To develop a natural antimicrobial agent, we investigated the antimicrobial activities of 13 species of edible herbal plant extracts against major Gram-positive foodborne bacteria. Among the 13 screened edible herbal plants, Caesalpinia sappan L. showed the highest antimicrobial activity. In the paper disc agar diffusion assay, Caesalpinia sappan L. extracts had strong antibacterial activities against most Gram-positive bacteria but did not have antibacterial activities against most Gram-negative bacteria. Minimum inhibitory concentrations of the ethanol extract were 0.06 mg/mL against Clostridium difficile and Listeria monocytogenes and 0.03 mg/mL against Staphylococcus aureus. Their inhibitory activities were not reduced by heat treatment or pH adjustment against C. difficile, L. monocytogenes, and S. aureus. Antimicrobial activities were higher in ethanol extract than in distilled water extract. These results support the potential use of Caesalpinia sappan L. ethanol extract as an antimicrobial agent or functional food components against Gram-positive bacteria.
In this study, the optimal conditions for hair dyeing using sappan wood (Caesalpinia sappan L.) powder were determined, and then the effect of chitosan treatment on the hair dyeing properties of sappan wood powder was investigated. Treatment of sappan wood powder and water with the ratio of 1:3 (w/w) at 40℃ for 60 minutes was found to be the most effective to hair dyeing. In addition, the pretreatment of low molecular weight (~30 kDa) chitosan at a concentration of 1% (w/w) prior to treatment with sappan wood powder effectively improved and maintained the hair dyeing properties of sappan wood. In conclusion, the low molecular weight chitosan is considered to be of great help as a mordant in improving the dyeing efficiency and retention of sappan wood powder in hair dyeing.
Antimicrobial activity of an aqueous extract from Caesalpinia sappan L. was investigated against skin flora such as Escherichia coli, Staphylococcus aureus, Cutibacterium acnes, and Malassezia furfur. The yield and polyphenol content of the aqueous extract were 14.01±0.81% and 487.5±19.69 ㎍/mg-extract, respectively. The minimum inhibitory concentration of the aqueous extract against E. coli, S. aureus, C. acnes, and M. furfur was 0.875, 1.750, 1.750, and 1.750 mg/mL, respectively. In disc diffusion test, the aqueous extract of C. sappan L. increased the clear zone in a dose-dependent manner. The aqueous extract inhibited the microbial growth in a concentration-dependent manner.

We studied the dyeability and functionality of silks dyed singly with Caesalpinia sappan, Dactylopius coccus, combination-dyed with Caesalpinia sappan and Phellodendron Amurense Rupr., and with Dactylopius coccus and Phellodendron Amurense Rupr.. The color of silk dyed singly with Caesalpinia sappan was Red with Hue of 2.2 R, the color of silk dyed singly with Dactylopius coccus was Reddish Purple with Hue of 3.0 RP, The color of silk combination-dyed with Caesalpinia sappan and Phellodendron Amurense Rupr. was 2.7YR being in the Orange family, and the color of silk combination-dyed with Dactylopius coccus and Phellodendron Amurense Rupr. was Red with Hue of 4.3R. In the case of antimicrobial activity, except for silk dyed only with Dactylopius coccus, the other three types of dyed silk showed excellent antimicrobial activity against Staphylococcus aureus achieving higher than 91.1% of bacteria reduction rate. Against Klebsiella pneumoniae, silk combination-dyed with Dactylopius coccus and Phellodendron Amurense Rupr., and with Caesalpinia sappan and Phellodendron Amurense Rupr. presented very excellent antimicrobial property achieving bacteria reduction rate of 99.9%.
This study was to study the antibacterial effect of Lignum sappan extract on S. mutans growth. The antibacterial activities of Lignum sappan's crude extract was measured 17.3 mm at 20 mg/ml concentration. The growth of S. mutans in control medium was the highest at 8hr, while the media of Lignum sappan extract added-medium (2 mg/ml) showed maximum growth at 16hr. The pH values of the control media was 5.08 at 8hr, but the media supplemented with Lignum sappan extract was 6.69 at 8hr. The amounts of total carbohydrate of the control media was 0.81 mg/ml at 8hr, but the media supplemented with Lignum sappan extract was 2.06 mg/ml at 8hr. In the protein change of culture medium, the control culture broth and the cultures supplemented with Lignum sappan extract was 8.39 mg/ml and 12.3 mg/ml at 8hr, respectively. The S. mutans polysaccharide contents of the control media and the media supplemented with Lignum sappan extract was 300 mg/100ml and 60 mg/100ml at 8hr, respectively.
