Air-cathode microbial fuel cell consisted of 4 unit cells were operated under batch condition and electricity generation and microbial community structure variation were investigated, depending on separator types and cathode characteristics: A) PEM(Proton Exchange Membrane)-30% Wet proofing Carbon Cloth(WC), B) AEM(Anion Exchange Membrane-WC, C) CEM(Cation Exchange Membrane)-WC, D) PEM-No Wet proofing Carbon Cloth(NC). Maximum power densities of PEM-WC, AEM-WC and CEM-WC were 510.9, 522.1 and 504.8 $mW/m^2$, respectively. But PEM-NC showed relatively lower maximum power density of 218.3 $mW/m^2$. And PEM-WC, AEM-WC and CEM-WC showed similar internal resistances(20.0-28.2 ${\Omega}$). PCRDGGE, PCA and diversity indices showed that uncultured bacteria which reported in previous MFC studies were detected in suspended growth bacteria and attached growth bacteria would be affected not by separator type but by cathode characteristic. Thus, cathode characteristic can be one of the critical factors for power generation in air-cathode MFC using PEM, AEM, and CEM as separator.
The relationships among total epiphytes, algal epiphytes and eelgrass ($Zostera$$marina$ L.) were studied at eelgrass medows from July, 1998 to July, 1999 in Yulim-ri, Yeosu, Korea. Epiphytic diatoms on eelgrass leaves were observed from July to December 1998. From the results of this study, we inferred the following three (3) conclusions : 1) As eelgrass grew older, biomass of epiphytes increased, according to relationships between the leaf length and area of eelgrass and biomass (DW, AFDW and Chl. $a$) of epiphytes. 2) According to the ratio of dry weight, ash-free dry weight and Chlorophyll of epiphytes, the algae with calcareous or siliceous skeletons, such as coralline algae or diatoms, were dominated in the epitphytic algae community. 3) The autotrophic index (AI) calculated from AFDW and Chl. $a$ of epiphytes varied from 151 to 375. However, the period of autotrophic community was shorter than heterotrophic community and the value of AI was high. From these results, we inferred that heterotrophic community, including detritus or microorganisms were dominated in the most of research period.

Coal-, coconut- and wood-based activated carbons and anthracite were tested to evaluate adsorption and biodegradation performances of chloral hydrate. In the early stage of the operation, the adsorption was the main mechanism for the removal of chloral hydrate, however as increasing populations of attached bacteria, the bacteria played a major role in removing chloral hydrate in the activated carbon and anthracite biofilter. It was also investigated that chloral hydrate was readily subjected to biodegrade. The coal- and coconut-based activated carbons were found to be most effective adsorbents in adsorption of chloral hydrate. Highest populations and activity of attached bacteria were shown in the coal-based activated carbon. The populations and activity of attached bacteria decreased in the order: coconut-based activated carbon > wood-based activated carbon > anthracite. The attached bacteria was inhibited in the removal of chloral hydrate at temperatures below 10$^{\circ}C$. It was more active at higher water temperatures(20$^{\circ}C$ <) but less active at lower water temperature(10$^{\circ}C$>). The removal efficiencies of chloral hydrate obtained by using four different adsorbents were directly related to the water temperatures. Water temperature was the most important factor for removal of chloral hydrate in the anthracite biofilter because the removal of chloral hydrate depended mainly on biodegradation. Therefore, the main removal mechanism of chloral hydrate by applying activated carbon was both adsorption and biodegradation by the attached bacteria. The observation suggests that the application of coalbased activated carbon to the water treatment should be the best for the removal of chloral hydrate.
Probiotics including Lactobacillus acidophilus, refer to a group of nonpathogenic organisms that protect the human host against gastrointestinal(GI) infections by pathogenic bacteria such as Shiga toxin-producing E. coli(STEC). In the study, the inhibitory effects of STEC ATCC 43894 adhesion by L. acidophilus A4 was investigated on the HT-29 epithelial cells. Specific proteins regulated by cell Iysates of L. acidophilus A4 on STEC ATCC 43894 were also characterized by proteomic analysis. Both cell mass and Iysate of L. acidophilus A4 have exhibited the profound inhibitory activity on the HT-29 cells(about 1.5 log scale reduction). Two-dimensional gel electrophoresis(2-DE) revealed seven proteins that were up-regulated by cell Iysates of L. acidophilus A4 and three proteins that were down-regulated. In addition, three protein spots were only detected in the presence of cell Iysates. These results suggest that inhibitory effects of STEC adhesion by L. acidophilus may be due to the regulation of specific protein of STEC.
For scrutinizing the changes of aggregated bacteria in Lake Paldang, the FISH method was applied by using the rhodamine labeled probes, and total bacteria, chloropyll a concentrations and nutrients concentrations were measured. The aggregates were collected with sediment traps. The T-N, T-P, chlorophyll a concentrations of aggregates were higher 5-15 times, 81-140 and 49-66 times than water samples, respectively. Also, the bacterial numbers of aggregates were 200 times higher than those of water smaples. The ratios of each groups of water sample were 2.1-7.4% for $\alpha$-group, 4.5-8.3% for $\beta$-group, 2.1-7.4% for $\gamma$-group, 2.1-6.1% for Cytophaga-Flavobacterium group and 0.1-2.5% for 'other'group, respectively. While, in aggregates, the ratios of $\alpha$-, $\beta$-, $\gamma$- and Cytophaga-Flavobacterium groups were very small and most abundant group was 'other' bacteria. With these results, the aggregated bacteria in Lake Paldang had a particular group composition of bacteria.
The objective of this study was to investigate community structure of attached algae and microbes in sedimentation basin of water and wastewater treatment plants by using respiratory quinone profile. There was an evident difference, in microbial community structure and attached algae species, between inclination plate settler and drainage canal in the sedimentation basin. The algae was composed of species in following order; Chlorophyceae>Bascillariophyceae>Cyanophyceae. The dominant quinone types of attached microorganisms in the wastewater treatment plant were plastoquinone (PQ)-9, vitamin(V)K-1 followed by UQ-8, but those for water treatment plant were VK-1, PQ-9 followed by UQ-8. These results suggests that nutrients, seasons and material of sedimentation basin have notable influence on composition of attached algae and microbial community structure in water and wastewater treatment plants.
초고압 가공처리는 별동의 화학 보존제를 사용하지 않고도 저온에서 식품유래 미생물을 사멸시킬 수 있기 때문에 식품분야에서 주목받는 새로운 가공기술이다. 이러한 초고압 처리의 장점 덕분에 관능적 특성이 우수하고 영양성분이 그대로 보존되는 고품질 식품의 제조도 가능하다. 고압 조건에서의 미생물 사멸정도를 측정하기 위해서는 흔히 실험실 규모의 장비(그림 1)을 사용하여 소량의 미생물 접종액을 처리함으로서 대량 처리시(그림 2)의 양상을 예측할 수 있다. 초기에 개발된 실험실 귬의 초고압 처리장비에는 일반적으로 고압용기 내부에 온도감지 장치가 부착되어 있지 않아, 압력 조건 하에서의 압축발열 및 순간 감압냉각 효과가 제대로 보고되지 않았다. 그러나 가열효과를 고려하지 않으면 초고압 처리기 특유의 가압 특성 대문에 실험 결과의 재현성을 얻기가 힘들고, 특히 대용량 생산설비의 경우 더욱 그러하다. 이론적으로 초고압 처리는 매우 예측 가능한 공정이다. 즉 고압요기 내부에서는 어느 지점이던 간에 압력이 고르게 분포되고, 가열 확산에 근거한 처리공정과는 달리 압력이 모든 지점에 순간적으로 공정상 불균일이 야기될 소지가 있는 부분은 오직 압축에 다른 발열과 열 전달에 의한 온도 편차에 기인한다. 실제로 처리 대상 제품과 압력 전달매체의 압축시 발열정도 차이와 시료, 매체, 고압용기 간의 열 손실 또는 열 흡수 대문에 고압처리 공정에서 온도가 일정하지 않을 수 있다.

