Effects of Dangkwisoo-San, Ginger and Curcumin on Transient Receptor Potential Melastatin 7 Channels (당귀수산, 생강, 커큐민의 대사성 질환과 관련된 일과성 수용체 전압 이온통로조절에 관한 연구)

  • Objectives: Metabolic syndrome is correlated with increased cardiovascular risk and characterized by several factors, including visceral obesity, hypertension, insulin resistance, and dyslipidemia. Several members of a large family of nonselective cation entry channels, e.g., transient receptor potential (TRP) melastatin 7 (TRPM7) channels have been associated with the development of cardiovascular diseases. The purpose of this study was to investigate the effects of Dangkwisoo-san, ginger and curcumin on TRPM7 channel. Methods: Human embryonic kidney (HEK) 293 cells stably transfected with the TRPM7 expression vectors were maintained in Dulbecco's modified Eagle's medium (DMEM) supplemented with 10% fetal bovine serum (FBS), 1% penicillin/streptomycin, $5{\mu}g/mL$ blasticidin, and 0.4 mg/mL zeocin in a humidified 20% $O_2$/10% $CO_2$ atmosphere at $37^{\circ}C$. Whole-cell patch clamp recordings were obtained using an Axopatch 700B amplifier and pClamp v.10.4 software, and signals were digitalized at 5 kHz using Digidata 1422A. Results: Dangkwisoo-san extract (100, 200, 300, 400, and $500{\mu}g/mL$) inhibited the outward and inward TRPM7 whole-cell currents at dose dependent manner and the half maximal inhibitory concentration $(IC)_{50}$ of Dangkwisoo-san was $218.3{\mu}g/mL$. Also, ginger extract (100, 200, 300, 400, and $500{\mu}g/mL$) inhibited the outward and inward of TRPM7 whole-cell currents in a dose dependent manner and the $IC_{50}$ of ginger was $877.2{\mu}g/mL$. However, curcumin had no effects on TRPM7 whole-cell currents. Conclusions: These results suggest that both Dangkwisoo-san and ginger have good roles to inhibit the TRPM7 channel, suggesting that Dangkwisoo-san and ginger are considered one of the candidate agents for the treatment of metabolic syndrome such as cardiovascular disease.

Conservation strategies and vegetation characteristics of Echinosophora koreensis of Korean endemic plants in DMZ (DMZ 일원 특산식물 개느삼의 생태적 특성 및 보전 방안)

  • 본 연구는 한국 특산식물이고, IUCN Red List의 EN(위기종) 등급에 속하는 개느삼(Echinosophora koreensis)을 대상으로 자생지 분포, 식생구조 등의 생태적 특성과 현지 내외 보전 방안을 제시하기 위해 수행되었다. 개느삼의 자생지 분포 조사 결과 강원도 양구군 13곳, 인제군 3곳, 춘천시 2곳, 홍천군 1곳 총 19곳에 분포하는 것을 확인하였다. 개느삼 자생지의 해발고도는 169-711m에 분포하는 것으로 나타났고, 평균 해발고도는 375m 인 것으로 조사되었다. 개느삼 자생지의 면적 조사 결과, $8,000-734,000m^2$인 것으로 분석되었고, 평균 $202,789m^2$으로 조사되었다. 개느삼 자생지의 입지환경 분석을 수행한 결과, 자생지 내 우점종은 교목층에서 소나무가 대부분 우점하였고, 아교목층에서는 신갈나무가 우점하는 것으로 분석되었다. 관목층은 생강나무가 우점하였고, 초본층은 모든 조사구에서 개느삼이 우점하는 것으로 분석되었다. 개느삼 자생지의 식생군집분석 결과, 신갈나무, 소나무, 굴참나무 우점 군집 3개 군집으로 분류되었다. 개느삼 자생지를 대상으로 관속식물상을 조사한 결과, 총 82과 204속 289종 4아종 43변종 5품종 341분류군으로 확인되었다. 개느삼의 출현지점을 이용하여 자생지 분포역 예측 model 중 하나인 MaxEnt 분석을 통하여 잠재 분포지 분석을 수행한 결과, AUC값은 0.9762로 분석되었다. 분포예측 자생지는 강원도 양구군, 인제군, 춘천시, 화천군 지역에 집중되어 분포하는 것으로 나타났다.

