Nematicidal Activity of Streptomyces flavogriseus KRA15-528 to Meloidogyne incognita (Meloidogyne incognita에 대한 Streptomyces flavogriseus KRA15-528의 살선충활성)

  • Plant disease caused by root-knot nematode is a major problem in crop production. Using of chemical pesticides, one of the most efficient methods to control nematodes, have raised issues in toxicity to humans and animals and environmental pollution. In this study, to select actinomycete strains that have potential to serve as a microbial agent for control of nematodes, we investigated nematicidal activity of culture broth from 670 Streptomyces isolates. A culture filtrate of KRA15-528 isolate that was identified as S. flavogriseus on the basis of 16S rRNA sequence analysis, showed strong nematicidal activity against second stage of juveniles of Meloidogyne incognita and inhibited egg hatching; exposure to 10% of culture filtrate resulted in 71% juvenile mortality at 48 hours afters treatment and suppressed egg hatching by 54% at 9 days after treatment. When the KRA15-528 culture filtrate was partitioned with ethyl acetate and butanol, ethyl acetate layer exclusively showed strong activity; 91%, 53%, 30% of mortality at 1,000, 500, $250{\mu}g/ml$, respectively. Additionally, the culture filtrate suppressed gall formation on cucumber plant by M. incognita with no phytotoxicity. These results suggest that S. flavogriseus KRA15-528 has potential to serve as a microbial nematicide for the control of root-knot nematode disease.

Characteristics and Virulence Assay of Entomopathogenic Fungus Nomuraea rileyi for the Microbial Control of Spodoptera exigua (Lepidoptera: Noctuidae) (파밤나방의 미생물적 방제를 위한 병원성 곰팡이 Nomuraea rileyi의 특성 및 병원성 검정)

  • To date, chemical control remains the most common way to reduce beet armyworm (Spodoptera exigua) populations. However, this insect has become more tolerant or resistant to many chemical insecticides and the insect larvae usually hide inside hollow, tube-like leaves of host plant so they were difficult to kill by spraying insecticides. The use of viral and bacterial insecticide to solve these problems has not been successful because of their novel feeding habit. To overcome these problems, in this study, the biological characteristics and virulence of an entomopathogenic fungus isolated from the cadaver of larvae beet armyworm were investigated. Isolated entomopathogenic fungus was identified as Nomeraea rileyi (Farlow) Samson by morphological examinations and genetic identification using sequences of the ITS, ${\beta}$-tubulin gene and EF1-${\alpha}$ regions. This fungus was named as N. rileyi SDSe. Virulence tests against 3rd larvae of beet armyworm were conducted with various conidial suspensions from $1{\times}10^4$ to $10^8$ conidia/ml of N. rileyi SDSe in laboratory conditions. Mortality rate of beet armyworm showed from 20 to 54% and the virulence increased with increasing conidial concentrations. Although N. rileyi SDSe showed low mortality rate against beet armyworm, it is expected that N. rileyi SDSe will be used effectively in the integrated pest management programs against the beet armyworm.

Biological Control of Anthracnose (Colletotrichum gloeosporioides) in Red Pepper by Bacillus sp. CS-52 (Bacillus sp. CS-52를 이용한 고추 탄저병 (Colletotrichum gloeosporioides) 방제 특성)

