Effects of Dietary Mugwort (Artemisia iwayomogi Kitamura) Powder Supplementation on Growing Performance in Pig (인진쑥(Artemisia iwayomogi Kitamura) 분말 급여가 돼지 생산성에 미치는 영향)

  • This study was conducted to investigate growth performance in weanling and growing pigs supplemented with mugwort powder as an antibiotic replacement. To examine the effects of antibiotic replacement, 0 (control, with and without antibiotics), 1, and 1.5% mugwort powder was supplemented into the basal diet. Pigs raised with a diet of 1.0% mugwort powder had improved average daily gain and feed conversion rate during 23~37 d feeding. During 40~59 and 63~97 d feeding periods, there were no differences between average daily gain in pigs fed no antibiotics and those given a 1% mugwort powder diet, whereas feed conversion rate of pigs given a 1.5% mugwort powder diet and average daily gain of pigs fed no antibiotics were lower than those of any other diet group. In conclusion, this study suggests that the 1.0% supplementation of mugwort in place of antibiotics is an invaluable feed additive as a physiologically activated material.

Improvement Effect of Cordycepin-Enriched Cordyceps militaris JLM 0636 Powder against Orotic Acid-Induced Fatty Liver in Rats (Cordycepin 고함유 동충하초(Cordyceps militaris JLM 0636)의 orotic acid 유발 흰쥐의 지방간 개선효과)

  • The concentration of cordycepin produced by crossbred Cordyceps militaris JLM 0636 (CM${\alpha}$) was 7.42 mg/g dry weight, which was 7-fold higher than that of C. militaris (CM). However, cordycepin was not detected in Paecilomyces japonica (PJ). The improvement effects of CM${\alpha}$, CM and PJ on orotic acid (OA)-induced fatty liver in male Sprague-Dawley rats was investigated. Rats were randomly divided into five groups (n=6) based on five dietary categories: normal (N), OA control (C), OA plus 3% (w/w) PJ (PJ), OA plus 3% CM (CM), and OA plus 3% CM${\alpha}$ (CM${\alpha}$). OA treatment induced the retardation of body weight gain and enlargement of the liver. The concentration of hepatic triglyceride was markedly increased by OA-feeding, whereas this rise was significantly reduced by simultaneous feeding of OA, PJ, CM, and CM${\alpha}$, and this phenomenon was more pronounced by cordycepin-enriched CM${\alpha}$. The contents of total lipid, triglyceride, and free fatty acid in the serum were significantly or slightly lower in the OA control group than those of the N group, but there were no significant differences among the OA treatment groups. The hepatocytes in the OA-induced fatty liver contained numerous large lipid droplets, but PJ, CM, and CM${\alpha}$ feeding prevented the OA-induced lipid droplet accumulation in the hepatocytes. This effect was more pronounced in cordycepin-enriched CM${\alpha}$ than in PJ or CM in OA treatment rats. Accordingly, cordycepin-enriched CM${\alpha}$ could be an ideal candidate material as a dietary supplement in healthy functional foods to improve the effects of fatty liver.

Comparative Molecular Phylogenetic Relationships in Different Strains of Pleurotus spp. (느타리속 버섯 계통의 분자생물학적 유연관계의 비교연구)

  • Pleurotus spp. have been used for edible and medicinal purposes in Asian countries for a long time. The fruiting bodies of the Pleurotus ostreatus, Pleurotus citrinopileatus and Pleurotus salmoneostramineus contained many physiologically beneficial substances for human health. Therefore, it is necessary to study the genetic diversity of Pleurotus mushroom cultivars commercially cultivated in Korea. Eleven strains of Pleurotus spp. were collected from different geographical regions in South-East Asia and ITS regions of rDNA and RAPD of genomic DNA were analyzed. The size of the ITS1 and ITS2 regions of rDNA from the different strains varied from 167 to 254 bp and 156 to 213 bp, respectively. The sequence of ITS1 was more variable than that of ITS2, and the 5.8S sequences were identical. A phylogenetic tree based on the ITS region sequences indicated that selected strains could be classified into 4 clusters. Eleven Pleurotus species were also analyzed by RAPD with 20 arbitrary primers. Ten of these primers were efficiently amplified the genomic DNA. The number of amplified bands varied with the primers and strains, with polymorphic fragments in the range from 0.1 to 2.0kb. The results revealed that genetic diversity of selected strains of P. ostreatus, P. citrinopileatus and P. salmoneostramineus is low.

