  • The present study was performed to develop a functional raw food material from hydrolyzed whey protein powder (23%-GNANA) medication containing sialic acid as a marker compound that is naturally occurring at 7% concentration in GMP (glycomacropeptide). GMP is used worldwide in foodstuffs for babies and infants and is obtained from the milk protein as safe food. While the purpose of our detailed evaluation was aimed to assess preliminary NOAEL values for and above 2,000 mg/kg/day, a clinical dose allowance for 23%-GNANA (as per characteristic of a functional health product, a highly refined test substance of 23% (v/v) sialic acid combined in GMP), at the same time we also wanted to assess the safety of GMP hydrolyzate lacking sialic acid but with identical properties as GMP. Animal safety evaluation was conducted using 23%-GNANA as the test substance, produced from hydrolyzed whey protein powder (product name: HELICOBACTROL-23; provided by Medinutrol Inc. [Korea]; composed of 23% sialic acid and GMP protein) after isolating the sialic acid using enzymes approved as food additives, with GMP as a raw material, and subsequently increasing the content of xx up to 23% through 80% (v/v) ethanol soaking and concentrating, in accordance with GLP Guideline. The animal safety evaluation mentioned above was made on the basis of toxicity in SPF Sprague-Dawley female and male rats dosed with 10 mL of the test substance diluted to 0, 1,250, 2,500, and 5,000 mg/kg directly into their stomachs for 90 d. This was determined in terms of the general symptoms and animal viability, weight and amount of feed intake, eye examination, uracrasia tests, hematological and blood biochemical disorder tests, blood coagulation test, abnormal intestine weight, abnormalities during postmortem and histopathological examinations. Statistical significance was set at P<0.05. Based on the toxicity determination, a certain minor effect associated with the test substance was observed in male rats with no major effects of the tested substance, in comparison with the control group dosed with sterilized water. Nevertheless, the NOAEL value, evaluated as per toxicity criteria, was verified as 5,000 mg/kg/day (P<0.05). Similarly, for female rats, a certain minor effect associated with the test substance was observed in 5,000 mg/kg/day dosed group, with no major effect, yet the NOAEL value (as assessed as per toxicity criteria) was determined to be 5,000 mg/kg/day (P<0.05), which was the same as for male rats. Accordingly, the NOAEL values of the test substances for all female and male rats were finally verified as 5,000 mg/kg/day (P<0.05). In conclusion, it was determined that the 23%-GNANA test substance exceeds 2,000 mg/kg/day, the clinical allowance characteristic for functional health food, and was finally evaluated to cause no safety concerns when used as a raw material in functional health food production, which was the ultimate goal of the present study.

Effects of Temperature and Irrigation Intervals on Photosynthesis, Growth and Growth Analysis of Pot-grown Cucumber Seedlings (온도와 관수 주기가 오이 포트 묘의 광합성, 생육 및 생장 해석에 미치는 영향)

  • This study was conducted in an indoor cultivation room and chamber where environmental control is possible to investigate the effect of temperature and irrigation interval on photosynthesis, growth and growth analysis of potted seedling cucumber. The light intensity (70 W·m-2) and humidity (65%) were set to be the same. The experimental treatments were six combinations of three different temperatures, 15/10℃, 25/20℃, and 35/25℃, and two irrigation intervals, 100 mL per day (S) and 200 mL every 2 days (L). The treatments were named 15S, 15L, 25S, 25L, 35S, and 35L. Seedlings at 0.5 cm in height were planted in pots (volume:1 L) filled with sandy loam and treated for 21 days. Photosynthesis, transpiration rate and stomatal conductance at 14 days after treatment were highest in 25S. These were higher in S treatments with a shorter irrigation interval than L treatments. Total amount of irrigation water was supplied evenly at 2 L, but the soil moisture content was highest at 15S and lowest at 25S > 15L > 25L, 35S and 35L in that order. Humidity showed a similar trend at 15/10℃ (61.1%) and 25/20℃ (67.2%), but it was as high at 35/25℃ (80.5%). Cucumber growth (plant height, leaf length, leaf width, chlorophyll content, leaf area, fresh weight and dry weight) on day 21 was the highest in 25S. Growth parameters were higher in S with shorter irrigation intervals. Yellow symptom of leaf was occurred in 89.9% at 35S and 35L, where the temperature was high. Relative growth rate (RGR) and specific leaf weight (SLA) were high at 25/20℃ (25S, 25L), RGR tended to be high in the S treatment, and SLA in the L treatment. Water use efficiency (WUE) was high in the order of 25S, 25L > 15S > 15L, 35S, and 35L. As a result of the above, the growth and WUE were high at the temperature of 25/20℃.