  • This study was carried out in order to develop a biological control of anthracnose of red pepper caused by fungal pathogens. In particular, this study focuses on the Colletotrichum species, which includes important fungal pathogens causing a great deal of damage to red pepper. Antagonistic bacteria were isolated from the soil of pepper fields, which were then tested for biocontrol activity against the Colletotrichum gloeosporioides anthracnose pathogen of pepper. Based on the 16S rRNA sequence analysis, the isolated bacterial strain CS-52 was identical to Bacillus sp. The culture broth of Bacillus sp. CS-52 had antifungal activity toward the hyphae and spores of C. gloeosporioides. Moreover, the substances with antifungal activity were optimized when Bacillus sp. CS-52 was grown aerobically in a medium composed of 0.5% glucose, 0.7% $K_2HPO_4$, 0.2% $KH_2PO_4$, 0.3% $NH_4NO_3$, 0.01% $MnSO_4{\cdot}7H_2O$, and 0.15% yeast extract at $30^{\circ}C$. The inhibition of spore formation resulting from cellulase, siderophores, and indole-3-acetic acid (IAA), were produced at 24 h, 48 h, and 72 h, respectively. Bacillus sp. CS-52 also exhibited its potent fungicidal activity against anthracnose in an in vivo test, at a level of 70% when compared to chemical fungicides. These results identified substances with antifungal activity produced by Bacillus sp. CS-52 for the biological control of major plant pathogens in red pepper. Further studies will investigate the synergistic effect promoting better growth and antifungal activity by the formulation of substances with antifungal activity.

  • This experiment was conducted to find out the changes in microflora of submerged soil uncultivated rice plant by application of CNP herbicide (2, 4, 6 - Trichlorophenyl-4-Nitrophenyl ether) under conditions applied with compost, rice straw, glucose or without organic material. The soil, sandy loam textured was incubated in green house for 66 days. Sampling and analysis of microorganisms were carried out during submergence periods. The results were summarilized as follows. 1. Number of aerobic total bacteria was increased by application of CNP herbicide during submerbed 50 days, afterthen, could not seen the difference. The application of rice straw increased number of aerobic bacteria regardless of CNP herbicide application or not, but glucose tended to decrease. 2. Number of Fungi was constantly maintained at $8-20{\times}10^3$ levels during period of submergence regardless of application of CNP herbicide and organic materials or not. 3. The CNP herbicide application tended to decrease the number of actinomycetes, particularly, in the treatments without organic substances and rice straw were remarkably decreased. 4. Anaerobic-and gram-negative bacteria populations were not showed any difference by application of herbicide and organic materials. 5. The ratios of aerobic bacteria to fungi and aerobic bacteria to actinomycetes appeared high values by application of herbicide and of organic substances. 6. At 66 days after submergence, the ratio of chromogenic actinomycetes to the total number of actinomycetes was lowered in application of herbicide. On the other hand, the percentage of both pretense-positive and cellulase-positive actinomycetes to the total isolates were higher in the treatment with herbicide than An without herbicide, particularly. The ratios of pretense-positive actinomycetes were high in the rice straw application regardless of herbicide application or not, but cellulase-positive actinomycetes was not remarkably difference.

  • The multicolored Asian ladybird beetle (Harmonia axyridis) has been commonly used with biological control agents for control of several kinds of aphids in agroecosystems. Also, environment friendly agricultural materials have been commonly applied in crop fields because the government held down pesticide application and environment friendly agricultures are gradually increased with consumer's desires. The multicolored Asian ladybird beetles may be directly or indirectly under the influence of environment friendly agricultural materials In crop fields. The insecticidal environment friendly agricultural materials (IEFAMs) might be saff against each developmental stage of multicolored Asian ladybird beetle. Fungicidal environment friendly agricultural materials (FEFAMs) had a miner effect to each developmental stage of multicolored Asian ladybird beetle with the exception of FEFAM A. Environment friendly agricultural materials contained useful soil microorganisms (EFAMSMs) C and H might be down the hatching rate of eggs, and EFAMSM A and F had a killing effect to 1st instar of lady beetles. Environment friendly agricultural materials contained plant extracts (EFAMPEs) A and D might be suffered effect a deathblow of egg hatching with lady beetles. Otherwise, there was a miner effect to lady beetles with the rest of tested environment friendly agricultural materials.

