Optimal condition for mycelial growth of Sparassis crispa (꽃송이버섯(Sparassis crispa)의 균사생장 최적화)

  • This study was carried out to obtain the basic data on artificial culture of Sparassis crispa. The mycelial growth and density were good in the PBA, PDA and PDM media, and especially prominent in the PBA medium. The optimum condition for the mycelial growth was $25^{\circ}C$ and pH 5.0, respectively. The carbon sources such as glucose, fructose, mannose, xylose(monosaccharide) and maltose(disaccharide) were favorable to mycelial growth of S crispa. The optimal concentration of glucose is 1.0~1.5%. As nitrogen sources, threonine, peptone, glycine, glutamine and valine appeared to be favorable to the mycelial growth and density. The optimal concentration of peptone is 0.3%.

Optimization of Culture Conditions for the Production of Antibacterial Activities by Paenibacillus polymyxa DY1 Isolated from Soil (토양에서 분리한 Paenibacillus polymyxa DY1의 항균활성물질 생산을 위한 배양조건 최적화)

  • In a previous study, a new antibacterial activity was found from Paenibacillus polymyxa DY1 isolated from Korean soil, which showed a good growth inhibition against various multidrug resistant enteric bacteria. Thus the effect of nutritional factors and physicochemical conditions on the growth of P. polymyxa DY1 cells and the production of antibacterial activity were evaluated in this study. For the growth of P. polymyxa DY1 cells the optimum condition reaches by incubating at $35^{\circ}C$ for 48 hours by shaking at 180 rpm in the basal medium containing 0.6% dextrose, 1.4% ammonium phosphate dibasic, 0.9% peptone, 2.4% glutamine, and 2 mM sodium molybdate with initial pH 8.0. A maximum level of antibacterial activity was obtained when P. polymyxa DY1 was incubated at $35^{\circ}C$ for 48 hours by shaking at 180 rpm in the basal medium with initial pH 8.0 containing 1% dextrose, 1.5% ammonium sulfate, 1.1% casamino acid, 1.9% aspartic acid, and 2 mM ferrous sulfate.

Cell Growth and Antioxidant Activity on Onion Juice Fermentation by Using Lactobacillus plantarum as Animal Probiotics (가축용 생균제 Lactobacillus plantarum을 이용한 양파즙 발효의 균체성장과 항산화 활성)

  • This study was conducted to establish the optimal medium condition for the animal probiotic Lactobacillus plantarum by using onion juice. Cell yield and antioxidant activity increased in proportion to high additive levels of onion juice in medium. Onion juice, sucrose and yeast extract were selected as media ingredient factors and the effects of their mixed ratio in medium were evaluated. The full factorial design consisted of 24 experimental runs and was employed to estimate the main effects of the factors and their interactions. Significant positive effects on cell yield and antioxidant activity was shown with yeast extract and onion juice, respectively. Significant interaction was found only between sucrose and yeast extract in antioxidant activity. Finally, we selected an optimal medium that was composed of (g/l) onion juice, 600; sucrose, 15; yeast extract, 5. The efficiency of this optimum medium was estimated by using a 5 l jar fermenter. As a result, the maximum cell yield was $9.7\;{\log}_{10}$ (CFU/ml) at 12 hr. Cell yield at the end of incubation (20 hr) was $8.9\;{\log}_{10}$ (CFU/ml) and it was very similar with the predicted value, $9.0\;{\log}_{10}$ (CFU/ml). Antioxidant activity of culture was maintained at about 60~65% during all incubation time, resulting in a higher-than-predicted activity of 47.1%.

The Medium Optimization through Continuous Culture of an Methanol Utilizing Bacterium for SCP Production (SCP 생산용 메탄올 자화균주의 연속배양에 의한 배지 최적화)

  • Methanol-utilizing bacterium isolated from sewage samples in Seoul showed optimal temperature and pH of $33^{\circ}C$ and 7.1 for growth, respectively. The maximum specific growth rate was $0.42hr^{-1}$. The minimum medium composition was reconstituted depending on the surplus and the deficit of each component in the basal medium at steady state. The optimal composition was given as(g/l); Methanol 40, $(NH_4)_2\;SO_42, \;KH_2PO_4\;1.5, \;K_2HPO_4\;0.2, \;H_3PO_4\;0.79, \;Na_2HPO_4{\cdot}12H_2O\;0.15, \;MgSO_4{\cdot}7H_2O\;1.5, \;FeSO_4{\cdot}7H_2O\;0.034, \;MnSO_4{\cdot}4H_2O\;0.005, \;CuSO_4{\cdot}5H_2O\;0.0027, \;CaCl_2{\cdot}2H_2O\;0.25, \;ZnSO_4{\cdot}7H_2O\;0.007, \;(NH_4)_6\;Mo_7O_{24}{\cdot}4H_2O\;0.00048, \;H_3BO_3\;0.00068, \;CoCl_2\; 0.00024$ Under the continuous culture with optimum medium the maximum cell productivity was 3.8g/1/hr at dilution rate $0.23hr^{-1}$. Maximum cell concentration and its protein content were 19.5g/l and 70% at dilution rate of $0.1hr^{-1}$, respectively.

