The results of the investigation of developmental characterisitics, leaf area, photosynthetic ability, chloroplast content for collective varieties are as follows. In the ecological characterisrics for five varieties of elephant food collected from domestic and foreign countries, appearance days for Japanese collective varieties was 60 days, which was two days earlier than domestic collective variety, Jechon collective variety. In appearance rate, Japanese collective variety was 90%, which was higher than Kumsan collective variety, 85%. In the development and yield of the ground portion, Japanese variety was the highest during all developmental period follwed by Chinese and Jechon collective varieties. The larger leaf area, the higher photosynthesis was found. In the amount of chlorophyll content, the higher intercepting light rate, the higher chlorophyll amount was found, which was 30, 50, 70% higher amount than non intercepting light rate.

  • Agromonia pilosa Ledeb. has been used as a medicinal plant in traditional folk remedy. There are few reports on classification, physiology, ecology and morphological studies of Agromonia pilosa L. in Korea. Therefore, advanced approaches on study and development with this plant would be done urgently. Present stndy was carried out to gain basic information on genetic resources and variation with collected domestic landraces through RAPD analysis in Agromonia pilosa L. Forty two collections of Agromonia pilosa L. from nation-wide area including USA one were analyzed by RAPD test. Molecular marker size by amplified DNA band pattern ranged from 300 to 2,100bp. Among the collection, two landraces of Hadong and Cheonghak-dong showed close relation in genetic similarity. Minimum and maximum value by matrix of 1-F among 26 collected landraces were figured out as 0.365 and 0.827 showing mean value for 0.624, respectively. Those landraces were classified into two groups with cluster analysis by Nei and Li's formula from RAPD-analyzed values, and considerable genetic differences were recognized between two groups.

Comparison of the Agronomic Characteristics of Perilla (Perilla ocimoides L.) Collected from Korean and Chinese Jilim Province (한국과 중국 길림성 재래종 들깨의 작물학적 특성 비교)

  • This study was conducted to evaluate the major agronomic characteristics Korean of 85 and Chinese Jilim Province of 5 local varieties of perilla at Chungbuk Agricultural Research and Extension Services in 2001. Perilla accessions examined were classified into three maturity groups, i.e, early, medium, and late maturity group of $19\;(23\%),\;57\;(67\%)$, and $9\;(10\%)$ accessions, respectively. The early and late maturity groups mainly consisted of collections from middle-northern area and southern area, respectively, while the medium group consisted of accessions from all over the country. But chinese Jilim collection wad all mediumgroup. Average 1,000-seed weight(TSW) was 2.7 g. TSW of the collection from Hamyang was the greatest a 3.9g, while the TSW of Pyungchang collection was the smallest as 1.7g, and most of collections produced medium and small seeds. Perilla accessions with greate 1,000-seed weight seemed to be belonged to the late maturity group. Chinese collection was 2.2 to 3.19g and overage was 2.6g in average. Accodingly TSW hadn't more variegation than Korean. Seed coat colors of gorilla accessions were dark brown$(30\%)$, brown$(55\%)$ and gray brown$(6\%)$, respectively. Among seed coat colors, brown color consisted of $91\%$. Chinese was dark brown$(20\%)$, brown$(80\%)$ in color. Positively correlated that stem height, the number of nodes, branches and flower clusters with Korean and Chinese collection, capsules per flower cluster and the length of flower cluster with Korean collection and 1,000-seed weight and maturing period, flower clusters, stem height and the number of nodes with Chinese collection. And negatively correlated that the number of capsules per flower cluster and 1,000-seed weight with Korean collection and the length of flower cluster, stem height, branches, the number of nodes and flower clusters with Chinese collection.

  • This study was carried out to obtain a basic informations for the improvement of human health and the development of variety through analysis of inorganic compounds, contents of amino acids and saccharids to three elephant- foots (Amporphophallus Konjac. K) collected from Kumsan, Japan and China. Mannan as a carbohydrate of major component of an elephant-food was slightly high in Kumsan than that of Japan and China variety. Degree of viscosity of an elephant-foot depends upon the soluble free sugar contents and amino acid, contents of these free sugars were high in the order of Kumsan, China and Japan elephant-foot powder. Results of analysis of inorganic compounds to an elephant-foot used; Kumsan variety was shown to be high than those of China and Japan, especially. K, P and Fe in Kumsan variety were high, while Na and Ca were appeared to be high in China elephant-foot.

