Identification of Antioxidative Substances in Allium fistulosum L. by GC-MS (GC-MS에 의한 대파의 항산화 물질 동정)

  • The ethyl acetate-soluble acidic fraction of juice, hot water, and MeOH extract of Allium fistulosum L. showed DPPH radical-scavenging activity. Each fraction was purified through silica gel adsorption column chromatography, and the active substances in the juice and hot water extract were identified as succinic acid, fumaric acid, 4-hydroxybenzoic acid, 4-hydroxy-3-methoxybenzoic acid, 4-hydroxycinnamic acid, and 4-hydroxy-3-methoxycinnamic acid. For the MeOH extract, 4-hydroxybenzoic acid, 4-hydroxy-3-methoxybenzoic acid, 4-hydroxycinnamic acid, and 4-hydroxy-3-methoxycinnmic acid were identified as the active substances by GC-MS. The contents of these compounds were determined by GC analysis, and their anti-oxidative activities were measured using the DPPH radical-scavenging assay. The results obtained showed that 4-hydroxy-3-methoxycinnamic acid was the dominant antioxidant in Allium fistulosum L.

Electrophoretic Characterization of Job′s tears (Yulmoo: Coix lachryma-jobi L. var. Ma-yuen stapf. & Yeomjoo: Coix lachryma-jobi L.) proteins (율무와 염주 단백질의 전기영동 특성)

  • This study was performed to examine the electrophoretic properties of Job's tears (Yulmoo: Coix lachryma-jobi L. var. Ma-yuen Stapf. & Yeomjoo: Coix lachryma-jobi L.) proteins. Albumins, globulins, gliadins and glutelins were extracted from the polished Yulmoo and brown Yeomjoo by the modified Osborne method. For a comparison, rice proteins were extracted and fractionated by the same method. The relative proportions of protein fractions were 17.4 : 19.6 : 55.2 : 7.7% in polished Yulmoo, 12.6 : 62.2 : 4.2 :21.0% in brown Yeomjoo and 14.2 : 57 4 : 0.77 : 27.8% in rice, in the order of albumis, globulins, gliadins and glutelins. Polyacrylamide gel electrophoresis (PAGE) and SDS-polyacrylamide gel electrophoresis (SDS-PAGE) were peformed to identify the subfractions of each protein fraction extracted from polished Yulmoo, brown Yeomjoo and rice. The electro-phoregrams of polyacrylamide gel electrophoresis showed that the same fractions of both polished Yulmoo protein and brown Yeomjoo protein had very similar electrophoretic patterns to each other respectively, but there were significant differences in the patterns between Job's tears proteins and rice proteins.

  • This study evaluates the erosive potential and effects of mixed alcohols by analyzing the pH, titratable acidity, and fluorescence loss degree (△F). Following alcohol groups were investigated: Soju, Calamansi+soju, Yakult+soju, Cola+soju, and Energy drink+soju. The ratio of soju:beverage in the alcohol mixtures was 7:3. Ed. Measurement of the pH and titratable acidity (the amount of 1M NaoH solution required to raise to pH 5.5 (TA5.5) and 7.0 (TA7.0)) of alcohols was achieved by stirring with pH meter. The erosive effect of the alcohol mixtures on bovine tooth (△F) after 1, 2, 4, and 6 hours exposure were analyzed by quantitative light-induced fluorescence (QLF-D). All the mixed alcohols in this study showed an acidic pH, lower than 4.5. The average pH of mixed alcohols was 3.17 ± 0.50 whereas the pH of Soju was 8.6 ± 0.01. The TA5.5 and TA7.0 values of the mixed alcohols were 0.5~18 and 0.5~23.5, respectively. △F of the three tested mixed alcohol groups (except yakult+soju group) were observed to increase in a time-dependent manner. The calamansi mixed alcohol had the highest acidity potential and erosive effect among the tested groups. Taken together, the results indicate that the mixed alcohols have a strong erosive effect and potential on dental enamel.

