Somatic Embryogenesis from Filament-Derived Callus of Paeonia lactiflora PALL. (작약(Paeonia lactiflora Pall.)의 화사 유래 캘러스로부터 체세포배발생)

  • This study was conducted to investigate the possibility of obtaining platelets via somatic embryogenesis as a means of in vivo mass propagation in Paeonia lactiflora Pall.. When cultured on MS medium with 0.5 mg/L 2, 4-D, filament explants formed calli. UPon transfer to basal medium the calli gave rise to somatic embryos at a frequency of 38%. Additional of 3g/L activated charcoal enhanced the frequency to 60%. Mature embryos preheated at 5$^{\circ}C$ for 2 weeks had 37% germination on medium with 0.3 mg/L GA$_3$. The germinated embryos developed to complete plantlet when cultured on medium with 2mg/L BA.

  • The objective of this paper is to compare and summarize the procedure and effectiveness of some bioassay systems and to point out ways to obtain reliable results from each bioassay. Detailed C:escriptions were given for those widely-adapted bioassay methods, such as mungbean rooting (auxin), Avena first internode straight growth (auxin), dwarf rice growth (gibberellin), dwarf pea epicotyl elongation (gibberellin), radish cotyledon expansion test (cytokinin), and tobacco stem pith callus growth (cytokinin), and the effects of various plant growth regulators including some recently introduced growth retardants (Paclobutrazol, Uniconazol, etc.) were also summarized.

Effect of Plant Growth Regulators in Minimizing Low Temperature Stress in Rice(Oryza sativa L.) (식물생장조절제(植物生長調節劑) 처리(處理)가 벼(Oryza sativa L.) 냉해경감(冷害輕減)에 미치는 영향(影響))

  • This experiment was conducted at Andong county cropping season to investigate rice response and consequent changes in the physiological activities and agronomic characters of rice as affected by growth regulator treatments, and to determine how low temperature controls some plant growth mechanisms. Plant growth regulators, particularly ABA, applied to 15 DAT regardless of concentration, gave higher tar plant height and tiller number after 10days of low temperature treatment than did the untreated plant. However, the differences in plant height and tiller number among treatments diminished to plant growth. Chlorophyll content seemed to be enhanced by growth regulators under low temperature condition. The 10-day treatment of low temperature decreased chlorophyll content by more than 17% compared with the untreated control. The oxidizing activity of roots decreased sharply to 52.2a after 10 days in low temperature condition. On the other hand, ABA at concentration of $10^{-4}M$, highly attains oxiding activity of roots. Generally, plant growth regulators applied at 15 DAT under low temperature reduced grain yield more than did the untreated control which can be attributed to decreased yield components. When growth regulators were applied to booting stage, the tiller number did not show significant difference, whereas the significant difference in culm length at harvest was observed in all treatment as a result of different concentration of growth regulators. The growth regulators generally exhibited a significant effect on yield component except on panicle length. Grain yield of low temperature treated plot at booting stage was significantly influenced by application of growth regulators. This was due to decreased number of spikelet and low filled spikelet percentage. However, TIBA treatment at booting stage showed no significant differences in grain yield.

Effect of Seeding Rates and Growth Regulator Application on Lodging Resistance and Yield in Direct Seeding on Flooded Paddy Surface (벼 담수표면직파재배에서 파종량과 생장조절제 처리에 따른 내도복성과 수량성)

  • Experiments were conducted to effect of seeding rates and growth regulator (KIM-112) application on lodging resistance and yield in direct seeding on flooded paddy surface. The number of seedlings per $m^2$ were ranged from 61 to 143, and seedling ratios were from 71.7 to 76.1%. The culm length was increased with the higher seeding rates. The culm length was significantly shortened by KIM-112 application, and the shortened effect of internodes were various with application times. The leaf angles with the culm and top leaves were errected and position of light reception by KIM-112 application, also the weight of culm base was increased. The bending moment at breaking of culm and culm diameter were decreased according to the higher seeding rates. The number of panicles per $m^2$ were increased according to the higher seeding rates. The ripened grain rate and 1, 000 grains weight became higher than the untreated control.

