Acetone-Butanol Fermentation of Rice Straw by Simultaneous Saccharification and Fermentation (동시당화 발효법에 의한 볏짚의 Acetone-Butanol 발효)

  • Rice straw was used in the acetone-butanol fermentation by semultaneous saccharification and fermentation (SSF) using Clostridium acetobutylicum and cellulolytic enzyme. Over 230 mM of solvent was produced from alkali treated rice straw of from ball-milled microcrystalline cellulose whilst only acidic fermentation products were formed from ball-milled rice straw. From the results it is concluded that rice straw used in the study contained an inhibitor for the solventogenesis by the organism which is insoluble in water and some organic solvent and destroyed by alkaline treatment.

Complete genome sequence of Celluosilyticum lentocellum WCF-2 isolated from cow dung (소 분변에서 분리된 Celluosilyticum lentocellum WCF-2의 유전체 염기서열 분석)

  • An anaerobic bacterial strain WCF-2 was isolated from cow dung in finding cellulose-degrading bacteria for use as silage additives. Strain WCF-2 showed a higher cellulolytic activity than Cellulosilyticum lentocellum DSM $5427^T$, the closest relative of strain WCF-2 (98.2% of 16S rRNA gene sequence similarity). We sequenced the complete genome of strain WCF-2 and compared it with that of C. lentocellum DSM $5427^T$. The OrthoANI value between the two strains was 97.9% thus strain WCF-2 was identified as C. lentocellum. The genome size of strain WCF-2 was 4,779,774 bp with a G + C content of 34.4%, 4,154 coding genes (CDS), 54 pseudo genes, and 142 RNA genes. Strain WCF-2 harbored seven cellulase genes, five of which showed low similarities with those of C. lentocellum DSM $5427^T$.

Effect of ${\alpha}-Amylase$ Treatment of Brown Rice(Goami) Alcohol Fermentation By-Product (현미(고아미) 알코올발효 부산물의 ${\alpha}-amylase$처리 효과)

  • To utilize the non-heat treated alcoholic by-products of brown rice(Goami) as food sources, the quality characteristics change according to the treatment conditions of ${\alpha}-amylase$ were evaluated. It resulted that the increase of hydrolysis temperature correspondingly increased the soluble solids, total dietary fiber and total sugar in the by-products of Goami, and the highest reducing sugar content was observed at $80^{\circ}C$. The free amino acids contents were tended to slowly decrease by the hydrolysis temperature more than $70^{\circ}C$, and the highest content of oligosaccharides were detected at the hydrolysis temperature of $80^{\circ}C$. The soluble solid according to the ${\alpha}-amylase$ concentration resulted to increase with the increase of the enzyme concentration and the total dietary fiber revealed similarly showing approximately 0.65%. The high content of reducing sugars was observed at the enzyme concentration around 0.08%(v/w). Total sugars and oligosaccharides contents tend to increase as the concentration of enzyme increased, and the content of oligosaccharides acquired at the enzyme concentration more than 0.10%(v/w) maintained to show rather similar contents. The soluble solids and total dietary fiber by hydrolysis time were found to show 6.66% and 0.65%, respectively at more than 60 min of hydrolysis, and the reducing sugars and total sugars were found to be 3,600 and 4,800 mg% in all treatment groups showing no significant difference. The content of oligosaccharides was increased with the increase of hydrolysis time, and the content was similar at more than 90 min of hydrolysis by ranging around 2,100 mg%. Based upon these results, the by-products of Goami are expected to be used as various food sources showing the highest dietary fiber and oligosaccharides contents by the hydrolysis at $80^{\circ}C$ for 90 min with the addition of 0.10%(v/w) of ${\alpha}-amylase$.

Monitoring of Feed-Nutritional Components, Toxic Heavy Metals and Pesticide Residues in Mushroom Substrates According to Bottle Type and Vinyl Bag Type Cultivation (버섯의 봉지재배 및 병재배 시 재배단계별 배지의 사료영양적 성분, 독성중금속 및 잔류농약 모니터링)

  • This study was carried out to monitor feed-nutritional components, toxic heavy metals (Cd, Pb and As) and pesticide residues through three cultivation stages (1st initial culture stage, 2nd mycelial growth stage, and 3rd fruit body-harvested stage) of king oyster mushroom (Pleurotus eryngii) produced by bottle type cultivation and oyster mushroom (Pleurotus osteratus) produced by vinyl bag type cultivation. For both cultivation types, compared with the initial culture, the weight reduction rate in spent mushroom substrates (SMS) after fruit body harvest was 29% for total wet mass, 21~25% for dry and organic matters and 19 ~22% for neutral detergent fiber. Two thirds to 3/4 of organic matter degraded and utilized by mycelia and fruit bodies was originated from fiber, of which the primary source (50~70%) was hemicellulose. The effect of mycelial growth stage on chemical compositional change in culture was little (P>0.05) for bottle type cultivation of king oyster mushroom but considerable (P<0.05) for vinyl type cultivation of oyster mushroom. Culture nutrients uptake by fruit bodies was very active for the bottle type cultivation. Compared with SMS, harvested fruit bodies (mushrooms) contained higher (P<0.05) crude protein, non-fibrous carbohydrate, and crude ash and lower (P<0.05) neutral detergent fiber. Regardless of stages, no culture samples were contaminated with toxic heavy metals and pesticide residues. In conclusion, the increase of fiber (neutral and acid detergent fibers) and indigestible protein contents and the decrease of true protein content in SMS indicated that the feed-nutritional value of SMS was significantly reduced compared with that of the initial culture and they were safe from toxic heavy metals and pesticide residues.

