A Study on the Artificial Intelligence-Based Soybean Growth Analysis Method (인공지능 기반 콩 생장분석 방법 연구)

  • Soybeans are one of the world's top five staple crops and a major source of plant-based protein. Due to their susceptibility to climate change, which can significantly impact grain production, the National Agricultural Science Institute is conducting research on crop phenotypes through growth analysis of various soybean varieties. While the process of capturing growth progression photos of soybeans is automated, the verification, recording, and analysis of growth stages are currently done manually. In this paper, we designed and trained a YOLOv5s model to detect soybean leaf objects from image data of soybean plants and a Convolution Neural Network (CNN) model to judgement the unfolding status of the detected soybean leaves. We combined these two models and implemented an algorithm that distinguishes layers based on the coordinates of detected soybean leaves. As a result, we developed a program that takes time-series data of soybeans as input and performs growth analysis. The program can accurately determine the growth stages of soybeans up to the second or third compound leaves.

Correlation of A Mating Type with Mycelial Growth Rate in Basidiospore-derived Monokaryons of Lentinula edodes (표고 담자포자 유래 단핵균사의 A 교배형과 생장 속도 상관관계)

  • Lentinula edodes is a tetrapolar basidiomycete and its mating type is determined by two unlinked genetic loci, A and B. Theoretically, one dikaryotic strain could produce basidiospores with four different mating types in a 1:1:1:1 ratio. Previous studies have described the skewed segregation ratio of mating types among basidiospores of L. edodes. However, they were based only on morphological characteristics, such as clamp connection, to determine mating types. To clarify whether the segregation distortion of mating types is a general phenomenon in L. edodes, we analyzed the mating types of basidiospores obtained from three cultivars of L. edodes using recently developed DNA markers. We found that the skewed segregation of mating types was strain-specific, as reported previously. Among the three cultivars, one cultivar showed balanced segregation, while the other two displayed distorted segregation. We also examined the relationship between mating type and mycelial growth rate of monokaryons derived from each basidiospore. It was found that the monokaryotic mycelial growth rate was related to the A mating type but not to the B mating type. Therefore, homeodomain transcription factor genes that reside on the A locus or other genes linked to the A locus affect the growth rate of monokaryotic mycelia. Considering the importance of mating types in mushroom breeding, this study is informative for establishing an efficient breeding strategy as well as for understanding the mechanism of monokaryotic mycelial growth.

Isolation and characterization of Auxin/indole-3-acetic acid 1 (Aux/IAA1) gene from poplar (Populus alba × P. glandulosa) (현사시나무에서 Auxin/indole-3-acetic acid 1 (Aux/IAA1) 유전자 분리 및 발현 특성 구명)

  • Auxin plays a crucial regulatory role in plant growth and development processes. Three major classes of auxin-responsive transcription factors controlled by the Auxin/indole-3-acetic acid (Aux/IAA), Gretchen Hagen 3 (GH3), and small auxin up RNA (SAUR) genes regulate auxin signaling. Aux/IAA, in particular, encodes short-lived nuclear proteins that accumulate rapidly in response to auxin signaling. In this study, we isolated a PagAux/IAA1 gene from poplar (Populus alba ${\times}$ P. glandulosa) and investigated its expression characteristics. The PagAux/IAA1 cDNA codes for putative 200 amino acids polypeptide containing four conserved domains and two nuclear localization signals (NLSs). Utilizing Southern blot analysis, we confirmed that a single copy of the PagAux/IAA1 gene was present in the poplar genome. The expression of this gene is specific to leaves and flowers of the poplar. PagAux/IAA1 expressed in the early exponential growth phase of cell-cultured in suspension. PagAux/IAA1 expression level reduced in drought and salt stress conditions, and the presence of plant hormones such as abscisic acid. However, expression enhanced in cold stress, cambial cell division, and presence of plant hormones such as gibberellic acid and jasmonic acid. Thus, these results suggest that PagAux/IAA1 participates in cold stress response as well as developmental processes in the poplar.

