Cytogenetic Radiation Adaptive Response Assessed by Metaphase Analysis and Micronuclei Test in Human Lymphocytes and Mouse Bone Marrow Cells (인체말초혈액 림프구와 마우스골수세포에서 중기염색체 분석법과 미소핵검사법을 이용한 방사선적응반응 평가)

  • Purpose: Radiation adaptive response in human peripheral lymphocytes and mouse bone marrow cells was investigated using both metaphase analysis and micronucleus assay. We assessed the correlation between both tests. Materials and Methods: Two groups of the human peripheral lymphocytes and mouse bone marrow cells were exposed to low dose (conditioning dose, 0,18 Gy) or high dose (challenging dose, 2 Gy) ${\gamma}$-rays. The other 4 groups were exposed to low dose followed by high dose after several time intervals (4, 7, 12, and 24 hours, respectively). The frequencies of chromosomal aberrations in metaphase analysis and micronuclei in micronucleus assay were counted. Results: Chromosomal aberrations and micronuclei of preexposed group were lower than those of the group only exposed to high dose radiation. Maximal reduction in frequencies of chromosomal aberrations were observed in the group to which challenging dose was given at 7 hour after a conditioning dose (p<0.001). Metaphase analysis and micronucleus assay revealed very good correlation in both human lymphocytes and mouse bone marrow cells (r=0.98, p<0.001 ; r=0.99, p=0.001, respectively). Conclusion: Radiation adaptive response could be induced by low dose irradiation in both human lymphocytes and mouse bone marrow cells. There was a significant correlation between metaphase analysis and micronucleus assay.

  • Surface ultrastructure of Pawctrema timonnauini developmental stages was studied using a scanning electron microscope. The metacercariae were collected from the marine clam, Topes phiLippinam and juvenile and adult worms were recovered at 1, 2, 3 and 7 days after experimental infection of mice. The metacercariae had a large oral sucker and characteristic lateral projections. Around the lip of the oral sucker many type I and type II sensory papillae were observed, and type III papillae were located symmetrically on the medial side of the lateral projection. Numerous type I papillae were grouped around the genital pore. The tegumental spines were distributed over the worm surface except the lip of the suckers and genital pore. The 1-day old worm had a well-developed ventral sucker, with 6 type II sensory papillae on its outer surface and another 6 type I papillae on the inner side. Two small type I papillae were seen on the anterior side of the ventral sucker. The genital pore was small and opened separately from the ventral sucker and 15 type I papillae were grouped around it. The 2-, 3-. and 7-day worms revealed that as they grew to be adults, the spine tips became multipointed, the genital pore formed a genital atrium, and the cytoplasmic process became well differentiated. In 2- and 3-day worms 10 type II papillae encircling the lip of the oral sucker, and additional 4 papillae at the dorsal side of 4 dorsal type II papillae were a characteristic feature. The distribution pattern of sensory papillae around the oral sucker and genital pore, and 2 type I papillae on the anterior side of the ventral sucker, was so peculiar in R timonnnuini, that they seem to be useful keys for taxonomic differentiation from other gymnophallids.

A Preliminary Study on Stratigraphy and Petrochemistry of the Okcheon Group, Southwestern Okcheon Metamorphic Belt (서남 옥천변성대 옥천층군의 층서 및 암석화학에 대한 예비연구)

  • The Okcheon Group in the southwestern part of the Okcheon Metamorphic Belt is subdivided into two distinct tectonostratigraphic units: the Boeun unit in the south and the Pibanryeong unit in the north. The Boeun unit consists of petites, psammites, carbonaceous petites, limestones and pebble-bearing quartzites. The Pibanryeong unit is composed of petites, well-sorted fine-grained psammites, carbonaceous psammites and quartzites. In order to outlining stratigraphy and depositional environments of the Okcheon Group, detailed stratigraphic sections were measured in three locations; one section(Gosan section) of the Boeun unit and two sections(Sorungjae and Hwangryeongzae sections) of the Pibanryeong unit. The Gosan section of the Boeun unit is interpreted to be deposited in the shallow marine environments, whereas the Sorungjae and Hwangryeonaiae sections of the Pibanryeong unit appear to be deposited in slope and deep basin environments. This result indicates rapid subsidence between deposition of the Boeun and Pibanryeong units in sedimentary environment. The trace of sedimentological environments in the Hwasan area was investigated by geochemical analysis of 109 metapelitic and psammitic rock samples. Distinct chemical variations of politic and psammitic rocks from the Boeun and Pibanryeong units in the study area are evident from plots of major elements and $A1_2O_3$/$SiO_2$ versus Basicity Index($Fe_2O_3{+}MgO$)/($SiO_2{+}K_2O{+}Na_2O$). The rocks show a progressive chemical trend from the Boeun unit to the Pibanryeong unit on these diagrams. They in the southern sector of the Boeun unit display lower values and a comparatively wide range of $A1_2O_3$/$SiO_2$ and Basicity Index, as compared with those from the northern sector of the Boeun and Pibanryeong units. The southern sector of the Pibanryeong unit including narrow staurolite-bearing zone is characterized by values that are transitional between the Boeun and Pibanryeong units. These data, combined with depositional environment progressively deepened towards the northwest, support a half-graben model for the Okcheon basin, as proposed by Cluzel et al.(1990)