Effects of the Dosing Regimen of Tissue-type Plasminogen Activator on Blood Coagulation System in Experimental Pulmonary Embolism (실험적 폐색전증에서 조직형플라스미노겐활성체의 투여방법이 혈액응고기전에 미치는 영향)

  • Background: As a physiologic plasminogen activator, tissue-type plasminogen activator (t-PA) could induce effective thrombolysis in massive pulmonary embolism, without the risk of systemic hemorrhage. However, therapeutic doses of t-PA has been associated with systemic lytic state, and fibrin selectivity may be influenced by the dosing regimen of t-PA. To investigate the effects of duration of t-PA infusion on blood coagulation system, we performed this study. Method: In a canine model of pulmonary embolism, which was induced by injection of autologous blood clots, we administered equal doses of t-PA (1 mg/kg) over 15 minutes in $t-PA_{15}$ group, over 180 minutes in $t-PA_{180}$ group, and only saline in control group. Then serial blood samplings were made to check complete blood count, prothrombin time, activated partial thromboplastin time, thrombin time, fibrin, plasminogen, ${\alpha}_2$-antiplasmin, coagulation factor V and VIII, and fibrin(ogen) degradation products. Results: 1) In all 3 groups, complete blood count showed same changes. Hemoglobin, hematocrit and platelet count decreased, but WBC count increased. 2) Prothrombin time, activated partial thromboplastin time, and thrombin time were prolonged during 15-60 minutes after t-PA administration in $t-PA_{15}$ group, and from 30 minutes through 180 minutes after administration in $t-PA_{180}$ gorup. 3) Fibrin, ${\alpha}_2$-antiplasmin, and cogulation factor V and VIII decreased in both $t-PA_{15}$ and $t-PA_{180}$ group, but returned to basal levels earlier in $t-PA_{15}$ group. 4) Fibrin(ogen) degradation products increased after pulmonary embolism in all groups, and further increased in both $t-PA_{15}$ and $t-PA_{180}$ groups after t-PA infusion. But more pronounced increment was noted in $t-PA_{180}$ gorup. Conclusion: In pulmonary embolism, the shorter (15 minutes) infusion of t-PA would have less risk of systemic hemorrhage than the longer (180 minutes) infusion when the doses is equal. And, this suggests that manipulating the duration of t-PA infusion can reduce the risk of major bleeding.

Changes in Characteristics of Brown Rice (Goami) Alcohol Fermentation By-Product by Cellulase (Cellulase처리에 따른 현미(고아미) 알코올발효 부산물의 특성 변화)

  • To utilize non-heat treated alcoholic by-products of brown rice (Goami) as food sources, the quality characteristics changes according to the treatment conditions of cellulase were evaluated. Results showed that the increase of hydrolysis temperature correspondingly increased the soluble solids and total sugar amounts in the by-products of Goami, and total dietary fiber amount was found to be around 0.67% Reducing sugar concentration was the highest at the hydrolysis temperature of $70^{\circ}C$. Maltooligosaccharides amounts were detected to be the highest at the hydrolysis temperature of $80^{\circ}C$ and were also, maltopentose and maltopentose were found. In the soluble solid, total dietary fiber, reducing sugar and total sugar according to the cellulase concentration, the content of hydrolysates with enzyme were higher than control, and the content of hydrolysates with enzyme was similar (6.30 and 0.69% 3,600 and 5,500 mg% respectively). The content of maltooligosaccharides was increased with the increase of enzyme concentration, and the content was similar at more than 0.6%(w/w) of enzyme concentration. The soluble solids and total dietary fiber by hydrolysis time were found to be 6.25% and 0.70%, respectively at more than 60 min. of hydrolysis. The content of reducing sugar, total sugar and maltooligosaccharides were increased with the increase of hydrolysis time, and the content was similar at more than 120min. of hydrolysis (3,800, 5,680 and 1,950 mg% respectively). Based upon these results, the byproducts of Goami are expected to be valuable as various food sources showing the highest dietary fiber and maltooligosaccharides contents by the hydrolysis at $80^{\circ}C$ for 120 min. with the addition of 0.6%(w/w) of cellulase.