Evaluation of No-tillage Rice Cover Crop Cropping Systems for Organic Farming (벼 유기농업을 위한 무경운 피복작물 작부체계 평가)

  • The objectives of this study were to evaluate no-tillage cover crop cropping systems for organic farming in paddy. The experiment was performed at Ihyeon series (silt loam: 9.1% sand, 73.0% silt and 17.9% clay) which affected by different management practices. Planthopper population per 20 plant was significantly higher 65.3 for conventional tillage with chemical compounds (control) compared with 3.4~9.6 for no-tillage treatments (without rice straw or green manure, amended with rice straw, hairy vetch, rape, rye, and Chinese milk vetch). Also, disease severity of sheath blight was significantly higher 10.5% for control compared to 0.7~2.9% for no-tillage treatments. Four weed species, namely Monochoria vaginalis, Ludwigia prostrata, Rotala indica, and Aneilema keisak occurred in no-tillage paddy, whereas Monochoria vaginalis occurred in control only. The pH, available phosphate, and microbial biomass C in paddy were steeply decreased in response to submerging, but increased at first heading stage. Soil $NH_4$-N content at first heading stage was significantly higher in no-tillage treatments compared with control. The grain yield was significantly higher in 4.30 Mg $ha^{-1}$ for control than other treatments. Meanwhile, rice productivity was significantly higher in 2.69 Mg $ha^{-1}$ for no-tillage amended with Chinese milk vetch compared to other no-tillage treatments. The number of panicle per plant, grain number perpanicle, and percent ripened grain were highly related for increasing the yield of rice. These results show that Chinese milk vetch was optimum cover crop for organic farming in no-tillage paddy.

Characterization of Bacillus thuringiensis subsp. aizawai CAB109 isolate with bioactivities to Spodoptera litura and Spodoptera exigua (Lepidoptera: Noctuidae) (담배거세미나방과 파밤나방에 활성이 있는 Bacillus thuringiensis subsp. aizawai CAB109 균주의 특성)

  • Bacillus thuringiensis subsp. aizawai CAB109 isolated in Korea is known active against Spodoptera sp.. Especially, B. thuringiensis aizawai CAB109 isolates showed 100% mortality against Spodoptera litura and Spodoptera exigua. To screen highly active B. thuringiensis, the pathogenicity of B. thuringiensis CAB109 was compared with that of commercialized B. thuringiensis products. $LC_{50}$ values of CAB109, product TB-WP and product SC strains of B. thuringiensis were $1.3{\times}10^5$, $2.3{\times}10^6$ and $5.2{\times}10^5\;cfu/ml$ against the 2nd larva of S. litura and $1.8{\times}10^4$, $1.3{\times}10^6$ and $1.5{\times}10^6\;cfu/ml$ against the 2nd larva S. exigua, respectively. To determine new gene's existence and absence, the plasmid DNA was extracted, and compared to that of B.t. aizawai HD-133. Both B. thuringiensis were not like plasmid DNA pattern. PCR technique was used to predict both plasmid DNA's cry gene. PCR products analysis showed that B.t. CAB109 harbor Cry1Aa, Cry1Ab, Cry1C and Cry1D and B.t. HD-133 has Cry1Aa and Cry1Ab, respectively.

Eco-Friendly Organic Pesticides (EFOP)-Mediated Management of Persimmon Pests, Stathmopoda masinissa and Riptortus pedestris (식물 및 미생물 유래 유기농자재 살충효과: 단감해충 감꼭지나방, 톱다리개미허리노린재)

  • Chemical pesticides have been used to control persimmon pests, however the overuse of the pesticides caused insect resistance, followed by failure in pest management and residual problems. Herein we investigate the potential of eco-friendly organic pesticides (EFOP) on the control persimmon pests, Stathmopoda masinissa (persimmon fruit moth) and Riptortus pedestris (bean bug). Ten commercially available plant-derived organic pesticides and one microbial pesticide were sprayed on the target insects in laboratory conditions. The chemical pesticide, buprofezin+dinotefuran wettable powder served as a positive control. In the first bioassay against persimmon fruit moth, alternatively Plutella xylostella larvae were used due to the lack of persimmon fruit moth population from fields, and three organic pesticides showed high control efficacy, such as pyroligneous liquor (EFOP-1), the mixture of Chinese scholar tree extract, goosefoot and subtripinnata extracts (EFOP-2) and Bacillus thuringiensis subsp. aizawai NT0423 (EFOP-11). When the three selected organic pesticides were treated on the persimmon fruit moths, the EFOP-2 treatment showed the highest control efficacy: 27.7% (5 days), 13.3% (7 days) and 6.7% (10 days) of survival rates. In the bioassay against bean bugs, the mixture of Chinese scholar tree, goosefoot and subtripinnata extracts (EFOP-2 and EFOP-9) and the extracts of sophora and derris (EFOP-10) showed high control efficacy, particularly the highest in the treatment of EFOP-2: 20.0% (5 days) and 16.7% (10 days) of survival rates. These results suggest that the mixture of Chinese scholar tree, goosefoot and subtripinnata extracts (EFOP-2) has high and multiple potential in the management of the persimmon pests.

