연안에 가까운 광양만 퇴적토를 대상으로 하여 PAHs 오염도와 말단제한절편 다형성 분석방법(T-RFLP method)을 이용하여 미생물 군집구조를 조사하였다. 조사된 10개 정점의 PAHs오염도는 80$\pm$20~5,690$\pm$580 (평균 1,499$\pm$1,716) ng/g dry wt.였으며 폐수종말처리장이 위치하는 하천 입구에 위치한 정점 (Wl)과 광양제철소 우수배출구 입구에 위치한 정점에서 상대적으로 높은 농도를 보였다. T-RFLP방법으로 조사한 미생물 군집은 10개 정점이 4개의 그룹으로 구분되었으며 정점 Wl에서 다른 정점과 다른 독특한 군집구조를 형성하였다. PAHs 농후배양시료에서의 미생물군집구조 변화와 비교해 볼 때 조사대상지역 퇴적토의 미생물 군집구조는 PAHs의 오염에 부분적으로는 영향을 받지만 입도, 유기물 등과 같은 환경요인이 군집구조를 결정하는 주된 요인인 것으로 생각된다.

Microorganisms are key-role player for stabilization of landfill sites. In order to evaluate the availability of T-RFLP(Terminal Restriction Fragment Length Polymorphism) for monitoring microbial community variations during stabilization of landfill sites, the phylogenic diversity of microbial community in the leachate from 4 different full-scale landfills was characterized by T-RFLP based on bacterial 16S rDNA. Main population of microbial community analyzed by T-RFLP was significantly similar with that of microbial community analyzed by clone library analysis. The results of T-RFLP analysis for main population of microbial community in the leachate from landfills with different landfill structures, waste types and landfill ages showed apparently different microbial diversity and structures. Therefore, long-term monitoring of microbial community in leachate from landfill sites by using T-RFLP is expected to be available for evaluation of landfill stability.
Effect of polycyclic aromatic hydrocarbons (PAHs) on the community structure of indigenous microorganisms in Gwangyang Bay sediments was investigated in Mar. & Aug.,2000. Microbial community structure was analyzed using 5'-terminal restriction fragment length polymorphism (T-RFLP) method. Microbial community structure based on T-RFLP method revealed that community differentiated by sampling period except station 1 located near the stream discharge site from Yeosu Industrial Complex. Even, microbial diversity was higher at stations showed relatively high concentrations of PAHs. The microbial community structure was severely changed during the enrichment culture with 1,000 ppm of PAHs mixture. It was also different between cultivated at 8$^{\circ}C$ and 30$^{\circ}C$. The results implied that temperature, poyosity, organic content and etc were more responsible than PAHs on the microbial community structure.
16S rRNA를 분석한 연구들은 자연 생태계에서 추출한 핵산을 이용하여 16rRNA 유전자의 염기서열을 분석하거나 특정 DNA probe를 이용한 hybridization 실험이 주류를 이루어 왔다. 특히 PCR 기법이 개발됨에 따라 적은 양의 시료를 대량으로 손쉽게 증폭시킬 수 있어 다양한 분야에 응용되고 있다. 세균 군집의 구조를 이해하는데 있어서 PCR 방법의 적용 대상은 주로 16S rRNA 유전자의 염기서열 해독분야이며 해양 생태계를 대상으로 많은 연구 결과가 보고되었다(11,13,21,26). 한편 자연 생태계의 개별적 미생물 분류룬들을 검출하기 위한 특정 oligonucleotide probe의 개발방법들은 미생물 군집의 유전적 다양성에 대한 정보 파악 이외에 배양이 어려운 혐기성 세균과 같은 특정 세균들의 동정에도 이용되고 있다(3,24,55). 본고에서는 세균 군집의 구조와 다양성을 연구하는데 적용 가능한 rRNA 분석방법들을 수계 생태계를 중심으로 살펴보고자 한다.

This study compared and analyzed the microbial composition and physicochemical characteristics of kimchi (gimjang kimchi) and mukeunji (aged kimchi). Commercial kimchi and mukeunji products were purchased through an online market. After an analysis of physicochemical characteristics, the pH of the mukeunji samples was found to be lower and the acidity higher than in the kimchi samples. There was no significant difference in salinity between kimchi and mukeunji, but the sugar content was higher in the kimchi samples. The phylogenetic diversity index, which incorporates phylogenetic difference between species, was significantly higher in mukeunji than in the kimchi. The most dominant order in both groups was Lactobacillales, but several lactic acid bacteria, such as the Pediococcus and Lactobacillus species, which may be more acid tolerant or more competitive, are relatively predominant in mukeunji. Beta set-significance analysis based on two different distance metric results revealed that microbial distributions of population were different at the statistical confidence level (p<0.001). We investigated the effect of respective species on total microbial community using the LEfSe (linear discriminant analysis effect size) mechanism. According to the results of LEfSe testing, a relatively higher abundance of Weissella kandleri in kimchi and a higher abundance of Pediococcus inopinatus in mukeunji have the greatest influence on the differences in microbial structure between the two groups.