  • Three growth regulators, TIBA (2. 3. 5-Triiodobenzoic acid). ABA(Abscisic acid) and DGLP were sprayed on soybean plants sown on April 25 and May 10 to investigate those effect on growth and yield of Hwangkeumkong cultivar. TIBA or ABA reduced stem length. and lodging. however. increased stem diamater podding rate. number of pods and seeds per plant, and seed yield. Among 3 growth regulators TIBA was most effective to healthy growth and to increase of seed yield. Optimum treatment method for healthy plant growth and higher grain yield was 2-3 times spray with 5-day interval from 6 leaf stage (V6) of soybean plants. Soybean seed yield in the plot of TIBA treatment with 3 times from 6 leaf stage was 20% higher both in early and ordinary seeding field than those of non-treatment plots.

  • 원예작물의 개화는 일장이나 생장조절제 처리 이외에도 광도, 관수량, 그리고 시비수준 등 재배환경에 따라 영향을 받는다. 차광처리는 광합성률을 낮추어 개화를 지연시키며, 관수처리는 건조시 양분흡수가 저해되거나, 수분부족 자체가 스트레스로 작용하여 종자생산을 촉진시키는 생식생장으로의 전환을 유도할 가능성도 있다. 쑥갓은 국내에서 엽채류로 4계절을 통해 많이 이용되나 개화생리에 대한 연구가 미비하다. (중략)

Effects of Bedsoil and Growth Regulator on Cutting Propagation of Cudrania tricuspidata Bureau (상토 및 생장조절제 처리가 꾸지뽕나무의 삽목번식에 미치는 영향)

  • This Study was carried out to select bed soil and growth regulator which are effective for the macro propagation by cutting of Cudrania tricuspidata Bureau. The rooting of Cudraniae tricuspidata showed good result when the bed soil was coarse soil, rooted hardwood cutting 56.9%, and softwood cutting 62.3%. The rooting of Cudraniae tricuspidata showed good result when treated by growth regulator IBA 100 ppm, rooted hardwood cutting 57.9%, and softwood cutting 89.2%.

  • The effect of nutrient concentration and plant growth regulators on rooting of Coleus stem cuttings were investigated. In contrast to sand cutting, the nutrient solution or plant growth regulator treatment increased the root numbers and root length. The concentration of nutrient solution were 1, l/2 and 1/4 strength of the balanced nutrient solution developed by Japanese Horticultural Experimental Station and the highest root growth was obtained from 1/4 strength nutrient concentration. NAA or IBA treatment in culture solution stimulated the rooting and 0.01mg/l of NAA or IBA was effective in root development. Furthermore, days for rooting were shortened and root growth were promoted by mixing nutrient solutions with plant growth regulators and by using cutting with leaves. The rooting of Coleus was observed under microscope and the adventitious root formation initiated at 3 days after cutting with nutrient and auxin mixture.

  • This study was carried out to investigate the effects of plant growth regulators on the growth and development of pot 'Pinto Rose' and 'Pinto Scarlet' geraniums. Plant height was increased by $GA_3$ treatments in both 'Pinto Rose' and 'Pinto Scarlet' geranium. However, the number of branches was not affected by $GA_3$ and daminozide treatments. The combined treatments of $GA_3$ and daminozide delayed the days to first flowering in 'Pinto Scarlet' geranium. Also, in 'Pinto Rose', $GA_3$ or the combination of $GA_3$ and daminozide treatments delayed the first flowering time. 'Pinto Scarlet' geranium was more sensitive to growth regulator treatments than 'Pinto Rose'. The number of flower stalks and branches were increased by uniconazole in 'Pinto Scarlet', while the flower stalk length and canopy diameter were severely decreased by uniconazole treatments in both cultivars. The days to first flowering tended to be delayed by all growth retardant treatments in geranium.

  • The experimental growth regulators Hoe78784 and PP-333 significantly reduced lodging in all rices. However, they did not significantly affect the crop vegetative characteristics and yield components, except by decreasing plant height when both growth regulators were applied at booting stage. Grain yield of IR21820-154-3-2-2-3 increased due to reduced lodging when Hoe78784 and PP-333 were applied at booting. These growth regulators increased the thickness and diameter of the culm. Applying CCC did not affect lodging in all test varieties. CCC-treated varieties and the control showed lower number of vascular bundles and culm thickness and diameter of the third and fourth internodes. Hoe78784 and PP-333 significantly reduced internode length from the second to the third internode in short-statured varieties. In tall varieties. reduction extended up to the fourth internode. Culm N content of the varieties did not significantly differ among treatments, except in IR8. Culm P content also did not differ significantly among treatments. although K and Si content was the highest than control when Hoe78784 was applied.
