Selenium and Human Health : from the view of anticarcinogenic effects (셀레늄과 인간의 건강 : 항암효과를 중심으로)

  • 필수 미량원소인 셀레늄은 인간의 건강에 기본적으로 중요한 역할을 하는 물질이다. 항산화 관여 효소의 작용과 구조역할을 담당하는 셀레늄이 함유된 단백질(selenoproteins)은 항산화 기능이 있을 뿐만 아니라, 갑상선호르몬의 활성화에 관여하는 것으로 밝혀졌다. 또한, 셀레늄은 정상적인 면역체계의 유지에 필요하며, HIV(human immunodeficiency virus)가 진행되어 AIDS로 발전되는 것을 억제하는 작용이 있다는 사실도 밝혀지고 있으며, 정자의 생성과 운동성에 관계하기도 한다. 우울증 환자가 셀레늄을 섭취하게 되면 효과가 있으며, 셀레늄이 순환계질환의 발생을 억제하는 것으로 알려지고 있다. 특히 최근에는 셀레늄의 섭취가 암발생을 예방할 수 있다는 역학조사결과를 토대로, 대규모 셀레늄 임상실험이 다국적 연구의 형태로 2개의 프로젝트(PRECISE, SELECT)가 수행 중이다. 한편, 셀레늄의 항암효과에 대한 세포수준에서의 연구가 활발하게 이루어지고 있으며, 어느 정도 가시적인 결과를 보이고 있다. 따라서, 셀레늄에 대한 재평가가 우리 나라에서도 이루어져야 한다. 그러기 위해서는 지역별로 셀레늄의 정확한 섭취량조차도 파악되어 있지 않을 뿐만 아니라 일일권장량 값조차 정확하지 않은 현실을 볼 때, 셀레늄의 기본적 연구에 대한 관심을 가져야 할 때라고 판단된다.

UVB Protective Effect of Yeast Originated Selenium Peptide on Fibroblast (효모 유래 셀레늄 펩타이드의 인간 섬유아세포에 대한 UVB 보호효과)

  • Selenium-containing peptide (Selenium peptide) was produced by autolysis of Saccharomyces cerevisiae which was cultured in inorganic selenium-supplemented medium. Selenium peptide showed antioxidant activity and protective effects on UVB irradiated human fibroblast. Minimal toxicity of selenium peptide was observed whereas selenium nitrate exhibited cell toxicity as low as $10^{-9}\;M$. Selenium peptide also increased human fibroblast growth, procollagen type I and also decreased MMP-1 (matrix metalloprotease-1). This result showed the potential of selenium peptide as a nontoxic antioxidant.

Production and Characterization of Selenium Peptide from Saccharomyces Cerevisiae (효모를 이용한 selenium peptide 생산 및 특성 연구)

  • Selenium containing peptide was produced by culturing yeast with selenium, Selenium was broadly incorporated in the various size of proteins based on the GPC analysis of the total yeast protein. The ratio of selenium to protein increased with the concentration of added selenium in the culture medium. Antioxidant activity (glutathione peroxidase-like activity) was proportional to the concentration of selenium concentration in the peptide. Different size of proteins were obtained by hydrolyzing the total yeast protein by protease XIV. Average molecular weight of selenium peptide was analyzed by GPC. Glutathione peroxidase (GPx) activity of the selenium peptide increased as the size of peptide decreased. Sodium selenite had strong inhibition on the yeast growth than sodium selenate. The ratio of selenium to protein was higher with sodium selenate than with sodium selenite. These results showed the potentials of selenium peptide production by yeast cultivation.

Antioxidant Activities of Selenium-Treated Spinacia oleracea L. (셀레늄 강화 시금치의 항산화 활성)

  • In the present study, we investigated the anti-oxidant activities of selenium-treated Spinacia oleracea L. by utilizing experiments in vitro assays. The selenium content of non-treated spinach in this study was noted at $61.19{\mu}g/kg$, whereby the selenium-treated spinach which was treated by a 2000 mg/kg selenium was 1000-fold diluted, and was reported to be about 4 times higher than that of non-treated spinach. In this case, the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity in the concentration of selenium-treated spinach, 0.1~1.0 mg/mL was measured as stronger than that of the identified non-treated spinach. By the same token, the DPPH radical activity of non-treated spinach and selenium-treated spinach was recorded as 46.05~52.75% and 49.52~59.09% respectively. It is emphasized that the 2,2'-azino-di-2-ethyl-benzthiazoline-sulphonate (ABTS) radical scavenging activity as revealed in the concentration of selenium-treated spinach, 0.1~1.0 mg/mL was noted as being stronger than that of non-treated spinach. The ABTS radical activity of non-treated spinach and selenium-treated spinach was 11.85~52.01% and 27.14~53.59% respectively. In this respect, the nitric oxide (NO) radical scavenging activity and reducing power activity in the concentration of selenium-treated spinach, 0.1~1.0 mg/mL was identified and noted as stronger than that of non-treated spinach. These results suggest that selenium-treated spinach could possibly be more useful as a potential antioxidant to improve human health outcomes, than the non-treated spinach.

