Application of Electron Energy Loss Spectroscopy - Spectrum Imaging (EELS-SI) for Microbe-mineral Interaction (생지구화학적 광물변이작용 연구에서 전자에너지 손실 분광 분석 - 스펙트럼 영상법의 활용)

  • The oxidation states of structural Fe in minerals reflect the paleo-depositional redox conditions for the biologically or abiotically induced mineral formation. Particularly, nano-scale analysis using high-resolution transmission electron microscopy (HRTEM) and electron energy loss spectroscopy (EELS) is necessary to identify evidence for the microbial role in the biomineralization. HRTEM-EELS analysis of oxidation states of structural Fe and carbon bonding structure differentiate biological factors in mineralization by mapping the distribution of Fe(II)/Fe(III) and source of organic C. HRTEM-EELS technique provides geomicrobiologists with the direct nano-scale evidence of microbe-mineral interaction.

Effects of Environmental Factors on Aeromonas spp. Population in Naktong Estuary (낙동강 하구 생태계의 환경요인과 Aeromonas spp. 분포와의 관계)

  • Population of Aeromonas and environmental parameters were investigated at three sites from August 1986, to December, 1986 in Naktong Estuary. The variation range of Aeromonas was $4.3\times10^{2}-4.6\times 10^{4}$ MPN/100ml. The result of ANOVA indicates significant differences among the populations of Aeromonas in each site. The highest population of Aeromonas occurred at site 2, and the lowest at site 3-B. To scrutinize the effects of environmental parameters on the distribution of Aeromonas spp, principal component analysis and multiple stepwise regression were used. The results showed that distribution of Aeromonas spp. was mainly influenced by outflow of freshwater and inflow of inorganic nutrients and correlated with heterotrophic bacteria, available nitrogen, fecal coliform bacteria, and temperature.

  • Sixty paddy soil samples were collected from the different Korean agricultural climatic area to find relationship between soil physicochemical properties and soil microorganisms. The results are summarized as follows : 1. The mean numbers of microorganisms in collected paddy soils were $121.8{\times}10^5$ in bacteria (B), $22.5{\times}10^5$ in actinomycetes (A) and $32.4{\times}10^3$ in fungi (F) per gr soil. The ratios of B/F, B/A, and A/F were 385, 5.1 and 82, respectively. 2. Number of soil microorganism was lowest in Mountainous area, lower in Eastern Coast, Gyeonggi Bay and Chungcheong Continental area, but higher in Honam Plain and Southern Part. 3. The significant positive correlation were obtained between the number of microorganisms and soil chemical properties, available phosphorous, $K^+$, $Mg^{++}$, T-C and soil pH. 4. The number of soil microorganism was in the order of Clay loamy soil > Clayey soil > Loamy soil > Sandy loamy soil.

Phylogenetic diversity of marine bacteria dependent on the port environment around the Ulleng Island (울릉도 항구의 해양환경에 따른 해양미생물의 분포 변화)

  • Pyrosequencing of 16S rDNA tags was used to obtain the bacterial diversity and community structure in the uncultured seawaters as well as in the cultured seawaters, which were collected from the 7 ports (Cheonbu, Hyunpo, Taeha, Namyang, Sadong, Dodong, and Jeodong) and 1 seashore (Guam) around the Ulleng island, Korea. Alphaproteobacteria were the most abundant group in the clean seawaters such as seawaters of Taeha and Sadong ports. Gammaproteobacteria proportion increased depending upon the wastewater amounts mixed with the seawaters such as seawaters of Namyang, Dodong, and Jeodong ports. The genuses of Alteromonas (from samples of Cheonbu, Taeha, Guam, Namyang, Sadong), Shewanella (from sample of Jeodong), and Vibrio (from samples of Hyunpo and Dodong) were dominant group in each of the cultured seawaters incubated in marine broth (Difoco). The results suggest that the incoming wastewaters to the port seawaters contribute to the dynamic change of the marine bacterial community around the Ulleng island.

