This study was carried out to investigate the anti-wrinkle effects of peptides derived from collagens isolated from Asterias amurensis, which was collected in the East Sea. The molecular weights of the peptides were between 10-50 kDa, as determined through sephadek G-75 gel. The cytotoxicities against CCD-986sk cells and HEL-299 cells were measured using the MTT assay. The cytotoxicity of all the fractions(F1: Fraction No. 4-13, 116 kDa; F2: Fraction No. 25-30, 100 kDa; F3: Fraction No. 45-55, 58 kDa; F4: Fraction No. 59-63, 43 kDa; F5: Fraction No. 79-90, 24 kDa) was less than 25%, by the addition of 1.0 mg/mL. These peptides did not show any adverse effects on human skin cells. In the presence of F1 at 1.0 mg/mL, matrix metalloproteinase-1 (MMP-1) expression of UVA-induced human normal fibroblasts was reduced to 34.8%. Overall, the results seem to suggest that peptides of approximately 20 kDa have superior anti-wrinkle effects.

The bacterial isopentenyl transferase (ipt) gene involved in cytokinin biosynthesis was fused with 35S promoter of cauliflower mosaic virus (CaMV) and introduced into tobacco plants (Nicotiana tabacum L. cv. Samsun) via Agrobacterium-mediated transformation. As expected, ipt gene was constitutively expressed in all tissues of transgenic plants. Several primary transgenic plants were obtained that expressed different level of transcripts for ipt gene. Three of transgenic plants with different expression level of ipt gene were selected and selfed to obtain homozygous line for further analysis. A number of interesting phenotypic changes such as viviparous leaves, delayed senescence, larger axillary shoots, an abundance of tiny shoots at the apex and a release of lateral buds, were observed in transgenic plants. Chlorophyll content was 1.5- t.o 4-fold higher in transgenic plants as compared with non-transformed plants. These results indicate that the cytokinin synthesized in transgenic plants could improve forage crop yield by delay of leaf senescence and increase of leaf number.

A new static contractor using capillary phenomena induced by a highly packed fiber bundle was developed for the solvent extraction. When two immiscible phases being cocurrently and forcedly fed into the packed fiber bundle, the contactor brings about a very large liquid-liquid contact area for mass transfer within a small definite space without any turbulence and drop phenomena. In order to test the characteristics and stability of the static contractor system, continuous extraction experiments of TBP-uranyl ion-nitric acid system were done and compared with the batch extraction experiment of the same chemical system. The performance of the static contractor were the same as that of the ideal batch extractor with the same extraction condition. For the increase of the extraction yield by the contactor, the increase of organic flow rate was required at a fixed aqueous flow rate, and a certain residence time of the aqueous phase flow within the contactor system had to be maintained to meet the performance of the batch system of the same phase ratio. The residence time in the case of TBP-uranyl ion-nitric acid system was about 1.9 minutes. This system was confirmed to be effective and stable enough for purposes of the kinetic study of solvent extraction as well as the mutual separation and purification of ordinary materials because of good reproducibility and the stable and large static liquid-liquid contact area.
This experiment was carried out to compare the change of muscle protein compositions, amino acid compositions of muscle protein, and free amino acid compositions by age of the Israeli strain of common carp, Cyprinus carpio nudus. Protein compositions of the muscle were: sarcoplasmic protein $25.8-27.2\%$, myofibrillar protein $62.3-56.2\%$, residual intracellular protein $9.6-13.2\%$ and stroma $2.3-2.9\%$. In between 1 year and 3 years, there were differences as follows; myofibrillar protein in 1 year was much than 3 years, and other proteins in 3 years were much than 1 year. By SDS-polyacrylamide gel electrophoresis, sarcoplasmic protein of the samples in 1 year an 3 years were composed of 11 subunits and 10 subunits, respectively. And appeared 210,000 dalton component in 1 year but did not appeared in 3 years. Myofibrillar protein was composed of 23 subunits in both 1 year and 3 years but the differences of subunits by age were not observed. No differences were observed by age in the composition of myofibrillar protein and residual intracellular protein. Amino acid composition of muscle protein in both 1 year and 3 years were no differences to each other, but the contents of glutamic acid, aspartic acid, lysine were higher than other amino acids. The amount of total free amino acid in 1 year was much than in 3 years.