Food materials for kimchi manufacturing were treated by ozone or gamma irradiation to reduce the number of contaminated microorganisms before the manufacturing of kimchi. Counts of total aerobic bacteria, yeast and mold were $10^6{\sim}10^7 and 10^3{\sim}10^4\;CFU/g$ in the food materials, respectively. After treatment with ozone $(3{\sim}6\;ppm), the total aerobic bacteria were reduced to 1 log cycle or more, in a dose-dependent manner. In comparison, gamma irradiation was superior to ozone treatment. Especially, irradiation of 5 kGy fully eliminated the yeast and mold attached in the food materials for kimchi manufacturing. The contents of ascorbic acid and total and reducing sugar were not affected by ozone or gamma irradiation. Taken together, it is suggested that microorganisms contaminated on materials for kimchi manufacturing could be substantially reduced by ozone (up to 6 ppm) and gamma irradiation (up to 5 kGy), without effect on the nutritional contents.

This study performed to investigate the probiotic properties of lactic acid bacteria 200 strains isolated from traditional fermented foods. Based on being higher tolerance to bile salts and showing higher acid resistance, 4 LAB Strains were selected in the screening experiment; Lactobacillus plantarum SRCM 102224, Lb. plantarum SRCM102227, Lb. paracasei SRCM102329, Lb. paracasei SRCM102343. Antibacterial activity against various pathogens, acid and bile salt tolerance, hemolytic phenomenon, cell surface hydrophobicity, and antibiotic resistance were examined. Among the tested strains, SRCM 102343 (95.9%) was highly observed hydrophobicity compared to Lb. rhmanosus GG (13.4%) as control. In this study, the in vitro adhesion properties of 4 strains of LAB was investigated using human intestinal caco-2 cell cultures. SRCM102329 and SRCM102343showed higher adherence to caco-2 cells than Lb. rhamnosus GG. The antibacterial activities of 4 strains LAB were investigated. the 3 strains showing strongly antimicrobial activity against Escherichia coli ATCC10798, Staphylococcus aureus KCCM11593, Listeria invanovii KCTC3444, Bacillus cereus ATCC11778 and S. enterica serovar. Typhi KCTC1926. These results suggest that selected strains have good probiotic potential for application in functional foods.
