Histological and Biochemical Studies on the Rooting of Hard-wood Cuttings in Mulberry (Morus species) (뽕나무 古條揷木의 發根에 關한 組織 및 生化學的 硏究)

  • Rootability of the hardwood cuttings of mulberry was related not only histological characteristics but dependent on biochemical properties. In this connection, the characteristics of the hardwood cuttings were histologically observed and the growth substances produced by the cuttings were also identified by means of mung bean bioassay. Amino acid, carbohydrate, nucleic acid contents, and the C/N ratio were also analysed. The results are summarized as follows. 1. There were differences in rootability of cuttings between mulberry species and varieties Among the three mulberry species tested, Morus Lhou Koidz. showed the highest rootability while M. bombycis showed the lowest one. In varietal differences in rootability, it was shown that the varieties could be grouped according to rootability: high varieties(above 80%), medium(41~79%), and low(below 40%). The higher varieties were Kemmochi, Nakamaki, Kosen, and Wusuba roso. 2. The histological characteristic of the hardwood cuttings most closely related to rootability was cell layer arrangement in the sclerenchyma tissue. The lower rootability varieties developed two or three overlapping cell layers in the bark tissue and in the higher rootability varieties they were scattered over the primary cortex. 3. In the higher rootability varieties, there was a positive correlation between the development of root primodia and rootability of the hardwood cuttings. It was also shown that there was a close relationship between the size of primodia and the surface area of the lenticel with rootability of the cuttings. 4. Effect of growth substances extracted from the hardwood cuttings were determined by mung bean bioassay. The higher rootability varieties usually showed higher activities of the growth substances, in contrast the lower rootability varieties showed higher activities of the inhibitory substances. 5. It was evident that the substance separated by paper chromatography was identified as indole acetic acid with $R_f$ value ranging from 0.3 to 0.5. The other substances detected at a $R_f$ value ranging from 0.8 to 1.0 and origin to 0.1 were also responsible for rooting. 6. There exists a quantitatively different distribution of growth substances in a synergistic system in the tissues of cuttings, and the balance between growth and inhibitory substances gives rise to the development of rooting. Particularly, no descent of the substances from winter buds resulted in no rooting of cuttings but these substances were produced a week after planting in a warm environment. 7. It was shown that there were positive correlations between carbohydrate ($r=0.72^*$) and total sugar ($r=0.67^*$) and rootability, respectively, but there were negative correlations between reducing sugars ($r=-0.75^*$) and rootability. 8. High C/N ratio gave rise to high rootability($r=0.67^*$). The latter therefore depended on high amount of carbohydrate rather than nitrogen in the cuttings. 9. The content of RNA and DNA in the cuttings was not changed for upto two weeks after the cuttings were planted. Then an increase in RNA content took place in only the high rootability varieties. 10. There were quantitative and qualitative differences in the compositions of the amino acids between the high rootability varieties and the low rootability varieties. More aspartic acid and cystine were found in the higher rootability varieties than in the low rootability varieties.

Effect of Several Herbicides in the Polyethylene - film Mulched Young Mulberry Field (P.E. 필름피복(被覆) 밀식(密植) 뽕밭에서의 수종(數種) 제초제(除草劑) 처리효과(處理效果))

  • Requirements in weed control in a mulberry field are much similar to those in orchards, but also feature a longer period of weed control of various kinds of persistent weeds, i.e., spring, summer, and winter annuals as well as perennials. In addition the mulberry tree is relatively more sensitive to herbicide injury. Hence, very few herbicides have been used in mulberry field. The present study was conducted to evaluate the usefulness of oxyfluorfen in comparison with alachlor and simazine, which are registered for ordinary mulberry field in Korea, for weed control efficacy in the new, rapidly increasing practice of transparent polyethylene-film mulched and densely planted younger mulberry culture. Dominant spring weeds were Galium spp., Erigeron spp., Polygonum senticosum, and Chenopodium spp. in the non-mulched interbed area in contrast to the Digitaria spp. and Potulaca spp, under mulch. Dominant summer weeds were Digitaria spp., Portulaca spp., Erigeron spp., Artemisia spp. and Calystegia japonica in the non-mulched interbed area while weeds did not occur significantly during summer under mulch which were shaded by vigorously growing mulberry trees. The weeds occurred under mulch in spring reduced shoot growth of young mulberry tree resulting in the reduced yield of mulberry leaves for silkworms. The weeds occurred in the interbed area did not affect until May, but interfered later summer- and fall-growth of mulberry tree. Early single spring application of alachlor(EC), simazine(WP) or oxyfluorfen(EC) at a rate of 650 g, 750 g or 350 g ai per ha, respectively, controlled most annuals satisfactorily to fall in the mulched bed area. In the nonmulched interbed area, however, thrice does of alchlor or simazine was necessary for satisfactory control of spring weeds, followed by summer application of alachlor or simazine at twice dose level as tank mixture with paraquat at 490 g ai per ha for satisfactory control of summer to fall weeds. Single spring application of oxyfluorfen at a rate of 1400 g ai per ha was persistently effective to control satisfactorily even summer and fall weeds. However, heavy rainfall splashed soil borne oxyfluorfen to the lower branch leaves causing some leaf burns. Spring application of oxyfluorfen at a rate of 350 g ai per ha followed by summer application of oxyfluorfen and paraquat tank mixture (350 g ai + 490 g ai) was the best choice for the non-mulched interbed area weed control among the treatments.

