Radiation Protection Effect of Mixed Extracts of Hottuynia, Perilla Frutescens, Camellia Sinnensis in the SD Rat (SD Rat에서 어성초, 자소엽, 녹차 혼합 추출물의 방사선 방호 효과 연구)

  • This study confirmed the radioprotective effects of a mixed extracts of Hottuynia, Perilla Frutescens, and Camellia Sinnensis as natural radioprotectors on the prostate, small intestine, and liver of SD rats. It is known that Hottuynia, Perilla Frutescens, and Camellia Sinnensis have antioxidant effects on the prostate, small intestine, and liver, respectively.In this study, SD rats were irradiated with 8 Gy of gamma rays to confirm the radioprotective effects of a mixed extracts of Hottuynia, Perilla Frutescens, and Camellia Sinnensis. After radiation irradiation, histological analysis of the prostate, small intestine, and liver was performed. After radiation irradiation, histological analysis of the prostate, small intestine, and liver was performed. In the case of the prostate, the HPC+IR Group had less prostate damage and better recovery due to radiation compared to the IR Group. It was confirmed that the prostate size of the HPC+IR Group was 11.48%p and 24.54%p higher than the IR Group on 1st and 7th days. In the case of the small intestine, the HPC+IR Group had less radiation-induced small intestinal damage and recovery was better than the IR Group. The length of small intestine villus in the HPC+IR Group was confirmed to be 23.73%p and 24.27%p higher than the IR Group on the 1st and 7th days. In the case of the liver, the HPC+IR Group had less liver damage due to radiation and had better recovery than the IR Group. This was confirmed through the hepatic portal vein and surrounding cells. The results of this study are considered to be used as basic data for research on natural radiation protection using mixed extracts.

  • 다량의 방사선에 피폭되는 경우 인체에 해로운 영향을 미치는 것은 틀림이 없으나 미량의 방사선인 경우에는 다량의 방사선인 경우와 다른 효과를 나타내며 심지어 정반대의 영향을 나타내기도 한다고 볼 수 있으며 여러 역학조사에서 이러한 사실이 증명되고 있다. 1991년의 국제방사선방어기구(ICRP) 보고서 및 1994년의 UNSCEAR 보고서는 미량의 방사선에 대한 적응반응을 인정하고 있다. 일부 학자들은 소량의 방사선에 지속적으로 피폭(연간 1 rem)됨으로써 암사망률을 감소시킬 수 있다고 주장하면서 현재의 개념을 수정하여 피폭한계 및 법적 제한 등을 변경하여 환경방사능을 증가시킴으로써 향후 인류의 생활의 질을 향상시켜야 한다고 주장하고 있다.

Effect of Tumor Hypoxia on Efficacy of Tirapazamine Combined with Fractionated Irradiation in Mouse Tumor (마우스종양에서 분할방사선조사와 병용된 Tirapazamine의 효과에 미치는 종양 저산소상태의 영향)

  • Purpose : Tumor hypoxia can be overcome with hypoxic cytotoxin. In mouse tumor, tirapazamine's efficacy of the potentiating radiation effect was tested by the tumor oxygenation status combined with hype facti on ated rad iotherapy .:The control and hypoxic mouse tumors we established by inoculation of RIF-1 tumor cells into the normal or previously irradiated back and thigh of C3H mice. When the tumors reached a proper size, both the control and hypoxic tumors were given hypefractionated treatments (8fractions/4 days) with saline (0.02 ml/g), tirapazamin (0.08 mM/0.02 ml/kg), irradiation (2.5 Gy), irradiation combined with tirapazamine given 30 minutes prior to each irradiation. The response was evaluated by the growth delay assay by measuring tumor size from day 0 (12 hrs prior to the first fractionation) to the day when the volume had 4-fold increase or cross sectional area had 2-fold increase. Results : Overall growth pattern showed that tirapazamine Potentiated radiation effect in back and thigh tumors grew in the normal and preirradiated tumor bed. With growth delay assay using reference point of initial tumor volume or cross sectional area, tirapazamine potentiated radiation effect 1.9 times for the control and 2.4 times for the hypoxic tumors in back, and 1.85 times for the control and 1.6 times for the hypoxic tumors. With reference of 4-fold increase of the initial volume or 2-fold increase of the cross sectional area, tirapazamine potentiated radiation effect 1.48 times for the control and 2.02 times for the hypxic tumors in back, and 1.85 times for the control and 1.6 times for the hypoxic tumors. Conclusions : Present result indicated that radiation response of hypoxic tumors was potentiated by tirapazamine in the back or thigh tumors grew in the control or preirradiated tumor bed, and potentiation of the hypoxic tumors was eDual to or greater than that of the control tumors in the back or thigh.

