$\text\tiny{D}$-Psicose 3-Epimerase (DPE)는 $\text\tiny{D}$-Fructose의 C3 Epimerase로써 $\text\tiny{D}$-Fructose를 $\text\tiny{D}$-Psicose로 전환해 주는 효소이다. $\text\tiny{D}$-Psicose는 설탕 대신 사용하는 감미료로 몸에 흡수되지 않아 칼로리가 없다고 알려져 있고 자연에서는 오로지 DPE에 의해서만 생산되는 희귀당이다. 이에 따라 DPE를 통한 $\text\tiny{D}$-Psicose 대량생산의 필요성이 대두되고 있는 등 이 분야에 대한 관심이 뜨거운 실정이다. 본 연구팀은 이 당과 관련된 작용기작 연구를 수행하기 위하여 아직 단백질 3차구조가 알려지지 않은 Clostridium hylemonae DPE (chDPE) 단백질의 3차 구조예측 연구를 수행 하였다. 우리는 HHsearch를 이용하여 agrobacterium tumefaciens의 DPE 외 2개의 구조를 호몰로지 모델링 연구를 위한 주형으로 선정하였다. 다음으로 PROMALS3D를 이용하여 주형들과 chDPE의 multiple sequence alignment를 수행하였고 이를 바탕으로 3차구조 예측 연구를 수행 하였다. 예측된 구조를 검증하기 위하여 ProSA와 Ramachandran plot분석을 이용하였고 Ramachandran plot에서 단백질의 94.8%에 해당하는 잔기들이 favoured regions에 위치하였다. ProSA에서는 Z-score값이 -9.3으로 X-선 결정학이나 핵자기 공명법으로 밝혀진 구조들에서 관측되는 범위 내에 위치하였다. 나아가 예측된 구조에 $\text\tiny{D}$-Psicose와 $\text\tiny{D}$-Fructose의 결합모드를 규명하기 위하여 도킹을 시도하였다. 이번 연구를 통하여 chDPE의 구조를 예측 할 수 있었고 이를 바탕으로 이 단백질의 기능을 이해하는데 도움을 줄 것으로 기대된다.

응력파의 기반한 비파괴 검사법은 비교적 실험 절차 및 실험에 필요한 장비가 단순하고, 인체에 해가 없으며, 비용이 저렴한 특성을 갖고있다. 따라서 건축/토목 구조물의 비파괴 검사에 매우 효과적인 방법으로 알려져 있다. 하지만 기존의 가속도계, 변위계, 지오폰과 같은 부착 센서를 사용할 경우 표면 처리, 센서의 부착 및 이동에 따른 추가적인 시간이 소요되고, 센서와 구조물의 불완전한 커플링으로 인한 측정 결과의 신뢰성 및 일관성을 유지하지 어렵다는 문제를 발생시킨다. 최근 이러한 문제의 해결책으로 Air-coupled sensor (ACS)의 사용이 각광받고 있으며, 여러 연구자들에 의해 ACS의 가능성 및 실용성이 증명되고 있다. 기존의 접촉센서와 비교하여 ACS를 사용했을 때 얻을 수 있는 가장 큰 장점은 센서의 커플링 문제를 근본적으로 해결하여 신뢰도가 높고 일관적인 측정이 가능하고, 대형 건축/토목 구조물의 표면을 음향스캔하여 결과를 신속하게 처리하여 실시간으로 시각화 할 수 있다는 점이다. 이론적으로 ACS를 이용하여 측정하는 물리값은 콘크리트 내부에서 발생된 응력파의 일부가 공기중으로 전파된 누설파 (Leaky wave)이다. 콘크리트 비파괴 검사에 주로 사용하는 100 kHz이하의 저주파를 측정할 경우 일반적으로 콘텐서마이크가 ACS로 사용될 수 있다. 기존 연구자들은 실험 및 이론적 연구를 통하여 응력파에 기반한 비파괴 방법에서 ACS가 기존의 접촉 센서를 대체할 수 있다는 점을 보여주고 있다. 현재 미국에서는 연방 도로청 (FHWA) 및 국가표준기술연구소 (NIST)의 연구비 지원으로 ACS의 실용성을 높이기 위하여 최적화된 음향반사판의 설계를 통한 ACS 의 민감도를 높이기 위한 연구, 다채널 센서 배열 및 데이터 통합을 위한 새로운 알로리즘 개발, 자동화 및 로봇 기술과 융합과 같은 연구가 활발히 진행되고 있다. 멀지 않은 미래에는 ACS를 장착한 무인 로봇이 다양한 종류의 건축/토목 구조물의 건전도를 평가하기 위하여 종횡무진 활약하는 모습을 현실 속에서 볼 수 있을 것이라 기대한다.