Evaluation of Biological Activity and Analysis of Functional Constituents from Different Parts of Mulberry (Morus alba L.) Tree (뽕나무(Morus alba L.) 부위별 생리활성 측정 및 기능성 물질 분석)

  • Evaluation of biological activity and analysis of functional constituents from water and ethanol extracts of four different parts of mulberry (Morus alba L.) tree were carried out to develop functional ingredients and foods using extracts of mulberry tree. The water and ethanol extracts of four different parts of mulberry tree were prepared and their biological activities and functional constituents determined by in vitro assays and HPLC, respectively. In general, ethanol extracts showed stronger biological activities and higher functional constituents than water extracts. Ethanol extracts of mulberry fruit, root bark, and twig showed stronger antioxidant ($IC_{50}=128.4{\mu}g/mL$), ${\alpha}$-glucosidase ($IC_{50}=12.0{\mu}g/mL$), and lipoxygenase ($IC_{50}=36.3{\mu}g/mL$) and tyrosinase ($IC_{50}=410.3{\mu}g/mL$) inhibitory activities, respectively, than those of other parts. Mulberry fruit and leaf showed the highest contents of anthocyanin (cyanidin 3-glucoside: 213.20 mg/100 g) and chlorogenic acid (514.97 mg/100 g), and especially ethanol extract of mulberry leaf contained higher quercetin 3-O-(6-O-malonyl)glucoside (143.25 mg/100 g) and kaempferol 3-O-(6-O-malonyl)glucoside (30.25 mg/100 g) contents without water extract of mulberry leaf. Meanwhile, mulberry twig contained both oxyresveratrol glycoside (48.90 mg/100 g) and its aglycone (21.88 mg/100 g), whereas mulberry root bark contained mostly oxyresveratrol glycoside (724.05 mg/100 g). Additionally, mulberry root bark and leaf contained much higher ${\gamma}$-aminobutyric acid (223.90 mg/100 g) and 1-deoxynojirimycin (86.07 mg/100 g) contents, respectively, than other parts of mulberry tree. These results suggest that high quality processed foods and functional foods using mixtures of mulberry fruits, leaves, twigs, and root barks should be developed for prevention and inhibition of several pathological disorders.

A Study on Sulwhaji (설화지(雪花紙)에 대한 연구)

  • Sulwhaji(雪花紙, meaning "snow-white flower paper") had been originally manufactured before 16th century until after 19th century in Pyunggang, Korea, which represented the typical characteristics of traditional Korean paper. Since 17th century, however, it had been also made in Namwon, Sunchang, Jinju and so on. Its material was mulberry bark tree and was bleached beautifully white by using snow. It was highly qualified mainly used for painting, caligraphy, and book printing. That had been used in the Royal palace and as cherished gift for foreign envoys.