Comparative evaluation on biological activities of Ziingiber officinale Roscoe extracted from different solvents (추출 용매를 달리한 생강 추출물에 대한 생리활성의 비교 평가 연구)

  • Objective : This study was designed to investigate anti-oxidative, anti-aging and anti-inflammatory activities of Zingiber officinale Roscoe water (ZD) and 50% ethanol extracts (ZE). Method : The antioxidant capacity of ZD or ZE was analyzed by DPPH, ABTS scavenging activities and reducing power, respectively. The anti-aging efficacy was performed with tyrosinase and collagenase inhibition activities. The anti-inflammatory activities were confirmed through inhibition of nitric oxide production and TNF-�� expression in LPS-treated Raw 264.7 cells. Result : As a result of measuring DPPH and ABTS radical scavenging activities, both ZD and ZE extracts increased in concentration-dependent manners. The ABTS radical scavenging activities of water and 50% ethanol extract were similar to that of positive control at high concentration. On reducing power, both ZD and ZE showed higher activities than the positive control at high concentration. On inhibition activities of tyrosinase and collagenase both ZD and ZE showed the large inhibition rates at low concentration. Also, the inhibitory efficacy of ZD on NO and TNF-�� production was better than ZE in RAW 264.7 cells. Conclusion : These results suggest that ZD and ZE have anti-oxidative, anti-aging and anti-inflammatory effects in vitro. Especially, ZD was more effective than ZE. Thus, Ginger extract is considered to be highly useful as a cosmetic or food material with anti-oxidative, anti-aging (prevention of whitening and wrinkle) and anti-inflammatory effects. Also these effects may vary depending on the extraction solvent.

  • This study selected 9 species of domestic native plants (Baechohyang, Red Pine, Turmeric, Ginger, San parsley seed, Bulsugam, Hwangchil, Tangja, Valerian) that have been proven effective for depression, and based on the analysis results of each fragrance component, 9 species were selected. Antioxidant (DPPH, ABTS), cytotoxicity (MTS), and anti-inflammatory (Nitric oxide) experiments were performed by combining essential oils. As a result of the analysis of fragrance components, DL-Limonene (38.44%), g-Terpinene (8.9%), Estragole (5.18%), and a-Pinene (1.73%), which were previously studied in combination oil, were identified. DPPH radical scavenging ability, which is an antioxidant activity, showed 75.4%, ABTS radical scavenging ability was 74.04%, and NO production inhibition was 33.14% at a concentration of 5uL/ml with no cytotoxicity confirmed. Through this, the effect of domestic blending essential oils on the improvement or prevention of depression is verified, and scientific efficacy and ingredient studies are conducted in a mutually cooperative manner to seek solutions to depression and provide basic data to confirm whether or not depressive symptoms are improved. want to do.

