Establishment of Analytical Method for Carpropamid in Agricultural Commodities using HPLC-DAD/MS (HPLC-DAD/MS를 이용한 농산물 중 살균제 Carpropamid의 정밀 잔류분석법 확립)

  • This study was performed to establish a single residue analytical method for determining fungicide carpropamid residues in various agricultural commodities. Korean cabbage, apple, brown rice and green pepper were selected as representative crops. Samples were homogenized, extracted with acetone and purified by liquid-liquid partition and Florisil column chromatography. Carpropamid residues were analyzed at 220 nm with reversed phase HPLC equipped octylsilyl and octadecylsilyl column and confirmed using mass spectrometry. ILOQ (Instrumental limit of quantitation) of carpropamid was 2 ng and MLOQ (Method LOQ) was 0.02 mg/kg. Mean recoveries from four kinds of crop samples fortified at three levels (MLOQ, 10LOQ, 100LOQ) in triplicate were in the range of 84~112%. Relative standard deviations of the analytical method were all less than 10%, irrespective of crop types.

Effect of Various Fungicides on Pollen Germination and Pollen Tube Growth in Some Fruit Trees (각종 살균제(殺菌劑)가 주요과수(主要果樹)의 화분발아(花粉發芽) 및 화분관신장(花粉管伸長)에 미치는 영향(影響))

  • The experiment was carried out in order to choose fungicides which are proper to be used in flowering time of fruit trees. The effect of fungicides on pollen germination and pollen tube growth was investigated in vitro. The results obtained are as follows : 1. Difolatan-Wp, Lime-sulfur, Captan-Wp and Bordeaux mixture greatly inhibited pollen germination and pollen tube growth. 2. The effect of Daconil-Wp and Zimaneb-Wp on pollen germination and pollen tube growth was not so great as that of Difolatan-Wp, Lime-sulfur, Captan-Wp or Bordeaux mixture. 3. Polyram-combi-Wp, Topsin-Wp and Benlate-Wp did not affect pollen germination but these fungicides slightly inhibited pollen tube growth. 4. Polyoxin-Wp significantly increased pollen tube growth but did not show promotive effect on pollen germinaiton.

  • Disinfection using UV irradiation has been considered to an alternative for chlorination because it does not suffer from disinfection byproducts problem. The aim of this study was to verify if UV irradiation with 368.6[nm] wavelength has a potential to be used a efficient disinfection. UV irradiation was given for 60 minutes to E. coli. which was used as a model microorganism. Remarkable decrease in number of microorganism was not observed for initial 30 minutes, however, 83.3[%] of disinfection efficiency was achieved after 30 minutes of irradiation. Inactivation constant, k was determined to 0.00868[$cm^2$]/([$mW{\cdot}s$]) using kinetic approach.

  • The disinfectivity of formalin fumigation was tested against silkworm nuclear polyhedrosis virus and yellow muscardine pathogen, Beauveria bassiana. The inactivation of the virus was acquired when it was fumigated by adding 30g of potassium permanganate to 75$m\ell$ of formalin per 3.3$m^2$ of rearing room area and viability of the yellow muscardine pathogen was also lost with the same treatment of fumigation. It was also proved that the fumigation didn't give and damage to silkworm larvae when it was applied 2 or 3 times to grown larvae.

Behaviors of the Fungicide Procymidone in Soils (살균제 Procymidone의 토양 중 동태)

  • This study was focused on adsorption, leaching, photolysis, and hydrolysis of the fungicide procymidone in soils. Adsorption type of procymidone on three different soil were well fitted to Linear and Freundlich isotherm. Distribution coefficients (Kd) were ranged from 2.75 to 12.18 and Freundlich isotherm Kf value $1.99{\sim}9.98$, 1/n value $0.74{\sim}0.89$. Desorption rates were $20.1{\sim}34.0%$ (Namgye), $26.3{\sim}44.6%$ (Jigog) and $31.6{\sim}50.9%$ (Baegsan series) and desorption hysteresis were $0.65{\sim}0.79,\;0.55{\sim}0.73\;and\;0.49{\sim}0.68$. Procymidone seemed to be stable to photolysis in acidic and neutral solutions but hydrolyzed rapidly in alkaline solution. Considering leaching properties procymidone mobility low in soils.