  • Authors started IORT for stomach cancer patient on 1988 and rectal cancer on 1991. We devloped various sized. shaped IORT cones for better clinical application and homogeneous surface and depth dose distribution. Authors obtained results as following. 1. The acryl cover fixed on the end for rectal IORT cone not only improvement of surface dose but also flattness of dose distribution. 2. Dose distribution of elliptical cone were shown almost 100% at inner field. 3. The output with acryl cone size were similar output of made electron cone.

  • 무독성 혼합안정제 Zn/Ca-stearate의 방사선 안정화 효과를 증진시키기 위해서 oxy-methylene, oxy-ethylene 및 oxy-porpylene 구조의 직쇄형 및 고리형 에테르 화합물을 PVC에 배합하여 방사선조사 하였을 때 PVC의 색차변화를 측정하여 이들 에테르 화합물의 방사선 안정화 효과를 비교 검토하였다. 에테르 화합물에서 oxy-ethylene 구조단위수 4이상인 직쇄형 에테르 또는 고리형 에테르와 oxy-propylene 구조단위수 7인 직쇄형 에테르는 매우 우수한 방사선 안정화 효과를 나타내고 있다. oxy-ethylene 또는 oxy-propylene 구조의 에테르의 방사선 안정화는 Zn/Ca-stearate에서 생성되는 ZnCl$_{2}$와 에테르의 주쇄구조 oxy-ethylene 또는 oxy-propylene의 착체반응에 의한 것으로 추정된다.

Synergistic Effects of Low Dose Gamma Irradiation and Growth Regulators on Seed Germination, Growth and Photosynthesis in Rice (Oryza sativa L.) (벼의 종자 발아와 생육 및 광합성에 대한 저선량 감마선과 생장조절물질의 상승작용 효과)

  • To investigate the synergistic effects of low dose gamma irradiation and growth regulators on the growth and photosynthesis in rice (Oryza sativa L.), laboratory and greenhouse experiments were conducted using 4-year-old rice seeds. In the laboratory experiment, the germination rate was increased in 0.001 ppm IBA treatment, showing the synergistic effect of gamma irradiation and growth regulators. The seedling growth was increased by treatment of GA₃ and IBA, the irradiated groups having higher than the non-irradiated ones. Particularly, it was remarkable in 0.001 ppm IBA. In greenhouse experiment, seedling growth was increased in response to a combination of gamma irradiation and 0.001 ppm IBA. Effective quantum yield of PSⅡ(Ф/sub PSⅡ/) and photochemical quenching (qP) were increased, while non-photochemical quenching (qN) was decreased by 0.001 ppm IBA. A synergistic effect of gamma irradiation and IBA was only found in seedling growth. The present results suggest that low dose gamma irradiation and growth regulator could synergistically stimulate seedling growth.

The Effect of Low Dose Radiation on the Neuronal Cell Proliferation in Diabetic Rats (당뇨병 유발 쥐에서 신경 세포 증식에 대한 저선량 방사선의 효과)