  • 꼭두서니과 커피속 식물의주요 2차대사산물인 카페인은 커피나무, 카카오, 차나무 등에서 관찰되는 알칼로이드 화합물로 다른 식물의 발아 억제, 살충 기능이 알려져 있다. 인체에 있어 카페인은 각성효과가 있어 수면 장애나 불안을 유발할 수 있으며 섭취를 중단 시 졸음, 두통 등 금단 증상을 나타나기도 한다. 최근 다양한 식물 종과 부위를 이용한 차 문화가 확산되고 있는 바, 본 연구는 시중에서 판매되고 있는 커피 2종류과 녹차 2종류를 비롯하여 꽃차 37종류, 잎차 15종류, 씨앗차 2종류, 열매차 1종류 및 뿌리차 2종류를 대상으로 메탄올 추출 후 gas chromatography-mass spectrometry를 이용하여 카페인 함량에 대한 정성, 정량 분석을 수행하였다. 대조로 사용된 시판 커피 2개사 제품은 각각 1.18 mg/g, 35.94 mg/g의 caffeine 함량을 나타냈으며 시판 녹차 티백 제품 2종은 1.3 mg/g, 3.42 mg/g의 함량을 나타내었다. 조사된 37종의 꽃차 중에는 차나무 꽃차에서만 1.50 mg/g 수준의 caffeine이 검출되었을 뿐 국내 소비가 확대되고 있는 구절초, 국화, 금계국, 금목서, 금어초, 금잔화, 노랑코스모스, 당아욱, 도라지, 동백나무, 뚱딴지, 라벤다, 마리골드(노랑), 마리골드(주황), 매화, 맨드라미, 무궁화, 백목련, 벚나무, 복사나무, 비단향꽃무, 생강나무, 수레국화, 아까시나무, 연꽃, 유채, 작약, 장미, 진달래, 천일홍, 캐모마일, 패랭이꽃, 팬지, 해당화, 홍화, 히비스커스 등 36종에서는 카페인이 검출되지 않았다. 잎차의 경우 레몬그라스, 레몬밤, 로즈마리, 뽕나무, 세인트존스워트, 스테비아, 쑥, 애플민트, 연잎, 적양배추, 조릿대, 차수국, 티트리, 파인애플민트, 페퍼민트 등 평가된 15종류 모두에서 카페인이 검출되지 않았으며, 씨앗차인 펜넬 및 작두콩, 열매차인 진피차, 뿌리차인 자색당근과 비트차에서 역시 카페인이 검출되지 않았다. 이상의 결과에 기초할 때, 시판되는 다양한 식물 종과 부위를 이용한 차의 경우 대부분 caffeine을 함유하지 않음을 알 수 있었다.

A Case Report of Post-COVID-19 Syndrome with Fatigue Treated with Saengkangsasim-tang based on Disease Pattern Identification Diagnostic System by Shanghanlun Provisions (『상한론(傷寒論)』 변병(辨病) 진단체계(診斷體系)에 근거하여 생강사심탕(生薑瀉心湯) 투여 후 호전된 피로를 주소로 하는 Post-COVID-19 Syndrome 1례)

  • Objective : Fatigue is the most common symptom in post-COVID-19 syndrome. We report the case of a patient with post-COVID-19 syndrome with fatigue treated using herbal medicine (Saengkangsasim-tang). Methods : A 64-year-old man had severe fatigue for 4 months after a severe acute respiratory syndrome-coronavirus-2 (SARS-CoV-2) infection. Saengkangsasim-tang was administered to this patient based on the disease pattern identification diagnostic system by Shanghanlun provisions (DPIDS). We used the Numeric Rating Scale (NRS) and Beck Depression Inventory (BDI) to evaluate the effectiveness of Saengkangsasim-tang for fatigue in the patient with post-COVID-19 syndrome. Results : According to DPIDS, the patient was diagnosed with the provision 157. After administration of Saengkangsasim-tang for 30days, the fatigue based on NRS and depression based on BDI were improved. Conclusions : The administration of Saengkangsasim-tang to patients with post-COVID-19 syndrome with fatigue, diagnosed according to the Shanghanlun provision 157, may be effective.

