Improving Efficiency and Effectiveness of Disinfection by Soaking Seeds before Rice Seed Disinfection (벼 종자소독 전 침종에 따른 소독효과 및 효율성 향상)

  • Seed disinfection is a necessary process to decrease Bakanae disease in rice and is carried out at 30℃ to increase disinfection efficiency. However, the length of sprout after seed disinfection is not suitable for sowing, so additional processes such as seed soaking is needed. For this reason, this study was conducted to develop the efficient and effective seed disinfection method. When the seed disinfection finished after soaking seeds in cold water for 2 days, length of sprout was about 1.0 mm and sowing can be done without additional processes. In addition, the incidence of Bakanae disease was greatly reduced by 64% compared to conventional method. In order to find out why the newly developed method (disinfection after soaking) is more efficient than conventional method, soaked and not soaked seeds were stained using methylene blue staining. It was found that soaked seeds were more easily stained with about 50% of internal and external endosperm stained. Therefore, pesticide can be absorbed easily into internal endosperm of soaked seeds and efficiently destroy the spore of pathogen. As a results of this study, newly developed method will be useful as it can improve efficiency of agricultural work and also effectively decrease Bakanae disease compared to conventional method.

An in Vitro Study of the Effects of Different Dentin Bonding Agents on the Prevention of Tooth Discoloration and the Sealing Ability of Calcium Silicate-Based Cement in Regenerative Endodontic Procedures (치수재혈관화 술식에서 다양한 상아질 접착제가 치아 변색과 칼슘 실리케이트 시멘트의 밀폐 효과에 미치는 영향에 대한 실험실적 연구)

  • This study aimed to compare the effects of applying different types of dentin bonding agents (DBAs) on tooth and on the sealing ability of CSC in regenerative endodontic procedures (REPs). Specimens were made from 120 human first premolars and categorized into the following groups based on the presence and the type of DBA: Group I, no bonding; Group II, One-Step; Group III, Clearfil SE Bond; and Group IV, Single Bond Universal. Subsequently, the groups were categorized into Subgroup a and Subgroup b, in which Endocem MTA® Premixed Regular and Biodentine were used, respectively. The CIE L*a*b* on the photos taken prior to TAP placement (S0), 1 week after TAP placement (S1), 2 weeks after TAP placement (S2), 4 weeks after TAP placement (S3) and 2 weeks after CSC placement (S4) was used to assess discoloration. The samples were submerged in a 2% methylene blue solution and magnified by 30 times under a stereomicroscope to assess microleakage. The application of DBA during REP reduced crown discoloration, while Biodentine showed minimal discoloration with or without DBA and regardless of the DBA type. Additionally, the application of DBA did not increase microleakage.

Effective Biodegradation of Polyaromatic Hydrocarbons Through Pretreatment Using $TiO_2$-Coated Bamboo Activated Carbon and UV ($TiO_2$로 코팅된 대나무숯 및 UV의 전처리를 통한 다환방향족탄화수소의 효율적 생분해)

  • $TiO_2$-coated bamboo activated carbon has been prepared and utilized under UV irradiation as a pretreatment method for an effective biodegradation of the recalcitrant polyaromatic hydrocarbons (PAHs). The anatase $TiO_2$ was successfully coated on the bamboo activated carbon (AC) and it showed the highest photoactivity against methylene blue. In the absence of the PAHs-degrading bacteria PAHs having low molecular weight (i.e., naphthalene, acenaphthylene, acenaphthene, and fluorene) were degraded by 9.8, 76.2, 74.1, and 40.5%, respectively. Higher molecular weight PAHs, however, maintained high residual concentrations of PAHs (400-1,000 ${\mu}g$/L) after the same treatment. On the other hand, the overall concentrations of PAHs became lower than 340 ${\mu}g$/L when the pretreated PAHs were subjected to biodegradation by a PAH-degrading consortium for a week. Herein, phenanthrene, anthracene, fluoranthene, and pyrene were removed by 29.3, 61.4, 27.0, and 44.3%, respectively, indicating the facilitated potential biodegradation of PAHs. Activated carbon coated with $TiO_2$ appeared to inhibit growth of PAH degraders on the surface of AC, indicating planktonic degraders were dominantly involved in the PAH biodegradation in presence of the $TiO_2$-coated bamboo AC. It was proposed that an effective remediation technology for the recalcitrant PAHs could be developed when an optimum pretreatment process is further established.

