Anaerobic Acid and Methane Fermentation using Paper Wastes (폐지를 이용한 혐기성 산 발효 및 메탄발효 특성)

  • This study was carried out to investigate the possibility of reusing newspaper or paperbox waste by methane fermentation. When 15 g of newspaper and box wastes were digested separately for 24 days by batch fermentation, the amount of total organic acids produced were 2461 and 4978 mg/L, respectively. The tCOD removal rates were found to be 60.9 and 62.4%, respectively. In addition, the removal efficiencies of total solid were 34.8 and 33.4%, and those of volatile solid were 40.0 and 39.2%, respectively. During this period, the amounts of biogas produced were 6.95 and 6.43 L. In a semicontinuous reaction, tCOD removal efficiencies for newspaper and box wastes were 64.7 and 65.0%, respectively, after 14 days of digestion. After 25 days, which were needed to stabilize the methane fermentation, the amounts of biogas produced daily were 0.31 and 0.30 L/g.dry wt, respectively. Methane contents were 57.3 and 56.2%, respectively, and the pHs in the anaerobic acidogenic and methanogenic fermenters were 5.0 and 7.5, respectively.

Establishment of a quality index of surfactin production during cheonggukjang fermentation (청국장 발효 중 surfactin 생산량 품질지표 설정)

  • Surfactin, one of the most powerful biosurfactants, can be widely applied in agriculture, food, and pharmaceutics. The purpose of the present study was to establish suitable indicators for a rapid detection method that can confirm the surfactin productivity of cheonggukjang. In the present study, changes in the total number of bacteria were quantified according to the fermentation time of chenggukjang. Furthermore, physicochemical factors, such as pH, color value, surface tension, refractive index, absorbance, and protein contents, were assessed in chenggukjang viscous substances, and a correlation analysis between the physicochemical factors and surfactin contents was performed. We found that the protein contents gradually increased up to 48 h (4.21±0.11 mg/mL) without a change in the surface tension. Furthermore, the refractive index, absorbance at 280 nm, and color value were significantly increased as the fermentation time increased; however, there were no statistically significant differences after 36 h of fermentation. Interestingly, this result exhibited a tendency similar to the surfactin content according to fermentation time. These findings suggest that surfactin can be used as a suitable quality indicator and may provide an experimental basis for the fermented food industry.

  • The feasibility of separating Pichia stipitis from a fermentation broth using a hollow fiber membrane was evaluated. The permeate flux was affected by such parameters as cell concentration, pH, content of antifoam agents, suction pressure, and recirculation rate. A minor effect of temperature on the flux loss was also observed. A microcomputer-aided backflush was proven effective in alleviating membrane fouling and allowing long term separation of P. stipitis from a fermentation broth.

Optimization of Fermentation Processes with Singular Approximation and Minimum Principle (Singular Approximation과 Minimum Principle을 이용한 발효공정의 최적화)

  • The two optimal control algorithms, singular approximation and minimum principle, were compared in this paper. The switching time with singular approximation was determined with mathematical derivation and the optimal control profile of specific growth rate was also calculated with minimum principle. The optimal control profiles were calculated by making simple model correlating the specific cell growth rate and specific product formation rate. The optimal control profiles calculated by singular approximation approach were similar to stepwise form of those calculatd by minimum principles. With the minimum principle, the product concentration was 8% more than that of singular approximation. This performance difference was due to a linearization of a nonlinear function with singular approximation. This optimal approaches were applicable to any system with different optimal cell growth and product formation.

  • The effects of cobalt chloride, methionine, and various carbon sources on the sisomicin production by M. inyoensis NRRL 3292 were investigated. It was found that both cobalt chloride and methionine exerted a greater stimulatory effect on sisomicin formation. Kinetic studies with various carbon sources revealed thai polysaccharide such as starch or dextrin was found io be better than glucose for sisomicin production Moreover, the relatively low concentration of dissolved carbon dioxide was one of the most important factors In accelerating sisomicin production during idiophase.

Strength of E. coli Floc as indicated by Shear Index (Shear Index로 표시된 E. coli Floc의 강도)

  • 침강제에 의해 형성된 E. coli floc들의 강도를 측정하기 위해 floc 의 shear index를 측정하였다. 형성된 E. coli floc은 10/sec 같이 낮은 shear rate에서도 분쇄되거나 변형되었 다. 측정된 shear index의 감소에서 보듯이 E. coli floc의 강도는 염의 농도가 증가함에 EK 라 감소하였다. E. coli floc의 shear index는 NaCl의 농도가 0에서 100 mM로 증가함에 따 라 0.47에서 0.09로 줄었다. 발효배지의 조성에서 형성된 E. coli floc들은(shear index=0.18-0.24 with BPA-1000. 0.13-0.22 with BPA-1050 and 0.37-0.42 with BPA-5020) 염이 없을 때 형성된 floc에(shear index=0.47 with BPA-1000 and 0.46 with BPA-1050) 비 해 약하였다. 따라서 발효배지에서 형성된 floc은 생물공정 중 쉽게 shear에 의해 분쇄되거 나 변형될것이다.

  • The sludge wastes fermentation process reactors were operated to produce the VFAs(volatile fatty acids) as supplemental carbon sources and to determine the optimum operating conditions. The experiment was carried out by varied mixture ration of 400:0 350:30 300:100 200:200 and operating temperature 2$0^{\circ}C$ 3$0^{\circ}C$ and 4$0^{\circ}C$ The results were as follows: Higher VFAs production rate observed at higher mixed ratio of primary sludge. When the mixed ratio of primary sludge and return sludge were 400:0 350:50 300:100 200:200 respectively. VFAs production are were 829.6mg/l 944.2 mg/l 597.9mg/ml an d441.6 mg/l , respectively. the yield of VFAs increased with temperature, but decreased with initial TSS concentration Because fermented sludge has relatively low nitrogen and phosphorus and relatively high VFAs it can be used as a substitute for external carbon in biological nutrient removal process.

  • An optical sensing membrane has been fabricated to measure the concentration of dissolved oxygen(DO) and pH value simultaneously. It has employed HPTS as a pH sensitive dye and a ruthenium(II) complex as a DO sensitive dye. The sensing membrane has been applied to wells in a 24-well microtiter plate. Using the 24-well microtiter plate the concentrations of dissolved oxygen and pH values have been on-line monitored during the cultivations of E.coli DH5${\alpha}$, B.cereus 318 and P.pastoris X-33. On-line monitoring of DO and pH in microorganism cultivation processes showed good performance of the sensing membrane containing 5 mM HPTS and 2 or 5 mg/mL Rudpp.

  • The studies were carried out to optimize a new starter for bread fermentation. Two strains of lactic acid bacteria and yeast were isolated from Kimchi. These strains showed good condition for quality bread fermented. The strains identified as Leuconostoc mesenteroides, Lactobacillus brevis, Saccharomyces fermentati and Saccharomyces cerevisiae. The mixed culture of four strains was due to the synergistic effect by interaction of these strains.

  • The computer interfaced by the necessary hardwares and the software developed in PARTs I and II were used to control the fermentation with recombinant cells in an on-line feedback-feed-forward manner. The recombinant cells were induced for expression either thermally or chemically. Very accurate controls with fast response and no oscillation could be performed.