The influence on cultivation characteristics of winter mushroom by carbon dioxide concentration (탄산가스 농도 변화가 팽이 버섯 재배에 미치는 영향)

  • This study was performed to determine the optimal concentration of carbon dioxide, which effects mushroom growth and yield. It was shown that the periods for fruiting initiation, growth and harvest of Flammulina velutipes were increased when the $CO_2$ concentration was raised. In general, those characteristics were less affected in brown strains than in white ones. Especially brown strain ASI4103 was susceptible to changes in $CO_2$ concentration. Yields per bottle and individual mushroom weight also decreased in most strains when $CO_2$ levels increased. We were unable to designate any tendency in the number of fruiting bodies due to the large variation within each respective strain. Finally, water contents in the fruiting bodies were found to decline under high $CO_2$ concentrations.

The influence on cultivation characteristics of fruiting body of winter mushroom by growing humidity (재배사의 습도가 팽이버섯 재배에 미치는 영향)

  • To elucidate the effect of humidity to the characteristics related to mushroom cultivation, five white strains and four brown strains of Flammulina velutipes were cultivated and investigated on their characters. The periods for fruiting initiation, growth and harvest were a little decreased when humidity increased after peak at 75%. Brown strains showed fast fruiting than white ones. ASI 4103 was the fastest and ASI 4166 and ASI 4153 were the latest. The productivity of ASI 4166 and ASI 4149 were the best in all conditions. In general, white strains showed high yields at 75% and brown ones did at 95%. Individual mushroom weight decreased and water content of mushroom increased in most strains when humidity increased.

Microclimatological Characteristics Observed from the Flux Tower in Gwangneung Forest Watershed (플럭스 타워에서 관측된 광릉 산림 소유역의 미기후학적 특징)

  • Microclimate of Gwangneung forest watershed is characterized by analyzing wind, radiation, profiles of air temperature and humidity, soil and bole temperature, precipitation and soil water content measured at and around the flux tower from April 2000 to September 2003. Mountain-valley wind was prevalent due to the topographic effect with dominant wind from east during daytime and relatively weak wind from west during nighttime. Air temperature reaches its peak in July-August whereas monthly-averaged incoming shortwave radiation shows its peak in May due to summer monsoon. Albedo ranges from 0.12 to 0.16 during the growing season. Monthly-averaged bole temperature is in phase with monthly- averaged air temperature which is consistently higher. Monthly-averaged soil temperature lags behind air temperature and becomes higher with leaf fall. With the emergence of leafage in April, maximum temperature level during midday shifts from the ground surface to the crown level of 15-20m in May. Profiles of water vapor pressure show a similar shift in May but the ground surface remains as the major source of water. Vapor pressure deficit is highest in spring and lowest in winter. Monthly averaged surface soil temperatures range from 0 to 20℃ with a maximum in August. Monthly averaged trunk temperatures of the dominant tree species range from -5.8 to 21.6℃ with their seasonal variation and the magnitudes similar to those of air temperature. Annual precipitation amount varies significantly from year to year, of which >60% is from July and August. Vertical profiles of soil moisture show different characteristics that may suggest an important role of lateral movement of soil water associated with rainfall events.

  • This study was carried out to examine the physicochemical and microbiological characteristics of Korean and other countries chicken circulated in Tokyo, Japan. The breast meats of Korean, Japanese, and Chinese chickens and thigh meats of Korean, Japanese, Chinese, American, and Thai chicken were used. In the proximate analysis of breast meat, Korean chicken had the lowest crude protein and crude ash(P<0.01), but showed the highest content of moisture(P<0.001). TBA values of Japanese breast meat were higher than Korean and Chinese one. In thigh meat, crude fat of American one was significantly higher than the others(P<0.001), and had very high bacterial counts. Korean chicken thigh meats showed the lowest VBN values.