  • 상전벽해(桑田碧海), 뽕나무 밭이 변해 푸른 바다가 된 것은 아니지만, 논 밭만 즐비하던 시골 땅에 하루 아침에 아파트들이 즐비하게 들어서고 공단이 들어선 것은 비단 어제 오늘의 일이 아니다. 교통의 발달로 경기도 근교는 서울과 출퇴근 생활권이 된지 오래다. 사정이 이렇다 보니 도시화의 물결은 점점 농촌 깊숙이 파고들고 있는 형편이다. 이러한 도시화로 피해를 입고 있는 많은 산업들이 있겠지만 그 중에서도 축산업은 최대 피해자라고 볼 수 있다. 축산업을 평생의 업으로 삼고 살아온 축산인들에게는 보상금 보다 산업을 지속적으로 영위할 수 있는 것이 가장 우선되기 때문이다. 모든 산업이 마찬가지겠지만 도시화가 가장 가속화되고 있는 경기도 일대에 가장 많은 분포를 보이고 있는 낙농업의 경우 상황은 더욱 심각하다. 공단과 신도시 개발 등으로 터전을 잃고 있는 낙농가들이 한해해만 몇십 농가가 된다는 것이 업계의 증언이다. 도시화로 인하 목장 이전의 문제점은 무엇이고 성공적인 이전을 한 농가의 성공요건을 낱낱이 분석한다.

실크의 비의류용 이용

  • 전통적인 고급 의류로서 견이 사용되기 시작한 이래, 섬유의 여왕으로서 실크는 지속적인 사랑을 받아왔다. 그러나 최근 실크에 가까운 합성섬유가 만들어져 견섬유의 위치를 위협하고 있는 현실이다. 이에 섬유재료로서 실크가 갖는 경쟁력은 어려운 위치에 처하게 되었고, 보다 경쟁력을 높이기 위해서는 실크제품의 제조공정에서 필연적으로 발생하는 부산물의 활용에 관심이 고조되기에 이르렀다. 부산물의 활용 분야로서는 화장용품, 의료용, 식품용, 산업용 등으로 그 응용분야가 광범위하게 확대되었고 지금도 속속 새로운 분야에의 응용이 시도되고 있다. 앞으로의 연구에서 견의 기능성 발현의 기구가 보다 자세히 규명된다면 실크분야에서 완전히 새로운 장이 열릴 것으로 기대된다. 그렇게 된다면 지금의 부산물 이용만으로는 수요를 감당할 수 없을지도 모를 것이다. 그때에는 직접 누에의 체내에서 견을 채취하게 될 것이다. 그렇게 된다면 과거의 풍요로운 뽕나무 밭도 되살아 나고 잠사 견업의 활성화에도 연관된다.

A New Hanji Manufacturing from Morus Resources (뽕나무자원의 새로운 한지제조 특성)

  • This study was carried out to investigate the sheet properties of Morus Hanji, made by different pulping methods, such as alkali, alkali-peroxide and sulfomethylated pulping, and different stocks composed to bast and whole stalk fibers. Two species of Morus, M. alba and M. lhou, were used. Pulp yield of the whole stalk parts, its brightness and opacity were higher than those of bast part. In particular sulfomethylated pulp has higher brightness of $62{\sim}63%$, than other pulps(average brightness $30{\sim}40%$). In mechanical properties, paper made of bast fiber has higher strength than those of the whole stalks. Hanji made from M. lhou, whole stalk, and sulfomethylated pulp gave better sheet formation and higher strength than those of the others. Hanji from M. alba bast fiber and whole stalk pulps were not so good sheet properties.

Inhibition of Lipid Autoxidation by the Extract of the Submerged-liquid Culture of Mushrooms in the Medium Containing Mulberry Tree Powders (뽕나무 첨가 배지에서 배양한 버섯균사체 배양물의 자동산화 억제 효과)

  • Effect of mulberry tree powders on the antioxidant activity of submerged -liquid culture of mushrooms was investigated. Agaricus blazei (AB), Hericicum erinacium (HE), Pleurotus ostreatus (PO), Phellinus linteu (PL) and Paecilomyces japonicus (PJ) were cultured in a shaking incubator (200 rpm, $25^{\circ}C$) for 7 days in culture media: 1) basal medium (BM) and 2) BM+1% mulberry tree powders (BMM). Hot water extracts from the submerged-liquid cultures of mushrooms and BMM were freeze-dried for the measurement of antioxidant activity, of which reaction mixture (25 mL: 10 mL of 0.2 M sodium phosphate buffer, pH 8.0; 4.5 mL distilled water; and 10.5 mL ethanol) contained 275 $\mu$mol linoleic acid and one mg test sample. The reaction mixture was incubated in a shaking incubator (200 rpm, 4$0^{\circ}C$) for 16 days. Peroxide value (POV) was measured for a period of over 16 days, and malonaldehyde (MA) was determined only for samples from the day 16 of incubation. Mycelial weight of mushroom strains cultured in BMM was greater than BM. The antioxidant activities of AB-cultured in BW (AB-BMM) and HE-cultured in BMM (HE-BMM) were superior to those of other mushroom strains-cultured in BMM or BM and of BMM. These results suggest that mulberry tree powders enhance the antioxidant activity of submerged-liquid culture of mushroom strains. The AB-BMM and HE-BMM were the most active cultures.