  • Purpose: To investigate the effect of low dose radiation on neuronal cell proliferation In diabetic rats. Materials and Methods: A group of rats (first group) were divided into three subgroups (nondiabetic control, nondiabetic 0.1 Gy and nondiabetic 10 Gy groups) to determine the effect of radiation on normal hippocampal neuronal ceil proliferation. A further group of rats (second group) were divided into six subgroups (nondiabetic control, diabetic control, diabetic 0.01 Gy, diabetic 0.1 Gy, diabetic 1 Gy and diabetic 10 Gy groups) to determine the effect of radiation on hippocampal neuronal cell proliferation under diabetic conditions. Using immuno-histochemistry for 5-bromo-2'-deoxyuridine (BrdU), the number of neuronal cells in the dentate gyrus of all the groups was counted. Results: The number of BrdU-positive cells in the dentate Gyrus of the nondiabetic control, nondiabetic 0.1 Gy and nondiabetic 10 Gy subgroups of the first group were $45.95{\pm}3.42,\;59.34{\pm}5.20\;and\;19.26{\pm}2.98/mm^2$, respectively. The number of BrdU-positive cells in the dentate gyrus of the diabetic control, diabetic 0.01 Gy, diabetic 0,1 Gy, diabetic 1 Gy and diabetic 10 Gy subgroups of the second group were $55.44{\pm}8.57,\;33.33{\pm}6.46,\;67.75{\pm}10.54,\;66.63{\pm}10.05,\;23.59{\pm}6.37\;and\;14.34{\pm}7.22/mm^2$, respectively. Conclusion: Low dose radiation enhances cell proliferation in the dentate gyrus of STZ-induced diabetic rats.

Protective Effects of Chitosanoligosaccharide on Gamma Ray-Induced Hepatic Damage in Mice (생쥐에서 키토산올리고당의 감마선 저항 효과)

  • The purpose of this study was to evaluate the effect the radiation-resistance of chitosan on the mice. A healthy male ICR mice were used for experiment. The SOD and MDA activity was measured from the liver of mice at 48 and 72 hours after the irradiation. The ultrastructural changes of the liver by irradiation was observed at 24, 48 and 72 hours after irradiation. The experimental groups were divided into three groups. Group 1 was the control group which was not treated with chitosanoligosaccharide before or after iradiation. Group 2 was the prefeeding group which chitosanoligosaccharide solution was supplied by feeding ad libitum for 30 days before irradiation. Group 3 was the postfeeding group which chitosanoligosaccharide solution was supplied by feeding after irradiation. In all groups 10 mice were used. The results were as follow: The SOD and MDA activity of the prefeeding group was decreased significantly (P<0.01). Control group - The nuclei were condensed. The mitochondria and rough endoplasmic reticulum (rER) were extended and the ribosome was dropped from the rER. Prefeeding group - The nuclei were rounded. The mitochondria was elongated. And the rER attached ribosomes. Postfeeding group - The nuclei were slightly condensed. The mitochondria and the rER were extended and the ribosome was dropped from the rER. It was concluded that the chitosanoligosaccharide was effective in the radiation-protection. So, chitosan would have the potential as the radiation-protection materials.

  • When the food poisoning bacteria Salmonella enteritidis and S. typhimurium were treated with radiation (cobalt-60 r-rays) and heat (10 minutes at $45^{\circ}C\;or\;50^{\circ}C$), their sterilizing effect was revealed differently depending on the order of treatments. Post-irradiation heating showed a synergistic effect whereas pre-irradiation heating revealed the opposite effect and the effects differed slightly with heating temperature.

Stability of Functional Properties and Chemical Components of Gamma-Irradiated Astragali Radix (감마선 조사 황기의 기능성 및 화학적 성분에 대한 안정성 평가)

  • Stability of functional properties and chemical components of gamma-irradiated Astragali Radix (AR) were investigated to study the application of radiation technology for preservation and hygiene of medicinal herbs. Thus, ethanolic extracts and ethyl acetate fractions were prepared from gamma-irradiated and gamma-nonirradiated AR, respectively. Functional properties were estimated by comparing antioxidant activity (DPPH radical scavenging activity, lipid peroxidation inhibitory activity) and protective effect against oxidative DNA damage in human lymphocytes. Chemical components were compared by HPLC analysis, comparing with calycosin ($_{t}$ R= 15.111 min), which was isolated from ethyl acetate fraction as a standard material. Their antioxidant activities and protective effects against oxidative DNA damage showed no significant difference at the concentrations tested. HPLC analysis also showed almost same pattem. These results suggest that the functional properties and chemical components of AR are not affected largely by gamma irradiation, and radiation technology can be applied to preservation and hygiene of medicinal herbs.s.
