Effect of Torilis Fructus on Procollagen Biosynthesis and Activity of Matrix Metalloproteinase-I(MMP-1) in Human Dermal Fibroblast (사상자(Torilis Fructus)가 섬유아세포의 Procollagen 생합성과 Matrix metalloproteinase-I(MMP-1)의 활성에 미치는 영향)

  • Skin wrinkle formations are associated with collagen synthesis and matrix metalloproteinase-I(MMP-1) activity. This study was carried out to find out skin wrinkle reducing components in Torilis Fructus. Torilis Fructus were extracted with 70% ethanol and the ethanol extracts were systematically fractionated with n-hexane, ethylacetate, n-butanol and distilled water. Among them, antiwrinkle component from n-hexane fraction was purified by several column chromatographies and HPLC, which identified as torilin by $^1H-NMR,\;^{13}C-NMR$ and ESI-MS. To determine cell viability, collagen biosynthesis and MMP-1 activity, human dermal fibroblast was treated with 1-5 ppm concentrations of Torilis Fructus extract fraction and torilin. Cell viability was showed 84-102% at all group treated with 1-5 ppm. Collagen synthesis was increased in all group, especially torilin-treated group was highest amount. Active forms of MMP-1 were decreased in all group. From these results, we consider that Torilis Fructus have several antiwrinkle components and torilin may be one of the effective components.

Antibacterial Effects of Oriental Herb Extract Against Gardnerella vaginalis (Gardnerella vaginalis에 대한 한약재의 항균활성)

  • To investigate the potential of treatment, antimicrobial activity of various oriental herb extracts were tested for Gardnerella vaginalis, which is the predominant organism in bacterial vaginosis. Among the tested 14 oriental herbs, water-extracts of Kalkeun, Kosam, Nuro, Pakjakyak, Sukchangpo, Shiyup, Junghyang and Hwangryun represented antibacterial activities against G. vaginalis. The minimal inhibition concentration (MIC) of Shiyup against G. vaginais was 0.63 mg/mL, and those of Pakjakyak and Hwangryun, Kalkeun and Nuro, Kosam, Sukchangpo and Junghyang were 1.25 mg/mL, 2.5 mg/mL, and 5 mg/mL, respectively. There-fore, the water-extracts of Kalkeun, Kosam, Nuro, Pakjakyak, Sukchangpo, Shiyup, Junghyang and Hwangryun were considered to be potential treatment of bacterial vaginosis caused by G. vaginalis.

  • On January 13th 2012 Italian flag Costa Concordia was partly sunk. The ship was carrying 4,252 passengers and crew; in the next few days the bodies of 30 people were found. Two are still missing. Two South Korean passengers and an Italian crew member was rescued from the ship on January 15th, more than 24 hours after the accident. As of June 18th 2012 (The date of presentation) around 32 people had not been accounted for. In this paper, it is viewed and pointed out the cause, problem, and implication for the future. As a result, intelligent innovative evacuation system for the VLPS (Very Large Passenger Ship) should be set up in terms of international maritime safety.

A Study of Evacuation in Elderly Care Facilities Fire (요양시설 화재 시 피난에 관한 연구)

  • The development of science and technology has increased the average life expectancy and resulted in a significant increase in the elderly population. Due to the preponderance of the nuclear family in modern society and the weakening of the spirit of filial piety and respect for the elderly caused by the influence of Western civilization, elderly people are usually not living with their family, but have become dependent on elderly care facilities and, consequently, the number of elderly care facilities has increased significantly. On May 28, 2014, a fire occurred in the elderly care facilities in Jangseong, South Jeolla Province, and despite the rapid extinction of the fire by firefighters, 21 of the 28 casualties died due to smoke inhalation. This study was conducted for the purpose of providing measures to prevent smoke from entering into the rooms from which elderly people have difficulty evacuating in the case of fire. The use of the air curtain method to prevent smoke inflow was proposed and an experiment was conducted in which smoke was generated with a smoke generator. As a result of the experiment, it was confirmed that more than 90% of the smoke inflow was blocked if the door was closed in the room where the air curtain was installed.