The Protective Effect of Selenium on Radiation in Rat (흰쥐에 있어 방사선에 대한 셀레늄의 보호효과)

  • In this study, the radiation protection effects of selenium, known as a powerful antioxidant, were studied. After 14 days of oral injection of selenium into the rat, a full body irradiation of 10 Gy was carried out. And after 1 day, 3.5 day, 7 day and 21 day, we observed blood cell components and superoxide dismutase(SOD) and small intestine changes. Compared to irradiation groups, there was a significant protection effect of reducing the hematopoietic immune system damage in the irradiated group after selenium administration(p<0.05). Then, selenium is a valid ingredient that increases the activity of the superoxide Dismutase(SOD), and it is confirmed that it has an effect of inhibiting apoptosis expression of small intestinal cells by irradiation. Based on these results, selenium is considered to be an essential ingredient for protecting living things from radiation.

Effects of Dietary Organic Selenium Levels on Performance and Selenium Retention in Broiler Chickens and Laying Hens (유기태 셀레늄의 첨가가 육계 및 산란계의 생산성 및 셀레늄 축적에 미치는 영향)

  • Two experiments were conducted to investigate the effect of dietary organic selenium levels on performance and selenium retention in broiler chickens and laying hens. In experiment 1, the effects of dietary organic selenium levels on the weight gain, feed intake, feed conversion, and selenium retention of meat and liver in broiler chickens were investigated. For each growth phase, the basal diet was supplemented with 0 (control), 0.60, 1.20, 1.80 and 2.40 ppm Se from selenium yeast(SY). Weight gain, feed intake, and feed conversion were not affected by the selenium addition in diets. Breast muscle Se levels were linearly increased (P<0.05) as dietary Se level increased by SY. Selenium concentration of liver tissue was increased (P<0.05) in supplemental SY compared to the control, and was increased (P<0.05) in supplemental 1.20, 1.80 and 2.40 ppm SY compared to the 0.60 ppm SY. In Experiment 2, 12-week-experiment using Hy-Line laying hens (68 wk of age) was conducted to examine the effects of dietary organic selenium on egg Production, egg weight, daily egg mass, feed intake, feed conversion, egg quality, and selenium concentration of eggs. A corn-soybean meal basal diet was supplemented with 0 (control), 0.30, 0.60, 0.90 and 1.20 ppm Se from selenium yeast (SY). Egg Production was significantly improved(P<0.05) in supplemental 0.30 and 0.90 ppm SY compared to the control and 0.60 ppm SY during week 1 to 12, but daily egg mass, feed intake, and feed conversion showed no difference in supplemental SY and control. Haugh unit, yolk color and eggshell breaking strength showed no difference in supplemental SY and control. Eggshell thickess was significantly (P<0.05) higher in supplemental 0.60 and 1.20 ppm SY compared to the 0.90 ppm SY in week 9. Egg Se levels were linearly increased (P<0.05) as dietary Se level increased by SY.

The Effects of Endurance Exercise and Selenium Treatment on Mitochondrial Transcription Factors Expression in Old GK Rats (지구성 운동과 셀레늄 투여가 노화 GK 흰쥐의 미토콘드리아 전사인자 발현에 미치는 영향)

  • The objective of this study is to identify the effects of endurance exercise and selenium on mitochondrial transcription factor in old Goto-Kakizaki (GK) rats. In this experiments, endurance exercise were treadmill-run at 24 m/min, 30 min/day, 5 days/week, 6 weeks and 5 umol/kg of sodium selenite was injected intraperitoneally. In exercise group, selenium group, and combination group, the mitohondrial biogenesis-related genes, including PGC-$1{\alpha}$, NRF-1, and Tfam expression level were significantly increased compared to control group. Consistent with the increased biogenesis-related genes, the cytochrome C in the treated groups, which was the indicator of mitochondrial content, was significantly increased compared to control group. Especially, combination of exercise and selenium may be effective in the increase of mitochondrial biogenesis, activity and insulin sensitivity. Therefore, exercise and selenium treatment is likely to promote diabeticmitochondrial malfunction and then improve diabetes.

Effect of Seleniferous Whole Crop Barley Silage on Growth Performance, Blood and Carcass Characteristics, and Tissue Selenium Deposition in Finishing Hanwoo Steers (셀레늄 강화 청보리 사일리지 급여가 비육기 거세한우의 생산성, 혈액성상 및 도체특성과 조직 내 셀레늄 축적에 미치는 영향)

