Effect of Vibrio alginolyticus on the Algicidal Activity of Shewanella sp. SR-14 (Vibrio alginolyticus가 Shewanella sp. SR-14의 미세조류 증식저해 활성에 미치는 영향)

  • The algicidal activity of Shewanella (formerly Alteromonas) sp. SR-14 against diatom, Chaetoceros calcitrans was reported in our previous papers. In this study, the effect of Vibrio alginolyticus on the algicidal activity of Shewanelia sp, SR-14 was examined under the optimum algicidal conditions, i.e., temperature ($21\pm1^{\circ}C$), light intensity (4,000 lux), and light: dark cycle (12 hour: 12 hour). Shewanella sp. SR-14 grew well in the presence or the absence of V. alginolyticus in Conwy medium. Algal growth was only inhibited by Shewanella sp. SR-14. V. alginolyticus did not show the algicidal activity, Growth of C. calcitrans increased synergistically with growth of V. alginolyticus. When the initial inoculum of V. alginolyticus was only 1 log cycle higher than that of Shewanella sp. SR-14, the effect of V. alginolyticus on the algicidal activity of Shewanella sp. SR-14 was insignificant during incubation of mixed culture, i.e., two bacterial species and the alga. However, when V. alginolyticus dominated Shewanella sp. SR-14 by 3 log cycles of bacterial counts, it was found that the strain SR-14 could not inhibited growth of C. calcitrans up to 5 days of incubation.

Attaching Nature and Community Variation of Epiphytic Diatoms on Leaf of Zostera spp. (Zostera속 해초에 부착하는 규조류의 부착 특성과 해초 종별 군집 변화)

  • Epiphytic diatoms are very important organisms in the seagrass ecosystem because their colonization on leaves increases microtopography and provides attachment sites that make the leaves more hospitable for other epiphytes. Epiphytic diatoms were attached to the leaves in the following 3 manners: (1) parallel to the cells of the seagrass leaf or by molding the shape of the diatom along the cell shape of the leaf; (2) with increasing diatom density toward the leaf tip; (3) Cocconeis species as attaching species than the Naviculoid species as the second attaching species on the leaf tip. In addition, the epiphytic diatom communities on Zostera marina leaves differed from those on the Zostera japonica leaves, but were very similar to the epiphytic communities on Zostera caespitosa leaves. Our results suggest that the epiphytic community on seagrass leaves varied according to the leaf shape such as leaf length and width, but the leaf cell shape or size did not influence the dynamics of the diatom communities.

  • Ballast water has been known as a major vector for global dispersal of toxic dinoflagellates and other microalgae. In this study, biodiversity in ships’ ballast water was examined using a dinoflagellate-oriented PCR primer set and species-specific real-time PCR. While motile dinoflagellates could be observe at very low cell densities by light microscopy,a wide range of dinoflagellate taxa including parasitic and phototrophic pico-dinoflagellates as well as harmful species to marine fish/shellfish was detected when techniques for cloning/sequencing of SSU rDNA of sample cells were used. Present result suggests that molecular methods including species-specific PCR primers may offer rapid and accurate detection of invasive species in ballast water.

Studies of Micro-Air Flotation for Removal of Turbidity (탁도제거를 위한 미세공기 부양법 연구)

  • In this study, efficiency of pre-treatment of turbid seawater was measured where micro-air bubbles were used to remove particles in seawater after input of natural coagulant PGA. Artificial seawater was prepared having the intended trubidity using marine sediments and microalgae. 73.7% of turbidity removal was achieved when 0.5g/L of $AlCl_3{\cdot}6H_2O$ was added in the artificial seawater, but 92.4% of turbidity removal was observed when 0.05g/L of PGA was added in the artificial seawater containing microalgae. In addition, much greater turbidity removal was achieved for microalage than sediments. For both cases, input of 0.1g/L PGA and following additional input of micro-air bubbles for 5 seconds resulted in the maximum removal efficiency where reaction time of coagulation was 1 min and flotation by micro-air bubbles was 10 min. From this study, we concluded that micro-air floation after coagulation could be a possible economical pre-treatment method for highly turbid seawater.

