Characteristics of Extracts from Stem and Shoot of Lycium chinense (구기자(拘杞子)나무 순과 줄기 추출물(抽出物)의 특성(特性))

  • The hunter values, pH, turbidity, brownness and solid yields of extracts in a shoot and stem of Lycium chinense were investigated. The results were as follows; The relationships between solid yield and turbidity according to extract solvents was not significant. The range of pH were 5.4 to 6.17 and the acidity of extracts were slitely low. Lightness of water extract was lower than that of ethanol extract, but red and yellowness of water extract was higher than that of ethanol extract. And also turbidity of water extract was higher in water extracts.

  • The extraction mode of orange-yellow pigment from Gardenia is depended upon the extraction time, extraction temperature and volume of solvent. The amounts of the extracted-pigment (C) is proportional to the log ${\theta}^{1.15}$ of extraction time $({\theta}:0{\cdots}{\cdots}{\cdots}60$ min.), the log $T^{3.73}$ of extraction temperature $(T:5{\cdots}{\cdots}{\cdots}60^{\circ}C)$ and the log $S^{3.7}$ of volume of solvent $(S:5{\cdots}{\cdots}{\cdots}50ml)$ at $18^{\circ}C$ for 10 minutes. Finally, the general emperical equation was derived as follows; C=1.15 log ${\theta}$+3.73 log T+3.7 log S-7.0

  • "The four types of temperament" (hereinafter "Sa-Sang Constitutional Medicine") is applied to not only medicine, but also various fields like management, politics, education, etc. Despite that "Sa-Sang Constitutional Medicine" is "Mind-Body Medicine" which is fundamentally based on "Controling Mind and Body", though, it was rarely used for psychotherapy in practical. Practical treatment on clinical medicine is limited to only medicine-treatment or acupuncture-therapy. However treatment on illness and pathological phenomenon suggested by "Sa-Sang Constitutional Medicine" keeps individual's nature in control, and it governs environmental facts: age, region, and socio-cultural factors: alcohol, debauchery, riches, authority(酒 色 財 權). Especially, "Moral Cultivation" in "Sa-Sang Constitutional Medicine" emphasizes the importance of properly understanding vice(邪心) and laziness(怠行) concealed in natural temperament of human being, and pursuing "Understanding Each Other(博通)" and "Upright Conduct by Oneself(獨行)". Furthermore, because "Sa-Sang Constitutional Medicine" is developing medical theory based on "Understanding Others(知人)" and "Straighten Oneself(正己)" doing try integral approach, it has great implications for the present generation that forms much of social connection. Similarly, Ken Wilber who is one of representatives of "World Philosophy" and "Integral Psychology" is suggesting "Integral Life Practice" (hereinafter ILP) - applies practical and heuristic "Integral Approach" to individual experiences. ILP is the only practical discipline for development of viable whole-area. ILP says that there are four core modules: body, mind, shadow, spirit(靈), and five auxiliary modules: morals(倫理), gender(性), work(일), emotion(情緖), relationships(關係性). These nine modules could apply mind-body treatment of "Sa-Sang Constitutional Medicine", and thereby more developed mind-body treatment would be found.

  • A bark comprises about 10 to 20 percents of a typical log by volume, and is generally considered as an unwanted residue rather than a potentially valuable resource. As the world has been confronted with decreasing forest resources, natural resources pressure dictate that a bark should be a raw material instead of a waste. The utilization of the largely wasted bark of genus Quercus grown in Korea can be enhanced by learning its anatomical structure and properties. In this paper, bark characteristics of Quercus grown in Korea are described. In bark anatomy, general features such as color of rhytidome, exfoliating form, color of periderm, arrangement of periderm, and thickness of the inner and outer hark. etc., arc discussed. Studies on the microscopic structure include sieve tube, companion cell, parenchyma, pholem fiber, ray, periderm(phelloderm, phelloogen, phellem), sclereid, and crystal, etc. The results may be summarized as follows: 1. In general characteristics of rhytidomes, exfoliating is not easy and sclereids are distint to the naked eye. Inner bark is thicker than that of outer bark except in case of Q. variabilis. 2. It is not clear to distinguish between phelloderm and phellogen in Quercus bark. The phellem is developed conspicuously in Q. variabilis but that of Q. accutissima is composed of thinwalled phellem and thickwalled stone cell. 3. Quercus Bark has sieve tube, companion cell, phloem fiber and sclereid. Sclereids of Quercus bark are the most distinguished characteristics comparing with pinus and populus. The volume percent of sclereids are higher than that of fiber. 4. Rays are 1~3 seriate, and multiseriate ranging with from 15 to 20. 5. Parenchyma cell contains two types, polygonal and druses crystal.