Evaluation of Database Comparison Methods for 18F-FDG Brain PET/CT (18F-FDG Brain PET/CT 검사를 위한 데이터 비교 방법의 평가)

  • Purpose Various database comparison methods(DCM) are used for analyzing functional neuro-imaging. It is possible to statistically evaluate decreased or increased metabolism of abnormal patient's brain by comparing with asymptomatic controls in DCM. And results of DCM are additionally used for easily explaining defect region. The aim of this study was to evaluate usefulness of statistical parametric mapping(SPM) and scenium. Materials and Methods Data of 15 patients($62.02{\pm}15.03year$) underwent $^{18}F-FDG$ brain PET/CT were collected and analyzed. Biograph TruePoint 40 with TrueV, (Siemens) was used as a PET/CT scanner. Scenium(version 4.0) in Syngo.via(version VA30A) and SPM99 were applied for statistical evaluation. Consistency between PET reading and result of DCM were evaluated by 5 nuclear medicine physicians through a questionnaire survey. SUV and SD changes were evaluated by changing iteration, gaussian filter and matrix size in scenium. And average required time for generating result of SPM99 and scenium was compared by 3 medical technologists. Results Consistency from the result of SPM99 and scenium showed 84% and 92.4% compare to PET reading. When iteration 4, FWHM 8 and matrix size 168, SUV and SD were decreased by 0.59%, 8.73%, 4.69%, 20.38% and 0.88%, 8.25% respectively compare to routine parameter(iteration 8, FWHM 2 and matrix size 336) of scenium. Average required time of SPM99 and Scenium took 282 seconds and 116 seconds to generate result. Conclusion Results of SPM99 and Scenium showed high consistency compare to PET reading. Various parameters can be controled by user when using SPM. However, normal database needs to be acquired. And it takes significant amount of time and effort for the first set up. On the other hand, Scenium provides normal database even though modifiable parameters are limited. Therefore, more informations could be provided for brain PET/CT if properly understanding and selecting each DCM.

Pathophysiological Regulation of Vascular Smooth Muscle Cells by Prostaglandin F2α-dependent Activation of Phospholipase C-β3 (Prostaglandin F2α 의존적 phospholipase C-β3 활성화에 의한 혈관평활근세포의 병태생리 조절 연구)

  • Atherosclerosis is an obstructive vessel disease mainly caused by chronic arterial inflammation to which the proliferation and migration of vascular smooth muscle cells (VSMCs) is the main pathological response. In the present study, the primary responsible inflammatory cytokine and its signaling pathway was investigated. The proliferation and migration of VSMCs was significantly enhanced by the prostaglandin $F_{2{\alpha}}$ ($PGF_{2{\alpha}}$), while neither was affected by tumor necrosis factor ${\alpha}$. Prostacyclin $I_2$ was seen to enhance the proliferation of VSMCs while simultaneously suppressing their migration. Both prostaglandin $D_2$ and prostaglandin $E_2$ significantly enhanced the migration of VSMCs, however, proliferation was not affected by either of them. The proliferation and migration of VSMCs stimulated by $PGF_{2{\alpha}}$ progressed in a dose-dependent manner; the $EC_{50}$ value of both proliferation and migration was $0.1{\mu}M$. VSMCs highly expressed the phospholipase isoform $C-{\beta}3$ ($PLC-{\beta}3$) while others such as $PLC-{\beta}1$, $PLC-{\beta}2$, and $PLC-{\beta}4$ were not expressed. Inhibition of the PLCs by U73122 completely blocked the $PGF_{2{\alpha}}$-induced migration of VSMCs, and, in addition, silencing $PLC-{\beta}3$ significantly diminished the $PGF_{2{\alpha}}$-induced proliferation and migration of VSMCs. Given these results, we suggest that $PGF_{2{\alpha}}$ plays a crucial role in the proliferation and migration of VSMCs, and activation of $PLC-{\beta}3$ could be involved in their $PGF_{2{\alpha}}$-dependent migration.