Effect of Organic Matter on the Occurrence of Fusarium Wilt in Cucumber (Fusarium oxysporum f. sp. cucumerinum에 의한 오이덩굴쪼김병의 발생에 미치는 유기물 시용의 효과)

  • Fusarium wilt caused by Fusarium oxysporum f. sp. cucumerinum has caused high damage in cucumber under greenhouse condition. This disease was suppressed $30\~55\%$ by application of organic matters compared with natural cropping soils. The suppression effect was the highest in the mushroom humic compost and fowl excretion matter among the various organic matters, varying with kinds of organic matters and degrees of humic resolvability. There was a slight difference in severity of fusarium wilt between sterilized organic matters and soils. The disease occurrence was delayed more in the nonsterilized organic matters and soils than in the sterilized ones. At 30 days after inoculation of F. oxysporum, numbers of Actinomycetes, fungi and bacteria were considerably increased, whereas F. oxysporum was decreased in the organic matter amended-soils compared with natural control soils.

Influence of Some Pollutants and Fertilizers on Degradation of Oxadixyl in Soil (몇 가지 오염물질과 비료의 처리가 살균제 Oxadixyl의 토양중 분해에 미치는 영향)

  • The degradation of fungicide oxadixyl in soil amended with manure, chemical fertilizers, heavy metals and detergent was studied. The degradation of oxadixyl in the soil was slow, but became to be fast after the lag phase of about 14 days. The half-life was 10.5 days. The degradation rate was accelerated largely by the amendment of manure. Potassium also promoted the degradation rate but nitrogen and phosphate did not. The heavy metals inhibited the degradation rate, in order of Ni, Cd, Cr, Cu, and Zn. The degradation rate was declined greatly with the addition of synthetic detergent. The microbial biomass and the respiration rate in the soil were increased by the amendment of manure and chemical fertilizers, but decreased by the addition of heavy metals and cleaner. The degradation rate of oxadixyl was positively correlated with the microbial biomass and the respiration rate.

Analysis of Half-life Time and Residual Concentration of Fungicide Iminoctadine Triacetate in Soils (토양에서 살균제 Iminoctadine Triacetate의 반감기와 잔류농도의 분석)

  • The optimum conditions for the half-life time and residual analysis of the iminoactadine triacetate fungicide on soils were investigated by using the gas chromatography. Iminoctadine triacetate(IOTA) was extracted from soils with 2.0 M-NaOH/methanol and chloroform. The extracted IOTA was derivatized to pyrimidine hexafluroacetylacetone by the acetylation and analyzed with GC/ECD after elimination of moisture and impurities on the Sep-Pak column. From the standard addition experiments with 0.1 and 1.0 ppm, the average recoveries were ranged from 83.8 to 93.2 % and the detection limit was 0.005 ppm. The half-life time of iminoctadine triacetate in the silty clay was 30 days in the laboratory and 19.5 days in the field test whereas it was 27 days and 17.5 days for each in case of silty loam.

Degradation of the herbicide dicamba under sunlight and in soil (제초제 Dicamba의 자연광 및 토양 중 분해)

  • In order to elucidate the degradation of the herbicide dicamba, the degradabilities of dicamba under sunlight and in soils were investigated. The photodegradation rate of the authentic dicamba under sunlight condition was only 3.3% after 9 weeks. 4-Hydroxy dicamba turned out to be the major product, and 5-hydroxy dicamba was also identified. Dicamba was degraded to 3,6-dichlorosalicylic acid by demethylation in the viable soil, which resulted in 14.7 to 23.2% degradation of the applied amount during 8 weeks of incubation. Meanwhile, the degradation was quite slow in the sterilize soil, which revealed that the soil microbes played a major role in dicamba degradation.

Develoment of the Control System against Shrimp Viral Disease (바이러스성 새우질병 방제법의 개발)

  • In order'to reduce loss of shrimp productivity by viral diseases on the cultured shrimp, we were develope the effective shrimp virus control system. Virus is inactivated by above 5 ppm of chloride and treatment of 50% fresh water. Mortality of infected shrimp is decreased by feeding of immunostimulants such as peptidoglycan and schizophyllan. Mg(OH)$_2$ was one of improvement agents which have best effect on pH, ignition loss(IL), chemical oxidation demand(COD) and total sulfide of water and sediment of shrimp farm.
