Effects of Allium hookeri Extracts on Glutamate-induced Neurotoxicity in HT22 Cells (글루타메이트로 유발한 세포독성에 대한 삼채추출물의 뇌세포 보호 효과)

  • Glutamate-induced oxidative stress results in neuro-degenerative disorders in many central nervous system (CNS) such as Alzheimer's disease, ischemia, Huntington's disease, and Parkinson's disease. Our study was performed to investigate neuroprotective effects of Allium hookeri extracts (leaf, root, and whole) on glutamate-induced HT22 cells. In this study, ethanol extract of A. hookeri showed the outstanding neuroprotective effect in HT22 cells. In addition, we found that ethanol extract of A. hookeri root increased heme oxygenase (HO)-1 in HT22 cells. Moreover, ethanol extract of A. hookeri root also upregulated nuclear accumulation of nuclear factor E2-related factor 2 (Nrf2) in HT22 cells. These results demonstrate that ethanol extract of A. hookeri root contributes neuroprotective effects against glutamate-induced oxidative stress in HT22 cells, via Nrf2-mediated HO-1 expression. Our study suggests that ethanol extract of A. hookeri root could be the potential agent for the treatment of many neuro-degenerative diseases.

Quality Characteristics and Storage Stability of Bread with Allium hookeri Powder (삼채가루 첨가 식빵의 제조조건 최적화 및 저장성 연구)

  • The purpose of this study was to determine the optimal mixing ratio of Allium hookeri powder and butter in the preparation of bread. The experiment was designed according to the central composite design for estimating the response surface, which demonstrated 10 experimental points, including 2 replicates for Allium hookeri powder and butter. Further, the mechanical and sensory properties of test materials were measured. A canonical form and perturbation plot conveyed the influence of each ingredient on the final product mixture. Overall, the measurement results of the mechanical properties showed a significant increase or decrease in dough pH, sweetness, volume, weight, height, specific volume, a & b-value of crumb and, springiness (p<0.05). Moreover, the sensory measurements demonstrated a significant improvement in color, appearance, texture, moistness and, overall quality. As a result, the optimum formulation from the numerical and graphical methods was calculated as 22.65 g of Allium hookeri powder and 47.77 g butter. After optimization, DPPH free radical scavenging activity, total phenolic content, and total plate counts over 10 days were evaluated. In sum, the results revealed the antioxidant and antibiotic actions of bread with Allium hookeri powder.

Lifespan-extending Effects of Allium hookeri Leaves in Caenorhabditis elegans (삼채 잎 추출물의 예쁜꼬마선충에 대한 수명연장 효과)

  • To evaluate the longevity property of Allium hookeri (Liliaceae) leaves, this study was performed. Ethanol extract of A. hookeri was successively partitioned as methylene chloride, ethyl acetate, n-butanol and $H_2O$ soluble fractions, and lifespan-extending effects of the fractions were checked using Caenorhabditis elegans model system. The most active ethyl acetate-soluble fraction elevated heat stress tolerance, and increased expression of stress resistance protein. Further studies were performed to investigate several aging-related factors such as reproduction, food intake, growth and movement of C. elegans. The results revealed that there were no significant changes in aging-related factors including reproduction and food intake, however, ethyl acetate-soluble fraction treatment led to up-regulation of locomotory ability and growth of aged worms, suggesting ethyl acetate-soluble fraction affected healthspan as well as lifespan of nematode.

Antioxidant and Cell Protection Effects of Ethanolic Extract from Cirsium setidens, Allium hookeri, Aronia melancocarpa (곤드레, 삼채, 아로니아 주정 추출물의 항산화 및 세포 보호 효과)

  • Cirsium setidens, Allium hookeri, Aronia melancocarpa, are popular dietary ingredients for Asian cuisine or beverage. this study attempted to search and report the antioxidant activity of extracts of aronia, gondre and samchae, which are used for food in Korea. As a result, total phenol content and DPPH radical scavenging activity tended to increase, and Gondre extract showed the highest DPPH radical scavenging activity effect. The results of a study on the ABTS radical scavenging activity of extracts of Gondre, Aronia, and Samchae showed a tendency for radical scavenging activity to increase as the concentration increased. Gondre hot water extract showed the highest ABTS radical scavenging activity. In this way, antioxidant activity was shown in the order of Gondre, Aronia, and Samchae, and the concentration showed a high activity effect at high concentrations. In the cell protection effect results, the concentration of Gondre extract was 500 ㎍/mL, the Samchae alcohol extract was used at concentrations of 2.5, 5, 7.5, and 10 ㎍/mL, and the Aronia extract was used at concentrations of 125 ㎍/mL. and showed a cytoprotective effect at 250 ㎍/mL. We hope that these research results will be used as basic data for the development of various functional foods.