The Effect of Glide Path on Canal Centering Ability in Reciprocating File System (Reciprocating 파일 시스템에서 Glide Path가 근관만곡도 유지에 미치는 영향)

  • The purpose of this study was to evaluate the influence of glide path on canal centering ratio after instrumentation with different single file systems; WaveOne and Reciproc. Reciproc R25 (VDW), WaveOne Primary (Dentsply Maillefer) and PathFile #13, 16, 19 (Dentsply Maillefer) were used in this study. In no glide path groups, Reciproc files and WaveOne files used for canal preparation without glide path. In glide path groups, the PathFile were used before canal preparation. Methylene blue dye was introduced into the canal to obtain a clear pre-instrumentation image. Pre-instrumentation images and post-instrumentation images were scanned using Epson Perfection V700 Photo scanner (Epson, Nagano, Japan). Transparencies of post-instrumentation images were changed and superimposed on pre-instrumentation images using Adobe Photoshop CS 3 (Adobe Systems Incorporated, San Jose, CA, USA). The centering ratio was calculated for each instrumented canal using the following formula: CR=|X1-X2|/Y. It was statistically analyzed using two-way ANOVA at 95% confidential level. The centering ratio in glide path groups were significant less than it in no glide path groups at 3, 4, 5 and 6 mm level. Except 1 and 6 mm level, WaveOne groups had significant less centering ration than Reciproc groups. At 6 mm level, there was no significant difference between WaveOne and Reciproc. In the limitation of this study, creation of a previous glide path before reciprocating motion instrumentation in curved canal appears to be appropriate and WaveOne system can be used for preparation of curved canal without severe aberrations.

Microleakage of endodontically treated teeth restored with three different esthetic post and cores (심미적 포스트 코어의 종류에 따른 미세누출에 관한 연구)

  • Statement of problem: At present, as the esthetic demands are on the increase, there are many ongoing studies for tooth-colored post and cores. Most of them are about fiber post and prefabricated zirconia post, but few about one-piece milled zirconia post and core using CAD/CAM (computer-aided design/computer-aided manufacturing) technique. Purpose: The objective of this study was to compare microleakage of endodontically treated teeth restored with three different tooth-colored post and cores. Material and methods: Extracted 27 human maxillary incisors were cut at the cementoenamel junction, and the teeth were endodontically treated. Teeth were divided into 3 groups (n=9); restored with fiber post and resin core, prefabricated zirconia post and heat-pressed ceramic core, and CAD/CAM milled zirconia post and core. After the preparation of post space, each post was cemented with dual-polymerized resin cement (Variolink II). Teeth were thermocycled for 1000 cycles between $5-55^{\circ}C$ and dyed in 2% methylene blue at $37^{\circ}C$ for 24 hours. Teeth were sectioned (bucco-lingual), kept the record of microleakage and then image-analyzed using a microscope and computer program. The data were analyzed by one-way ANOVA and Scheffe's multiple range test (${\alpha}=0.05$). Results: All groups showed microleakage and there were no significant differences among the groups (P>.05). Prefabricated zirconia post and heat-pressed ceramic core showed more leakage in dye penetration at the post-tooth margin, but there was little microleakage at the end of the post. Fiber post and resin core group and CAD/CAM milled zirconia post and core group indicated similar microleakage score in each stage. Conclusion: Prefabricated zirconia post and heat-pressed ceramic core group demonstrated better resistance to leakage, and fiber post and resin core group and CAD/CAM milled zirconia post and core group showed the similar patterns. The ANOVA test didn't indicate significant differences in microleakage among test groups. (P>.05)

MARGINAL LEAKAGE OF COMPOSITE RESIN AND COMPOMER RESTORATIONS CURED WITH THREE DIFFERENT LIGHT SOURCES (광중합기 종류에 따른 복합레진과 콤포머의 미세누출에 관한 연구)