Photosynthetic Characteristics and Growth Performances of Containerized Seedling and Bare Root Seedling of Quercus acutissima Growing at Different Fertilizing Schemes (시비 처리에 따른 상수리나무(Quercus acutissima) 용기묘와 노지묘의 광합성 및 생장특성)

  • This present study was conducted to investigate photosynthetic characteristics, chlorophyll fluorescence, chlorophyll contents, and growth performances of containerized seedling and bare root seedling of Quercus acutissima growing under different fertilizing schemes. Both of containerized seedling and bare root seedling of Quercus acutissima showed good photosynthetic capacity and apparent quantum yield in the seedlings applied with one gram refining fertilizer (each of N, P, and K is 19%) diluted in one liter water. And chlorophyll contents also showed higher in the seedlings applied with the above fertilization scheme. The seedlings also showed good relative growth performances of root collar diameter, seedling height, total biomass, and leaf area in the some treatment scheme. In most of the treatments, containerized seedlings showed better photosynthetic capacity, apparent quantum yield, chlorophyll contents, and growth performances than bare root seedlings.

Effects of Fertilization Treatments on Growth of Container and Bare Root Seedlings of Pinus densiflora (시비처리가 소나무 용기묘와 노지묘의 생육에 미치는 영향)

  • This study was conducted to investigate effects of fertilization treatments (non-fertilizing, deep-fertilizing 20g, 50g, 100g, and surface-fertilizing 20g) on survival rate, growth performances, and seedling quality index (SQI) of container seedling and bare root seedling of Pinus densiflora in the field. There were no significantly differences in survival rate among fertilization treatments in the field. Besides, there was no toxic effect on seedling by over 100g fertilization in deep-fertilizing treatment. The root collar diameter and height of P. densiflora in both seedling types were the highest at 100g fertilization in deep-fertilizing treatments, and the biomass products and SQI were the same as above growth of root collar diameter and height. In most of the treatments, container seedlings showed better growth performances than bare root seedlings. In optimal fertilization, effect of fertilization was higher in container seedling than bare root seedling.

Identification of Key Metabolites Involved in Quantitative Growth of Pinus koraiensis (잣나무의 생장특성과 관련있는 주요 대사물질 인자 구명)

  • A metabolomic study was conducted to identify key metabolic components, which are correlated with the growth of 4-year-old Pinus koraiensis seedlings harvested at actively height growing season (May 18th). Among 105 individual metabolites identified by GC/MS analysis, alanine, threonine, oleic acid, and butanoic acids were negatively correlated with both height and weight of 4-year-old seedlings, while malic acid, xylose, glucose, d-turanose and inositol had positive correlation with various growth parameters. During the actively growing season, the concentrations of both amino acids and organic acids in the main stem of Superior seedling group were lower but the photosynthates such as mono-saccharide and sucrose were higher than in other seedling groups such as Intermediate and Inferior. Interestingly, d-turanose, an analogue of sucrose that is not metabolized in higher plants but used as carbon source by many organisms including numerous species of bacteria and fungi, showed the highest correlation (r=0.896, p<0.001) with height of 4-year-old seedlings, indicating that possible interaction with mycorrhizal organisms. Therefore we suggest that several metabolites selected in this study may be used as metabolic markers for complex traits in P. koraiensis.

Effects of the Repression of sphpr1 Expression on Growth and mRNA Export in Fission Yeast (분열효모에서 sphpr1 유전자의 결실이 생장 및 mRNA Export에 미치는 영향)

  • THOC1/Hpr1 is one subunit of THO complex that is an evolutionally conserved assembly involved in the mRNP packaging and mRNA export during transcription elongation. In fission yeast Schizosaccharomyces pombe, an ortholog (spHpr1) of THOC1/Hpr1 was identified based on sequence alignment. A deletion mutant in a diploid strain was constructed by replacing one of spHpr1-coding region with a $kan^r$ gene using one-step gene disruption method. Tetrad analysis showed that the sphpr1 is essential for growth. Over-expression of sphpr1 from strong nmt1 promoter caused no defects of growth and mRNA export. However, repression of the sphpr1 expression resulted in growth inhibition accompanied by accumulation of poly$(A)^+$ RNA in the nucleus. These results suggest that spHpr1 is involved in mRNA export from the nucleus to cytoplasm.