To constrain the depositional ages of the Gyeongsang sedimeantary formations, SHRIMP U-Pb ages were determined from detrital zircons in three samples: (1) a pebble-bearing sandstone from the lowermost Jinju Formation of the Sindong Group and (2) two conglomerates from the Silla Conglomerate of the Hayang Group. Their concordia ages are $112.4{\pm}1.3(2{\sigma})$ Ma and $110.4{\pm}2.0(2{\sigma})$ Ma respectively. Such ages represent the maximum deposition ages for the lowermost Jinju Formation and Silla Conglomerate, indicating the deposition of the Jinju Formation started from late Aptian and lasted to early Albian, then deposition of the rather thin Chilgok Formation and Silla Conglomerate was followed during the Albian. The age distribution of the analyzed detrital zircons indicates the presence of protoliths, or zircons derived from them, regarding a wide span of igneous activities from Mesozoic to Archean. Among such ages, there are Mesoproterozoic, Neoproterozoic and Paleozoic igneous activities, which have not been known or seldom reported from Korean peninsula. These ages further suggest the possible presence of rocks with such ages during the deposition periods or their derivation through a long river system developed into the continents at the time of deposition.
Railway bridges with continuously welded rail have a limitation of span length due to track-bridge interaction. In order to overcome this, a sliding slab track system has been developed that comprises with a low-frictional sliding layer between the bridge deck and the track slab to isolate the longitudinal behavior between the bridge and the track. In this study, a real scale track system is prepared to experimentally evaluate the longitudinal frictional behavior. Applied loading rates were 0.2, 1.0, 5.0 and 10mm/min; vertical mass on the track are track slab only, 5,000 and 10,000kg added mass, respectively. Test results showed that the resulting frictional coefficients varied from 0.22 to 0.33. In addition, 10,000 cycle loadings were applied to simulate repetitive sliding to represent 30 years of service life. The frictional coefficient increase was measured and found to be 7% of that of the initial loading stage, which means that the sliding layer is adequate to provide low-frictional behavior for the sliding slab track system. Effects of changes of the frictional coefficient of the sliding layer were analyzed by rail-structure interaction analysis.

We measured rare earth element and Zr concentrations of USGS granite standard material GSP-2 and GSJ granite standard material JG-1a to clarify the effect of zircon during rare earth element analysis using ICP-MS. We also measured rare-earth element and zirconium (Zr) contents of zircon from granite by acid-digestion methods using conventional teflon vial and pressure-bomb. The results show that acid-digestion using teflon vial dissolved ca. 50% of zircon compared to pressure-bomb method. The Zr contents of JG-1a and GSP-2 gave ca 50% of reference value. However, rare-earth element abundance of JG-1a and GSP-2 were similar to those of reference values. This suggests that the decomposition degree of zircon might give a negligible effect on a petrological and geochemical interpretation using chondritenormalized REE pattern.
To investigate a quantitative evaluation of the actinobacteria, we have collected samples from various kinds of bamboo forest soil. Each different layers contained $2.7{\times}10^6-2.7{\times}10^8$ CFU/g of actinobacteria which was the highest in litter layers of Sasa boreali forest soil. We obtained 330 actinobacteria from different layers of bamboo forest soil; litter (100 strains), humus (70 strains), and rhizosphere soil (160 strains). Based on the colony morphology (aerial mycelium, substrate mycelium, and soluble pigment), isolates were divided into thirty-six groups and we selected 50 representative isolates. 16S rRNA gene sequence analysis showed Streptomyces was major actinobacteria (94%) and they were categorized as cluster I (2 strains), II (35 strains), III (6 strains), and IV (7 strains), respectively. The diversity index of 50 Streptomyces collected from the bamboo forest soil was calculated with the Shannon-Wiener method. Bamboo litter showed higher diversity index level of 3.33 than that of humus and rhizosphere soil. Also, antibiotic activities of our isolates were investigated against Botrytis cinerea, Xanthomonas campestris, Xanthomonas axonopodis pv. vesicatoria, and Bacillus cereus and found in 74, 16, 25, and 24 strains, respectively.