  • Several mutants were isolated from the parent strain of Aspergillus niger CF: the first mutant strain CF-11 was obtained by UV irradiation, and the second mutant strain CF-21 and CF-22 were from NTG (N-methyl-N'-nitroso-N-nitroso-guanidine) treatment on the CF-11. These mutants were characterized, and their enzyme and acid production on wheat bran Koji and wheat flour Koji were studied. Asp. niger CF-22 mutant appeared to be tan type which conidial heads were discolored. It's glucoamylase activity was inreased approximately two times and its ${\alpha}-amylase$ about 50 percent as compared with that of the parent strain of Asp. niger CF, when grown on wheat bran Koji under the optimal conditions. Asp. niger CF-21 mutant showed slower growth rate and poor sporulation than the wild type, although its conidial heads were not discolored. Approximately 4-fold increment in its acid production was observed as compared with the weld type. The activities of glucoamylase and ${\alpha}-amylase$ of the Asp. niger CF-22 and CF-21 mutants were higher than those of the wild type, but their protease activity was rather lower. The maximum production of glucoamylase by the Asp. niger CF-22 mutant was obtained after 2 to 3 days incubation on wheat bran at 30 to $35^{\circ}C;$ ${\alpha}-amylase$2 days incubation at 30 to $35^{\circ}C$. The maximal levels of acid production by the mutant CF-21 was appeared after 2 days incubation on wheat bran Koji, and after 3 days on wheat flour Koji at $30^{\circ}C$. Little differences in the levels of acid production were observed between on wheat bran and flour Koji.

Studies on the Raw Starch Saccharifying Enzyme from the Aspergillus niger and Its Mutants (Aspergillus niger 및 그 변이주(變異株)의 생전분당화효소(生澱粉糖化酵素)에 관(關)한 연구(硏究))

  • Aspergillus niger IFO 8541 (NRRL 3112) was investigated through a series of UV rays and N-Methyl-N'-Nitro-N-Nitrosoguanidine (NTG) treatments to induce mutants that produce highly active raw starch saccharifying enzyme, and two mutants with strong enzymatic productivity were obtained. The mutants obtained were investigated for their fungal characters, condition of enzyme production, and other activities. Furthermore, the raw starch saccharifying enzyme was purified and the characteristics of purified enzyme were studied. The results obtained were summarized as follows; 1. The color of conidial head of UV-46 mutant obtained from UV rays treatment was changed to tan type and the gelatinated starch saccharifying enzyme productivity and the raw starch saccharifying enzyme productivity increased up to twice and 1.8 times compared to the productivities of original Aspergillus niger IFO 8541 cultured on the wheat bran, respectively. 2. The conidial head color of NG-41 mutant obtained from NTG treatment became lighter than that of parent strain. The gelatinated starch saccharifying enzyme productivity and raw starch saccharifying enzyme productivity increased about 1.8 times, and twice over the Aspergillus niger IFO 8541 parent strain cultured on wheat bran, respectively. The productivity of ${\alpha}$-amylase increased about 3 times more than the parent strain. 3. Two peaks of glucoanlylase and a peak of ${\alpha}$-amylase were obtained when enzyme solution of mutants and parent strain were passed through DEAE-Sephadex A-50 column chromatography. Glucoamylase I showed only gelatinated starch saccharifying enzyme activity. However, glucoamylase II (raw starch saccharifying enzyme) showed both raw starch saccharifying enzyme activity and gelatinated starch saccharifying enzyme activity. 4. Mutant, UV-46 was strengthened in glucoamylase II productivity and mutant NG-41 was strengthened in ${\alpha}$-amylase productivity. 5. Glucoamylase II of mutants and parent strain were appeared to have the same enzymatic properties. 6. Glucoamylase II of mutants and parent strain were recognized as simple enzyme through electrophoresis. 7. The glucoamylase II crystallized showed rhombic board type. 8. The molecular weight, isoelectric point, optimum pH, and optimum temperature of the glucoamylase II crystallized were estimated as 76,000, 3.4, 3.5 and $60^{\circ}C$, respectively.