Degrading and Flocculating Property of A Bacterium Isolated from the Extract of Earthworm (지렁이로부터 분리한 Bacillus pumilus JS-01 균주의 유기물 분해능 및 응집능)

  • To develop the microbial agents for the environmentally-friendly treatment and recycling of food waste, useful microorganisms, which showed higher degradation activities to various organic compounds and possessed flocculating activities, were isolated from the earthworm. One of the isolated strains, named JS-01, was further selected due to its higher flocculating activity against 0.5% Kaolin clay. JS-01 was identified as Bacillus pumilus sp. by 16s rDNA analysis. The optimal temperature and pH for the growth of JS-01 to express the flocculating activity was found to be at $37^{\circ}C$ in pH 7.0 of EPS broth medium. JS-01 also expressed good degrading activity against cellulose, which is one of the representative organic materials in food waste. We propose that JS-01 will be a good candidate for the efficient treatment of food waste and leachate due to the property to degrade cellulose and flocculating activity.

Roughage Value of Deepstacked Rice Hulls-bedded Broiler Litter in Sheep (퇴적발효 육계분의 면양에서의 조사료적 가치 평가)

  • This study, in which sheep were used as models for beef cattle, was conducted to determine the effect of replacing 100% of rice straw with deepstacked broiler Iitter(BL) as a roughage source on total tract apparent digestibility, digestible nutrient intake, ruminal and blood parameters, and N balance of sheep. Under the conventional formulated mix-rice straw(60: 40) feeding system(control), replacement of rice straw with BL resulted in similar(P> 0.05) total tract apparent digestibilities of fiber and organic matter, similar(P> 0.05) intake of digestible NDF, ADF and organic matter, and higher(P <0.05) intake of digestible crude protein. Feeding BL instead of rice straw resulted in 10wer(P <0.05) ruminal pH, higher(P <0.05) $NH^3-N$ concentration and similar(P> 0.05) rominal volatile fatty acid percentage and blood urea concentration. In a N balance study, increased(P<0.05) N intake for the BIAed group induced higher(P <0.05) quantities(g/d) of fecal and urinary N excretion, bodily N absorption, and N retention. It was concluded that deepstacked BL fiber was comparable to rice straw fiber as a roughage source and BL protein was also favorably utilized within the body of ruminant.

Quality characteristics of the bread with sprouted brown rice flour (발아현미분을 첨가한 식빵의 품질 특성)

  • Breads were prepared with the addition of sprouted brown rice flour(SBRF) as a funtional ingredient up to 50%, and their quality characteristics were evaluated through analyses of proximate composition amino acid by HPLC, fatty acid by GC, and sensory evaluation. The optimum amount of SBRF for bread preparation was 30%(w/w). The more SBRF was added, the greater the proximate components such as ash, crude protein, crude fat, fiber and amino acid content were, although the baking quality such as volume decreased. The results of sensory evaluation for bread showed that the more SBRF was added, the lower the scores of color, appearance. texture and the feelings in the mouth were, but the higher the score of taste.

Effects of Yeast Culture Supplementation on Rice Straw Digestibility and Cellulolytic Bacterial Community in the Rumen (볏짚 조사료에 대한 효모 배양물 첨가가 반추위 소화율 및 섬유소 분해균의 군락 변화에 미치는 영향)

  • In vitro and in situ incubation studies were conducted to determine effects of yeast culture supplements (Saccharomyces cerevisiae) on cellulolytic bacterial function and fiber digestion in rice straw. In vitro dry matter digestibility of rice straw gradually increased according to supplemental levels of yeast culture (0.0, 0.2, 0.4, 0.6, 0.8 and 1.0%). Digestibility of rice straw started to increase apparently when yeast culture was added more than 0.6% level (p<0.05). Also, we reconfirmed that in vitro dry matter digestibility was significantly increased by 0.6% of yeast culture addition in 4% NaOH treated and non-treated rice straws (p<0.05). When in situ dry matter digestibility was tested in Korean native goats fed basal diet or experimental diet which contained 1.0% of yeast culture, the yeast culture feeding improved in situ dry matter digestibility in both 4% NaOH treated and non-treated rice straws (p<0.05). In case of real-time PCR monitoring cellulolytic bacterial function, the bacterial population attached on rice straw showed the increasing trends with higher level of yeast culture spraying on rice straw. F. succinogenes and R. flavefaciens were significantly increased in accordance to spraying levels of yeast culture (0.0, 0.1 and 0.3%) at both 12 and 24 hrs of in situ incubation (p<0.05). R. albus was significantly higher population in yeast culture spraying than non-soraying at 12 hrs of in situ incubation (p<0.05). These bacterial populations were showed the increasing trends with digestibility enhancement of rice straw according to the higher levels of yeast culture supplement. Overall, these results clearly suggest that the presence of yeast culture result in noticeable increase of rice straw digestion, which is modulated via good effect on cellulolytic bacterial attachment to fiber substrates.
