  • Low molecular weight endo-glucanase was partially purified from cellulase complex using Sephadex G-100 gel chromatography. Biochemical properties of the purified component was investigated. Optimum pH and temperature determined were 6.0 and 5$0^{\circ}C$, respectively. Enzymatic hydrolysis of four cellulosic substrates having varying crystallinity was evaluated. It was found that hydrolysis of amorphous region was followed by the hydrolysis of crystalline region. In order to examine the effect of adsorption of the enzyme onto the cellulosic substrates on the hydrolysis kinetics, adsorption studies were carried out. Time course of adsorption of low molecular weight endo-glucanase onto various cellulostances was observed for 25 min. The rate and amount of adsorption to amorphous cellulose was greater than those to the crystalline cellulose. This result suggested that the role of endo-glucanasc was more important to the hydrolysis of amorphous cellulose than to the crystalline region of the cellulose.

  • Decomposition rate of organic matiter in the mud flat of Sunchon Bay was estimated. Physicochemical parameters, cellulose degradation rate. distribution of heterotrophic bacteria, and extracellular enzymatic activities were measured from August 1997 to July 1998. Soil temperatures, water contents, concentration of $PO_4$-P and organic matter were -1-~$30^{\circ}C$, 42.1-53.1%, 0.0779-0.1961 mgig and 1.99-7.64%, respectively. Decomposition rate of cellulose film ranged from 7.7 to 100%imonth, high in summer and low in winter. The number of heterotrophic bacteria ranged from $0.87{\times}10^6 to 3.6{\times}10^7 $CUFsIg dq soil. Enzymatic activities of phosphatase, $\alpha$-D-gluEosidase, $\beta$-D-glucosidase and cellobiohydrolase, which were measured as decomposition rate of methylumbelliferyl(MLiF)-substrate, were 152.23-1779.80 nMIhr, 2.67-202.18 nM/hr, 5.03-258.26 M h r and 3.42-63.07 nM/hr, respectively Cellulose degradaaon rate and extracellular extracellular enzymatic activities were conelated with each other, and showed high correlation coefticiency with soil temperature.

  • Colony morphology, growth responses on some simple carbon sources and taxonomic position were established for three stock cultures of National Collection of Industrial Bacteria, Scotland. It was confirmed that NCIB 8077 belonged to the Cellulomonas species and that NCIB 8633 and NCIB 8634 belonged to the Pseudomonas species. Taxonomy of other cellulolytic bacteria published on various journals was also discussed.

  • Characteristics of TNP-cellulose which prepared from carboxymethyl cellulose powder, CM32, as substrate for cellulase activity assay were investigated. Enzymatic hydrolysis of TNP-cellulose occured on the cellulose moiety but not on amide bonds, following Michaelis-Menten kinetics. Three cellulase preparations from Trichoderma viride, Aspergillus niger, and Cellulomonas sp. were tested for their pH and temperature dependences and compared with the method determining the increase in reducing power. The enzyme activity was found to have the same temperature range in both methods, however the pH range was broadened in the case of using TNP-cellulose as substrate. The colorimetric method for cellulase assay using TNP-cellulose as substrate was compared with the other methods: one based on determination of the increase in reducing power; and the other based on determining the decrease in viscosity of Na-CM-cellulose solution. The activities measured by the colorimetric method showed a linear correlation with the enzyme concentration of certain range in all three enzymes tested, and the activity values were proportional to those obtained from the other methods. Depending on the enzyme, however, the activity values from this method were not always in proportion to those from the viscometric method. suggesting that this method was not specific for determination of the endo-type cellulase.

  • During the cultivation of Penkillium uerrmulosum in the media containing cellobiose octaacetate (COA), avicel, or KC-flock as an inducer and as a sole carbon source for 21 days, cellulolytic activity and SDS-PAGE pattern of proteins in the culture broth were investigated. Protein concentration and cellulolytic activity were highest in the COA medium. As cultivation period was increased, protein content and avicel hydrolytic activity of culture broth were increased as similar extent but neither $\beta$-glucosidase nor CMC hydrolytic activity was correlated to protein content. When crude proteins from the culture broth were separated on DEAE column by HPLC, distribution of avicel-hydrolytic activities were well correlated with that of major proteins. From those results it was suggested that three major proteins having 60 K, 68 K, and 76 K of Mr. were avicel-hydrolytic enzymes.

  • The patterns of fungal growth and fiber digestion under the microscope, and tile productions of fibrolytic enzyme were studied in an in vitro culture with Neocallimastix frontalis SA when either filter paper or rice straw was provided as sole energy source. Under the microscopic observation, active zoospores attachment, sporangium development and complex rhizoidal system were founded on the surface and at the edge of filter paper. After 7 days of incubation, a reduced fiber mass, a decreased fiber cohesion and a weakened fiber structure by fungal digestion were clearly observed. Similar fungal development was observed with rice straw, but fungal growth and digestion took place mostly on the damaged and exposed portion of rice straw. Although there were some differences in absolute concentration and pattern, the concentration of both cellulase and xylanase increased with incubation time with the higher activity being obtained with filter paper. Their differences were large especially after 48 and 96hr of incubation(P< 0.05). The filter paper was more good inducer of cellulolytic and xylanolytic enzymes compared with complex substrate, rice straw. These findings suggest that the filter paper is the better energy source for N frontalis than the complex substrate, and structural disintegration by physical process is able to help rumen fungal growth on the lignified roughage although anaerobic rumen fungi have mechanical and enzymatic functions for fiber digestion.

  • In this study, five different Lentinula edodes cultivar (Chamaram, Sanbaekhyang, Sanjo 713ho, Sanjo 715ho, Sanjo 718ho) were evaluated for their ability to decolorize Remazol Brilliant Blue R (RBBR) in MEB medium, respectively. Chamaram and Sanjo 713ho decolorized RBBR rapidly in MEB medium within 3 and 5 days. The activities of manganese peroxidase (MnP) and laccase were determined on the MEB medium with and without lignin. Sanjo 713ho resulted the highest ligninolytic enzyme activities on incubation day 1, indicating of 1,213 U/mg of MnP activity and 1,421 U/mg of laccase activity.

  • The negative effects of unburned fabric in municipal solid waste incinerator (MSWI) ash on the performances of cement composite were investigated. The chemical and physical characteristics of MSWI ash powder containing high volume of unburned fabric were analyzed. The workability and compressive strength of mortar-type cement composites with the ash powder were evaluated. The workability of the cement composites was decreased mainly by unburned fabric in the ash, while the compressive strength was decreased by MSWI ash itself. From the experimental results, the critical limits for both MSWI ash and their powder containing unburned fabric in the cement composites should be limited from standpoint of the workability of the composite.