A Study on the Theory of Human Nature in Daesoon Thought (대순사상의 인간 본성론(本性論) 연구)

  • This study aims to understand human nature in Daesoon Thought. In addition, it seeks to determine the characteristics of human nature and how the theory of human nature in Daesoon Thought relates to the direction of the Basic Works and the Three Major Societal Works of Daesoon Jinrihoe. In order to understand the theory of human nature in Daesoon Thought, I have classified various theories of human nature that appear in the Confucian tradition and have analyzed and reviewed them. I used this framework of types as a methodological criterion to study the theory of human nature found in Daesoon Thought. In Daesoon Thought, human nature is defined clearly as the conscience that is honest and truthful. Also, it accepts humans' basic biological needs, the pursuit of clothing, food, and sexual desires as part of human nature. This view can be evaluated as quite similar, although not perfectly consistent, with the theory of human nature posited by Wang Fuzhi (王夫之), who defined human nature as a combination of morality and biological desires. The theory of human nature in Daesoon Thought emphasizes human morality, but at the same time, it fully accepts that biological needs are basic elements of human life. In addition, it shows a characteristic that strongly implies a religious tendency as it recognizes that human nature is internalized through divine commands issued by the Supreme God of the Ninth Heaven (九天上帝 Gucheon Sangje), the transcendent and absolute ruler. The Basic Works of Daesoon Jinrihoe, which are Propagation, Edification, and Cultivation, aim to restore human nature to its innate state of honesty and truthfulness. Among the Three Major Societal Works, we can see that the works of Social Welfare and Charity Aid are implemented in accordance with the accepting acknowledgment of basic human needs (the pursuit of clothing, food, and sexual desires), and helping people meet their needs in appropriate ways and to appropriate degrees.

Principal Characteristics of Pinus parviflora S. et Z. Native to the Dagelet Island (울릉도(鬱陵島) 섬잣나무의 특성(特性)에 관(關)한 연구(硏究))

  • In order to examine the taxonomic difference between the type of Pinus parviflora S. et Z. native to the Dagelet Island and the type of the species introduced to a number of places of the inland of South Korea, investigations on principal characters of needle, cone and seed were made with a hope to obtain informations on the evaluation of the species for possible use in the reforestation program in Korea in the future. Pinus parviflora is belonged to the Sub-genus Haploxylon of Genus Pinus and it has been speculated among dendrologists that this speoies is not monotypic. 308 rendomly selected trees from 8 different elevations of a natural stand of P. parviflora in the Dagelet Island, and 168 trees of P. parviflora growing at 15 different locations of the inland of South Korea were employed as samples along with 300 trees of P. koraiensis as control. The results obtained are summarized as follows: 1. The needle length of the Pinus parviflora of the Dagelet Island is longer than that of the species growing in the inland by 21-35 percent with statistical significancy. (Table 2) 2. In the cross section of needle, no resin canal was observed in about 50-70 percent of the sample trees of the Dagelet Island, whereas the resin canals appearing at external in most cases were observed in all sample trees from the inland. Consequently, the number of resin canals per needle was 0.4-0.9 with the Dagelet Island type and 2.0-2.7 with the inland type and these differences were statistically significant. (Table 3, Fig.2) 3. The Pinus parvviflora type of the Dagelet Island bas yellowish brown cones, and the Pinus parviflora type of Suwon and Kwangyang has redish brown cones. In both the length of cone and the number of cone scale, the difference between the type of the Dagelet Island and the type of the inland was also statistically significant. The cone scales of the Dagelet Island type are slightly opened, whereas the cone scales are widely opened with both of Suwon and Kwangyang type. (Table 4, Fig. 3) 4. the seed color, of the Dagelet Island type is yellowish brown, while it is greyish brown with Kwangyang and Suwon type. In the length and width of seed, the Dagelet Island type showed significantly larger values than that of the inland type. The length of seed was longest with the Kwangyang type being followed by Suwon and the Dagelet Island type in ordar. The seed wing of the Kwangyang type are longer than the seed, while that of the Dagelet Island type is degenerated to be shorter than the seed. (Table 5, Fig. 4) 5. The Pinus parviflora type of the Dagelat Island is similar in many respects to the southern type of Pinus parviflora of Japan except that many has no resin canals in the needle. 6. On the basis of the results obtained in this study, it may be concluded that the type of Pinus parviflora of the Dagelet Island is significantly different from the type of the species introduced to the inland and that there is no recognizable variation between the population of the different altitude of the Dagelet Island and the individual variation within population is also negligible. In the light of the high value of the tree not only as an ornamental tree but as an economical tree, The type of Pinus parviflora of the Dagelet Island is considered to be recommendable to be used for the future reforestation program of Korea.