  • This study was undertaken in order to examine the effect of ginger, gerlic and red pepper, usually used as the ingredients of Kimchi, on the growth and acid production of Lact. plantarum ATCC 8014 and Lact. fermenti ATCC 9338 during incubation in the medium containing different amount of each extract. The results obtained are as follows: 1. The effects of ginger extract a. The growth of Lact. plantarum ATCC 8014 and Lact. fermenti ATCC 9338 tended to be stimulated as the amount of extract added into the medium increased in a certain amount (3.64mg soluble solid/ml). b. The pH of the culture medium of Lact. plantarum ATCC 8014 became lower to some degree and acid production tended to be stimulated but acid production of Lact. fermenti ATCC 9338 tended to be suppressed as the amount of extract added in a certain amount (3.64mg soluble solid/ml) increased. 2. The effect of garlic extract a. The growth of Lact. plantarum ATCC 8014 and Lact. fermenti ATCC 9338 tended to be stim ulated as the amount of extract added in a certain amount (31.7 mg soluble solid/ml) increased. b. The acid production of Lact plantarum ATCC 8014 was suppressed (p<0.01) and drop in pH was suppressed (p<0.05) as the amount of extract added in a certain amount increased. In case of Lact. fermenti ATCC 9338, the acid production t ended to be suppressed also. 3. The effects of red pepper extract a.   • A study was made on the effect of anti-nutritional factors found in some Korean beans : soybean, red bead, mung bean and kidney bean. Two animal experiments were conducted to investigate the nutritional value of the beans. The first experiment, in which the diet contained 15% protein from raw beans, compared the intensity of inhibition caused by methionine deficiency or a total amino acid deficiency. In the second experiment, the conditions were the same as in experiment I, except that heated beans were substituted for raw beans. Severe growth inhibition and high mortality was found in the raw kidney bean and red bean groups than in the soybean and mung bean groups. As no effect on the growth inhibition of raw bean groups was shown by methionine and protein supplementation, the inhibition could be ascribed mainly to the low feed intake and the low protein digestibility caused by toxic factors. Pancreatic enlargement was obserbed in all the raw bean groups. A increase in body weight, a decrease in mortality and a decrease in the weight pancreases were found in the heated bean groups. But the digestility of the diet and of the protein and the PER by heating did not increase as markedly as weight, except in the heated red bean groups. Even with heat treatment, the whole inhibitory action could not be eliminated.

  • To determine the effects of condiments upon Proteinase Activity, condiments such as welsh onion, garlic, ginger, black pepper, red pepper, mi-won (glutamic acid natrium), sugar, mustard and horse-radish were ground by a homogenizer, and each of them was dosed by 0%, 1%, 5% and 10% into Pancreatin Solution of 0.2% for storage at the temperature of 15 degrees Cels. The Enzyme Solution thus obtained then was measured at a certain interval of time by the Fuld Gross Method, and the following results were obtained. 1) The condiments that kept Proteinase Action of Pancreatin checked below 75% were mustard, horse-radish, red pepper and welsh onion. The centrol power of welsh onion, in particular, became stronger as storage time became longer. 2) The condiments that kept Proteinase Action of Pancreatin checked below 50% were sugar, black pepper and ginger. 3) Mi-won and garlic showed a strong checking powor over Proteinase Action at an early stage of storage, but as time passed, their control power gradually diminished to naught. In short, it may be concluded that ail of the condiments used in this experiment demonstrated their checking power over Proteinase Action.

  • We got these results after studying about modification of Gejitang(桂枝湯) that showed on sympton and prescription about Soumin(少陰人). 1. There are totally eleven prescriptions related to Gejitang appeared on Dongyi-Soose-Bowon(東醫壽世保元). Among them, one is prescription of Danguisayoktang(當歸四逆湯) in prescription of Discussion of Cold Induced Disorder(傷寒論) and another is Gejibujatang(桂枝附子湯) in Won-Myung dynasty period(元明時代). There are nine prescriptions in LeeJae-Ma(李濟馬)'s pre-scription and we can see there are many prescriptions used prescriptions related to Gejitang for Mang Yang(亡陽). 2. Pre-scriptions related to Gejitang has seven prescriptions in Extra-disease(表病證), and two prescriptions in Intra-disease(裏病證) which are analyzed as modified prescription from Gejitang. 3. Although 藿香正氣散, 香蘇散, 芎歸香蘇散, 八物君子湯 etc. are not modified prescriptions of Gejitang, We can see that its are brought up to be on the same with alternative remedy of Gejitang in each step of symptoms. 4. Symptoms are exactly classified in Soumin, the prescription which is used in different symptoms of diseases is similar in raw materials. It's because the remedy of Acending and Descending(升降) is used under the same purpose that maintain Yangnan Qi(陽暖之氣). 5. We could realize that Panax gienseng(人蔘), Cinnamomum Cassia(肉桂) and Aconitum Carrnichali(附子) are commonly important to reinforce Yangnan Qi because the more disease got worse, the more patient lost Yangnan Qi on both Extra-disease and Intra-disease in the Soumin's disease.