Experimental and analytical studies on the behavioral characteristics of a pre-stressed (PS) steel girder are conducted to investigate the effects of deviators on the non-linear inelastic properties of the PS system. In this regard, 4 test specimens consisting of a steel H-beam, a straight cable with eccentricity, anchorages, and deviators are built and failure tests are performed under two-point loading. In addition, in-plane elastic deformation theories for the PS system without a deviator, and with three deviators at regular intervals are analytically formulated and solved using a symbolic calculation technique. To verify the validity of the experimental and the proposed analytical theories, the results obtained using FEM models composed of beam elements, rigid beam elements, and truss cable elements, are compared to the experimental results and the analytical solutions. As a result, it is determined that externally installed un-bonded deviators inhibit flexural deformation of the deformed beam to such an extent that their elastic stiffness, and failure strength are significantly improved compared to those of the PS system without deviators.

This study introduces a technique for improving the elastomeric-isolator performance using modular devices. The modular devices are shear resistance block, polymer spring, displacement acceptance guide, and anti-falling block. They are installed on the elastomeric isolator as a supplementary device. Each modularized device improves the isolator performance by performing step-by-step actions according to the seismic intensity and displacement. The PRB isolation device works in four stages, depending on the seismic magnitude, to satisfy the target performance. It is designed to accommodate design displacement in the first stage and large magnitude of earthquakes in the second and third stages. This design prevents superstructures from falling in the fourth stage due to large-magnitude earthquakes by increasing the capacity limit of the elastomeric isolator. In this study, the PRB isolation device is analyzed using finite element analysis to verify that the PRB isolation device works as intended and it can withstand loads corresponding to large-magnitude earthquakes. The performance of the PRB isolation device is validated by the analysis, which is further corroborated by actual experiments.
Anisotropic characteristics of deformation are important to understand the particular behavior in the pre-failure state of soils. Recent experiments show that cross-anisotropic moduli of granular soils can be expressed by functions of normal stresses in the corresponding directions, which is closely linked to micromechanical characteristics of particles. Granular soils are composed of a number of particles so that the force-displacement relationship at each contact point governs the macroscopic stress-strain relationship. Therefore, the micromechanical approach in which the deformation of granular soils is regarded as a mutual interaction between particle contacts is one of the best ways to investigate the anisotropic elastic deformation of soils. In this study, a numerical program based on the theory of micromechanics is developed. Generalized contact model for the irregular contact surface of soil particles is adopted to represent the force-displacement relationship in each contact point far the realistic prediction of anisotropic moduli. To evaluate the model parameters, a set of analytical solutions of anisotropic elastic moduli is derived in the isotropic stress condition. A detailed procedure to determine the model parameters is proposed with emphasis on the practical applicability of micromechanical program to analyze the elastic behavior of the granular soils.

