1-β-D-Arabinofuranosyl-cytosine Induces Chromosomal Breaks in vitro (In vitro에서 1-β-D-arabinofuranosyl-cytosine의 염색체 파열 유도)

  • Purpose : Fragile sites are points on chromosomes which tend to break non-randomly when exposed to specific chemical agents or conditions of tissue culture. The chromosomal break induced by the antineoplastic drug, 1-${\beta}$-D-arabinofuranosyl-cytosine(Ara-c), was investigated to study the laboratory conditions in which the incidence of chromosomal break could be enhanced. Besides, the fragile sites induced by Ara-C were investigated and compared to the already known locations of the specific chromosomal alterations observed in specific neoplasms. Methods : T-lymphocytes from theree normal males and three females were cultured for 48 hours. Cells from each individual were exposed to the Ara-C for an additional 24 hours. After the caffeine was added during the last six hours culture, the metaphase chromosomes were prepared following the conventional method. A site was considered fragile if it was found to break two or more per 100 chromosomal breaks in more than four of six individuals tested. Results : Ara-C induced 252.1 chromosomal breaks per 100 mitotic cells and this result was significantly higher than that of the control, which induced 25.2 breaks(P<0.05). The incidence of the chromosomal break by Ara-C was higher, if cultured in the MEM-FA, which has no folic acid, than in the RPMI 1640 which contains enough folic acid(P<0.05). The most common break site by Ara-C was 3p14.2(FRA3B). There were 20 fragile sites induced by Ara-C. Among these 20 fragile sites, seven coincided with the locations of the mapped oncogenes, JUN, SKI, REL, N-MYC, FHIT, MET, ETS-1, and FOS. Conclusion : S phase specific chemotherapeutic agent, Ara-C, induced the expression of the chromosomal fragile sites effectively using the T-lymphocyte in vitro. Some of the fragile sites by Ara-C highly coincided with the oncogenes and neoplasm specific chromosome breakpoints. In this regard, the fragile sites reported here could provide the unknown neoplasm related chromosomal alternation points.

  • 중수로용 핵연료봉의 봉단마개 부위는 열.기계적 작용에 의해 핵연료봉 파손이 쉽게 발생할 수 있는 취약 부위로 알려져 있다. 따라서, 핵연료봉 설계시 봉단마개 부위의 열.기계적 거동을 해석하고, 이 결과를 설계에 반영하여 파손 가능성이 없음을 확인하여야 한다. 여기에서는 중수로용 개량 핵연료인 CANFLEX-NU 핵연료봉 봉단마개에서의 열.기계적 거동을 해석하였는데, 이 결과 CANFLEX-NU 핵연료봉은 출력이 매우 낮아서 열중성자속 집중을 고려하더라도 봉단마개 부위 건전성을 충분히 유지하는 것으로 나타났다.

  • 본 연구에서는 다양한 작업역과 동작범위 설정에 관련이 많은 인체특정 부위와 손, 팔에 해당하 는 부위 49개 항목을 선정하여 18세-65세 사이의 한국인 성인 남자 179명, 여자 236명을 대상으로 측정하였다. 측정된 자료의 분석은 4개 연령그룹으로 구분하여 주요부위에 대해 연령그룹에 따른 차이를 보았으며, 이를 일본인의 측정 자료와 비교하여한국인과 일본인의 발육차이를 비교하였다.

Chemical Properties of Sea Tangle (Saccharina. japonica) Cultured in the Different Depths of Seawater (양식 다시마의 수심과 부위에 따른 화학적 성분 특성)

  • To develop the technology for cultivation of high-density seaweed, sea tangle was cultured from varying depths(0.5 m, 3 m) of seawater at Gijang and Wando area. Proximate composition, component sugar, total amino acid, fatty acid composition, and element composition of different parts of sea tangle (Saccharina japonica) have been examined. Significant differences were found in the amount of crude protein and ash content in lower, middle, and upper parts of algal blades. The upper parts of the sea tangle was rich in crude protein, while lower parts was rich in crude ash. Crude lipid content was higher in the middle parts than those of the other parts. The component sugars were not significantly different from all parts of algal blades. The highest content of most of the amino acids were found in the upper parts of the blades. The amount of saturated fatty acids concentrated mostly in lower parts of blades, while the content of polyunsaturated fatty acids concentrated in the upper parts. The highest N element contents were found in upper part of algal blades. However, the contents of those chemical component were not affected by the depths of seawater.

  • We proposed a robust body part tracking system for intelligent environment that will not limit freedom of users. Unlike any previous gesture recognizer, we upgraded the generality of the system by creating the ability the ability to recognize details, such as, the ability to detect the difference between long sleeves and short sleeves. For the precise each body part tracking, we obtained the image of hands, head, and feet separately from a single camera, and when detecting each body part, we separately chose the appropriate feature for certain parts. Using a calibrated camera, we transferred 2D detected body parts into the 3D posture. In the experimentation, this system showed advanced hand tracking performance in real time(50fps).

