Anatomical Ultrastructure of Spermatozoa of Platichthys stellatus (Pleuronectidae, Pleuronectiformes) from Korea (강도다리(Platichthys stellatus) 정자의 미세해부학적 구조 (가자미목, 가자미과))

  • The spermatozoa of Platichthys stellatus is relatively simple cells composed of a spherical head, a short midpiece, and a tail, as in most Pleuronectiformes. The ultrastructure is characterized by the following features: a round nucleus with a deep nuclear fossa, the centriolar complex located at a right angle to each other, a short midpiece, a tail with paired lateral ribbon and no acrosome. However there are some minor morphological differences, including the appearance and number of the mitochondria, the shape and size of the nuclear fossa and the structure of the basal body. Especially the basal body structure consisting of a basal foot, a rootlet and nine alar sheets structures varies considerably in different species. It can be used as indicator of relationships in Pleuronectiformes because minute morphological differences might have functional and evolutionary significance. In conclusion, the spermatozoa of P. stellatus show a certain structural homogeneity and provide support for the concept that ultrastructural features of spermatozoa can be useful in taxonomic studies of Pleuronectiformes.

Comparative Study on Biological Effects of Gamma-Radiation and Bisphenol A with Tradescantia Micronucleus Assay (자주달개비 미세핵 분석법을 이용한 비스페놀 에이 및 감마선의 생물학적 영향 비교 연구)

  • Some of synthetic chemicals can net as an endocrine disrupting substance in higher animals. Dioxins, DDT, PCBs and bisphenol A (BPA) are classified into endocrine disruptors and aye under a strict control in many countries. This research was designed to compare the clastogenic effects of BPA to those of ionizng radiation to establish the relaltive effectiveness of BPA by means of Tradescantia micronucleus assay. For the uptake of the BPA through the stems, groups of fresh cuttings of Tradescantia BNL 4430 weve placed in BPA solutions of 0 to 4 $\mu$M for 6 hours under continuous aeration. The other groups of the cuttings were irradiated with 0 to 0.5 Gy of gamma- rays. The frequencies of micronucleus showed a positive dose- response relationship in the range of 0 to 0.5 Gy, and a clear concentration-response relationship in the experimental range of BPA concentrations. By comparing the two experimental results, it is possible to estimate the BPA concentration and its equivalent radiation dose for a fixed value of MCN frequency. BPA of $ll.8\mu{M}$ can give rise to 53.3 MCN/100 tetrads, which is the same frequency induced by 25 cGy of gamma- rays. It is of biological importance that clinical symptoms start to develop after a whole body exposure to radiation higher than 25 cGy. The results indicate that the pollen mother cells are an excellent biological end- point for toxicity test of suspected endocrine disrupting chemicals such as bisphenol A, cotylphenol and nonylphenol.

Adaptive Response Induced by Low Dose Ionizing Radiation in Human Lymphocytes (인체 말초 혈액 림프구에서 저선량 방사선 조사에 의해 유도되는 적응 반응)

  • Adaptive response induced by low dose ${\gamma}-ray$ irradiation in human peripheral lymphocytes was examimed. Human lymphocytes were exposured to low dose of ${\gamma}-ray$ (priming dose, 0.01Gy) followed by high dose (challenging dose, 1.5Gy) after various time intervals (4, 7, 20 hours). Frequencies of micronuclei were enumerated in both primed and unprimed groups. Maximum reduction in frequency of micronuclei was observed when challenging dose irradiation was followed by priming dose after 4hr incubation period. When challenging doses were irradiated 7 or 20hr after priming dose, frequencies of micronuclei were reduced slighty. However, these reduction were not statistically significant. In this study, human peripheral lymphocytes were irradiated at Go phase and they showed adaptive response induced by low dose radiation. Since micronucleus assay is relatively simpler and faster than other methods, it may be a good tool for evaluating radiation-induced adaptive responses.

The Comparison of Volatile Organic Compounds (VOCs) Analysis and Tradescantia Micronucleus (Trad-MCN) Bioassay for Evaluation of Hazardous Materials in Chemical Workplace Field (화학공장 실내 작업장에서의 유해물질 평가를 위한 VOC 분석법과 자주달개비 미세핵 분석법의 비교)

  • This research examined the presence of hazardous materials in chemical workplace field using an integrated chemical/biological monitoring. Chemical workplace field air for volatile organic compounds (VOCs) analysis was collected using a collection tube packed with Tena.x TA adsorbent 400 mg. Workplace field air samples were analyzed by gas chromatography/mass spectrometry (GC/MS). Simultaneously, Tradescantia BNL 4430 clone was exposed in situ to monitor hazardous materials in chemical workplace field. GC/MS analysis showed the presence of various VOCs such as trichloroethylene, toluene, ethylbenzene, (m,p,o)-xylenes, styrene, 1,3,5-trimethylbenzene, and 1,2,4-trimethylbenzene. The results showed that in situ monitoring of VOCs with the Tradescantia-micronucleus (Trad-MCN) assay gave positive results in chemical workplace field and negative response at outdoor air. In conclusion, inhalation of these field air by workers may affect chronic demage to their health by inducing micronuclei formation in Tradescantia pollen mother cells. The combination of chemical/biological monitoring is very effective to evaluate hazardous materials in workplace field and can be alternatively used for screening hazardous materials.