Seasonal Variations of Microphytobenthos in Sediments of the Estuarine Muddy Sandflat of Gwangyang Bay: HPLC Pigment Analysis (광합성색소 분석을 통한 광양만 갯벌 퇴적물 중 저서미세조류의 계절변화)

  • Seasonal variations of microalgal biomass and community composition in both the sediment and the seawater were investigated by HPLC pigment analysis in an estuarine muddy sandflat of Gwangyang Bay from January to November 2002. Based on the photosynthetic pigments, fucoxanthin, diadinoxanthin, and diatoxanthin were the most dominant pigments all the year round, indicating that diatoms were the predominant algal groups of both the sediment and the seawater in Gwangyang Bay. The other algal pigments except the diatom-marker pigments showed relatively low concentrations. Microphytobenthic chlorophyll ${\alpha}$ concentrations in the upper layer (0.5 cm) of sediments ranged from 3.44 (March at the middle site of the tidal flat) to 169 (July at the upper site) mg $m^{-2}$, with the annual mean concentrations of $68.4{\pm}45.5,\;21.3{\pm}14.3,\;22.9{\pm}15.6mg\;m^{-2}$ at the upper, middle, and lower tidal sites, respectively. Depth-integrated chlorophyll ${\alpha}$ concentrations in the overlying water column ranged from 1.66 (November) to 11.7 (July) mg $m^{-2}$, with an annual mean of $6.96{\pm}3.04mg\;m^{-2}$. Microphytobenthic biomasses were about 3${\sim}$10 times higher than depth-integrated phytoplankton biomass in the overlying water column. The physical characteristics of this shallow estuarine tidal flat, similarity in taxonomic composition of the phytoplankton and microphytobenthos, and similar seasonal patterns in their biomasses suggest that resuspended microphytobenthos are an important component of phytoplankton biomass in Gwangyang Bay. Therefore, considering the importance of microphytobenthos as possible food source for the estuarine benthic and pelagic consumers, a consistent monitoring work on the behavior of microphytobenthos is needed in the tidal flat ecosystems.

  • 최근 세균을 이용한 적조 방제연구가 활발히 진행되면서 해양에서 다수의 미세조류 저해균이 분리되고 있다. 전보에서 저자들은 규조류 Chaetoceros spp.의 생육을 저해하는 해양세균 Shewanella sp. SR-14를 분리하였으며, 이 균은 대사산물을 생성하여 그 생육을 저해하는 것으로 보고한 바 있다. 그런데 Shewanella sp. SR-14에 의한 Chaetoceros calcitrans의 생육저해는 대사산물(배양여액)만을 사용한 경우보다 세균이 공존하는 경우에 그 활성이 휠씬 강하여 세균이 생성하는 저해물질에 의한 것으로만 이해하기 어려웠으며, 다른 미지의 기구가 관여하고 있을 가능성이 시사되었다. (중략)

  • The objective of this paper is to investigate into the design of photbioreactor for mass production of microalgae. Characteristics of previously developed photobioreactors were examined to obtain design factors, including light transfer, fluid dynamics and metabolic reactions, of the photobioreactor. Design technology of components related to the design factors, such as light sources, photobioreactor cases, spargers, mixer, etc., was discussed to improve the viability and the growth rate of microalgal and productivity of the photobioreactor. Finally a principle direction of the design for an airlift flat plane photobiorectors was investigated.

  • Recently, microalgae have been focused on potential biomass for bio-diesel and biorefinery process. The aim of this paper is to review the biorefinery process including biodesel using microalgae as a microreactor. The state-of-the-art of biodiesel and biorefinery research such as extraction and reaction process as well as byproducts utilization is described. In addition, we suggest possibility for develop bioactive substances and their industrial products from byproducts of microalgae massively obtained after bio-diesel extraction.

  • A study on the fine structure of the marine diatom has been carried out for taxonomical purpose during the period from Fib. 1982 to Feb. 1986 in Korean coastal waters. Fine structure of small Thalassiosira (Bacillariophyceae) was studied by means of light and scanning electron microscopy. Firstly, four species of Thalassiosira were identified and described. Of these Thalassiosira weissflogii, T.minima and T.oestrupii var venrickae were new records for Korean coastal waters.

  • Biofuels produced from biomass can be substituted for petroleum fuels due to GHG reduction, sustainability and environmental friendly. The process technologies that convert biomass into biofuels are varied and depend on the feedstocks. Microalgae are considered to be one of the most promising alternative source to the conventional feedstocks for biofuel. Microalgae can be converted to biodiesel, bioethanol, biogas and biojet fuel via thermolchemical and biochemical production technologies. This reviews discusses recent advance in understanding the effects of the characteristics of various processes on the production of biofuels using microalgae. The performances of microalgae based biofuel are compared.