Medium optimization for growth of Bacillus amyloliquefaciens ISP-5 strain and evaluation of plant growth promotion using lettuce (Bacillus amyloliquefaciens ISP-5 균주의 배지 최적화 및 상추를 이용한 식물 생장 촉진 평가)

  • Bacillus sp. is a useful strain for agriculture because it promotes plant growth and controls plant pathogens through a variety of mechanisms. In this study, we obtained a microbial preparation with a high number of viable cells by culturing newly isolated soil bacteria on an optimized medium. Subsequently, we applied this preparation to lettuce to enhance its growth and yield. First, B. amyloliquefaciens ISP-5 was isolated from soil. Next, optimization of culture medium was carried out using 5 L scale fermenters. When culturing B. amyloliquefaciens ISP-5 on this optimized medium, the number of viable cells was approximately 1000 times higher than that obtained from culturing on the commercial medium. Afterwards, the plant growth promotion properties of the ISP-5 strain were evaluated using lettuce as a test plant. Foliar spray treatment of lettuce was carried out by inoculating half the standard concentration suspension (0.5 × 107 cfu/ml). As a result, leaf width increased by 8.6% and leaf length increased by 12.9% compared to the control group. Live weight also increased by 24.2% and dry weight by 23.9%. Considering the results from field test, B. amyloliquefaciens ISP-5 showed potential as a plant growth-promoting bacteria.

  • Medium components for lactic acid production were optimized with a strain of Lactobacillus sp., isolated by our Lab. Nitrogen source was the key component and manganese ion was also important for lactic acid production in this strain. Optimal concentration of manganese ion was 0.03 g/l as MnSO$_{4}$ 4 - 5 H$_{2}$O base. Other mineral elements, however, had little effect on it. Among the nitrogen sources we examined, yeast extract showed the highest productivity. Yeast extract, the exellent but very expensive medium component, could be partially replaced by soytone until 60% dry base with higher productivity, or by tryptone enforced with vitamines and nucleic acids. In order to replace yeast extract completely, we examined several inexpensive nitrogen sources and their enzymatic hydrolyzates. The hydrolyzate of vital wheat gluten was the best of them.

  • Entomogenous fungi which attack living insects are powerful means for microbiological insecticide. The purpose of this study is to establish the culture conditions and media for mass production of Beauveria sp. C208 which has a broad host range as a potential microbiological pesticide. The temperature and pH range for optimal cultivation of this strain were 28$circ$C and pH 5.0-7.0. For Beauveria sp. C 208, 2% rice straw and 0.6% tryptone were found as the proper carbon and nitrogen sources, considering cell mass, enzyme activities such as chitinase, protease and lipase, and spore concentration.

  • In order to maximize the riboflavin production by a mutant strain Ashbya gosspyii, the optimization of medium and fed-batch fermentation were performed. As carbon sources, glucose and soybean oil were necessary for the riboflavin overproduction. Optimal concentrations of glucose and soybean oil in the flask cultures were found to be 3.0% and 0.5%, respectively, in a complex medium containing corn steep liquor(CLS) 1%. Among the various organic nitrogen sources tested, CSL was the most effective one both for the cell growth and riboflavin overproduction.

Characteristics of a Mutant of Trehalose-producing Micrococcus luteus and Optimization of Production Conditions (Trehalose를 생산하는 Micrococcus luteus 변이주의 특성 및 생산배지의 최적화)

  • For the production of trehalose, microorganisms capable of producing trehalose extracellularly were screened from the stock cultures in our laboratory. among them, Micrococcus luteus IFO 12708 showed the highest productivity of trehalose. For the increase of productivity, the mutant strai Hs-208 having higher trehalose production was selected with NTG(N-methyl-N'-nitrosoguanidine) mutagenesis, which led to the decrease of the specific activity of trehalose phosphorylase(3.2-fold) as compared to the wild strain. The optimum condition for the trehalose production was established as follows: 20g/l of glucose and 6g/l of tryptone were used as a sole carbon source and nitrogen source, respectively, and cultivations were carried out at 3$0^{\circ}C$ and pH 6.0. After 20hrs cultivation, addition of 20unit/ml penicillin G led to the higher conversion yield of trehalose. Under the optimum condition, 6.547g/l trehalose was produced with conversion yield of 32.7%.