Studies on the Regional Distribution and Some Morphological Characters of Buckwheat, Fagopyrum esculentum Moench, Grown in Chungnam Province (충남지역(忠南地域)에서 재배(栽培)하는 메밀종자(種子)의 몇가지 특성(特性)과 그 분포(分布)에 관(關)한 조사연구(調査硏究))

  • An investigation was made to find regional differences and seed characteristics of buckwheats collected from 36 areas in Chungnam province in 1984, and the results obtained are summarized as follows; 1. Grain types of the collected buckwheats were classified as winged, semi-winged and common types. All collections were on the average composed of 50.1 % of common grain types, 26.8 % of semi-winged type and 23.1 % of winged types, respectively. However, there was a great regional difference in components of each grain type among collections. 2. The mean 1000 grain weight was 24.9 gr. The regional differences in 1000 grain weight showed that a collection (collection no. 30) from Susan was 17.4 gr and a collection (collection. no. 16) from Suchon was 31.9 gr. There wasn't any significant relationship between grain types component and 1000 grain weight. 3. Germination of seeds was accelerated as temperature goes up from $5^{\circ}C$ to $30^{\circ}C$. However, interesting germination pattern was observed. That is, seeds from Suchon (collection no. 16), Hongsung (collection no. 23), Cheonweon(collection no. 35), and Yesan (collection no. 27) showed abrupt decrease of germination rate at the temperature of $15^{\circ}C$. The seeds collected from Suchon (collection no. 29), Dangjin (collection no. 32) and from Chungyang (collection no. 21) showed decrease of germination rate at $25^{\circ}C$. These seeds showed, however, the increase of germination percentage at temperature higher than $15^{\circ}C$ and $25^{\circ}C$, respectively. The germination rate for the most collections at $5^{\circ}C$ was less than 10%. However, seeds from Asan (collection no. 33) and Daeduk (collection no. 3) showed 20% and 30% of germination rate at $5^{\circ}C$, respectively. 4. Color of seed coat could be classified into two maj or colors, dark and dark brown. Based on the seed coat color and grain types, all the collections could be classified into the following six categories: winged-black, semi-winged-black, common-black, winged-dark brown, semi-winged dark brown and common-dark brown. The different light absorption rate was found within the UV light zone (190-390 nm) depending upon the two different major seed coat color.

  • This experiment was carried out to study the characteristics and composition of the oil extracted from elder berry seeds. The results obtained were summarized as follows. 1. The elder berry fruits are composed of $1.56%{\sim}2.50%$ crude lipid, $2.50{\sim}6.10%$ crude protein, $0.7{\sim}1.3%$ crude ash, 0.73% pectin and the fresh whole fruits bore $6{\sim}7%$ of its seeds on weight basis. 2. The hot pressed oil had similar flavor and taste to sesame oil, and cold, pressed one, to olive oil. Its iodine value ranged $95{\sim}110$, and its oil extraction rate from seed $26.18{\sim}26.55%$. 3. The elder berry seed oil contained a high level of essential fatty acids, and the oil seed cakes, $12.65{\sim}18.65%$ of crude protein. These may be utilized as concentrated fodder for livestock.