리포좀을 이용한 형질전환 닭 생산에 관한 연구

  • 본 연구는 1세포기 닭 수정란에서 외래 표지유전자(EGFP)와 리포좀(liposome)을 사용하여 외래유전자의 핵 전이의 효율성을 검증하고자 하였다. 실험은 리포좀과 혼합된 표지유전자와 naked 유전자 두 그룹으로 나누고, 유전자 미세 주입방법을 이용하여 배반엽 단계(stage X)와 1세포기 수정란의 세포질에 미세 주입하고 지속적으로 배양하면서 GFP의 발현 양상들을 관찰하였다. 실험결과, 배반엽 단계와 1세포기 수정란 모두에서 리포좀과 외래 유전자의 혼합물을 미세 주입한 경우 일주일 정도 지속적으로 GFP가 발현되었으나, 외래 유전자만을 주입한 경우 GFP의 발현이 관찰되지 않았다. 본 연구결과는 리포좀이 효율적으로 외래 유전자를 닭 수정란의 핵으로 이동시킴을 보여주고 있다.

Processing of Polyurhane Microcellular Foam for Thermal Insulation (단열재용 폴리우레탄 미세포 포움의 가공)

  • 단열용도의 폴리우레탄 미세포 포움의 가공에 대한 연구를 수행하였다. 미세포 구조 를 얻기 위해서는 핵생성율을 증진시키고 균일한 분포의 기포를 생성시켜야 한다. 이를 위 해 이산화탄소 기체를 풀리올과 이소시아네이트에 각각 과포화시키고 충돌혼합하여 초음파 가진을 적용하였다. 이산화탄소 기체가 수지 내부에서 기포 내부로 확산함에 따라 기포의 성장이 조절된다고 가정하고 금형이 충전되는 동안에 금형 내부에서의 기포성장기구를 이해 하기 위하여 수치적인 방법으로 이론적 연구를 수행하였다. 경화 시간과 확산 경계를 고려 하여 최종적인 기포의 크기를 계산하였으며 반응속도론을 고려하여 중합반응동안의 폴리우 레탄의 점도의 변화를 예측하고 경화 시간을 결정하였다. 실험적으로 결정된 기체 분자수를 기준으로 하여 이론적으로 확산경계를 예측하였다. 화학적 발포제인 물과 함께 물리적 발포 제인 이산화탄소를 각각 1,2,3기압의 포화압력으로 변화시키면서 폴리올과 이소시아네이트에 포화시켜 폴리우레탄 포움을 제작하고 제작된 포움의 밀도, 열전도도, 및 기포의 수와 지름 을 측정하였다. 측정된 결과로부터 이산화탄소의 포화압력과 초음파 가진이 포움의 기포핵 생성에 미치는 영향을 살펴보았다.

Structure of Egg Envelope and Oogenesis of Kichulchoia multifasciata (Pisces, Cobitidae) (수수미꾸리 Kichulchoia multifasciata의 난자형성과정과 난막의 구조)

  • Histological study on egg envelope and oogenesis of Kichulchoia multifasciata (Pisces, Cobitidae) was carried out by light microscopes and scanning electron microscopes. Various developmental cells appeared in ovary of the specimen catched during November 2010. The cytoplasm of oogonia was acidic and many nucleoli were located at the inner side of nucleus membrane. The size of the oogonia was $103.9{\pm}24.7$ ${\mu}m$ with nucleus size $42.9{\pm}6.9$ (31.1~50.3) ${\mu}m$. Primary oocyte having $277.5{\pm}60.5$ (216.7~354.9) ${\mu}m$ in diameter began to accumulate yolk vesicles. As the developmental stages proceed, secondary oocyte grows larger to $617.6{\pm}85.1$ (503.4~723.6) ${\mu}m$, and eosinophilic yolk granules yolk granules appeared between the yolk vesicles occupying most cytoplasm, and there are some yolk mass formed already. There are some yolk mass formed already. Envelope of fertilized egg investigated by a scanning electron microscope had plenty of microvilli (2~3 ${\mu}m$ in length) over the entire egg surface and a micropyle. Especially, the microvilli surrounding the micropyle were longer than those of egg surface with $5.26{\pm}1.22$ ${\mu}m$.