Microbial Distribution as an Environmental Factor for the Conservation of Cultural Properties in the War Memorial in Korea (전쟁기념관내 문화재 보존을 위한 환경요인으로서의 미생물의 분포)

  • Two isolation methods using the open plate and air sampler were applied to examine microbial distribution as an environmental factor for conservation of cultural properties in the War Memorial in Korea. The numbers of fungi were the abundant in the air of entrance and inside of the exhibition room where visitors were crowded, compared with inside of repository rooms. Fungal and bacterial distributions in the exhibition room during exhibition period were higher than during nonexhibition period, These results are due to the inflow of air current carrying microoganisms to the exibition rooms from outside with visitors. Fungi isolated from the War Memorial were identified as 19 species and one thermophilic fungus, Neurospora crassa. The majority of the isolated fungi are seemed to cause damage to cellulose materials in the Memorial.

Biofilter Treatment of Waste Air Containing Malodor and VOC: 1. Pressure Drop and Microbe-population Distribution of Biofilter with Improved Design (악취 및 VOC를 함유한 폐가스의 바이오필터 처리: 1. 개선된 바이오필터설계에 의한 압력강하와 미생물 population 분포)

  • In this study, both pressure drop and microbe-population-distribution were observed while running a novel biofilter system with improved design in which the biofilter system is composed of two, upper and lower biofilters with both equal feed-rates of up-flow and down-flow, respectively. Then they were compared with the pressure drop and microbe-population-distribution observed in a conventional biofilter of the same effective volume with unidirectional flow. The pressure drop-value of biofilter system with improved design turned out to be less at the incipient stage of run or steady-state long term operation by more than 40~80% of that of the conventional biofilter. The microbe-population-distribution was observed to be lower and higher at higher and lower column of biofilter, respectively, for both the conventional biofilter and the biofilter system with improved design. The microbe-media of waste-tire crumb showed much greater CFU counts than GAC. In the biofilter system with improved design, the $bottom{\rightarrow}up$ feeding of waste air showed greater microbe-population growth than the $top{\rightarrow}down$ feeding for both the microbe-media of waste-tire crumb and GAC. However, it was more prominent for the former than the latter. Comparing the microbe-population-distributions of both of the conventional biofilter and the biofilter system with improved design, the microbe-population of latter was distributed ca. 15 and 2.5 times more evenly for GAC and the media of waste-tire crumb, respectively, than that of former.

  • 갈대(Phragmites communis Trin.)란 염분이 있는 곳에서 자라는 염생식물로서 우리나라 전역에 분포하고 있다. 지난 50년 동안 우리나라의 여러 습지에 걸쳐서 갈대는 우점종으로 자라왔고, 육지와 수중서식지에서 갈대의 확산범위는 증가하고 있다. 갈대의 확산은 다른 습지 식물의 서식지를 파괴하고, 갈대가 번식하면 동 식물들의 번식 자체가 어려울 뿐 아니라 갈대숲에 포식자가 늘어나 살아가기 어려운 환경으로 변하기때문에 갈대를 체계적으로 관리할 수 있는 방안이 마련되어야 한다. 본 연구는 우점종이 다른 두 습지에서 갈대군집의 성장률을 관찰하고, 토양의 화학적 분석과 식물의 생리적 분석을 통해 갈대군집 성장에 미치는 영양염류의 영향을 규명하였다. 연구 대상지는 한강하구에 위치한 장항습지와 성동습지로서 동일하게 갈대가 분포하며, 장항습지에는 줄 군락이 성동습지에는 새섬매자기 군락이 우점하고 있다. 분석 항목은 이화학적 항목을 비롯하여 용존유기탄소(DOC, dissolved organic carbon), 체외미생물효소활성도(Extracellular enzyme activities), 암모니아성 질소($NH_4^+$), 질산성 질소($NO_3^-$)을 분석하였다. 실험결과, 두 습지 갈대의 성장은 7월부터 9월에 증가하였고 성동습지의 토양성분이 점토질로 형성되어 높은 수분함량과 유기물함량을 유지하고 있기 때문에 갈대의 밀도가 높고 성장률이 활발한 것으로 나타났다. 또한 미생물활성과 환경인자간 양의 상관관계를 보아 환경인자들이 미생물 활성을 자극하고 미생물들은 식물의 성장을 촉진하여 영향을 주며, 반면 식물 뿌리는 enzyme을 생성하는 미생물에게 C 삼출물을 공급해 enzyme 활성에 영향을 미칠 것으로 사료된다.