Physicochemical Properties of Forest Soils Related to Sulfate Adsorption (황산이온의 흡착에 관여하는 산림토양의 물리화학적 특성)

  • Sulfate adsorption in forest soils is a process of sulfur dynamics playing an important role in plant uptake, cation movement, acid neutralization capacity and so on. The relationship between sulfate adsorption and some physicochemical properties of four forest soils was investigated. Extractable sulfate contents and sulfate adsorption capacity (SAC) in the forest soils varied much among study sites. Extractable sulfate contents were more in sub-surface soils with lower organic matter and greater Al and Fe oxides than in surface soils. The average contents of $Al_d$ and $Fe_d$ in the sub-surface soils were 8.49 and $12.45g\;kg^{-1}$, respectively. Soil pH, cation exchange capacity and clay content were positively correlated with the extractable sulfate contents and SAC. Organic carbon content, however, was negatively correlated with the extractable sulfate contents, implying the competitive adsorption of sulfate with soil organic matter. Considerably significant correlation was found between inorganic + amorphous Al and Fe oxides and the sulfate adsorption, but crystalline Al and other fractions of Fe oxide showed no correlation. Relatively close relationship between the adsorbed sulfates and soil pH, cation exchange capacity, or amorphous Al oxides indicates that the accelerated soil acidification may substantially reduce the potential for sulfate adsorption contributing to sulfur flux in forest ecosystems.

  • Contents and distribution of amino acids in the hydrolysates of humic acids extracted from straw of rice and barley at three different dates during decomposition were examined. The results obtained from this study may be summed up as the following: 1. There are differences between the humic acid hydrolysates from rice straw and barley straw in regards of composition of humic acids and distribution of amino acids. 2. Neutral amino acids as a group occupy the largest share, followed by acidic amino acids and basic amino acids. 3. The total amount of amino acids per gram of humic acid is greater in straw of rice than in straw of barley. 4. With the humification progressing the content of lysine increases, but the content of histidine decreases. In general glycine, glutamic acid, aspartic acid, alanine and leucine constitute the 5 predominant amino acids in all hydrolysates. 5. Arginine is not detected at all in any of the hydrolysates of humic acids obtained from humified materials. 6. The presence of phenylalanine and tyrosine is an evidence for the aromatic characteristics of humic acids.

Improvement of Hygienic Quality of Vegetable Mixed condiments Using Gamma-Irradiation (식물성 혼합조미료의 품질개선을 위한 감마에너지의 이용)

  • Vegetable mixed codiments, commercial products prepared using soybean,paste and natto, respectively as the ingredients, were used ina study to evaluate the efficacy of gamma irradiationas a means of decontamination and the emphasis was placed upon the determination of the effect of irradiation on the microbiological and some physicochemical properties of the samples. The number of microorganisms contaminated ranged from $10^{6}\;to\;10^{7}$ cells per gram in mesophilic total bacteria, which were composed of thermophilts and acid tolerant bacteria by over 90%. They were reduced by 3 to 4 log cycles with irradiation at 10 kGy. Gamma irradiation at 5 kGy could eliminate the microbial populations of yeasts and molds ($10^{2}\;to\;10^{3}$ cells per gram) and coliforms ($10^{6}\;to\;10^{6}$ cells per gram of natto condiments). However, total destruction of microorganisms in soybean-paste and natto condiments was shown to be possible at a dose-range more than 10 kGy. Irradiationup to 10 kGy was not detrimental to the physicochemical properties of the sample, such as pH. amino nitrogen, rancidity and color, even though some change was brought about in the content of sulfur-containing amino acids.

  • We investigated biofilm formation in various media, growth in low pH, and the hemolytic activity of 14 strains of Cronobacter spp. (Enterobacter sakazakii) isolated from a variety of foods including powdered infant formula (n=75), infant cereal (n=100), honey (n=30), and other infant foods (n=100) in Korea. The Cronobacter spp. adhered and formed biofilms on polyethylene, and a greater extent of biofilm was observed in nutrient-rich media. No clear difference in biofilm-forming ability was noted among the media constituents and the pattern of biofilm formation was strain-dependent. Seven strains out of 14 strains (50%) grew at pH 4.1, indicating that the acid resistance of these Cronobacter spp. isolated in Korea was relatively low. Hemolytic activity was not observed in any of the strains. This study provides basic information for the physiological and biochemical characteristics of Cronobacter spp. isolated from a variety of infant foods in Korea.