제4기 만빙기에서 후빙기 사이의 기후변화는 인류문명에 있어 정착생활 및 농경활동과 더불어 잉여생산물 저장을 위한 토기제작 등과 같은 생활상의 변화를 가져 온 계기가 되었다(F.M. Chambers, 1993; Guoyu Ren, 1998; Mori Y, 2002; Martin Bell et al., 2004). 식물화석, 화분, 식물규소체 등의 대리자료(proxy data)를 이용하여 이시기의 식생환경을 밝히는 것은 고생태복원분야 뿐 아니라 당시의 환경변수들 을 추정하여 인간활동에 어떠한 영향을 미쳤는지에 대한 중요한 정보를 제공해준다. 그러나 홀로세 초기 환경은 인간에게 지대한 영향을 미쳤으나 후기로 올수록 인간 스스로가 환경을 변화시키는 주체가 되었다(Martin Bell et al., 2004). Meyer&Turner(1994)는 인간활동이 자연적인 영향보다 생물구에 시계열적인 변화를 가져왔다고 하였으며, R.B Singh(1995)는 현재뿐 아니라 과거까지 다양한 인위 적인 간섭이 전지구적인 환경변화를 야기 시키는 주된 원인이라고 하였다. 따라서 환경을 복원한다는 것은 바로 그 지역을 생활 터전으로 삼았던 당시 선사인들의 생활상을 간접적으로 들여다 볼 수 있는 기회를 제공해준다. 현재까지 국내 고고학 유적지에서 이루어진 대부분의 발굴조사는 유물과 유구를 토대로 당시 선사인의 생활상을 밝히는데 중점을 두었으나 최근 인간과 영향을 주고 받았을 환경에 관심을 갖고 이를 통해 역으로 인간활동을 추적하려는 다수의 연구들이 제4기학, 고고학, 지리학, 인류학, 생물학, 농학, 기후학 등 다양한 분야에서 학제간 연구로 진행되고 있다(황상일 등, 1999; 조현종, 2000; 김주용 등, 2002; 최정민, 2004; 윤순옥 등, 2005). 그러나 이러한 연구에 있어서 어떠한 대리자료(proxy data)를 어떻게 고고학 유적지에 적절하게 적용시킬 것인지 대리자료의 조성변화나 값의 해석에 있어서 기후인자와 인간영향을 어떻게 분리해 낼 것인지에 대한 문제점이 남는다. 예를 들어 화분분석의 경우 1차적인 환경변화에 따른 식생조성변화를 반영한다. 인간이 활발한 농경활동을 하게 되면서 주변식생을 제거하게 되고, 제거된 나대지에는 재배작물과 잡초들이 자라게 된다. 그리고 이들 화분은 주변 소택지에 퇴적물과 함께 퇴적되어 화분분석 결과 벼과(科)(Gramineae)와 문화지표수종이라 부르는 여뀌속(屬)(Persicaria), 국화과(科)(Composite), 쑥속(屬)(Artemisia), 쐐기풀속(屬)(Urtica) 등의 화분에 있어 확연한 조성변화를 보여준다. 그러나 화분을 이용한 분석은 토탄층이나 유기질 퇴적물이라는 한정된 토양에서만 분석이 가능하며, 조성변화에 있어서 역시 기후와 인간활동의 영향 모두를 반영하기 때문에 이를 구분해 주는 명확한 기준이 필요하다. 따라서 본 연구에서는 이러한 다양한 대리자료 중 고고학 유적지에 적용 가능한 동식물 화석 및 미화석, 지리, 지질학적 자료에 어떠한 것들을 적용할 수 있는지를 살피고, 몇 가지 대리자료를 중심으로 선사인의 농경활동에 대해 살펴보고자 한다.
The tuber of Liriope platyphylla was extracted with 80% aqueous MeOH and solvent-fractionated with EtOAc, n-BuOH and $H_2O$. Evaluation of growth inhibitory activity of each fractions on various carcinoma cells, A549, SK-OV-3, SK-Mel-2, XF-498 and HCT-15, indicated the n-BuOH fraction to be the highest in the activity. From the fraction, two saponin compounds were isolated through Amberlite XAD-II and silica gel column chromatographies, repeatedly, and their chemical structures were elucidated as spicatoside A and B by interpretation of spectral data, NMR and IR, and adaptation of acid hydrolysis. Spicatoside A showed growth inhibitory activity on carcinoma cells, and the $IC_{50}$ values against A549, SK-OV-3, SK-Mel-2, XF-498 and HCT-15 cells were determined to be 17.3, 21.7, 14.9, 18.8 and $15.6\;{\mu}g/ml$, respectively.