Characteristics of Mycelial Growth for Artificial Cultivation in Termitomyces albuminosus (흑피흰개미버섯(Termitomyces albuminosus) 인공재배를 위한 균사배양 특성)

  • The investigation of mycelial growth for artificial cultivation in T. albuminosus determined that within PDA, YM, MEA medium, both KNCAF 9001 and 9002 strains showed the highest mycelial growth in MEA media with 89, 81 mm, respectively, followed by YM, PDA in respected order. Furthermore, optimal mycelial growth of 89 and 77 mm in 9001 and 9002, respectively, at 25 ℃. Mycelial growth rapidly decreased at 30 ℃, but it was confirmed that it survived at 40 ℃. At pH 6, mycelial growth was fast with growth of 89 and 88 mm for both 9001 and 9002, respectively. In the case of 9001, the addition of with cotton seed hull(80%) and rice bran (20%) each other resulted in maximum productivity of 99 mm/10 days. The result differed in order of corn cob(97 mm), oak sawdust(96 mm) and poplar sawdust(93 mm). Ultimately, the result confirmed that addition of rice bran in comparison to bran resulted in higher yield. Even with identical medium component, 9002 showed the same tendency as 9001 but its growth was relatively slower than 9001. The effect of mycelial growth on the moisture content of the medium was best at 101. 94 mm/10 days when the moisture content of 9001 and 9002 was 65%. Mycelial growth rate tended to be slower when the moisture content was higher or lower than 65%.

Characteristics of Germination and Early Growth of Parasenecio firmus in Container by Shading Treatment (차광처리에 따른 병풍쌈의 용기 내 발아 및 초기생장 특성)

  • This study was carried out in order to investigate the germination and the early growth characteristics of Parasenecio firmus. Seed pre-treatment before the each experiment was carried out by shading treatment (with drying at room temperature (DRT), drying at low temperature (DLT) and water soaking (WS) for 48 hours). Experiment was performed by shading treatment (full sunlight, 35%, 50%, 75%, and 95% shading). Seeds of Parasenecio firmus were surveyed the highest germination rate (61.1%) in full sunlight with WS (overall 25.7~61.1%). Height was surveyed the highest under 95% shading. And root collar diameter was surveyed the highest in full sunlight. Fresh weights (leaf, shoot, root and total) were the highest under 50% shading. Dry weights (leaf, shoot and total) were the highest under 75% shading. It was indicated the lowest leaf, shoot, root and total under 95% shading. Leaf growth (leaf area, leaf length, leaf width and leaf thickness) and root growth (total root length, root project area, root surface area, root diameter and root volume) were good under 35%~75% shading, but the lowest under 95% shading. As a result of surveying the whole experiment, seed of Parasenecio firmus grows well under 50%~75% shading after germination by high sunlight with water soaking (WS).

Monitoring Soil Characteristics and Growth of Pinus densiflora Five Years after Restoration in the Baekdudaegan Ridge (백두대간 마루금 복원사업지에서의 5년 경과 후 토양특성 및 소나무 생장 모니터링)

  • This study was conducted to monitor the soil characteristics and growth of Pinus densiflora and to determine the effect of soil characteristics on growth rate five years after an ecological restoration project in Baekdudaegan ridge including Ihwaryeong, Yuksimnyeong, and Beoljae sites. The ecological restoration project was executed with the forest of P. densiflora in 2012-2013. In April 2018, we collected soil samples from each site and measured the height and the diameter at breach height (DBH) of P. densiflora. Although there was no significant change of soil pH compared to the early stage of restoration (one year after the project), it was high in Ihwaryeong, and Beoljae with values of 7.7 and 6.4, respectively. Also, the organic matter decreased by 70-80%, and the available phosphorus (P) was unchanged in three restoration sites. The decreased organic matter can be attributed to restriction of inflow and thus decomposition of litter in the early stage after the restoration. The tree height growth rate ($m\;yr^{-1}$) of P. densiflora in Yuksimnyeong was the highest at 1.02, followed by Beolja at 0.75 and Ihwaryeong at 0.17. The height growth rate showed negative relationships with soil pH and cations, including Na and Ca concentrations and a positive relationship with available phosphate. The low growth rate in the Ihwaryeong site, in particular, might result from the poor nutrient availability due to high soil pH and the decrease in water absorption into the root due to high Na and Ca concentrations. The substantial reduction of organic matter after five years indicates that the need for soil improvement using chemical fertilizer and biochar.