Precipitation Behavior of Ammonium Vanadate from Solution Containing Vanadium (바나듐 함유 수용액의 암모늄바나데이트 침전거동 고찰)

  • In this study, the precipitation reaction of vanadium and ammonium chloride in aqueous solution was investigated in order to recover vanadium. Ammonium metavanadate having a crystal structure of [$NH_4VO_3$] was precipitated from aqueous solution containing vanadium at pH 9.2 ~ 9.4, and ammonium polyvanadate having a crystal structure of [$(NH_4)_2V_6O_{16}$] was precipitated when the pH of the aqueous solution containing vanadium was adjusted with sulfuric acid. Ammonium polyvanadate [$(NH_4)_2V_6O_{16}$] precipitated at a temperature of $80{\sim}90^{\circ}C$ and pH 2, and at a temperature of $40^{\circ}C$ and pH 6 ~ 8 of aqueous solution. In the acidic region of aqueous solution pH 2, the vanadium content of the aqueous solution should be at least 3,000 mg/L and the precipitation temperature should be maintained at $80^{\circ}C$ or higher in order to obtain a precipitation ratio of 99% or more. When the ammonium vanadate was precipitated in the alkaline region, the vanadium content was more than 10,000 mg/L and the precipitation temperature was maintained at $40^{\circ}C$ to increase the precipitation ratio. Aluminum was not precipitated regardless of the vanadium content and pH of the aqueous solution. However, the iron component reacts with ammonium chloride to precipitate into ammonium jarosite. Therefore, Fe component must be preferentially removed in order to increase the recovery of vanadium.

  • We investigated the properties of Korean traditional animal glue which are associated with the use of different parts of cattle hide and extraction conditions. Both average molecular weight(Mw, Mz) and polydispersity (PDI) of the animal glue increased with extraction time, whereas only PDI decreased after 48h. There were no differences in the average molecular weight and PDI for the different cattle hide parts used, although a individual difference was observed with regard to total molecular weight. Differences in extracts over time were compared by investigating the amide regions representing the gelatien component of the animal glue. The triple helix structure of the belly skin deteriorated in a manner proportional to the extraction time, and differences were observed for each cattle hide part. The yellowness increased with the extraction time; however, the viscosity was not proportional to the average change in molecular weight. This study has some limitations because of difficulties in the quality control of cases where small amounts of glue were extracted as small differences might impact the overall results considerably. Further research exploring various extraction conditions is required to ensure the prodiction of traditional animal glue with optimum qualities.

A Pilot Test for the Utilization of Road Subsoil of the Tertiary Mudstone in Pohang Basin (포항분지 제3기 이암의 도로 노체 활용을 위한 현장시험)

  • The purpose of this study is to present the possibility a utilization of the tertiary mudstone in Pohang as road subsoil material through pilot experiments on the road embankment structure. This mudstone is an unconsolidated rock that is distributed in the soft rock sedimentary layer, the tertiary layer of the Cenozoic, and causes physical problems such as slaking, swelling, and reduced shear strength and chemical problem like acid drainage. In order to solve various complex problems, an laboratory mixing test was conducted, and the optimal mixing conditions of the tertiary mudstone (90%), composite slag (steel making 70%, blast furnace 30%), and neutralization and coating agent treatment were derived. In order to prove its utilization, a real-scale road embankment structure was constructed and tests were conducted for each section. The pre-processing section is stable due to the design of optimal mixing conditions, while in post-processing section, natural weathering proceeded rapidly, and structural problems were concerned. Since the effect of neutralizing and coating agents was confirmed in temporary-staking section, the neutralizing and coating agents can be applied during the temporary storage period.