배양액이 마우스 핵이식 재구축배의 초기발생에 미치는 영향

  • 본 실험은 Piezo-미세조작기(PrimeTech Ltd., Japan)를 사용하여 마우스 핵이식 후 재구축배를 CZB와 KSOM 두가지 배양액을 사용하여 체외배양성적을 비교 검토하였다. MII의 미수정란은 성숙한 4~5주령 B6D2Fl에 hCG 주사 후 14시간째에 과적 방법을 통해 난관의 팽대부로 부터 회수하였고, metaphase II chromosome-spindle complex와 최소량의 세포질을 내경이 10$\mu\textrm{m}$인 피펫으로 흡입하여 탈핵하였다. 핵이식에 사용된 난구세포(8-l0$\mu\textrm{m}$)는 3시간동안 12% PVP 에처리 하여 piezo-미세조작기를 이용하여 세포질에 세포의 핵을 직접 미세주입 하였다. 핵이식 후 생존한 재구축배는 2시간동안 배양한 후 10mM SrC1$_2$와 5$\mu\textrm{g}$/$m\ell$의 cytochalasin B가 첨가된 $Ca^{2+}$-free CZB에서 6시간 활성화 처리하였다, 활성화 처리 후 위전핵이 관찰된 재구축란을 CZB 와 KSOM 배지에서 배양하면서 발달률을 비교하였고, 상실배 및 배반포배로 발달한 재구축배를 day 3 대리모에 이식하였다. 표 1에서 보는 바와 같이 재구축배의 2-cell로의 발달률에 있어서 KSOM이 CZB에 비하여 유의적으로 높게 나타났으며(P<0.05), 또한 4-cell과 상실배/배반 포배로의 발달률에 있어서도 KSOM이 CZB에 비하여 유의적으로 높은 발달률을 나타내었다(P<0.01). 또한 KSOM 배지에서 배양된 상실배/배반포배를 대리모에 이식한 경우에 11.5 d.p.c에 생존한 태아가 관찰되었다. 이상의 결과로 핵이식 재구축배의 활성화 처리 후의 발생에는 KSOM 배지가 CZB 배지에 비하여 유효함을 확인 할 수 있었다.그와 같은 배양 기술을 이용하여 외래유전자를 도입한 일련의 결과에 관하여 보고 하고자한다., 이것은 세포내 유전자가 transfection되지 않은 세포도 neo selection에서 선발된다는 것을 제시하고 있다. 따라서 체세포를 이용한 형질전환동물 생산을 위해서는 세포내 유전자 도입과 선발 과정에서 나타난 colony에 대하여 보다 엄격한 screen을 하는 것이 필요한 것으로 생각된다.로 우점하였다. 여름철 식물플랑크톤 대발생에 영향은 수온과 직산염이 중요하였으나, 부유물질 크게 기여하지 못하였다.애를 확인하고 지도 관점을 파악하는 것을 포함한다. 그러나 본 논문은 역사발생적 수학 학습-지도 원리의 실제적인 적용에 관하여는 기초적인 연구에 지나지 않기 때문에, 역사발생적 원리를 학교수학에 실제적으로 적용하기 위해서는 각각의 내용에 대한 철저한 역사적 분석을 바탕으로 하는 후속 연구가 필요하다./TEX>구성교육${\lrcorner}$이 조선총독부의 관리하에서 실행되었다는 것을, 당시의 사범학교를 중심으로 한 교육조직을 기술한 문헌에 의해 규명시켰다.nd of letter design which represents -natural objects and was popular at the time of Yukjo Dynasty, and there are some documents of that period left both in Japan and Korea.   • Onuf's nucleus, which is located in the ventral horn, has been known to innervate the striated muscles of the urethral and anal sphincter muscles via the pudendal nerve Onuf's nuclei are resistant to pathologic condition such as poliovirus. The reason why the motor neurons in Onuf's nucleus are less degenerated is not certain until now. The present study aims at updating the microscopical characteristics including its location the Onuf's nucleus innervating the external urethral sphincter, and ultrastructures of the zinc-enriched (ZEN) terminals synaptically-contacting with Onuf's motor neurons in the rat spinal gray matter by using HRP tracing method and zinc selenium autometallography, respectively. Based on HRP tracing method, Onuf's nuclei were located adjacent lateral dendritic projections of the ventral horn. Their shape was almost round at lumbar level, but oval at sacral segment of spinal cord. In size, their somata were smaller than that of other motor nuclei. In AMG stained sections, Onuf's nuclei were innervated by highly concentrated ZEN terminals, and contained small and middle-sized ZEN, but totally void of large ZEN terminals. AMG silver grains were confined to presynaptic ZEN terminals against dendritic elements and somata of the Onuf's motor neurons. A majority of the ZEN terminals contained flattened synaptic vesicles and made symmetrical synaptic specializations.

A Theoretical Study of GaAs Nucleation in GaAs/Si Heteroepitaxy Structure (GaAs/Si Heteroepitaxy 구조에서 GaAs의 초기 핵생성에 관한 이론적 고찰)

  • Early stage of GaAs nucleation on Si substrate was theoretically studied by computer simulation. Compared to the constant ledge interaction energy in conventional nucleation theory, functional behavior of ledge-ledge interaction resulted in small size clusters depending on the cluster size and shape. Among various kinds of clusters, the multilayer pyramidal shape GaAs cluster requires smallest excess free energy due to the formation of Ga(111) facet planes. There this result suggests that the defects involved in GaAs/Si are originated from the early stage nucleation.