Accuracy Evaluation of Alternative Concept Joint Models (결합부위 단순모델의 정확성 평가 방법의 개발)

  • The concept models are used for the analysis of joints because they are simple to use and accurate. The modeling parameters of concept models are estimated using the results of experiments performed on the joints. The concept joint models accurately describe the behavior of joints under the loads which are used in the experiments for the estimation of parameters. However, they may not be accurate under the loads which are not used in the experiments. The accuracy can be dependent on the loads which are used in the evaluation of accuracy. In this study, antioptimization is presented to find the worst possible loads, under which the accuracy of concept joint models can be evaluated. The procedure was applied to the accuracy evaluation of concept joint models in an isolated 3-D joint and 2-D joints of a vehicle structure.

  • Activities of several hydrolytic enzymes in the rusted pear loaves were estimated. Tested enzymes were cellulase (Cx), invertase $\beta-amylase$, pectinase (pectinmethylesterase and polygalacturonase), and phosphatase. Enzyme activities represented by pH value-enzyme activitycurve. Activities of Cx. invertase. and $\beta-amylase$ were higher in the healthy part than those in the near-lesion and lesion parts. and showed the maximal activities around pH 7.0. On the other hand, activities of pectinase and phosphatase were higher in the lesion and near-lesion parts than in the healthy part. Pectinmethylesterase activities in the healthy and diseased parts were different with different curve-pattern as the pH of treating solution changed.

  • The purpose of this study is to estimate elemental costs of an apartment building. The adapted research method includes a case study from $^{\circ}{\AE}H^{\circ}{\phi}$ construction company. The results of this study are as follows: 1) An elemental cost format of an apartment building is proposed, 2) An elemental cost table based on a case study of $^{\circ}{\AE}H^{\circ}{\phi}$ construction company is suggested.

The Effect of Progesterone on Differentiation of Implantation Sites during Early embrynic development of the Rat (흰쥐 초기배아 발생과정 중 착상부위 분화에 미치는 Progesterone의 영향)

  • 포유류 배아의 착상기작을 규명하고자 본 연구에서는 착상시기에 수정된 배아가 착상부위를 어떻게 인식하는 지 그리고 착상부위의 분화유발요인이 무엇인지를 조사하였다. 착상시기(임신 제6일)와 착상후 초기배아 발생기간(임신 제9일)중 혈청내 estradiol (E2)과 Progesterone(P4)의 농도를 측정하였으며, 자궁내막 조직을 착상부위 (antimesome trium)와 비 착상부위(mesometrium)로 분리하여 P4의 수용체 농도를 측정하였다. 혈청내 E2의 농도는 임신 제6일군에서 E2처리군에서 가장 높았으며, 임신 제9일에서는 intact 대조군에서 가장 높고 제9일의 모든 실험군의 농도는 임신 제6일군보다 높았다. 혈청내 P4 농도는 임신 제6일과 9일에서 다같이 대조군에서 가장 높으며 처리군 중에서는 P4처리군에서 높아지고 있다. P4 농도 역시 임신 제9일의 모든 실험군에서 제6일의 실험군보다 높았다. P4수용체 농도는 착상부위가 비 착상부위보다 높으며, 대조군(P < 0.01)과 P처리 군(P < 0.05)에서는 유의한 차이로 비 착상부위보다 높았다. 자궁내막조직 착상을 위한 분화에는 P4의 영향이 크며 P4의 혈청내 농도와 핵의 수용체 농도는 모든 실험군에서 상응하는 관계를 나타내었다. 이 결과는 앞서 일차적으로 발표한 알카리성 phosphatase (ALPase) 활성과도 상응하는 것이다.

A Study on the Differentiation of the Implantation Sites in the Pseudopregnant Rat Uterus (가임신 흰쥐 자궁조직의 착상부위분화에 관한 연구)

  • 포유류 배아의 착상기전을 규명할 목적의 일환으로 자궁내막조직의 탈락막 형성과 분화에 미치는 난소 스테로이드 호르몬의 영향과 인위적 자극등의 영향을 조사하였다. 가임신을 유도시킨 흰쥐와 정상임신군의 자궁내막조직을 착상부위와 비 착상부위로 분리하여 자궁내막조직의 분화에 지표가 되는 Alkaline phosphatase(ALPase)의 활성을 측정하였다. 가임신군에서 인위적 자극(trauma)을 가한 자궁이 자극을 받지않은(control) 자궁에 비해 Progesterone(P)만을, 또는 progesterone과 estradiol(E + P)을 동시 처리받은 실험군에서 ALPase 활성이 유의한 차이(p<0.01)로 높게 나타났다. 정상임신군에서는 착상시기인 임신 제6일군의 착상부위에서 I처리군과 P처리군의 ALPase활성이 비착상 부위에 비해 유의한 차이(P < 0.05)로 크게 나타나며, 특히 임신 제6일군에서는 착상, 비착상 부위의 P처리군의 활성이 Intact군보다 유의하게(P<0.05) 높아졌다. 정상임신 제 9일 Intact군의 ALPase활성은 임신 제3일, 6일의 Intact 군의 활성에 비하여 착상(3, 6일 :P<0.01), 비 착상부위 (3일 :P<0.05, 6일 :P<0.02)에서 다같이 유의한 차이로 높아졌다. 본 연구결과에서 배아가 분화, 발생해 감에따라 자궁내막조직 분화도 활발해지고, 착상부위 분화는 착상시기부터 현저해지며, 특히 P호르몬의 영향이 크다는 것과 배아 아닌 인위적 자극으로도 탈락막 형성 유도가 가능하다는 것을 확인할 수 있었다.