Analysis of Acquisition Parameters That Caused Artifacts in Four-dimensional (4D) CT Images of Targets Undergoing Regular Motion (표적이 규칙적으로 움직일 때 생기는 4DCT 영상의 모션 아티팩트(Motion Artifact) 관련된 원인분석)

  • The aim of this study was to clarify the impacts of acquisition parameters on artifacts in four-dimensional computed tomography (4D CT) images, such as the partial volume effect (PVE), partial projection effect (PPE), and mis-matching of initial motion phases between adjacent beds (MMimph) in cine mode scanning. A thoracic phantom and two cylindrical phantoms (2 cm diameter and heights of 0.5 cm for No.1 and 10 cm for No.2) were scanned using 4D CT. For the thoracic phantom, acquisition was started automatically in the first scan with 5 sec and 8 sec of gantry rotation, thereby allowing a different phase at the initial projection of each bed. In the second scan, the initial projection at each bed was manually synchronized with the inhalation phase to minimize the MMimph. The third scan was intentionally un-synchronized with the inhalation phase. In the cylindrical phantom scan, one bed (2 cm) and three beds (6 cm) were used for 2 and 6 sec motion periods. Measured target volume to true volume ratios (MsTrueV) were computed. The relationships among MMimph, MsTrueV, and velocity were investigated. In the thoracic phantom, shorter gantry rotation provided more precise volume and was highly correlated with velocity when MMimph was minimal. MMimph reduced the correlation. For moving cylinder No. 1, MsTrueV was correlated with velocity, but the larger MMimph for 2 sec of motion removed the correlation. The volume of No. 2 was similar to the static volume due to the small PVE, PPE, and MMimph. Smaller target velocity and faster gantry rotation resulted in a more accurate volume description. The MMimph was the main parameter weakening the correlation between MsTrueV and velocity. Without reducing the MMimph, controlling target velocity and gantry rotation will not guarantee accurate image presentation given current 4D CT technology.

Study on Effect of Convection Current Aeration System on Mixing Characteristics and Water Quality of Reservoir (대류식 순환장치의 저수지수체 유동특성 및 수질영향)

  • This study examines the operational effectiveness of a Convection Current Aeration System (CCAS) in reservoir. CCAS was run from June, 2008 when the thermocline begun forming in the reservoir. This paper reviews the influence of stratification, dissolved oxygen dynamics and temperature in the lake's natural state from June to October 2008. The survey was done on a week basis. Upwelling flow effects a radius of $7{\sim}10m$ at a surface directly and was irrelevant to the strength of thermocline. On the other hand, it was affected the number of working days, and strength of thermocline at vertical profiles of the reservoir. Longer CCAS run, the deeper was the vertical direct flow area. However it didn't break the thermocline during summer season of 2008. The operating efficiency of the CCAS in the reservoir depends on hydraulics and meteological conditions. Computational Fluid Dynamics (CFD) is a very useful tool for evaluating the operating efficiency of fluid dynamics. The geometry for CFD simulation consists of a cylindrical vessel 25 m radius and 40 m height. The CCAS is located in center of domain. The non-uniform tetrahedral meshes had a bulk of the geometry. The meshes ranged from the coarse to the very fine. This is attributed to the cold water flowing into the downcomer and rising, creating a horizontal flow to the top of the CCAS. The result of CFD demonstrate a closer agreement with surveyed data for temperature and flow velocity. Theoretical dispersion volume were calculated at 8m depth, 120 m diameter working for 30 days and 10 m depth, 130 m diameter working for 50 days.