Comparison of Bacterial Diversity in the Water Columns of Goseong Deep Seawaters (고성 심해에서 수심에 따른 해양미생물의 다양성 비교)

  • Microbial diversities in the 300 m and 500 m deep seawaters near Goseong, Gangwon Province (South Korea), were investigated. Pyrosequencing of 16S rRNA genes of marine microbes resulted in 19,474 reads from the 300 m deep seawaters, which consisted of Alphaproteobacteria (57.41%) and Gammaproteobacteria (38.85%), and 82,806 reads from the 500 m deep seawaters, which consisted of Gammaproteobacteria (99.64%) mostly. Rhodobacterales (57.31%) were dominant in the 300 m deep seawaters, but Alteromonadales (45.65%) and Oceanospirillales (34.61%) were dominant in the 500 m deep seawaters. On the bases of operational taxonomic units and diversity indexes (Shannon and Simpson), biodiversity of marine bacteria in the 500 m deep seawaters was shown to be higher than that in the 300 m deep seawaters.

Comparative Characterization of the Bacteria Isolated from Market Milk Treated with ESL and Conventional System (ESL 생산공정에 따른 시유 유래 미생물의 분포 비교 연구)

  • This study was conducted to investigate the characteristics of strains which were isolated from market milk treated with ESL(extended shelf life) and conventional system, and to compare the microbiological quality of ESL milk with conventional milk. In order to characterize the isolated strains, purification, Gram staining, spore staining, catalase, oxidase, motility test, and identification by means of automatic identificator were performed. The results obtained are as follows: total 364 selected strains were analyzed in this study. Depending upon the isolated source, the number of strains from conventional milk was found to be Higher than ESL-milk. By means of grouping of total strains, Bacillus ssp. and Staphylococcus ssp. showed to be predominant. But most of strains were distributed with various groups except Lactobacillus ssp. When the isolates were compared with milk process methods, Enterococcus ssp. was detected much on market milk treated with LTLT pasteurization. Also, Pseudomonas ssp. was detected much on conventional milk treated with UHT pasteurization. By comparison with genus groups depending upon storage temperature of market milk, the higher milk storage temperature increased, the most frequency detected Bacillus ssp. increased. Also, Pseudomonas ssp. was detected most frequently at 10$^{\circ}C$ storage condition. Generally this genus derived from post-contamination during milk processing and related to the quality of market milk during chilled system. In conclusion, it was shown that ESL system reduced post-contamination during milk process, following the improvement of product quality and life cycle during the distribution of market milk.

Distribution of Microorganisms and Foodborne Pathogens in Yukae (육회 중에 분포하는 미생물과 주요 식중독 세균의 조사)

  • People can be exposed to various microorganisms when they eat yukae(seasoned raw meat) because yukae is eaten raw. The purpose of this study was to find out microbial distribution in yukae. In this experiment, 5 restaurants serving yukae were chosen in Daegu area. Mesophilic microorganisms and coliforms were measured by Korean Food Standards Codex. Analyses of Salmonella spp., Listeria monocytogenes, Staphylococcus aureus and Escherichia coli O157:H7 were conducted. As results, mesophilic microorganisms ranged $6.6{\times}10^3-2.7{\times}10^5\;CFU/g$ and coliforms ranged $8.9{\times}10^1-2.1{\times}10^5CFU/g$. Salmonella spp., Listeria monocytogenes and Escherichia coli O157:H7 were not detected in all 5 samples. However, Staphylococcus aureus was detected in 1 sample out of 5 samples. Hygiene practice during production, processing and cooking process is required.