Variation analysis of Atractylodes japonica Koidzumi ex Kitamura Based on Quantitative Characters (삽주(Atractylodes japonica Koidzumi ex Kitamura)의 정량적 형질에 의한 변이분석)

  • Sixteen Atractylodes japonica Koidz. accessions were collected in Jeonbuk, Kyeongnam, Kangwon and Kyeonggi province, and their quantitative characteristics were compared with each other. The relationship of variation between Atractylodes japonica Koidz. accessions was also investigated by means of multivariate analysis. Hamyang 1 accession had the most desirable quantitative characteristics among the accessions: plant height was 86.2cm, number of leaves, 117.2 and number of stems, 34.3. Sudo 1 accession had 12 stems, the most among the accessions. The length of leaf margin serras found in all of them was on average 1.6mm. Most accessions had simple or palmately compound leaf with oval or elliptical type of the leaf shape. The accessions could be classified into three groups by average linkage cluster method. The most remote relationship was found between Group 1 containing Jinan 1, Jinan 2 and Group 3 containing Jeongsu, and Geoje.

Agronomic Characteristics and Path-coefficients of Safflower (Carthamus tinctorius L.) Collections (잇꽃 수집종의 주요특성과 경로계수 분석)

  • This study was carried out to obtain the basic information on mating parents and selection for high yielding safflower breeding. Thirty nine world safflower lines, which had collected from 13 countries of origjn, were evaluated for major agronomic characters, correlation and path-coefficients. The results obtained were summarized as follows. Days to flowers ranged from 83 days to 100 days. Days to flower of most foreign lines were later than that of domestic lines. Plant height ranged from 75 cm to 162 cm. The plant height and stem diameter of most foreign lines were higher than those of domestic lines. The leaf number and size of domestic lines were much more than those of foreign lines. The total branch numbers of foreign lines were more than domestic lines. Particularly, the number of third branch ranged from 0 to 25 and there were none in domestic lines. The number of corolla of foreign lines was much more than that of domestic lines. The yield of foreign lines collected from Central Asia and United States of America were more than any others. Significantly positive correlations were observed between yield and days to flower, stem diameter, the number of branch and the number of corolla. In the analysis of path-coefficient, the highest direct effects on yield was the number of corolla followed by the number of total branch, whereas those of the number of second branch and stem diameter were negative.

Analysis of Genetic Diversity in Echinochloa Species Using Random Amplified Polymorphic DNAs(RAPDs) Markers (RAPD Marker를 이용한 피 수집종의 유연관계 분석)

  • Echinochloa species maintained by selling for more than 10 years were classified using random amplified polymorphic DNAs(RAPDs) analysis. Seventy-four decamer of randomly sequence markers were used to classify intraspecific variation irt Echinochloa species. The number of amplification products increased with increasing GC content of the primer in the range between 60% and 70% GC. Single-base substitutions of a primer altered amplification, providing new polymorphisms. The size of amplified DNA was mostly between 0.40kbp and 1.4kbp with the most common bands at 1.1kbp. Echinochloa species were detected with 6 primers which generated 26 polymorphic amplified DNAs. By hierarchical cluster analysis, Echinochloa species collected in Korea were divided into three groups. These results revealed that RAPD markers are useful tools for the determination of genetic variations in Echinochloa species.

Analysis of Major Traits for Native Ginseng(Panax ginseng C.A. Meyer) Collected from Poonggi Area in Korea Using DNA Marker (DNA marker를 이용한 풍기 재래인삼의 주요 특성 분석)

  • In this study, a total of 25 lines consisting of five native ginseng collections from Poonggi area, five lines from Geumsam area and 15 varieties were analyzed and clustered for the selection of Poonggi native ginseng in Korea using DNA markers. The results indicated that the long cluster distance were observed between the collections of 331002, 331004, 331005, 331007 and 331026 from Poonggi area, and the collections of 332009, 332021, 332046, 332050 and 332066 from Geumsan area because of the sensible differences on the number of leaves per stem, stem color and petiole color. Thus, the collections from Poonggi area with specific characters consisting of one stem per plant, five leaves per stem and broad elliptic leaflet shape were finely classified using nine primers including OPD05, OPD20, OPG17, OPH05 and so on. In this study, the collection of 331007 from Poonggi area was considered as the respective collection with above characters. Thus, the nine primers such as OPD05 and so on, will be used to select the Poonggi ginseng in the future studies.