A Study on the Simcho of Wooden Pagodas in Baekjae (백제의 심초 및 사리봉안)

  • Recently, there has been an increase in excavation studies of wood pagodas from the Three Kingdoms and Unified Shilla periods and new data related to wood pagoda erection are being found bringing about progress in research on this field. In other words, studies on wooden pagodas in Korea were composed mainly of flat, axis construction techniques and sarijangeomgu, but by acquiring new data, it has now become possible to study not only the stylobate construction procedure and transition, but also studies on restoring wooden pagodas. Furthermore, pagoda sites similar to this were found in China and Japan as well, making it possible to make comparative studies among ancient wooden pagodas possible. In this paper, the main remains were set as Baekjae wooden pagodas, which were the most frequently studied and among the wooden pagodas, the simcho (central base stone) and sarira housing locations. In result, simcho can be found changing its position from underground ${\rightarrow}$ halfway underground ${\rightarrow}$ above ground. Baekjae wooden pagodas up until the mid sixth century located at Neungsan-ri saji (AD 567) and Wangheungsaji (AD 577) had its simcho located underground and later it was constructed halfway underground and then above ground. It was confirmed that in the 7th century, it became customary to place above ground as seen in the Jaeseoksaji (AD639) and Hwangnyongsaji (AD645) wooden pagoda sites. The sarira was usually located on the south side of the simcho, but gradually changed to the center. In particular, sarira were combined in the simcho in the mid sixth century at the Wangheungsaji. This is approximately 11 years earlier than the Bijosa (AD 588) simcho found in Japan and this was not found even in the simcho of wooden pagodas in Yeongnyeongsa (AD 516) and Jopaengseong temple (AD 535~561) of China showing that the Wangheungsaji simcho was the earliest of its kind.

Evaluation of the usefulness of the method according to changes in patient breathing during chest 4D CT imaging (흉부 4D CT에서 호흡 변화에 대한 일시 중지 및 재개 방법의 유용성 평가)

  • Purpose : In order to evaluate the usefulness of clinical application of the Pause & Resume methods by comparing and analyzing the data stability and dose reduction effect when repeat scan assuming irregular breathing and using the Pause & Resume method during chest 4D CT using QuasarTM Phantom. Materials and Methods : Using the QuasarTM Phantom, set the breathing rate per minute to 15 BPM and 7.5 BPM, and set the S15 point as an irregular breathing section, and then placed OSLD to this point and use the Pause & Resume method to measure the dose of S15. CTDIvol, DLP, and ALARA-CT were used for comparative analysis of radiation dose between Pause & Resume method and Repeat-scan. In order to evaluate the stability and usability of the data applying the Pause & Resume method, the captured images were sorted by Advanced Workstation Volume Share7 and then sent to EclipseTM, the diameter and volume were analyzed by forming a contour on the iron ball in the QuasarTM Phantom Results : When using Pause & Resume, the dose of OSLD measurement increased by 1.97 times in the section of S15. As a result of image evaluation, the average value of all volumes measured with and without the Pause & Resume method at 15 BPM and 7.5 BPM was 15.2 cm3±0.5%.Allthemeasuredvaluesfor the radius of iron ball were 3.1 cm regardless of whether Pause & Resume method was used or not. In the case of using Pause & Resume, 33% decreased from the lowest DLP value and 38% decreased from the highest DLP value of repeat scan, and the effective dose also decreased 32.1% from the minimum value and 37.6% from the maximum value. Conclusion: Irradiation dose was increased by Pause & Resume method because of the repeat scan on the S15 site where assuming irregular breathing occurred, However Pause & Resume method led to a significant reduction in dose on overall scan range. It also proved the usefulness of clinical application of the Pause & Resume method as a result of similar diameters and volumes of iron ball measurement.

Sprouting Inhibition after CIPC Spraying on Early and Mid-season Potato Varieties during Storage in Semi-underground Warehouse at Room Temperature in Summer (CIPC 처리한 조·중생종 감자의 반지하 저장고를 이용한 하계 실온저장 중 맹아 억제 효과 비교)

