The Bacterial Community Structure in Cheonho Reservoir Dominated by Cyanobacteria (봄철 Cyanobacteria 가 우점한 천호지에서 세균군집구조의 변화)

  • The composition of bacterial communities was detected in surface water of Cheonho Reservoir dominated by cyanobacteria, using fluorescent in situ hybridization (FISH) method. Total bacterial numbers were very high ranging from 0.6~$1.3{\times}10^7 \cells{\cdot}ml^-1$, whereas the ratio of Eubacteria to total bacteria was 29.8~45.8%, which was lower than that in other freshwater ecosystems. On average only 2.1% of DAPI-stained bacteria were detected by FISH with probes for $\alpha$, $\beta$, and $\gamma$-groups, respectively. Unknown eubacteria which was not bound to any probes except EUB 338, was relatively high. On the other hand, the Cytophaga-Flavobacterium group increased following the change of dominant species from Anabaena sp. to Microcystis sp. This result showed that bacterial communities could be affected by phytoplanktons, especially cyanobacteria.

Bacterial Community of Free-living and Aggregated Bacteria at Thawing Period in Lake Baikal (해빙기 바이칼호에서 부유세균과 Aggregates에 부착한 세균의 군집구조)

  • Fluorescent in situ hybridization (FISH) with rRNA-targeted oligonucleotide probes was used to compare the community structures of free-living and aggregated bacteria at thawing period in Lake Baikal. Targeted groups were Eubacteria, $\alpha$-, $\beta$-, $\gamma$- proteobacteria groups, Cytophaga-Flavobacterium group and Planctomycetales. Total bacterial numbers of free-living bacteria were ranged from $0.2{\times}10^6\cells{\cdot}ml^-1$ to $3.2{\times}10^6\cells{\cdot}ml^-1$, which were decreasing with depth, while the aggregated bacterial numbers were dramatically increasing from $0.4{\times}10^4 to 3.3{\times}10^4 \cells{\cdot}ml^-1$ with depth. The ratios of EUB probe binding cells to DAPI counts were ranged from 52.3 to 74.1% in free-living bacteria, and from 39.6 to 66.7% in the aggregated bacteria, respectively. Community structures of the aggregated bacteria were very different from each free-living bacteria at every depth. At 25 m depth, where the chlorophyll a concentration was highest, both structures were quite different from those of surface layers, rendering the fact that the community structures might be affected by phytoplankton. The vertical profile of community structure of aggregated bacteria is particular. The proportion of $\beta$-proteobacteria group was increasing with depth and it was 51.8% at 100 m, but the dominant group was $\gamma$-pro-teobacteria group at 250 m. Taken together, the biodiversity and succession of aggregated bacteria are quite different from free-living bacteria.

Bacterial Community Profiling during the Manufacturing Process of Traditional Soybean Paste by Pyrosequencing Method (Pyrosequencing을 이용한 전통된장 제조과정 중 세균군집구조의 분석)

  • In order to evaluate the diversity and change of bacterial population during the manufacturing process of traditional soybean paste (doenjang), bacterial communities were analyzed using 16S rRNA gene-based pyrosequencing. In rice straw, the most important inoculum source for fermentation, the bacterial sequences with a relative abundance greater than 1% were assigned to four phyla, Proteobacteria (71%), Actinobacteria (20.6%), Bacteroidetes (4.2%), and Firmicutes (1.3%). Unlike bacterial community composition of rice straw, a different pattern of bacterial population in meju was observed with predominantly high abundance (99.1%) of Firmicutes. Phylum composition in young doenjang was almost same as that of meju. Major genera in young doenjang were Bacillus (81.3%), Clostridium (6.9%) and Enterococcus (6.3%) and the predominant species among bacterial population was B. amyloliquefaciens (63.6%). Abundance of the phylum Firmicutes in mature doenjang was 99.98%, which was even higher value than those in meju and young doenjang. Predominant species in mature doenjang were B. amyloliquefaciens (67.3%), B. atrophaeus (12.7%), B. methylotrophicus (6.5%), B. mojavensis (3.2%), and B. subtilis. (2.5%), which were also identified as major species of the microbial flora in meju. These results suggested that rice straw was a primary source for supplement of Bacillus species in manufacturing the traditional doenjang and that some species of Bacillus strains were mainly involved in the fermentation process of traditional doenjang.