Dietary effect of red ginseng extracts mixed with torilis fructus and corni fructus on the epidermal levels of ceramides and ceramide related enzyme proteins in uv-induced hairless mice (자외선이 조사된 무모생쥐에서 홍삼, 사상자, 산수유 혼합 추출물 섭취가 표피 세라마이드 함량 및 관련 효소의 발현에 미치는 영향)

  • UV-irradiation is a major factor of photo-aged skin, by which pigmentation, wrinkles and laxity are increased. In addition, the epidermal barrier is disrupted, ultimately causing dryness in photo-aged skin. As an effort to search dietary sources for improving the dryness of UV irradiated skin, the dietary effect of red ginseng based functional foods on the epidermal level of ceramides, a major lipid maintaining epidermal barrier, was determined in this study. Albino hairless mice were fed either a control diet [group UV (UV-irradiated control)] or diets with 0.5% (group M0.5) or 1% (group M1.0) of red ginseng extracts mixed with Torilis fructus and Corni fructus (66.7% red ginseng) in parallel with UV irradiation for 5 wks. A normal control group (group C) was fed a control diet without UV irradiation for 5 wks. The epidermal level of ceramides in group UV was significantly lower than that in group C, in which ceramidase, an enzyme involved in ceramide degradation, was highly expressed. In group M0.5, the epidermal level of ceramide was significantly increased to the level even higher than in group C. In addition, protein expression of serine palmitoyl transferase (SPT), a key enzyme involved in de novo ceramide synthesis, was increased in group M0.5. However the epidermal levels of ceramides as well as of ceramidase protein expression in group M1.0 did not differ from those in group UV. In conclusion, we demonstrate that dietary supplementation of red-ginseng extracts mixed with Torilis fructus and Corni fructus at a level of 0.5% level in diet increased the epidermal level of ceramides coupled with the elevated expression of SPT protein.

  • Many local governments are reducing speed limits according to the "Safe Speed 5030" policy implemented by the government to reduce traffic accident deaths. In the urban road sections in many metropolitan cities where speed limit reduction is applied as a pilot project, the decrease effect of the traffic accident number and death count is shown; however, no study of the effect analysis on the road sections of medium and small cities (basic local government unit) is found. Therefore, this study investigated the travel speed, number of traffic accidents, and death count before and after the implementation of speed limit reduction, targeting some urban road sections in Pyeongtaek city and some provincial road sections in Icheon city among basic local governments, namely among medium and small cities in Seoul Metropolitan Area.

  • 최근 세계 각지에서 발생하는 대규모 터널 화재사고는 많은 사상자를 동반하고 이에 따른 경제적, 사회적 손실 또한 방대하게 진행되는 실정이다. 터널 구조물의 화재 특성상 외부에 쉽게 노출되지 않기 때문에 화재 발생 시 화재에 노출된 표층이 박리되거나 비산해서 단면결손이 생기는 폭렬 현상(explosive spalling)이 발생하게 된다. 이러한 폭렬 현상은 붕괴와 같은 대형 참사로 이어질 가능성이 크다. 따라서 본 연구에서는 터널 내 화재 발생 시 콘크리트 구조물의 폭렬에 의한 붕괴를 예방하기 위하여 이액형 상온경화 실리콘 고무와 인체에 무해한 친환경 첨가제인 펄라이트를 일정한 혼합비(5wt%, 10wt%, 15wt%, 20wt%)로 혼합하여 고성능 난연 복합체를 제조하고, 열적 특성과 난연 특성을 연구를 진행하였다. 열적 특성에 관한 시험으로 TGA를 측정하였으며, 난연 특성에 관한 시험으로는 화염 시험, 내화로 시험, 탄화로 시험을 진행하였다. 우선 TGA 시험은 $20^{\circ}C/min$ 승온 속도로 $800^{\circ}C$까지 실험을 하였고, 화염 시험은 제작한 시편과 gas torch($1200^{\circ}C$)의 화염 거리를 약 10cm로 하여 약 1시간 동안 시험을 하였다. 내화로 시험은 내화로 장치를 이용하여 RABT curve(5분만에 $1200^{\circ}C$도달 후 한 시간 동안 유지 후 냉각, 총 시험 시간 180분) 조건을 만족하는 환경에서 제작한 시편을 콘크리트에 부착하여 콘크리트의 내부온도를 측정하였다. 탄화로 시험은 탄화로 장치를 이용하여 $2^{\circ}C/min$ 승온속도로 $900^{\circ}C$까지 실험을 하여 외부 형태 변화를 관찰하였다. 각각의 시험 결과 TGA 열분해 결과 순수한 실리콘 고무보다 난연제인 펄라이트를 첨가했을 때 더 높은 온도에서 초기 분해 거동을 보였으며, 최종 잔류량은 80%를 보였고, 5 wt%의 펄라이트가 혼합된 시편의 최종 잔류량이 높은 것으로 보아 열분해에 가장 강한 조성임을 알 수 있었다. 화염 시험 결과 펄라이트가 혼합된 모든 시편에서 $300^{\circ}C$가 넘지 않은 결과를 보였다. 이는 제조된 복합체가 화염에 직접적으로 장시간 노출이 되어도 안전하다는 것을 알 수 있다. 내화로 및 탄화로 시험 결과 펄라이트가 15wt%와 20wt%가 첨가된 시편들보다 5wt%와 10wt% 첨가된 시편들이 고온에서 안정하다는 것을 보였다.