  • This study was carried out to determine the sprouting period of early and mid-season varieties, which includes 'Atlantic', 'Chubaek', and 'Superior', during the summer storage period in a semi-underground warehouse without cooling system. And also it was investigated the effect of chlorpropham [Propan-2-yl N-(3-chlorophenyl)carbamate, CIPC] treatment on the sprouting inhibition for the varieties. This study was conducted to figure out a sprout inhibitory effect when CIPC was applied to 1kg of the potato tubers at concentrations of 10 mg and 20 mg which are lower than the treatment concentrations of ca 30 mg prescribed by the positive list system (PLS). The internal temperature of the warehouse used in this experiment was lowered by 5℃ or more than the outside temperature. The difference between the lowest and highest temperature during the experiment throughout the day was 5℃. It showed the effect of reducing to 1/2 of the difference in outdoor temperature. As for the sprouting of potatoes, the extremely early variety 'Chubaek' sprouts appeared at the 6th week of storage of control and it was the fastest sprouting potato among the control groups of the varieties. Sprouting began to appear in the Superior at the 6th week of storage, while the 'Atlantic' sprouted at the 8th week of storage. The appearance of sprouts was suppressed in all treatment groups of 'Atlantic' and 'Superior' varieties in CIPC treatments. Sprouts were observed in all treatment groups of 'Chubaek' after the 7th week, but the elongations of the sprouts in tubers were completely inhibited until the 8th week of storage. 'Atlantic' and 'Superior' seemed to have a sprouting inhibitory effect even with a low CIPC concentration of 10 mg·kg-1, with the exception of extremely early variety 'Chubaek' that breaks out of the dormancy quickly. Although weight loss occurred continuously during storage, it was minor loss of 0.7-1.6%. There was no consistent trend for changes of the loss in the varieties and CIPC treatments. Most common pathological disorder was the dry rot during the experiment, but only few were affected. The use of the tubers treated at 18℃ and 90% RH for 10 days and the rack of refrigeration system which lead to lack of convection seemed to have suppressed the spread of pathogens.

The Vascular Plants in Mt. Seungma (Gyeonggi-do) (승마산(경기도)의 관속식물상)

  • This study was carried out to investigate the flora of Mt. Seoungma (Gyeonggi-do). The vascular plant were collected 13 times (from March to October 2011), and 384 taxa in total were identified, including 94 families, 260 genera, 340 species, 5 subspecies, 37 varieties and 2 forms. Korean endemic plants of this area were nine taxa; Clematis brachyura, Rhododendron mucronulatum, Forsythia koreana, Paulownia coreana, etc. Rare plants (IUCN) by the Korean Forest Service and Korean National Arboretum three taxa were recorded; one Critically Endangered species (CR), Prunus yedoensis, one Vulnerable species (VU), Epimedium koreanum, one Least Concemed species (LC), Phacelurus latifolius. The floristic regional indicator plants found in this area were 15 taxa comprising two taxa of grade V and III, one taxa of grade II, 10 taxa of grade I. Furthermore, therophytes (28.3%), hemicryptophytes (23.0%), hydrophytes (14.3%) showed high proportional ratio in life form spectrum. The naturalized plants were identified as 43 taxa and the percent of naturalized index (NI) was 11.2% of total 384 taxa vascular plants. The vegetation status of investigated area was comparatively well conserved, but the degree of damage was increasing rapidly by the disturbances of human beings. Therefore, a long-term monitoring of vegetation changes and movement grasp must become accomplished.

The Vascular Plants in Mt. Gilsangsan (Ganghwa-Isl.), Korea (길상산 (강화도)의 관속식물상)

  • This study was carried out to investigate the flora of Mt. Gilsangsan (Ganghwa-Isl.). The vascular plants were collected 14 times (from March to November 2012), and were identified as 595 taxa; 122 families, 359 genera, 529 species, 7 subspecies, 51 varieties, 2 forms and 3 hybrids. The plant formation of Mt. Gisangsan is the deciduous broad-leaved and conifer mixed forest which is the common one in the middle part of Korean peninsula. All most mountain covered with young secondary forest which is mainly composed of Pinus and Quercus. The family represented by the largest number of species diversity were Asteraceae (11.%), Poaceae (10.7%), Cyperaceae (6.38%), Fabaceae (3.86%) and Rosaceae (3.52%) respectively. Korean endemic plants of this area were 12 taxa; Asarum misandrum, Clematis brachyura, Philadelphus schrenkii, Forsythia koreana, Polygonatum infundiflorum, etc. The red list of vascular plants according to IUCN valuation basis were examined, endangered species (EN) Abies koreana, Near Threatened species (NT) both Wikstroemia trichotoma and Sparganium erectum, Least Concern species (LC) Platycladus orientalis, Swertia diluta var. tosaensis and Iris minutoaurea and by Not Evaluate species (NE) Polygonatum infundiflorum. The floristic regional indicator plants found in this area were 45 taxa comprising two taxa of grade V and VI, nine taxa of grade III, three taxa of grade II, 29 taxa of gradeI. The naturalized plants were identified as 64 taxa and the percent of naturalized index (NI) was 10.7% of total 595 taxa vascular plants. Ecosystem disturbance wild plants were consisted of eight taxa; Rumex acetosella, Sicyos angulatus, Solanum carolinense, Ambrosia artemisiifolia, Ambrosia trifida, Aster pilosus, Hypochaeris radicata and Lactuca scariola. Furthermore, therophytes (26.5%), hemicryptophytes (26.0%), Geophyte (14.0%) showed high proportional ratio in life form spectrum.