A three dimensional (3D) method of analysis is presented for determining the free vibration frequencies and mode shapes of thick, circular and annular plates with nonlinear thickness variation along the radial direction. Unlike conventional plate theories, which are mathematically two dimensional (2D), the present method is based upon the 3D dynamic equations of elasticity. Displacement components u/sub s/, u/sub z/, and u/sub θ/ in the radial, thickness, and circumferential directions, respectively, are taken to be sinusoidal in time, periodic in θ, and algebraic polynomials in the s and z directions. Potential (strain) and kinetic energies of the plates are formulated, and the Ritz method is used to solve the eigenvalue problem thus yielding upper bound values of the frequencies by minimizing the frequencies. As the degree of the polynomials is increased, frequencies converge to the exact values. Convergence to four digit exactitude is demonstrated for the first five frequencies of the plates. Numerical results we presented for completely free, annular and circular plates with uniform linear, and quadratic variations in thickness. Comparisons are also made between results obtained from the present 3D and previously published thin plate (2D) data.

Unlike conventional shell theories, which are mathematically two-dimensional (2-D), the present method is based upon the 3-D dynamic equations of elasticity. Displacement components μ/sub Φ/, μ/sub z/, and μ/sub θ/ in the meridional, normal, and circumferential directions, respectively, are taken to be sinusoidal in time, periodic in θ, and algebraic polynomials in the Φ and z directions. Potential (strain) and kinetic energies of the hemispherical shells are formulated, and the Ritz method is used to solve the eigenvalue problem, thus yielding upper bound values of the frequencies obtained by minimizing the frequencies. As the degree of the polynomials is increased, frequencies converge to the exact values. Novel numerical results are presented for solid and hollow hemispheres with linear thickness variation. The effect on frequencies of a small axial conical hole is also discussed. Comparisons are made for the frequencies of completely free, thick hemispherical shells with uniform thickness from the present 3-D Ritz solutions and other 3-D finite element ones.
An analytical solution of dynamic responses for seabed in finite and infinite thicknesses including shallow has been developed under flow and partial standing wave with arbitrary reflection ration coexisting field at a constant water depth condition. In the analytical solution, a field was simply transited to a coexisting field of progressive wave and flow when reflection ratio was 0 and to a coexisting field of fully standing wave and flow when reflection ratio was 1. Based on the Biot's consolidation theory, the seabed was assumed as a porous elastic media with the assumptions that pore fluid is compressible and Darcy law governs the flow. The developed analytical solution was compared with the existing results and was verified. Using the analytical solution the deformation, pore pressure, effective and shear stresses were examined under various given values of reflection ratio, flow velocity, incident wave's period and seabed thickness. From this study, it was confirmed that the dynamic response of seabed was quite different depending on consideration of flow, which causes changing period and length of incident and reflection waves. It was also confirmed that dynamic response significantly depends on the magnitude of reflection ratio.
In this paper, an improved four-node Reissner-Mindlin plate-bending element with enhanced assumed strain field is presented for the analysis of isotropic and laminated composite plates. To avoid the shear locking and spurious zero energy modes, the transverse shear behavior is improved by the addition of a new enhanced shear strain based on the incompatible displacement mode approach and bubble function. The "standard" enhanced strain fields (Andelfinger and Ramm, 1993) are also employed to improve the in-plane behaviors of the plate elements. The four-node quadrilateral element derived using the first-order shear deformation theory is designated as "14EASP". Several applications are investigated to assess the features and the performances of the proposed element. The results are compared with other finite element solutions and analytical solutions. Numerical examples show that the element is stable, invariant, passes the patch test, and yields good results especially in highly distorted regimes.

A method analizing contact pressure between plate and elastic half space is presented by using F.E.M. With the method, the pressure intensities at surface nodes of half space cae be directly calculated by using flexibility matrix of half space. The method is originally presented by Y.K. Cheung et al.(3) Insted of Y.K. Cheung's method, which use a conception of equi-contact pressure area around each surface nodes of half space in the noded rectanqular element area. We use the equi-contact pressure area around the Gaussian integration points of half space surface in the noded isoparametric element area. Numarical examples are presented and compared with other's studies.