Antimicrobial activity of Caesalpina sappan L. extract (CS extract) against animal husbandry disease-related bacteria was studied. The CS extracts showed a significant antimicrobial activity against Gram(+) bacteria and this antimicrobial activity was most significant against Staphylococcus epidermidis. Minimum inhibitory concentrations (MIC) of CS was in the range of 0.8∼16 mg/ml and 0.8∼10 mg/ml, in the case of MeOH extracts and EtOH extracts, respectively. In addition, the antimicrobial activity of each solvent fraction was most significant with EtOAc layer. The antimicrobial activities of CS extracts against most microbial strains were stable by either heat treatment or acid treatment, whereas those against S. aureus and Sal. typhimurium were reduced by acid treatment. The inhibitory effect of CS extracts on microbial cell growth was further examined by the addition of 0, 100, 300, and 500 ppm of CS extracts into growth medium. The growth of gram(+) bacteria, S. aureus and S. epidermidis, was inhibited for 72 hours in all ranges of CS extracts added, but the growth of gram(-) bacteria was only inhibited when at least 100 ppm of CS extracts were added. Taken together, the antimicrobial activities of CS extracts were more effective against gram(+) bacteria compared to those against gram(-) bacteria.
To evaluate the antioxidative activity of the ethanol extracts from 116 domestic medicinal plants, each extract was added to palm oil and lard in 1,000 ppm level, and the induction time was measured by Rancimat method. Almost all plant extracts tested showed more or less antioxidative activity. The extracts which had strong antioxidative activity to palm oil and lard were Caesalpinia sappan L., Paeonia japonica Miyabe et Takeda, Dendrobium moniliforme (L.) Sw and Crcumae longa L. These extracts were fractionated with chloroform, ethyl acetate, butanol and water. Ethyl acetate layer of Caesalpinia sappan L. and Paeonia japonica Miyabe et Takeda showed marked antioxidative activity, and chloroform layer of Dendrobium moniliforme (L.) Sw and Crcumae longa L. ethanol extract had stronger antioxidative activity than all the other layers.
Caesalpinia sappan L. is an oriental medicinal plant distributed in the Asia Pacific region including India, Malaysia, and China. The dried heartwood of Caesalpinia sappan has been traditionally used as an analgesic and anti-inflammatory drug. In this study, the effects of extract methods of C. sappan on contents of total polyphenols and flavonoids, antioxidant activity, and cytotoxic activity were evaluated. As a result, hot water extract from C. sappan (CSWE) significantly exhibited contents of total polyphenols and flavonoids (22.6 mg GAE/g and 14.5 mg QE/g) higher than 70% ethanol extract (CSEE) (17.6 mg GAE/g and 13.2 mg QE/g). However, CSEE showed greater antioxidant activity than CSWE in both DPPH and ABTS. Also, the cytotoxicity of C. sappan against three kinds of cancer cell lines was higher in CSEE than in CSWE. These results show that ethanol extract is a better extract method than hot water method to maintain antioxidant and anti-cancer activities.
In this study we have investigated the effect of Caesalpina sappann L. extracts on the apoptosis in NIH3T3 cells exposed to methylmethan sulfonate (MMS), an alkylating agent. MTT assay study showed that Caesalpina sappan L. extracts potentiate the MMS-induced viability. Cell morphology studies, acridine orange (AO) staining, and DNA fragmentation analysis indicated that the postincubation of Caesalpina sappan L. extracts increase the nuclear condensation of MMS-induced apoptotosis. These results suggest that Caesalpina sampan L. extracts contain components potentiating MMS-induced apoptosis of NIH3T3 cells.
Somok, the heart wood of Caesalpinia sappan is used in traditional Chinese medicine. This study was performed to investigate the effect of growth and culture conditions of brazilin from C. sappan against S. mutans ATCC 25175. The bacteria were cultured in brain heart infusion (BHI) broth, and then incubated under 5% $CO_2$ at $37^{\circ}C$ for 18-24 hours. The effect of brazilin against S. mutans was confirmed under the changes of the culture conditions, such as growth curve and the change of pH, protein, and total carbohydrate. The growth of S. mutans in control medium was the highest at 24 hr, while brazilin-added medium (0.3 mg/ml) showed maximum growth at 32 hr. The pH values of the control medium was 5.25 at 16 hr, but the media supplemented with brazilin (0.3 mg/ml) was 7.0 at 16 hr. The amounts of total carbohydrate of the control medium was 11 mg/ml at 8 hr, but the brazilin-added media (0.3 mg/ml) was 18 mg/ml at 8 hr. In the protein change of the culture medium, the control culture broth and the brazilin supplemented-cultures was 2.4 mg/ml and 2.54 mg/ml at 24 hr, respectively. Polysaccharide contents of the control medium and test media were 3 mg/ml and 2 mg/ml at 8 hr, respectively. Thus, the application of C. sappan can be considered a useful and practical material for the prevention of dental caries.