  • This study was conducted to investigate the effects of feeding seleniferous whole crop barley silage (WCBS) on the growth performance, blood and carcass characteristics, and tissue selenium deposition infinishing Hanwoo steers. A total of 20 growing Hanwoo steers were selected and assigned to one of the following feed groups: control (0.1 ppm Se), T1 (0.3 ppm Se), T2 (0.9 ppm Se), and T3 (0.9 ppm of inorganic Se). Five steers were allocated to each treatment group, and the trial lasted for 90 days. All experimental diets combined seleniferous and/or non-seleniferous WCBS up to a ratio of 30%. For the T3 diet, 0.9 ppm Se in the form of sodium selenite was added to the control diet. Dietary level and type of selenium did not affect feed intake and body weight gain. Blood total lipid and triglyceride concentrations were significantly (p<0.05) lower in the T2 group than in control. Blood immunoglobulin G concentration increased significantly (p<0.05) with increasing levels of dietary selenium; concentrations were significantly (p<0.05) higher in T2 and T3 than in control. Blood selenium concentration was the highest (p<0.05) in T2. No significant difference was observed in dressing rate, ribeye area, marbling score, meat color and fat color. Further, no association was found between levels and chemical form of dietary selenium and quality and quantity of meat. To the contrary, tissue selenium content in kidney, liver, and hind leg increased significantly (p<0.05) with increasing levels of selenium; however, feeding inorganic selenium did not introduce a significant increase in tissue selenium content of finishing Hanwoo steers. The results indicated that the selenium present in seleniferous WCBS was an effectively absorbable selenium source, suitable for increasing tissue selenium content in beef. Approximately 100 g of hind leg muscle from steers fed on the T2 diet met approximately 69% of the recommended daily selenium requirements.

  • 나노 입자 분무 기법을 이용한 $Cu(In,\;Ga)Se_2$ (CIGS) 광흡수층 제조 기법은 고진공 장치를 사용하지 않는다는 점에서 대면적 저가형 CIGS 태양전지 양산에 적합한 차세대 기술로 인식되고 있다. 그러나 일반적으로 스프레이 된 상태의 CIGS충 자체는 태양전지 제조에 적합하지 않은데 이는 스프레이 막의 다공성 구조 때문이다. 본 연구에서는 나노입자 분무 기법을 이용하여 증착한 CIGS 광흡수층막을 질소 또는 셀레늄 분위기에서 열처리함으로써 태양전지 제조에 적합한 치밀한 구조의 CIGS 광흡수충을 제조하고자 하였다. 실험 결과, 질소 분위기 $500^{\circ}C$의 온도에서 1시간 열처리하여도 CIGS 나노 입자의 성장은 거의 일어나지 않는 것으로 나타났다. 반면 셀레늄 분위기 $500^{\circ}C$의 온도에서 30분 열처리시 입자 크기가 $1{\mu}m$이상인 치밀한 광흡수층을 얻을 수 있었다. 본 결과는 CIGS 나노 입자의 입자 성장 반응에서 열에너지 단독에 의한 표면 에너지 감소 효과는 미미하며 셀레늄 증기의 역할이 더욱 크다는 것을 의미하는 것이다.

Studies on Selenium-fortified Functional Hanwoo-Beef by Utilizing Spent Mushroom Composts II. Effects of Spent Composts of Se-Enriched Mushrooms as the Dietary Se Source on Selenium Deposition in the Muscular Tissue and Plasma Glutathione Peroxidase Activity in the Finishing Hanwoo Steer (버섯폐배지를 이용한 셀레늄강화 기능성 한우고기 생산에 관한 연구 II. 셀레늄강화 버섯폐배지 첨가가 한우의 근육조직 내 셀레늄 축적과 혈중 glutathione peroxidase(GSH-Px)활성에 미치는 영향)

  • This study was conducted to investigate effects of spent composts of Se-enriched mushrooms as the dietary Se source on muscular Se deposition and plasma glutathione peroxidase (GSH-Px) activity in the finishing Hanwoo steer. Twenty Hanwoo steers were used in the experiment and they were divided into four groups in a randomized complete block design with five replicates. Treatments were four levels (0.1, 0.3, 0.6 and 0.9ppm as fed basis) of dietary Se from the combination with spent composts of Se-enriched mushrooms and/or Se non-enriched mushrooms, in which each treatment was formulated with corn and corn gluten meal and so forth. Treatment diets were fed to Hanwoo steers for 90 days until the slaughter. Dry matter intakes had no significant differences among treatments and there were no significant differences for performances such as total BW gain and ADG among treatments. The Se concentration in blood was linearly increased with increasing dietary selenium levels and reached a plateau level after 8 weeks (p<0.001). Plasma GSH-Px activities had the similar trends to blood Se concentrations by showing that the increased dietary Se level significantly increased plasma GSH-Px activities of both total and Se-dependent (p<0.001). Muscle Se contents of Se-supplemented groups were linearly increased by 1.35 ~ 1.68 folds compared with the control group (0.1ppm; 0.273㎍/dry g) and especially those of the hind legs for 0.9ppm treatment showed the highest Se content as shown 0.457㎍ per dry gram (p<0.01) corresponding to approximately 70% increase of the control group. Se retention rate in the muscle of dietary Se originated from spent composts of Se-enriched mushrooms was estimated of maximum approximately 30% and dietary Se content showed the significant correlation with plasma GSH-Px activities and muscle Se contents (p<0.01). Accordingly, Se present in spent composts of Se-enriched mushroom as the dietary Se source not only had great bioavailabilities showing higher blood Se concentration and plasma GSH-Px activities, but also increased Se deposition in the muscle for Hanwoo beef cattle.
