Confirmation of Chestnut Powdery Mildew, Erysiphe castaneigena, in Korea with Morphological Characteristics and Molecular Analyses (형태적 특징 및 분자적 분석에 의한 밤나무 흰가루병균 Erysiphe castaneigena의 확인)

  • The powdery mildew on Castanea crenata in South Korea was first recorded as Microsphaera alni in 1958. On the basis of its morphological characteristics and host range, the mildew was determined as Microsphaera sinensis in 1988. According to the rules of The International Code of Botanical Nomenclature, M. sinensis was renamed Erysiphe castaneigena in 2006. Nevertheless, taxonomic re-consideration of the morphological and molecular characteristics for the Castanea powdery mildew in Korea has not been performed. In the present study, we studied 34 powdery mildew samples collected from C. crenata in the Korea University Herbarium. On the basis of microscopic examinations of the holomorph and phylogenetic analysis of the internal transcribed spacer regions, we confirmed the powdery mildew fungus as E. castaneigena. In addition, sequence comparison between E. castaneigena and E. alphitoides ex Castanea sp. suggested a close phylogenetic affinity.

Isolation of $\alpha$-Amylase Hyperproducing Strain HG4 from Bacillus sp. and Some Properties of the Enzyme ($\alpha$-Amylase 생산성이 높은 Bacillus sp. HG4의 분리 및 효소 특성)

  • An $\alpha$-amylase producing bacterium, strain 2B, was isolated from soil and identified to genus Bacillus. To enhance $\alpha$-amylase productivity, strain 2B was mutagenized successively with nitrosoguanidine. For an efficient selection of a-amylase hyperproducers, mutants which produced $\alpha$-amylase in the presence of glucose were isolated. The resultant mutant HG4, which was classified as constitutive and catabolite derepressed hyperproducer of a-amylase, produced about 30 folds more $\alpha$-amylase than parental strain in medium containing lactose as carbon source. The strain HG4 grew rapidly and produced enzyme in parallel with cell growth. Moreover, its cell lysis did not occur until time of maximal yield of enzyme, which was considered to be a favorable characteristic for the production and purificiation of enzyme in industrial scale. The enzymatic properties of parental strain 2B and mutant strain HG4 were almost the same. The optimal temperature and pH for enzyme reaction was $70^{\circ}C$ and pH 6.0, respectively, in 'the presence of 0.6mM $Ca^[2+}$ as an effective stabilizer.

Environmental Impact Evaluation for the Power Generation System Using the LCA Methodology (LCA 기법을 이용한 발전시스템의 환경성 평가)

  • In this study, life cycle assessment(LCA) for the nuclear power generation system and the thermal power generation system, which make a great distribution of the electric power supply in Korea, has been carried out to compare the environmental impact between two power generation systems. In system boundary of this study, the stage of construction, operation and demolition & disposal were included. For life cycle impact assessment(LCIA), three cases were considered; the single environmental impact for the $CO_2$ emissions, the 8 major global environmental impact, and the major global environmental impact categories including radioactive impact. As the results, it was found that the nuclear power generation system is environmentally superior to the thermal power generation system as 10,000 times in the evaluation for the $CO_2$ emissions, 90 times in the evaluation for the 8 major environmental impact categories, and 40 times in the evaluation for the environmental impact categories including radioactive impact.

A New Image Completion Method Using Hierarchical Priority Belief Propagation Algorithm (계층적 우선순위 BP 알고리즘을 이용한 새로운 영상 완성 기법)

  • The purpose of this study is to present a new energy minimization method for image completion with hierarchical approach. The goal of image completion is to fill in missing part in a possibly large region of an image so that a visually plausible outcome is obtained. An exemplar-based Markov Random Field Modeling(MRF) is proposed in this paper. This model can deal with following problems; detection of global features, flexibility on environmental changes, reduction of computational cost, and generic extension to other related domains such as image inpainting. We use the Priority Belief Propagation(Priority-BP) which is a kind of Belief propagation(BP) algorithms for the optimization of MRF. We propose the hierarchical Priority-BP that reduces the number of nodes in MRF and to apply hierarchical propagation of messages for image completion. We show that our approach which uses hierarchical Priority-BP algorithm in image completion works well on a number of examples.

Cross-layer Optimization for Multichannel Multiradio Multisession Wireless Networks with Network Coding (멀티채널 멀티라디오 멀티세션 무선 네트워크를 위한 네트워크 코딩 기반 계층간 최적화 기법)

  • Network coding has been extensively studied to increase the network throughput by exploiting the broadcast and overhearing capabilities. In this paper, we formulate a network utility maximization problem to improve the network utility in multichannel, multiradio and multisession wireless multihop network with intrasession network coding. To find the solution of this problem, we propose a congestion control, distributed rate control, and heuristic resource allocation algorithm. We find the network utility and evaluate the performance in multichannel, multiradio and multisession environment by using MATLAB. Finally, the results show that the proposed schemes can achieve throughput improvement by performing the network utility optimization in wireless multihop network.