  • The purpose of this study was to evaluate the microleakage of composite resin($Z250^{(R)}$) and compomer(Dyract AP) cured with various curing lights(Elipar $TriLight^{(R)}$, $Flipo^{(R)}$, and Elipar FreeLight $2^{(R)}$). Box-shaped class V cavities were prepared on the buccal and lingual surfaces of extracted human third molars. The sectioned specimens were evaluated for dye penetration and following results were obtained. 1. Regarding microleakage in the enamel margin with different curing light sources, there was no significant difference in Dyract AP whereas $Z250^{(R)}$ with $Flipo^{(R)}$ showed higher microleakage score than those of FreeLight $2^{(R)}$ or $TriLight^{(R)}$(p<0.05). 2. Regarding microleakage in the dentin margin with curing light sources, there were no significant differences in both Dyract AP and $Z250^{(R)}$(p>0.05). 3. Regarding microleakage in the enamel margin with filling materials. there were no significant differences in both filling materials with $Flipo^{(R)}$ whereas $Z250^{(R)}$ showed less microleakage than Dyract AP with FreeLight $2^{(R)}$ or $TriLight^{(R)}$ (p<0.05). 4. Regarding microleakage in the dentin margin with filling materials, there wer no significant differences in both filling materials with $TriLight^{(R)}$ whereas $Z250^{(R)}$ showed less microleakage than Dyract AP with FreeLight $2^{(R)}$ or $Flipo^{(R)}$ (p<0.05). 5. Enamel margin showed less microleakage than dentin margin when filled with Dyract AP and cured with $Flipo^{(R)}$ or FreeLight $2^{(R)}$ (p<0.05), but there were no statistically significant differences between the enamel and the dentin in the rest groups. This study suggested that $Z250^{(R)}$ showed lower microleakage score than Dyract AP. and that $Flipo^{(R)}$ showed higher microleakage score than FreeLight $2^{(R)}$ and $TriLight^{(R)}$ in the enamel margin filled with $Z250^{(R)}$.

The Correlation between the Number of Fungiform Papillae and Electrogustometric Thresholds in Adults (젊은 성인에서 심상유두개수와 전기미각역치의 상관성)

  • The aims of this study were to establish the effect of gender and age on number of fungiform papillae and electrogustometric thresholds of the tongue, and to determine the relation between number of fungiform papillae and electrogustometric threshold. Sixty-two healthy subjects of 20 to 39 years old were enrolled in this study. Examination areas were defined as center, anterior, left lateral, right lateral, and posterior on the dorsal surface of the tongue, which was stained with methylene blue dye and photographed with a scale bar on it. The number of fungiform papillae within $1\; cm^2$ square area of each examination site was counted twice. The counted values were averaged to produce means. Taste thresholds were measured three times on each examination area using an electrogustometer. Means of the threshold values were calculated. 1. The number of fungiform papillae was the largest at the anterior examination area, followed by right lateral, left lateral, posterior and central. Women had more fungiform papillae than men at anterior, right lateral, and center areas. Subjects in twenties showed more fungiform papillae than those in thirties at anterior, right lateral, and left lateral areas. 2. Anterior area of the tongue measured the lowest electrogustometric threshold, followed by left lateral, right lateral, posterior and center. The threshold was higher in men at anterior and right lateral areas. The subjects in thirties had higher threshold only at right lateral area than those in twenties. 3. Electrogustometric threshold was inversely correlated with the number of fungiform papillae at anterior and posterior areas. The results showed that the number of fungiform papillae and taste threshold varied according to tongue locus, and they were influenced by age and gender. The results also suggested that the number of fungiform papillae was related to electrogustometric threshold inversely.