Degradation of Poultry Feathers by Bacillus amyloliquefaciens Y10 With Plant Growth-promoting Activity and Biological Activity of Feather Hydrolyzates (식물 성장 촉진 활성을 가진 Bacillus amyloliquefaciens Y10에 의한 가금 우모의 분해 및 생산된 우모 분해산물의 생리활성)

  • This study was conducted to characterize strain Y10, isolated from discarded chicken feathers. Strain Y10 was identified as Bacillus amyloliquefaciens through phenotypic and 16S rRNA gene analysis. B. amyloliquefaciens Y10 exhibited plant growth-promoting activities, including the production of fungal cell-degrading enzymes (cellulase, lipase, protease, and pectinase), siderophores, ammonia, and indoleacetic acid. Furthermore, strain Y10 was able to inhibit the mycelial growth of several phytopathogenic fungi. When 0.1% sucrose as a carbon source and 0.05% casein as a nitrogen source were added to the basal medium, adjusted to pH 10, and cultured at 35℃, the degradation rate of chicken feathers by strain Y10 was about two times higher than that of the basal medium, with the feathers almost completely degraded in four days. Strain Y10 also degraded various keratin substrates, including duck feathers, wool, and human nails. It was confirmed that the feather hydrolyzates prepared using strain Y10 exhibited antioxidant activities, such as 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity (EC50 = 0.38 mg/ml) and superoxide dismutase-like activity (EC50 = 183.7 mg/ml). These results suggest that B. amyloliquefaciens Y10 is a potential candidate for the development of bioinoculants and feed additives applicable to the agricultural and livestock industries, as well as the microbiological treatment of keratin waste.

Selection and appropriate culture conditions of antagonistic bacterium Bacillus altitudinis HC7 against button mushroom cobweb disease caused by Cladobotryum mycophilum (양송이버섯 솜털곰팡이병균(Cladobotryum mycophilum)에 대한 길항미생물 Bacillus altitudinis HC7의 선발 및 적정 배양조건)

  • This study was conducted to selection and investigate appropriate conditions for mass production of antagonistic microbes to control cobweb disease caused by Cladobotryum mycophilum. A grampositive bacterium was isolated from spent substrate of Agaricus bisporus and showed significant antagonistic activity against Cladobotryum mycophilum. The bacterium was identified as Bacillus altitudinis. based on the cultural, biochemical and physiological characteristics, and 16S rRNA sequence. The isolate is saprophytic, but not parasitic nor pathogenic to cultivated mushroom whereas it showed strong inhibitory effects against C. mycophilum cells in vitro. The control efficacy of B. altitudinis HC7 against cobweb disease of C. mycophilum was up to 78.2% on Agaricus bisporus. The suppressive bacterium may be useful for the development of biocontrol system. To define the appropriate conditions for the mass production of the Bacillus altitudinis HC7, we have investigated appropriate culture conditions and effects of various nutrient source on the bacterial growth. The appropriate initial pH and temperature were determined as pH 6.0 and 30℃, respectively. The appropriate concentration of medium elements for the growth of pathogen inhibitor bacterium(Bacillus altitudinis HC7) was determined as follows: 3.0% soluble startch, 10% soytone, 1.0% (NH4)2HPO4, 1.0 mmol KCl, and 0.5% L-asparagine.