Characteristics of Hydrogen Generation from Methanol and Ethanol using Cylindrical Barrier Discharge (실린더형 무성방전을 이용하여 메탄올과 에탄올로부터 수소발생 특성)

  • Hydrogen is sustainable energy without environment pollution. In this study, experiments and analysis of hydrogen generation from gases methanol and ethanol using cylindrical barrier discharge reactor was carried out. The discharge reactor to generate hydrogen molecules used in this work is one type of Non-thermal Plasma (NTP) reactors and neon-transformer as power source to make a plasma was used. Hydrogen concentrations were measured as parameters of applied voltage, concentrations of methanol and ethanol, and flow rates of carrier gases($N_2$). Hydrogen generation increased according to applied voltage and produced largely in case of methanol compared with ethanol. It is thought that the reason is deeply related with those different chemical structures. Energy yield of hydrogen generation in case of ethanol decreases according to increasing applied voltage, but that in case of methanol has a peak at applied voltage of 22[kV] and decreased. Specifically, hydrogen generation increased with increasing applied voltage, but low voltage was better, which is the best parameter in the aspects of energy efficiency.

Identification of Podosphaera xanthii as the causal agent of powdery mildew disease affecting Echinacea purpurea in Korea (에키나시아 흰가루병을 일으키는 Podosphaera xanthii 동정)

  • During the extensive forays for discovering the biodiversity of phytopathogenic fungi in Korea, powdery mildew-affected purple coneflowers (Echinacea purpurea) were frequently found. Since 2004, nine samples have been deposited in the Korea University Herbarium. The initial symptoms include white, evanescent mycelia and irregular patches on mature leaves. Mild symptoms in the form of discolored lesions are also noticed on the stem and petal of the affected plants. Based on the morphological characteristics of the anamorph and the phylogenetic analysis of the internal transcribed spacers (ITS) and large subunit (LSU) rDNA sequences, the causal fungus was identified as Podosphaera xanthii. This is the first report that identifies the causal agent of powdery mildew disease affecting purple coneflower in Korea.

  • This study was conducted to analyze the pattern of growth and phenological characteristics of Mankyua chejuense B.-Y. Sun, M.H. Kim & C.H. Kim, which belong to the Ophioglossaceae family. M. chejuense asexually reproduced using rhizomes, and a clone of the species grew up to 52 cm based on root growth and new leaves was produced from rhizomes. The development of leaves were divided into four stages; leaf emergence-separation and growth of leaf-sporophyll maturation-senescence. Most leaves emerged in July and August with high temperatures and precipitation and most leaves reached full expansion during September-October and died during April-May next year. The life span of leave was 10 months, from July to April, and the start of leaf senescence varied depending on the habitat environment and this might result from micro environmental differences among the habitats.

  • Aspergillus niger van Tieghem was cultured by the method of synchronous and submerged culture. Throughout the culture, sporulation was occured. Highly phosphorylated nucleotides in sporulating mycelia were detected to assure whether the eucaryotic Aspergillus niger produce these substances or not during the differentiation. Phosphorylated nucleotides were extracted from the conidiophore, bearing mycelia and spore forming body, these nucleotides were identified by TLC with P.E.I. cellulose. Guanosine tetraphosphate was found in both phialide forming mycelia and spore forming body. The contents of free amino acids were assayed and its level was found to increase at early stage of sporulation. The effects of 8-azaguanine examined, it was found to prevent spore formation and to made abnormal structure. The effects of inosinic monophosphate and guanosine monophosphate on spore formation were examined, spore formation was enhanced by these nucleotides.

Effect of Extract of Fermented Dropwort on Intestinal Bacteria and Enzymes In Vitro (미나리발효액이 장내 유해세균 및 유익균의 In Vitro 생육 및 효소활성에 미치는 영향)

  • Effect of extract of fermented dropwort (Oenanthe stolonifera) on growth of intestinal harmful/useful bacteria and enzyme activity were investigated in vitro. The extract showed strong inhibition on harmful microbes including Vibrio and Salmonella, but mild inhibition on Bifidobacterium longum in both agar plate and liquid cultivation. Minimum inhibitory concentration (MIC) value of B. longum was the highest among tested microbes. Inhibition effect of fermented extract on harmful microbes increased according to fermentation period. Extract of fermented dropwort showed inhibitory effects on activity of microbial ${\beta}$-glucuronidase and tryptophanase. The inhibitory effects were also proportional to fermentation period. As consequence, it is assumed that the uptake of fermented dropwort might be useful for human intestinal health.