INFLUENCE OF REBONDING PROCEDURES ON MICROLEAKAGE OF COMPOSITE RESIN RESTORATIONS (복합레진 수복 시 재접착 술식이 미세누출에 미치는 영향)

  • During a composite resin restoration, an anticipating contraction gap is usually tried to seal with low-viscosity resin after successive polishing, etching, rinsing and drying steps, which as a whole is called rebonding procedure. However, the gap might already have been filled with water or debris before applying the sealing resin. We hypothesized that microleakage would decrease if the rebonding agent was applied before the polishing step, i.e., immediately after curing composite resin. On the buccal and lingual surfaces of 35 extracted human molar teeth, class V cavities were prepared with the occlusal margin in enamel and the gingival margin in dentin. They were restored with a hybrid composite resin Z250 (3M ESPE, USA) using an adhesive AdperTM Single Bond 2 (3M ESPE). As rebonding agents, BisCover LV (Bisco, USA), ScotchBond Multi-Purpose adhesive (3M ESPE) and an experimental adhesive were applied on the restoration margins before polishing step or after successive polishing and etching steps. The infiltration depth of 2% methylene blue into the margin was measured using an optical stereomicroscope. The correlation between viscosity of rebonding agents and mciroleakage was also evaluated. There were no statistically significant differences in the microleakage within the rebonding procedures, within the rebonding agents, and within the margins. However, when the restorations were not rebonded, the microleakage at gingival margin was significantly higher than those groups rebonded with 3 agents (p < 0.05). The difference was not observed at the occlusal margin. No significant correlation was found between viscosity of rebonding agents and microleakage, except very weak correlation in case of rebonding after polishing and etching at gingival margin.

Comparison of apical sealing ability of continuous wave of obturation technique using EndoTwinn and System B (EndoTwinn과 System B continuous wave of obturation units를 이용한 근단부 근관충전 효율비교)

  • The purpose of this study was to compare the apical leakage of the root canal filled with the System B and the EndoTwinn(the combined application of heat and ultrasonic vibration). Sixty extracted premolars with straight root were cleaned and shaped to size 35. Group SB was obturated using System B and Group ET was filled with EndoTwinn. A size 35 of 0.06 tapered gutta-percha and Adseal were used and the plugger which could be introduced to 4 mm short of working length was selected in the obturation procedure. As the positive control, Group PC was not filled. In Group SB, ET and PC, all external surfaces of each tooth were coated with nail varnish leaving only 1 mm area around the apical foramen. In the negative control of Group NSB and Group NET, all of external tooth surface including apical foramen was coated with the nail varnish. The specimens were immersed in methylene blue dye solution for 2 days. Then the specimens were sectioned at each 1 mm from apex to 5 mm level. The final score of one specimen was given by summing up of the points at all levels. The dye leakage of Group ET was significantly less than that observed in Group SB (p < 0.05). And the frequency of gutta-percha pulling out from root canal when the plugger was removed was more often with the System B than with EndoTwinn but there was no significant difference.

Preparation and Characterization of Bamboo-based Activated Carbon by Phosphoric Acid and Steam Activation (인산 및 수증기 활성화에 의한 대나무 활성탄 제조 및 특성 연구)

  • Bamboo is an evergreen perennial plant, and it is known as one of the most productive and fastest-growing plants in the world. It grows quickly in moderate climates with only moderate water and fertilizer. Traditionally in Asia, bamboo is used for building materials, as a food source, and as versatile raw materials. Bamboo as a biomass feedstock can be transformed to prepare activated carbon using the thermal treatment of pyrolysis. The effect of process variables such as carbonization temperature, activation temperature, activation time, the amount of steam, and the mixing ratio of phosphoric acid and bamboo were systematically investigated to optimize the preparation conditions. Steam activation was proceeded after carbonization with a vapor flow rate of $0.8{\sim}1.8mL-H_2O\;g-char^{-1}\;h^{-1}$ and activation time of 1 ~ 3 h at $700{\sim}900^{\circ}C$. Carbon yield and surface area reached 2.04 ~ 20.59 wt% and $499.17{\sim}1074.04m^2\;g^{-1}$, respectively, with a steam flow rate of $1.4mL-H_2O\;g-char^{-1}\;h^{-1}$ for 2 h. Also, the carbon yield and surface area were 24.67 wt% and $1389.59m^2\;g^{-1}$, respectively, when the bamboo and phosphoric acid were mixed in a 1:1 weight ratio ($700^{\circ}C$, 2 h, $1.4mL-H_2O\;g-char^{-1}\;h^{-1}$). The adsorption of methylene blue into the bamboo activated carbon was studied based on pseudo first order and second order kinetics models. The adsorption kinetics were found to follow the pseudo second order model, which is governed by chemisorption.