A Study on the Problems and Character for Management of Fire Prevention at the Wholesale Markets (도매시장 화재안전관리 측면에서의 특성 및 취약요소에 관한 연구)

  • There are lots of buildings with their deterioration in wholesale markets which have high concentration that can cause big fire due to difficulty of installing fire protection system in fire compartment, absence of safety awareness, access by many and unspecified people. There had been big fire that caused by these kind of fire weakness such as Seomun Market's in 2005 which costed tens of millions dollar, Dongmun Shopping District’s with 7 casualties. Now counter-measure against these fire is urgently needed. This study drew characteristic of current condition of wholesale markets and the one classified by main agent, fire cases and prevention of fire through statistical data. Also, current condition of fire prevention management and actual state were investigated by conducting a survey of person in charge of fire prevention and current practician. As a result, it was shown that securing professionalism of fire prevention, improvement of structural weakness of buildings, obtaining reliability on performance of fire protection system and internal fire education against practician in the market are needed.

  • In the smart home era, fire safety is very important for human life and facility safety. Casualties and property damage from the fire would be a huge national loss. In this paper, we propose to predict the risk by determining the operating time of the fire detector according to the fire in the smart home. Among IoT fire detectors, heat detectors and smoke detectors, the risk can be predicted due to the difference in the operating time depending on the fire. Based on the results of this experiment, the ion-type smoke detector shows very fast characteristics, so it would be good to use the results in future fire prevention facility.

Effects of Torilis japonica Extract on DNCB-induced Contact Dermatitis in BALB/c Mice (사상자 추출물이 DNCB로 유도된 BALB/c 마우스의 접촉성 피부염에 미치는 효과)

  • 목적: 본 연구에서는 TJ 열수추출물이 아토피 피부염에 미치는 효과를 확인하고자 하였다. 방법: 2, 4-dinitrochlorobenzen (DNCB)으로 BALB/c 마우스에 1차 감작 후 3주간 2차 감작을 시행하여 아토피 유사 피부병변을 나타내는 접촉성 피부염을 유발한 뒤 동결건조 처리된 TJ 분말을 (PBS/EtOH/Cremophor=6:1:3)에 용해시켜 쥐의 등 피부에 10, 50 mg/ml 농도로 3주간 도포하였다. 결과: TJ 열수추출물은 아토피 피부 병변의 염증세포 침윤을 억제하였고 (10, 50 mg/ml ) 표피와 진피 두께를 회복시켰으며 (10, 50 mg/ml ), 혈청에서 히스타민 방출을 억제하였다 (00 mg/ml ). 또한 Th2 세포와 관련된 cytokine인 interleukin (IL)-4, IL-5, IL-13과 thymic stromal lymphopoietin (TSLP)의 mRNA 발현 양을 감소시켰고 (10, 50 mg/ml ), Th2 chemokine인 thymus- and activation-regulated chemokine (TARC or CCL17)의 mRNA의 발현 양을 감소시켰다 (10, 50 mg/ml ). 결론: TJ 열수추출물을 BALB/c 마우스에 외용하였을 때 항아토피 효과가 있음을 확인하였으며 따라서 TJ가 아토피 피부염 치료의 외용제로 활용될 수 있을 것으로 사료된다.