Bacterial- and Archaeal Communities in Variously Environmental Conditioned Basins of Several Wastewater Treatment Plants (다양한 환경 조건의 하수처리시설 반응조 내 세균 및 고세균 군집)

  • To investigate the differences of bacterial- and archaeal communities depending on kind of wastewater (municipal/livestock) and on treating conditions of basins, sludges were sampled from 10 basins of 3 municipal wastewater treatment plants(WWTP) with A2O and a activated sludge sample from a livestock WWTP. The metagenomic DNAs of the sludge samples were extracted and amplified with primers, 27F/518R for bacteria and Arch519F/A958R for archaea, and pyrosequenced with Roche 454 GS-FLX Titanium. As results, the bacterial communities in basins of municipal WWTPs were quite different from those of livestock WWTP, but within the same municipal WWTP their community structures were similar to each other regardless of different environmental conditions such as O2. And their archaeal communities resulted from anaerobic·anoxic basins were clustered only within communities originated from the same WWTP. Furthermore Seo-bu WWTP with high bacterial diversity and species richness performed better N/P-removal compared to the orther WWTPs.

Effect of Salinity on the Bacterial Community in the Sewage Treatment System (하수 처리 과정에서 염분이 세균 군집에 미치는 영향)

  • For elucidating the effect of salinity to the effect of wastewater treatment, the heterotrophic bacterial numbers, total bacterial numbers, and the bacterial community structure by FISH method were analyzed. The total bacterial numbers were not significantly changed by the salinity. But the heterotrophic bacterial numbers and bacterial community structures were drastically changed by the increase of salinity. In case of 1% salinity, the heterotrophic bacterial numbers and structure were slightly changed comparing to those of contol. In case of 2% and higher salinities, the numbers of heterotrophic bacteria and the proportions of Eubacteria, Proteobacteria $\alpha$-group, $\rho$-group and Cytophaga-Flavobacterium groups were deceasing. By these results, the salinity stress to bacterial community in waste water treatment was unveiled, and for sustaining the waste water treatment system, the salinity should be lower than 1%.

수계 생태계에서의 세균 군집 구조의 분자생물학적 분석

  • 16S rRNA를 분석한 연구들은 자연 생태계에서 추출한 핵산을 이용하여 16rRNA 유전자의 염기서열을 분석하거나 특정 DNA probe를 이용한 hybridization 실험이 주류를 이루어 왔다. 특히 PCR 기법이 개발됨에 따라 적은 양의 시료를 대량으로 손쉽게 증폭시킬 수 있어 다양한 분야에 응용되고 있다. 세균 군집의 구조를 이해하는데 있어서 PCR 방법의 적용 대상은 주로 16S rRNA 유전자의 염기서열 해독분야이며 해양 생태계를 대상으로 많은 연구 결과가 보고되었다(11,13,21,26). 한편 자연 생태계의 개별적 미생물 분류룬들을 검출하기 위한 특정 oligonucleotide probe의 개발방법들은 미생물 군집의 유전적 다양성에 대한 정보 파악 이외에 배양이 어려운 혐기성 세균과 같은 특정 세균들의 동정에도 이용되고 있다(3,24,55). 본고에서는 세균 군집의 구조와 다양성을 연구하는데 적용 가능한 rRNA 분석방법들을 수계 생태계를 중심으로 살펴보고자 한다.