Antioxidant and Longevity Properties of the Root of Allium hookeri in Caenorhabditis elegans (삼채 뿌리의 항산화 및 수명연장 효과)

  • Allium hookeri (Liliaceae), native to India is widely cultivated in Korea lately as a medicinal plant. This study was performed to evaluate the antioxidant and lifespan extending effects of the root of A. hookeri. Methanol extract from the root of A. hookeri was successively partitioned as methylene chloride, ethyl acetate, n-butanol and H2O soluble fractions. Antioxidant effects of the fractions were measured by measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH), and superoxide quenching activities. Lifespan extending effects of the fractions were investigated using Caenorhabditis elegans model system. In addition, stress resistant, superoxide dismutase (SOD) and catalase activities, and intracellular reactive oxygen species (ROS) levels were also studied. Furthermore, we had investigated aging-related factors such as reproduction, food intake, growth and movement of C. elegans to find any sample-induced significant change. Our results revealed that ethyl acetate soluble fraction from the root of A. hookeri showed the most potent antioxidant and lifespan extending effects, and this fraction elevated the most potent SOD and catalase activities of worms, and reduce intracellular ROS accumulation.

The Hypolipidemic Effect of Allium Hookeri in Rats Fed with a High Fat Diet (삼채가 고지방식이를 급여한 흰쥐의 고지혈증 개선 효과에 미치는 영향)

  • This study evaluated the effect of Allium hookeri roots on lipid metabolism of the serum, liver, and adipose tissues induced by a high-fat diet in male Sprague-Dawley rats. The rats were divided into four groups with 8 rats per group for 4 weeks: the normal-diet group (N), the high-fat diet (HFD) group, the HFD containing 3% Allium hookeri (HFD-A3) group, and the HFD containing 5% Allium hookeri (HFD-A5) group. The results showed that the body weight gain and food intake of rats in the HFC-A3 and HFC-A5 groups were significantly decreased compared with those in the HFD group. The epididymal adipose tissue weight in the HFD-A5 group was significantly decreased compared with those in the HFD group, and adipose tissue weights of liver and mesenteric adipose tissues in the HFD-A3 and HFD-A5 groups were significantly decreased compared with those in the HFD group. Serum triglyceride, total cholesterol and LDL cholesterol concentrations, atherogenic index, and cardiac risk factor were significantly lower in the HFD-A3 and HFD-A5 groups than in the HFD group. Serum lipid profiles, as well as ALT and AST activities did not show any difference in all groups. Serum ALP and LDH activities were suppressed in the HFD-A5 group compared with those in the HFD group. The hepatic triglyceride and total cholesterol levels of rats in the HFD-A5 group was significantly lower than those in the HFD group. Moreover, triglyceride and total cholesterol in the epididymal and mesenteric adipose tissues were significantly lower in the HFD-A5 group than in the HFD group. These results demonstrated that the intake of Allium hookeri showed a hypolipidemic effect, changing the lipid metabolsim of a high-fat diet induced rats.

Effect of Starter Cultures on the Antioxidant Activities of Allium hookeri Root-Hot Water Extract (발효 균주에 따른 삼채뿌리 열수추출물의 항산화능 비교연구)

  • Allium hookeri, a member of the onion family, has long been mainly cultivated for food and medicinal use in Southeast Asian countries, owing to its various biological properties. However, no studies of the anti-oxidative effects of fermented A. hookeri root extracts have been conducted to date. Therefore, this study investigated the effect of different starter cultures on the antioxidant activities of hot water extract of A. hookeri root by using the following five strains: Lactobacillus acidophilus, Bifidobacterum longum, Enterococcus faecium, Streptococcus thermophilus, and Saccharomyces cerevisiae. DPPH and ABTS radical scavenging activity, total phenolic acid contents, and total antioxidant capacity were higher in the hot water extract of A. hookeri root fermented with starter cultures than those of A. hookeri root. Among hot water extract of A. hookeri root fermented with starter cultures, fementing with S. cerevisiae showed the highest antioxidant activities. The results of this study provide new evidence of the anti-oxidative properties of A. hookeri root with starter cultures, indicating that it may be highly valuable as a natural product owing to its high-quality functional components.