  • The self-diffusion coefficients of chloride and tritiated water ranged from 4.8 $\times$ 10-7 to 7.2 $\times$ 10-7 cm2/sec and 5.5 $\times$ 10-5 to 1.6 $\times$ 10-4 cm2/sec for three different depths of soil constituents at about 50% water content by volume, respectively Mobility of solute and water was conducted under steady-state flow conditions in a field soil consisting of 70 cm of clay to silty clay over a medium sand. A steady-state water flow conditions was maintained by applying irrigation water at a constant flux of 2cm per day. The water labeled with chloride and tritium was leached into the plot during the steady-state condition for 87 days. The positions of tritium and chloride as a function of soil depth and the time was measured by extracting samples of the soil solution with suction probes. Extremes in solute displacement occurred at equal and different depths within the plot. An analysis of these measurements indicated the observations of the pore-water velocity and the apparent diffusion coefficient were log normally disturbed. Twenty-four soil suction probes, used to identify the rate at which a solute was displaced in the soil, will yield an estimate of the mean pore-water velocity of this soils within a range of approximately 5% of its true value providing the effects of potential solute-soil interaction are taken into account.

  • This study was carried out to investigate the effects of aging and soil texture on composting of diesel-contaminated soil. The soils used for this study were silt loam and sand. Target contaminant, diesel oil, was spiked at 10,000mgTPH/kg of dry soil. Aging times of diesel-contaminated soils were 15days and 60days, respectively. Fresh diesel-contaminated soil was also investigated. Moisture content was controlled to 70% of soil field capacity. Mix ratio of soil to sludge was 1:0.3 as wet weight basis. Temperature was maintained at $20^{\circ}C$ Volatilization loss of TPH was below 2% of initial concentration. n-Alkanes lost by volatilization were mainly by the compounds of C10 to C17. Diesel in contaminated soil was mainly removed by biodegradation mechanism. First order degradation rate constant of TPH in sandy soil was ranged from 0.081 to 0.094/day, which is higher than that in silt loam(0.056-0.061/day). From fresh to 60day-aged soils, there was little difference of TPH biodegradation rate between the soils. Carbon recovery ranged from 0.61 to 0.89. TPH degradation rate was highly correlated with $CO_2$ production rate.

  • Such fruits as nut persimmons, mandarin oranges and apples stored in the far-infrared radiated chamber at 5$^{\circ}C$ under 90% of relative humidity had been maintained in the fresh state for longer times than the control stored only in the cold chamber. Fruits stored in the far-infrared radiated chamber showed lower values in weight loss rate, microbial colony count and decay ratio and higher ascorbic acid content than the control stored only in the told room through the storage period. We confirmed that fruits stored in the far-infrared radiated chamber under low temperature and high humidity showed least changes in quality properties through the storage period and the far-infrared radiated facilities could be a good storage system.

  • Yongchonppong is liable to withering of leaf and it is lowering leaf value for the feed. In this respect, covering of the vinylon gauze over the feed has been carried out in the branch rearing system to sustain leaf value as itself. The covering of the vintlon gauze over Yongchonppong marks 7% and 9% increment of cocoon yield in dry season, spring rearing season, as compared to the non-covering of the vinylon gauze over Kaeryangppong and Yongchonppong, respectively. There was no significant differences in cocoon shell percentages of them. In more humid environment than average relative humidity in Korea the rearing results of "Yongchonppong" was improved by covering with vinylon gauze after feeding compared with the non-covered rearing. However, the vinylon guage covering was not effective for improving the rearing results of "Kaeryangppong". No effect of the vinylon gauze covering was seen in the area where it frequently rains and raining season lasts. In case of the non-covering of the vinylon gauze, Yongchonppong reduces repidly in the first three hours of exposure to the air, as compared to Kaeryangppong. The water content reductions rate of Yongchonppong in 6 hours of exposure to the air marks 7% to so when the dry vinylon gauze cover and 17 to 26% when the wet vinylon gauze covers, as compared to 58.8% of it for the noncovering of the vinylon gauze.f the vinylon gauze.