Recovery of Several Mulberry Varieties from Late Frost Damage (늦서리 피해 뽕나무 품종별 회복정도)

  • The recovery of mulberry from a late frost (April 24 to 27) was investigated for four varieties : Kaeryangppong, Cheongilppong, and Jamsang 21 in Cheongju area, Chungbuk Province. 1. Unfavourable meteorlogical conditions for recovery of mulberry followed the early late frost : lower than normal temperatures in mid-April, less sunshine in early and mid-Jay, lower temperatures and early and less precipitation in late-May, and less precipitation than normal in early June. 2. Jamsang 21 grew to the 2-3 leaf stage first, at which time Kaeryangppong and Jamsang 22 were at the 1-2 leaf stage, and Cheongilppong was at the swallow-bill shape-1 leaf stage. 3. The first leaf stage occurred on May 12, 15 days after the frost and, 19 days later in an average year. The difference lasted until the harvesting season, the 6 leaf stage. 4. Young shoot length was 50-58% of that in 1989. Kaeryangppong was the shortest at 50% and the Jamsang 21 highest at 58% of 1989 values. 5. Leaf number was 52-67% that in 1989. Kaeryangppong was the lowest at 52% and the Jamsang 21 highest at 67% of 1989 values. 6. Young shoot and leaf weight was 44-63%, and leaf weight 39-61%, compared to 1989. Kaeryangppong and Cheongilppong were the lowest, Jamsang 21 and Jamsang 22 were the highest, compared to 1989 data. 7. Young shoot length was 50-58% compared with 1989. Kaeryangppong was the lowest as 50%, the highest Jamsang 21 as 58%. 8. Number of leaf was 52-67% compared with 1989. Kearyangppong was the lowest as 52%, the highest Jamsang 21 as 67%. 9. Young shoot and leaf wight was 44-63%, and leaf weight was 39-61% compared with 1989. Kaeryangppong was the lowest, lowest, Jamsang 21 was the highest, Cheongilppong was similar with Kaeryangppong and Jamsang 22 was similar with Jamsang 21. 10. Rate of young shoot and leaf weight to total weight decreased from 75% in 1989 to 55-61% in 1990. Rate of laef weight to young shoot decreased from 72-82% to 67-71%, showed 5-10% decrease. 11. Leaf yield decreased highly in Jamsang 22 as 57% compared with 1989. Kearyangppong 61%, Jamsang 21 64%, Cheonglppong 72%. Leaf yield was 827kg/10g in Kaeryangppong, 1, 000-1, 050kg/10a in the other varieties.

Characterization of Phytoplasmal Disease Occurred on Floricultural Crops in Korea (우리나라 화훼류 파이토플라스마병의 특성)

  • Seven phytoplasma diseases have been occurred on floricultural crops in Korea : Ph-ch1 and Ph-ch2 of chrysanthemum, Ph-lily of lily, petunia flat stem-Korean (PFS-K) of petunia, poinsettia branch inducing- Korean (PoiBI-K) of poinsettia, statis witches' broom-Korean (SWB-K) of statis and azalea witches broom (AWB). Classification of the seven phytoplasmal diseases based on 16S ribosomal RNA (rRNA) sequences showed that floricultural crop phytoplasma disease were widespread in order of aster yellow (AY), stolbur and X-disease in Korea. In phenotypic characters, the fasciation was occurred in both monocotyledon plant of lily and dicotyledon plants of petunia and poinsettia. Besides, the fascination was occurred in Ph-lily of stolbur, petunia PFS-K of AY and PoiBI-K of X-disease. This result indicated that phytoplasma classification based on 16S rRNA and symptoms are not consistently related. The comparison of 16S rRNA sequence of the seven floricultural crop phytoplasma with five tree phytoplasmal diseases of jujube witches' broom, paulownia witches' broom, wild jujube witches' broom, mulberry dwarf, golden rain phytoplasma occurred in Korea showed as high as 88.5-99.9% homology. Among them, especially mulberry dwarf showed the highest homology with the seven floricultural crop phytoplasms. Based on this result, floricultural crop phytoplasmas were assumed to be transmitted by insect vectors from tree phytoplasmas in Korea.