Despite the importance of traditional Korean fermented foods, little is known about the microbial communities and diversity of fermented soy products. To gain insight into the unexplored microbial communities of both Doenjang (DJ) and Ganjang (GJ) that may contribute to the fermentation in Korean traditional foods, we carried out next-generation sequencing (NGS) based on the V3-V4 region of 16S rDNA gene analysis. The alpha diversity analysis results revealed that both the Shannon and Simpson diversity indices were significantly different between the two groups, whereas the richness indices, including ACE, CHAO, and Jackknife, were not significant. Firmicutes were the most dominant phylum in both groups, but several taxa were found to be more abundant in DJ than in GJ. The proportions of Bacillus, Kroppenstedtia, Clostridium, and Pseudomonas and most halophiles and halotolerant bacteria, such as Tetragenococcus, Chromohalobacter, Lentibacillus, and Psychrobacter, were lower in DJ than in GJ. Linear discriminant effect size (LEfSe) analysis was carried out to discover discriminative functional biomarkers. Biomarker discovery results showed that Bacillus and Tetragenococcus were identified as the most important features for the classification of subjects to DJ and GJ. Paired-permutational multivariate analysis of variance (PERMANOVA) further revealed that the bacterial community structure between the two groups was statistically different (p=0.001).

This study examined microbial community structures (MCSs) according to environmental factors through DGGE analysis and comparison in various soils collected from near army bases and gas stations. As a result, the similarities based on DGGE band profiles showed the closer relationship in regional properties than in pollution characteristics, probably due to the degree of weak contamination. The highly contaminated samples with oil revealed low MCS similarities with others in the same region and very low with all the other samples in the other regions. Thus the microbial community structure would more be affected by region-based natural factors than by contamination factors in case of minor pollution. All the dominant culturable bacterial species were involved in firmicutes or high GC Gram+ in a major portion of soil samples and the highly oil-contaminated samples contained Arthrobacter, Bacillus, Methylobacterium, Clavibacter, Streptomyces and Nocardia as reported genera, and Leifsonia as a unreported genus.
In this study, the structure and diversity of bacterial community were investigated in the surface and subsurface marine sediments using a NGS method (i.e. illumina sequencing technology). The bacterial community in the surface was distinct from that in the subsurface of marine sediment; with the exception of the phylum Proteobacteria, the relative abundance of Bacteroides phylum were higher in the surface than subsurface, whereas the sequences affiliated to the phyla Chloroflexi and Acidobacteria were relatively more copious in the subsurface than surface sediment. Moreover, interestingly, we observed that the phyla Nitrospinae and Nitrospirae contribute to nitrogen cycle in the marine sediment. This study may present the possibility for the presence of novel microorganisms as unexplored sources and provide basic information on the microbial community structure.

Global climate changes including elevated $CO_2$, drought, and global warming may influence greenhouse gas emissions in wetlands. A variety of microbial communities including denitrifiers and methanogens play a key role in determining such processes. In this paper we summarize current knowledge on the effects of climate changes on $CO_2,\;CH_4$, and $N_2O$ production and microbial communities mediating those processes in wetlands. Elevated atmospheric $CO_2$ and warming generally increase gas emissions, but effects of droughts differ with gas type and drying level. The responses of microbial community to climate changes in terms of composition, diversity and abundance are still in question due to lack of studies in wetlands. Based on the present review, it is suggested that future studies on microbial processes should consider microbial community and relationships between microbial function and structure with diverse environmental factors including climate changes. Such knowledge would be crucial to better understand and predict accurately any shifts in ecological functions of wetlands.
This review inquired the recently applied molecular biological and biochemical methods analyzing the microbial community structure of groundwater and, as a result, summarized the functional or taxonomic groups of active microorganisms with major contaminants in groundwater. The development of gene amplification through PCR has been possible to figure out microbial population and identification. Active microbial community structures have been analyzed using a variety of fingerprinting techniques such as DGGE, SSCP, RISA, and microarray and fatty acid analyses such as PLFA and FAME, and the activity of a specific strain has been examined using FISH. Also, this review included the dominant microflora in groundwater contaminated with fuel components such as n-alkanes, BTEX, MTBE, and ethanol and chlorinated compounds such as TCE, PCE, PCB, CE, carbon tetrachloride, and chlorobenzene.