The Bacterial Communities Structure and Its Environmental Determinants in Lake Soyang (소양호 세균군집구조와 그 구조에 영향을 주는 환경요인)

  • The temporal variation of bacterial community and environmental factors, affecting on bacterial community structure were estimated monthly kom April, 1998 to May, 1999. Bacterial community structures were determined by in situ hyblidization with rRNA-targeted fluorescently labeled oligonucleotide probes (FISH) and epifluorescence microscopy; and the statistical analysis was done by SPSS program. The oligonucleotide probes used in this study were EUB338, ALFlb, GAM42a, and CF. In surface water, $\alpha$-group was related to only DOC (-0.538, p<0.05) and Chlorophyll a concentration was related to y-group (-0.630, p$\beta$-group and Cytophaga-Flavobacterium group were related to water temperature as 0.665, and 0.685 @<0.05). Between pH and $\beta$-group, there was a positive relationship (0.541, p<0.05), and Cytophaga-Flavobactevizim group was represent to correlation (0.672, p

Community Analysis of the Bacteria in Sponges of Lake Baikal by FISH Method (바이칼 호 Sponge에 서식하는 세균 군집 구조의 분석)

  • The bacterial community structures at 2 sponge species belonging to the genus Baikalospongia and Lubomirskia in Lake Baikal were analyzed with fluorescent in situ hybridization (FISH) method. The total bacterial numbers in the genus Baikalospongia ranged from $7.2{\times}10^{7}\;to\;4.2{\times}10^{8}\;cells/ml$, and those in Lubomirskia from $1.2{\times}10^{8}\;to\;1.6{\times}10^{8}\;cells/ml$, while those of lake water were from $2.3{\times}7.7{\times}10^{5}\;cells/ml$. Total bacterial numbers in the sponges were $10^{3}-10^{4}$ times higher than those of lake water. In the genera Baikalospongia and Lubomirskia, the proportions of other unidentified bacterial groups to the Bacteria were 42.0-60.3% and 40.7-51.9%, respectively. These proportions were similar to those in lake water (22.6-46.3%). These results suggest that bacterial compositions in Lake Baikal water and sponges are highly unique.

The Change of Attached Bacterial Community on Aggregates in Lake Paldang (팔당호에서 Aggregates에 부착한 세균군집구조의 변화)

  • For scrutinizing the changes of aggregated bacteria in Lake Paldang, the FISH method was applied by using the rhodamine labeled probes, and total bacteria, chloropyll a concentrations and nutrients concentrations were measured. The aggregates were collected with sediment traps. The T-N, T-P, chlorophyll a concentrations of aggregates were higher 5-15 times, 81-140 and 49-66 times than water samples, respectively. Also, the bacterial numbers of aggregates were 200 times higher than those of water smaples. The ratios of each groups of water sample were 2.1-7.4% for $\alpha$-group, 4.5-8.3% for $\beta$-group, 2.1-7.4% for $\gamma$-group, 2.1-6.1% for Cytophaga-Flavobacterium group and 0.1-2.5% for 'other'group, respectively. While, in aggregates, the ratios of $\alpha$-, $\beta$-, $\gamma$- and Cytophaga-Flavobacterium groups were very small and most abundant group was 'other' bacteria. With these results, the aggregated bacteria in Lake Paldang had a particular group composition of bacteria.

Structure of Bacterial Communities in Biological Nitrogen Removal System (Biological Nitrogen Removal System의 세균 군집 분석)

  • To understand the efficient process of biological nitrogen removal (BNR) system, the structure of bacterial communities in nitrification reactors was analyzed using PCR and terminal restriction fragment length poly morphism (I-RFLP) methods. In this study, we used an advanced treatment system with plotting media, Nutrient Removal Laboratory system, or the rumination type sequencing batch reactor (SBR) system. The terminal restriction fragments of ammonia-oxidizing bacteria (AOB) and other $\beta-proteobacteria$ were observed in all of three BNR systems. The nucleotide sequence analysis of terminal restriction fragments showed that Nitrosomonas and Nitrosolobus were major populations of AOB in SBR system, whereas uncultured $\beta-proteobacteria$ and Cardococcus australiensis were the predominant groups in other two BNR systems. Also the SBR system may be more efficient to enrich AOB. These results indicate that the different structure of bacterial community may be developed depending on the wastewater treatment systems, although the same influent is used.