Anti-oxidant and anti-inflammatory effect of Allium Hookeri water extracts in RAW 264.7 cells (삼채(三菜) 물추출물이 RAW 264.7 세포의 항산화 및 염증반응에 미치는 영향)

  • Objectives : Allium hookeri is a well-known traditional herbal remedy and its root used for treatment of inflammation and tumor. However, the mechanism of anti-inflammatory effect of Allium hookeri is still unknown. This study aims to examine the mechanism of anti-inflammatory effect of Allium hookeri on mouse macrophage cell line, RAW 264.7 cells. Methods : Anti-oxidant effect of water extract of Allium hookeri (WEAH) was measured by 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) assay. 3- (4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay was performed to determine the effect of WEAH on cell viability in RAW 264.7 cells. In addition, anti-inflammatory effect of WEAH was investigated in RAW 264.7 cells. Inflammation of RAW 264.7 cells induced by lipopolysarccharide (LPS) treatment and expression levels of inflammatory cytokine interleukin 1 β (IL-1β) and interleukin 6 (IL-6) gene were analyzed using quantitative reverse transcription PCR (qRT-PCR) analysis. Furthermore, the phosphorylation of inhibitor of nuclear factor kappa B (IκBα) after LPS treatment with WEAH-treated RAW 264.7 cells was confirmed by immunoblot analysis. Results : WEAH showed a strong anti-oxidant effect and no cytotoxicity to RAW 264.7 cells up to 2 mg/㎖ concentration. The LPS-induced mRNA expression levels of IL-1β and IL-6 were decreased by WEAH treatment. Furthermore, the LPS-induced phosphorylation of IκBα is attenuated by WEAH treatment. Conclusions : Through experimental demonstration of anti-oxidant and anti-inflammatory effects of WEAH, we suggest that Allium hookeri is a valuable material for prevention and treatment of various inflammatory diseases.

Effect of Allium hookeri Root on Plasma Blood Glucose and Fat Profile Levels in Streptozotocin-Induced Diabetic Rats (당뇨 유발 흰쥐에서 삼채뿌리 첨가 식이가 혈당과 지질 수준에 미치는 영향)

  • This study was designed to examine the effects of Allium hookeri (AH) on plasma blood glucose and fat profile levels in diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats through injection of streptozotocin (STZ) dissolved in citrate buffer into tail veins at a dose of 45 mg/kg of body weight. Sprague-Dawley rats were then fed for 4weeks, with the experimental groups receiving a modified diet containing 5% or 10% powder derived from AH roots. The experimental groups were divided into four groups, consisting of a control group, STZ-control group, and diabetic fed with AH 5% & 10% treated groups. Rats' body weights, blood glucose, total cholesterol, HDL-cholesterol, triglyceride (TG), and free fatty acid (FFA) values in plasma were measured along with hematocrit (Hct) values and aminotransferase activities. Body weight losses were observed in the STZ-control group, whereas the STZ-AH group of diabetic rats gained weight. There was a significantly decrease in brain weight of the STZ-AH group but no significant differences in kidney and liver weights of the STZ-AH 5% & STZ-AH 10% groups compared to the STZ-control group. Blood glucose was significantly reduced in the STZ-AH 5% & STZ-AH 10% diabetic groups. There were no significant differences in total cholesterol and TG levels among the diabetic groups. HDL-cholesterol significantly increased while FFA significantly decreased in the STZ-AH 5% & STZ-AH 10% diabetic groups. The Hct level of the STZ-AH group was lower than that of the STZ-control group. Aspartate aminotransferase activity was significantly reduced in the STZ-AH 5% & STZ-AH 10% diabetic groups. These results indicate that supplementation with Allium hookeri root may have beneficial effects on diabetic complications as a potential therapeutic candidate.

Anti-oxidative Effects of Allium hookeri Leaves in Caenorhabditis elegans (삼채 잎의 예쁜꼬마선충 내의 항산화 효과)

  • As an ongoing study about Allium hookeri (Liliaceae), this study was performed to evaluate the anti-oxidative effect of the leaves of this plant. Ethanol extract of A. hookeri leaves was successively partitioned as methylene chloride, ethyl acetate, n-butanol and $H_2O$ soluble fractions. The ethyl acetate soluble fraction showed the most potent DPPH radical scavenging and superoxide quenching activities among those fractions. To prove antioxidant activity of ethyl acetate fraction of A. hookeri leaves, we checked the activities of superoxide dismutase (SOD) and catalase, and intracellular ROS level and oxidative stress tolerance in Caenorhabditis elegans. In addition, to verify if increased stress tolerance of C. elegans by treating of ethyl acetate fraction was due to regulation of stress-response gene, we checked SOD-3 expression using transgenic strain. As a consequence, the ethyl acetate fraction increased SOD and catalase activity of C. elegans, and reduced intracellular ROS accumulation in a dose-dependent manner. Besides, the ethyl acetate fraction-treated CF1553 worms showed higher SOD-3::GFP intensity.
